Pengembangan Media Transpor untuk Koleksi Sampel Preputium, untuk deteksi Bovine Genital Campylobacteriosis
Campylobacter fetus subsp. Venerealis (Cfv) is bacteria causing contagious genital diseases in cows called Bovine Genital Campylobacteriosis (BGC) or vibriosis. Isolation of Cfv is difficult, because the bacteria are fragile and need specific nutrients and oxygen (5-10%). The transport media is very important to maintain Cfv survival before culturing in laboratory. The aim of this study was to modify a new transport media as an alternative media for Cfv. Developed media capability was compared to Weybridge media, and Phosphate Buffered Saline (PBS). All transport media was contaminated by Cfv with concentrations of 105,104,103,102,101 (CFU/ml), and was stored for <6, 24, 48, 72, and 96 hours in each transport medium before culturing on blood agar, all in triplicate. The quality of transport media was analyzed based on bacterial growth on blood agar. PCR test was used as a confirmatory test of growing bacteria cultured on blood agar. Based on culture results, Cfv stored in three transport mediums for <6 hours, Cfv grew on blood agar from all concentration levels provided. Cfv stored for 24, 48, 72 and 96 hours on PBS did not grow, whereas on developed media and Weybridge media, the bacteria could grow and enrichment occurs at all concentration levels given. This study indicated the developed media can be used as an alternative transport medium for Cfv bacteria
Deteksi Staphylococcus aureus dan Staphylococcus sp. Secara Langsung Dari Susu Segar Kambing Peranakan Etawa dengan Teknik PCR
Genus Staphylococcus merupakan salah satu patogen bakteri penyebab mastitis yang menyebabkan kerugian ekonomi pada kambing Peranakan Etawa. Diantara Staphylococcus sp. yang dapat tumbuh dengan baik dalam susu segar, diketahui Staphylococcus aureus dapat membahayakan kesehatan manusia yang mengkonsumsi (food borne disease) karena kemampuannya dalam memproduksi enterotoksin yang tahan terhadap enzim pencernaan maupun pemanasan. Tujuan penelitian ini adalah mendeteksi Staphylococcus sp. dan S. aureus secara langsung dari susu kambing peranakan etawa dengan teknik PCR.Metode yang dilakukan adalah mengekstraksi DNA dari 60 sample susu segar dengan prinsip spin column-based nucleic acid purification dan kemudian dilakukan amplifikasi gen spesifik 23S rRNA Staphylococcus sp. dan S. aureus. Hasil PCR diketahui 37 (61%) sampel susu positif mengandung Staphylococcus sp. dan hanya 1 (1,6%) sampel mengandung S. aureus. Efektifitas Terapi Asam Urat dengan Poliherbal Ekstrak Bawang Merah (Allium ascalonicum L.) dan Jahe Merah (Zingiber officinale var rubrum) pada Tikus Hiperurisemia
Uric acid cause inflammation of acute gout arthtritis and other complications. Giving chemical drugs has side effects. Flavonoids in shallots (Allium ascalonicum L.) can inhibit the xanthine oxidase enzyme as antihyperuricemia. Gingerol in red ginger (Zingiber officinale var rubrum) as antihyperuricemia with antiinflammatory effect. The research aims to determine the effectiveness of the polyherbal extract of shallots(Allium ascalonicum L.) and red ginger (Zingiber officinale var rubrum) in hyperuricemia rats. Twenty five of male rats were divided into five goups, namely negative control, positive control, (P1) 25% red ginger extract and 75% shallot, (P2) 50% red ginger extract and 50% shallot, (P3) 75% red ginger extract and 25% shallot. Potassium oxonate 250 mg / kg BW was induced intraperitoneally on the 7th day. Uric acid has been measured on 14th, 21th and 28th days. On the 15th to 28th days, herbs/chemical drugs were administered according to the groups. Data were analyzed using Anova Multifactorial Randomized Design, continued with Post Hoc with the Least Significant Different test. The results showed that administration of potassium oxonate, chemical drugs and combination of herbs extracts had a significant effect (p <0.05) on uric acid levels compare with negative control. The conclusion show that the administration of polyherbal shallot extract (Allium ascalonicum L.) and red ginger (Zingiber officinale var rubrum) indicates as antihyperuricemia. The most effective dose is red ginger extract 450 mg/200 g and shallot 150 mg/200 g BW/day for two weeks of administration
Pemberian Ekstrak Etanol Daun Jambu Mete (Anacardium occidentale) dalam Reduksi Kadar IL-6 dan Peningkatan Kadar SOD pada Mencit Fibrosis Hepar yang Diinduksi CCl4
Liver fibrosis is the excessive accumulation of extracellular matrix. The prevalence of liver fibrosis in small animal was reported by 10% of all systemic diseases occured in the United States. One of the toxic materials that cause liver fibrosis is carbon tetrachloride (CCl4). This study aims to determine the preventive effect of ethanol extract of leaves of cashew (Anacardium occidentale) against the production of Interleukin-6 (IL-6) and increase the levels of superoxide dismutase (SOD). Leaves cashew contain of flavonoids which have anti-inflammatory and antioxidant effects. This research is an experimental study, post-test control only design using a completely randomized design. Mice (Mus musculus) were divided into 5 groups, namely the negative control group, positive control group, preventive group dose of 500 mg/kg BW, 1000 mg/kg BW, and 1500 mg/kg BW. Interleukin-6 production is calculated using flowcytometry, while SOD was measured using spectophotometer. Data were statistically analyzed within one way ANOVA (α=0.05) with advanced test Post Hoc Tukey. Conclusions from this research that the preventive use of ethanol extract of leaves of cashew to prevent an increase of IL-6 production and increasing the levels of SOD in mice models of liver fibrosis. Production of IL-6 in group of 1500 mg/kg BW is 2.58%. SOD levels in group of 1500 mg/kg BW is 4.23%.
Penilaian Organisasi Perangkat Daerah (OPD) tentang Otoritas Veteriner dalam Kebijakan Penanggulangan Rabies di Provinsi Banten
Veterinary Authority (Otovet) is a government institution, formed in order to deal with decision making of anything related to animal health, by involving veterinarians and mobilizing all related professional lines. The purpose of this study was to describe the Regional Organizations’ (OPD) assessment of the implementation of Veterinary Authority (Otovet) in rabies management policies. Research material included 36 Regional organizations (OPD), 9 Technical services, supervising animal health (keswan) and veterinary public health (kesmavet), 9 Health services, 9 Regional development planning services, 9 Regional staffing services, Banten Province. The results comprised eight questions: 1). Are the organization bureaucratic? 2). Positions, roles, and authority of veterinarians? 3). Is the veterinary authority regarding rabies control already in line with the implementation? 4). Strategic policy 5). Cooperations OPD, 6). Regional Policy 7). Veterinarian openings, and 8). Enforcement and improvement of otovet institutions. The answers were: Technical services of animal health and veterinary public health: 0%, 33,33%, 77,78%, 100%, 33,33%, 44,44%, 77,78%, 100% answered yes; 100%, 66,67%, 22,22%, 0%, 66,67%, 55,56%, 22,22%, 0% answered no. Health services: 22,22%, 66,67%, 88,89%, 44,44%, 55,56%, 22,22%, 0% , 77,78% answered yes; 77,78%, 33,33%, 11,11%, 55,56%, 44,44%, 77,78%, 100%, 22,22% answered no. PLATELET RICH PLASMA (PRP) DARI LIMBAH DARAH SAPI SEBAGAI OBAT LUKA BAKAR PADA TIKUS PUTIH (Rattus norvegicus)
Luka bakar adalah kerusakan jaringan pada kulit akibat terpajan panas tinggi, bahan kimiawi maupun arus listrik. Salah satu hewan yang disembelih di Rumah Potong Hewan (RPH) adalah sapi. Seekor sapi dapat menghasilkan limbah darah kurang lebih sebanyak 28 liter. Komponen darah terdiri dari plasma, sel darah merah, sel darah putih dan platelet. Platelet mengandung growth factor. Platelet Rich Plasma (PRP) atau plasma yang kaya akan platelet terbukti dapat mempercepat penyembuhan tulang dan jaringan lunak. Pembuatan PRP dari darah sapi dilakukan dengan metode sentrifuge. Hewan coba yang dipakai pada penelitian ini adalah tikus putih sebanyak 10 ekor. Besi panas berbentuk bulat dipanaskan dengan api, lalu ditempelkan pada kulit. Kelompok perlakuan diberi PRP dengan konsentrasi 10%, 20% dan 30% dengan salep vaselin album, kelompok kontrol negatif yang tidak diberi perlakuan dan kelompok kontrol positif yang diberi obat yang terbuat dari ekstrak plasenta sapi 10%+neomycin sulfate 0,5%. Parameter yang dipakai untuk mengukur tingkat kesembuhan luka adalah waktu kesembuhan dan pemeriksaan histopatologi. Pada hasil pengukuran waktu kesembuhan dianalisis dengan metode statistik One-way Analsisis of variance (Anova) tingkat signifikansi 95% dengan Kruskal-wallis, kesembuhan luka terdapat perbedaan signifikan antara PRP konsentrasi 10%, 20%, 30%, dan kontrol positif dengan kontrol negatif. Konsentrasi PRP yang optimal adalah 20%. Analisa tipe jaringan yang terbentuk dilakukan dengan pembuatan preparat histopatologi, hasil menunjukkan bahwa luka pada kelompok perlakuan dan kontrol positif tampak sembuh, sedangkan kelompok kontrol negatif epitelisasi belum tertutup jelas. PRP merupakan obat penyembuh luka bakar yang efektif sebagai pengganti obat komersil yang sudah tersedia
Effectiveness of Gout Therapy with Polyherbal Extract of Shallot (Allium ascalonicum L.) and Red Ginger (Zingiber officinale var rubrum) in Hyperuricemia Mice
Gout can cause inflammation of acute gout arthtritis, as well as other complications. Provision of chemical drugs in the long term have side effects. Shallots (Allium ascalonicum L.) contain flavonoids which can inhibit the xanthine oxidase enzyme, so as to reduce uric acid levels. Red ginger extract (Zingiber officinale var rubrum) is rich in gingerol to reduce uric acid with anti-inflammatory effect. Polyberbal combination is expected to increase the effectiveness of therapy. The research aims to determine the effectiveness of polyherbal combinations. 25 male rats (2-3 months) were divided into five groups, namely negative control, positive control, (P1) 25% red ginger extract: 75% shallot, (P2) 50% red ginger extract: 50% shallot, ( P3) 75% red ginger extract: 25% shallot. Induction of potassium oxonate 250 mg / kg BW intraperitoneally on the 7th day. The first measurement of uric acid on the 14th day with Urid Acid Toos, the 15th to 28th days was administered herbal / chemical drugs according to the group. Measurement of second and third uric acid on days 21 and 28. Data were analyzed by one-way Anova (RAL), further significance tests using Least Significant Different (LSD). The results showed that administration of potassium oxonate and combination of herbal extracts had a significant effect (p <0.05) on uric acid levels in all groups. The most effective herbal combination doses are (P3) 75% red ginger (450 mg / 200 gr) and 25% shallot (150 mg / 200 gr BW). The conclusion of this study is the combination of red ginger extract and shallot can be recommended as a uric acid-lowering agent
Gambaran Leukosit Kucing Penderita Feline Panleukopenia
One of the diseases in cats with high morbidity and mortality is feline panleucopenia (FPL). This study aims to evaluate the total leukocytes to determine the prognosis in cats suffer Feline Panleukopenia. This study used 27 male and female cats of various ages, that have been diagnosed as FPL based on the feline parvo virus Ag test. As much as 1 ml blood sample were collected from all of cats to examinated the total of leukocytes, then analyzed descriptively. The results showed that a total of 19 FPL (70.4%) had a total of leukocytes 2,500 cells/mm3. The incidence of FPL was more in cats ≤ 6 months, ie 21 (77,8%) compared to > 6 months, ie 6 cats (22,2%). Feline Panleukopenia wassuffered by more male (59.3%) than females (40.7%) cats. Based on the results of this study it was concluded that the majority of feline panleukopenia have very low leukocytes with infausta prognosis, especially in male and young cats
Profil Reseptor Gonadotropini Releasing Hormone (GnRH) dari Hipothalamus Sapi
Gonadotropine Releasing Hormone (GnRH) is one of the recommended hormones to overcome ovulation problems and it can increase pregnancy rate so that it is used in government programs to increase cattle population in Indonesia, although the results are not yet optimal. Hormone and receptor compatibility is the main key to succesful hormone application while data on the composition of the Indonesian cow GnRHreceptors is not yet known. The purpose of this study was to obtain the composition of GnRH receptors cattle in Indonesia and compare them to the GenBank sequence reference so that is known whether genetic diversity occurs at GnRH receptors cattle in Indonesia. Samples in the form of 3 female cow hypothalamus stored at -800C temperatue freezer. The methods used are mRNA isolation, cDNA synthesis, implification of GnRH gene using Polymerase Chain Reaction (PCR) process by Promoter F and Exon 1 R primers and then elektrophoresis and sent for sequencing. Sequencing product were further aligned with the reference sequences of Bos TaurusGnRHR mRNA GenBank using the MEGA X program. The results of the analysis found the presence of Single Nucleotide Polymorphism (SNP) in the coding region of 1st cow position 38(A> T), 261(C > T), 342(C >T), 411(C >T) and 495(C > T) and 2nd cow positions 261(C >T). Changes in amino acids were also detected in 1stcowposition 13 (H> L). The presence of SNP was found to indicate genomic variation between individuals at GnRH receptors cattle in Indonesia
Platelet Rich Plasma(Prp) dari Limbah Darah Sapi sebagai Obat Luka Bakar pada Tikus Putih (Rattus Norvegicus)
The burn is tissue damage to the skin caused by exposure to high heat, chemicals or electric current. Skin or tissue that burns will become necrotic tissue. Platelet Rich Plasma (PRP) is a platelet that is concentrated in a little plasma. PRP is used to stimulate and accelerate the healing of bone and soft tissue because it contains growth factor. PRP is obtained by blood centrifugation to get the most of platelets by minimizing plasma. The purpose of this study was to prove the ability of PRP from the waste cow’s blood to accelerate the healing of rat burns. Experimental animals used in this study were 10 white rats. An iron with round shape heat in fire, then stick into rats skin to make wound. The experimental design were used 5 groups. Groups 1-3 were animal with 10%, 20%, 30% PRP concentration treatment. The other 2 groups would be the negative control (not treated) and the positive control (placenta extract 10% + neomycin sulfate 0,5%). The parameters used to measure the rate of wound healing is the healing time and the type of tissue that is formed. The data were analyzed using One-way Analysis of variance (Anova) with Kruskal-Wallis statistical method in 95% significant rate. The result showed there is a significant difference between treatments. The results shown by the best healing wounds treated with PRP concentration of 30%. Tissues analysis were done by making preparations of histopathological from the skin. The results showed that the treated wounds healed but the untreated wound, ephitelization were not formed yet
