    Pavlovian impatience: The anticipation of immediate rewards increases approach behaviour

    Contains fulltext : 311792.pdf (Publisher’s version ) (Open Access)People often exhibit intertemporal impatience by choosing immediate small over delayed larger rewards, which has been implicated across maladaptive behaviours and mental health symptoms. In this preregistered study, we tested the role of an intertemporal Pavlovian bias as possible psychological mechanism driving the temptation posed by immediate rewards. Concretely, we hypothesized that the anticipation of immediate rewards (compared with preference-matched delayed rewards) enhances goal-directed approach behaviour but interferes with goal-directed inhibition. Such a mechanism could contribute to the difficulty to inhibit ourselves in the face of immediate rewards (e.g., a drug), at the cost of long-term (e.g., health) goals. A sample of 184 participants completed a newly developed reinforcement learning go/no-go task with four trial types: Go to win immediate reward; Go to win delayed reward; No-go to win immediate reward; and No-go to win delayed reward trials. Go responding was increased in trials in which an immediate reward was available compared with trials in which a preference-matched delayed reward was available. Computational models showed that on average, this behavioural pattern was best captured by a cue-response bias reflecting a stronger elicitation of go responses upon presentation of an immediate (versus delayed) reward cue. The results of this study support the role of an intertemporal Pavlovian bias as a psychological mechanism contributing to impatient intertemporal choice.19 p

    Determinants of Self-Management Behavior in Gout: A Scoping Review.

METHODS: Four databases (Medline, Embase, PsycINFO, and CINAHL) were searched using terms related to gout, self-management, and determinants of behavior as described in the Theoretical Domains Framework (TDF). Two reviewers independently selected relevant studies via screening of title/abstract and full text. Thematic synthesis was performed for qualitative data; quantitative data were summarized using cross-tabulation displaying the investigated associations of determinants with self-management behavior. The TDF facilitated identification and grouping of determinants. RESULTS: From 2,087 unique articles found, 56 studies were included in this review, of which there were 27 qualitative and 29 quantitative studies. Eight themes were identified: knowledge and skills for self-management, acceptance of disease, beliefs about necessity of self-management to improve gout-related health, resistance and reluctance for medication adherence and dietary alteration/changes, negative emotions influencing self-management, social support and interactions, environmental context, and self-regulation of behavior. Quantitative determinants associated with self-management behavior, predominantly medication adherence, were mapped to 12 of the 14 domains of the TDF. No determinants regarding skills and goals have been identified in quantitative research. CONCLUSION: Intervention targets for self-management behavior in patients with gout mainly included determinants related to knowledge, implicit and explicit beliefs and attitudes, the environmental context and resources, and (social) support and reinforcement.01 april 202

    Collaborating for gifted students with complex educational needs

Through interviews with students, parents, educational professionals, and care providers, both successful and unsuccessful collaboration strategies were examined. Subsequently, a model was proposed to enhance effective collaboration for this unique, but heterogenous group of students. This model describes overarching collaboration conditions that preferably should be discussed beforehand with actors involved to establish a functional collaborative culture. Furthermore, this model describes specific tasks for all actors involved that could be applied accordingly to enhance collaboration practices. Lastly, a case coordinator should be assigned that ensures continuity, stability, and a constant flow of information. By following this model, educational professionals, care providers, parents, and policy makers may find ways to enhance their collaboration practices to support these students more adequately in their development.13 p

Congenital disorders of glycosylation (CDG) currently encompassing over 160 subtypes, are characterized by glycan synthesis and/or processing defects. Despite the increasing number of CDG patients, therapeutic options remain very limited as our knowledge on glycan synthesis is fragmented. The emergence of CDG resulting from defects in ER/ Golgi homeostasis makes this even more difficult. SLC10A7 belongs to the SLC10 protein family, known as bile acid and steroid transport family, exhibiting a unique structure. It shows a ubiquitous expression and is linked to negative calcium regulation in cells. The mechanisms by which SLC10A7 deficiency leads to Golgi glycosylation abnormalities are unknown. The present study identifies major O-glycosylation defects in both SLC10A7 KO HAP1 cells and SLC10A7-CDG patient fibroblasts and reveals an increased ER and Golgi calcium contents. We also show that the abundance of COSMC and C1GALT1 is altered in SLC10A7-CDG patient cells, as well as the subcellular Golgi localization of the Ca(2+)-binding Cab45 protein. Finally, we demonstrate that supraphysiological manganese supplementation suppresses the deficient electrophoretic mobility of TGN46 by an aberrant transfer of GalNAc residues, and reveal COSMC Mn(2+) sensitivity. These findings provide novel insights into the mechanisms of Golgi glycosylation defects in SLC10A7-deficient cells. They show that SLC10A7 is a key Golgi transmembrane protein maintaining the tight regulation of Ca(2+) homeostasis in the ER and Golgi compartments, both essential for glycosylation

    Contains fulltext : 316833.pdf (Publisher’s version ) (Open Access)PURPOSE: The aim of this study was to validate simplified methods for quantifying [(68)Ga]Ga-FAPI-46 uptake against full pharmacokinetic modeling. METHODS: Ten patients with pancreatobiliary cancer underwent a 90-min dynamic PET/CT scan using a long axial field of view system. Arterial blood samples were used to establish calibrated plasma-input function from both continuous arterial sampling and image-derived input function (IDIF). Lesional [(68)Ga]Ga-FAPI-46 kinetics were described using conventional non-linear plasma-input tissue-compartment models. Logan plots using 30-90 min and 30-60 min post-injection (p.i), image-based target-to-whole blood ratio (TBR), mean standardized uptake values (SUVmean) normalized to body weight, lean body mass, and body surface area, at 20-30 min, 60-70 min and 80-90 min p.i were assessed. RESULTS: One patient was excluded due to discontinued scan acquisition and missing arterial sampling. Thirteen tumoral lesions and 11 non-tumoral lesions were included. A reversible 2-tissue-compartment model showed most preferrable fits for all types of [(68)Ga]Ga-FAPI-46 positive lesions. The distribution volume (V(T)) results obtained using arterial sampling plasma-input function and those using plasma-IDIF (V(T_plasma_IDIF)) showed an excellent correlation (Spearman rank correlation coefficient (r(s)) = 0.949). Logan V(T) using both time intervals were highly correlated with V(T_plasma_IDIF) (r(s) ≥ 0.938). The correlation values with V(T_plasma_IDIF) for image-based TBR and SUVmean parameters were higher at 80-90 min (r(s) ≥ 0.839) and at 60-70 min (r(s) ≥ 0.835) p.i than at 20-30 min p.i (r(s) ≤ 0.774). CONCLUSION: Image-based TBR and SUVmean at 60-70 min p.i are suitable for quantifying [(68)Ga]Ga-FAPI-46 uptake. TRIAL REGISTRATION: EudraCT, EudraCT 2022-001867-29. Registered 02 November 2022
