Advancing cardiac safety and drug discovery screening using human stem cell-derived cardiomyocytes
This thesis delves into the expansive utilization of both 2D and 3D human cardiac stem cell models for assessing cardiac safety and efficacy through a combination of in vitro and in silico methodologies. Various technological innovations are described in this thesis that led to cardiac in vitro models that are more scalable, reproducible and in which the cardiomyocytes are more mature.The final chapter of this thesis presents a groundbreaking demonstration of how 3D human cardiac stem cell models can consistently reveal a disease phenotype that can be rescued through drug treatment. This represented a significant milestone in the use of these models for disease modeling, high-throughput screening, and potential regulatory approval.Overall, the advancements detailed in this thesis have elevated the "technology readiness level" of these models, making them well-suited for use within the pharmaceutical industry. With continued progress, as outlined in this thesis, human cardiac stem cell models hold the potential to revolutionize drug discovery for cardiovascular diseases, safety testing, and personalized medicine.Ncardia Services B.V.;

Whole genome sequencing and contact screening: in the surveillance and infection control of methicillin and borderline oxacillin resistant staphylococcus aureus
Dit proefschrift onderzocht hoe ziekenhuisinfecties met resistente bacteriën zoals MRSA, LA-MRSA en BORSA beter kunnen worden voorkomen en opgespoord.Uit het onderzoek bleek dat MRSA zich zelden verspreidt in het ziekenhuis als de standaardmaatregelen goed worden nageleefd en er geen extra risicofactoren zijn. De kans op overdracht neemt echter toe als patiënten een kamer delen of bijvoorbeeld katheters, wonden of een infectie hebben. Daarom is het verstandig om bij een mogelijke besmetting vooral mensen met een verhoogd risico eerst te onderzoeken.Met behulp van moderne DNA-technieken (whole-genome sequencing en cg/wgMLST) kon nauwkeuriger worden vastgesteld hoe deze bacteriën zich verspreiden. De DNA-analyse hielp bij het herkennen van uitbraken, bijvoorbeeld van BORSA onder dermatologische patiënten. De gedetecteerde cluster alert in het geval van een recente transmissie was tussen de 0 en 2 allelen. Daarnaast werden hiermee meer ziekmakende bacteriestammen gevonden die lijken op vee gerelateerde MRSA(LA-MRSA) , maar eigenlijk afkomstig zijn Azië en bekend staan als hypervirulente PVL-positieve CC398 HO-MRSA L2 stammen. Omdat in de huidige richtlijn bij een LA-MRSA infectiepreventiemaatregelen mogen worden versoepeld leidde deze bevinding tot een nuancering van de richtlijn: alleen als mensen aantoonbaar in contact zijn geweest met vee, mogen infectiemaatregelen versoepeld worden.LUMC / Geneeskund

Coalition formation and co-sponsorship in the United Nations security council
Exploring how coalitions among member states are formed in international organisations is important to understanding ongoing global governance. Analysis of co-sponsorship patterns offers valuable insight into decision-making processes in the United Nations Security Council (UNSC). Our study utilises recently compiled data to investigate co-authorship of resolutions in the UNSC and compares them with co-sponsorship information of United Nations General Assembly (UNGA) resolutions. We observe an alignment between China and Russia among the permanent UNSC member states and a gradual shift towards a more individualistic approach among Western powers in the Security Council. The observed trends for the Security Council, however, are mitigated by the practice of ‘penholdership’, in which P3 members (France, UK, and USA) play an important agenda-setting role. Our analysis of co-sponsorship patterns in the UNGA supports these findings, demonstrating a convergence between Russia and China, and a gradual divergence of Moscow from the P3 countries in UNGA-supported resolutions, while France and the UK are still often aligned. Our research provides valuable insights into the strategic priorities and alliances that influence global governance, notably based on evolving patterns of UNSC resolution co-sponsorship.Security and Global Affair
Conclusion: the UNSC, multilateralism, and patterns of global governance
This conclusion synthesizes the book Multilaretalism and the United Nations Security Council (UNSC)'s key contributions, which examine the interplay between the UNSC and multilateralism. It highlights UNSC voting patterns, coalition dynamics, veto use, and the roles of both global powers and emerging states. While great power rivalries—especially among the P5—pose challenges, and its procedures resist criticism and reform questions, the resilience of multilateralism is evident in the UNSC's ability to facilitate cooperation. The book underlines the growing influence of small and emerging powers, the strategic role of regional blocs like the European Union, and the increasing focus on non-traditional threats such as climate change and attacks on cyber security. The Chapters highlight and also advocate for reforms to ensure inclusivity and representation within the UNSC, addressing structural inequalities while maintaining multilateralism's core principles. Despite the challenges presented, the analysis reaffirms the UNSC's centrality to global governance and the lasting necessity of multilateral collaboration in addressing complex, transnational issues. The reception of translated vaccination information: evidence from a reading and stops-making-sense judgment task
Reception-oriented research on health communication, especially when focusing on migrant populations, allows for an exploration of what it means to provide access to health information, shedding light on migrants’ communicative needs with practical implications for translators’ work. Adopting a multi-method approach, this study explores the potential for reading difficulty and for misinterpretation of translated vaccination-related public health information. For this purpose, we conducted a reception-oriented study that used a participant-paced, region-by-region reading method coupled with an incremental stops-making-sense judgment task and a post-hoc comprehension questionnaire with two participant groups (native and non-native English speakers). Three main findings stand out from our analysis. First, the triangulation of multiple data sources indicates that reading difficulty and misinterpretation were not exclusive to the non-native group but affected both participant groups. Second, while participants generally responded correctly to comprehension questions, average reading times showed that both groups took longer to read most texts than expected. Third, medical language was the most challenging aspect for our participants, as indicated by data from online and offline subjective measures.Theoretical and Experimental Linguistic
