    Analysis of Stiripentol Enantiomers on Several Chiral Stationary Phases : A Comparative Study

    A comparative study was developed for the separation of stiripentol enantiomers on several chiral stationary phases which were CyclobondI 2000 , S,S Whelk O1 , R,R Whelk O1 , Chiralcel OB , Chiralcel OF , Chiralcel OB-H and Chiralpak AD-RH . The best separation was achieved on Chiralpak AD-RH chiral column where a simple, rapid and validated method for the determination of stiripentol enantiomers was developed. Stiripentol was separated and quantitated on Chiralpak AD-RH chiral column using a mixture of water/acetonitrile (30/70 v/v) as a mobile phase (t 1 , t 2 = 5.626, 6.891, α = 1.22, R = 2.53) at 25 °C and a flow rate of 1 mL min −1 s . The UV-detector was set at 254 nm. The applied HPLC method allowed the separation and quantification of stiripentol enantiomers with good linearity (r > 0.999) in the studied range. The relative standard deviations (% RSD) were 0.723 and 0.692 for the stiripentol enantiomers with accuracy of 98.40 and 98.53. The limit of detection and limit of quantification of stiripentol were found to be 10 and 30 µg mL −1 , respectively. The method was validated through the parameters of linearity, accuracy, precision and robustness. The HPLC method was applied for the quantitative determination of stiripentol in pharmaceutical formulations

    Alfuzosin Treatment Improves The Rate and Time for Stone Expulsion in Patients with Distal Uretral Stones: A Prospective Randomized Controlled Study

    evaluate the safety and efficacy of alfuzosin treatment on rate and time of stone expul- sion in patients with uncomplicated distal ureteral stones. PATIENTS AND METHODS Prospective, randomized, open-label, controlled study. Patients > 18 years of age presenting to the outpatient urology clinic with uncomplicated radio-opaque stones located in the distal third of the ureter and of size ≤ 10 mm were included. Patients were randomly assigned to either a control group (n=26) and received standard of care management (daily oral hydration and diclofenac 75 mg IM on demand) or to the alfuzosin group (n=28) and received alfuzosin SR 5 mg twice daily in addition to standard of care management. Patients were followed weekly at office visits and twice weekly by telephone for 4 weeks or until stone expulsion. Assessments included stone passage rate and time, and patients were monitored for occurrence of adverse drug events, complications, number of pain episodes, analgesic consumption, and number of hospital revisits. Mann–Whitney, v2, and Fisher exact test were used for data analysis.RESULTS All 54 patients completed the study. Stone expulsion rate was higher in the alfuzosin arm (53.6%, 15/28) compared to the control arm (26.9%, 7/26, p=0.04). Median stone passage time was lower in the alfuzosin group than in the control group (9 vs 19 days, respectively, p=0.006). Ureteral sepsis, uncontrollable pain, and hospitalization readmissions were reported in the control group only. There were no differences between groups in number of pain episodes, pain scores, or analgesic consumption. Alfuzosin therapy was tolerable with only minor adverse effects (headache, dizziness, mild postural hypotension, and rhinitis).CONCLUSION Alfuzosin is safe and effective in increasing stone expulsion rates and shortening stone passage times for uncomplicated distal ureteral stones

    RP-HPLC with Time Programmed Fluorescence Detection for Quantitation of Avanafil and Dapoxetine Hydrochloride; Application to Pharmaceutical Dosage Form and Biological Fluid

    Avanafil (AVN) was recently co-formulated with dapoxetine HCl (DAP) for treatment of erectile dysfunction and premature ejaculation. Sensitive and simple reversed-phase (RP) high-performance liquid chromatographic method (HPLC) was developed and validated for their simultaneous determination using tadalafil (TAD) as an internal standard. Isocratic separation was achieved within run time of only 7.0 min on Eclipse C18 column (150 mm × 4.6 mm, 5 µm particle size) using a mobile phase composed of acetonitrile: 0.15% triethylamine (40:60, v/v) at pH = 4.0 adjusted with o-phosphoric acid. The analysis was performed at a flow rate of 1.0 mL/min with fluorescence detection at 236/370 nm for AVN, 236/410 nm for DAP, and 236/330 for TAD using time programming. The analytes were determined by their native fluorescence and the response was linear over concentration ranges of 0.05–40 and 0.01–30 µg/mL for AVN and DAP, respectively, with limits of detection of 0.043 and 0.007 µg/mL in a respective order.
