Determination of sparfloxacin and besifloxacin hydrochlorides using gold nanoparticles modified carbon paste electrode in micellar medium
A gold nanoparticles modified carbon paste electrode (AuCPE) was used to study the electrochemical behavior of sparfloxacin HCl (SPAR) and besifloxacin HCl (BESI) using cyclic and differential pulse voltammetry modes in the presence of micellar medium. Effect of different surfactants on peak current was studied in Britton–Robinson buffer solution of pH 2. Sodium dodecyl sulphate is the optimum surfactant based on the enhancement of the peak current. The modified electrode shows highly sensitive sensing giving an excellent response for SPAR and BESI. The peak current varied linearly over the concentration ranges from 1.1 × 10−7 mol L−1 to 3.3 × 10−6 mol L−1 and from 2.2 × 10−6 mol L−1 to 5.5 × 10−5 mol L−1 with determination coefficients of 0.9976 and 0.9984 in case of SPAR and BESI, respectively. The recoveries and the relative standard deviations were found in the following ranges: 99.97–101.4% and 0.63–1.48% for SPAR and 99.89–101.1% and 0.85–1.76% for BESI. The detections limits were 2.87 × 10−8 and 3.76 × 10−7 mol L−1 for SPAR and BESI, respectively. The proposed method has been successfully applied to determine SPAR and BESI in biological fluids
Simultaneous determination of 200 pesticide residues in honey using gas chromatography–tandem mass spectrometry in conjunction with streamlined quantification approach
A sensitive, accurate and reliable multi-class GC–MS/MS assay protocol for quantification and confirmation of 200 common agricultural pesticides in honey was developed and validated according to EU guidelines. A modified extraction procedure, based on QuEChERS method (quick, easy, cheap, effective, rugged and safe) was employed. Mass spectrophotometric conditions were individually optimized for each analyte to achieve maximum sensitivity and selectivity in MRM mode. The use of at least two reactions for each compound allowed simultaneous identification and quantification in a single run. The pesticides under investigation were separated in less than 31 min using the ultra-inert capillary column (DB-35MS). For all analytes, neat standard calibration curves in conjunction with correction for matrix effect were successfully employed. The detection limits of the assay ranged from 1.00 to 3.00 ng mL−1 for the studied pesticides. The developed assay was linear over concentration range of 10.00–500.00 ng mL−1, with correlation coefficient of more than 0.996. At the LOQ, 81% of the studied pesticides were efficiently recovered in the range of 70.00–120.00%, with CV% less than 15.00% while 99.3% compounds had mean percentage recovery of 60.00–140.00%, with CV% less than 21.00% (N = 18, over three different days). The proposed assay was successfully applied for the analysis of the studied pesticide residues in one PT sample and 64 commercial honey samples collected over 1 year from different districts around Egypt. Results revealed that only one honey sample out of the 64 analyzed samples was contaminated with tau-Fluvalinate (10.00 μg kg−1). This wide scope assay protocol is applicable for monitoring pesticide residues in honey by national regulatory authorities and accredited labs; that should help ensure safety of such widely used product

Effect of oralistat (antiobesity drug)on rat submandibular salivary gland (histological and ultrastructural study)
The present study was performed to investigate the effect of antiobesity drug orlistat on the rat submandibular salivary glands using histological and ultrastructural techniques. Statistical analysis was also performed to monitor changes occurred in animal’s body weights during the experiment.
The experiment was done on thirty healthy adult male albino rats. The animals were divided into three groups as follows:
1- Control group (group I): This group received their diet contained ground barley and drank water adlibitum.
2- Experimental group (group II): This group received the above mentioned diet contained 200 mg orlistat / kg barley for 15 days.
3- Experimental group (group III): This group received the control diet containing 200 mg orlistat / kg barley for 45 days.
The average body weight for each animal was calculated every week till the end of the experiment.
At the end of the experiment the animals were scarified. The submandibular salivary glands of both sides were dissected and separated.
In all groups the specimens of one side were immediately fixed in 10 % calcium formol, dehydrated in alcohol, cleared in xylol, embedded in paraffin wax, sectioned and stained with Hx. and E. for histological examination.
The specimens of the other side in the control and experimental group III were processed for electron microscopic examination, small pieces of average size 1mm³ were cut and fixed for primary fixation in 3% phosphate buffered glutraldhyde then post fixation, dehydration and embedding in oven dried gelatin capsules took place. Then semithin sections were cut and stained to be examined with light microscope. Afterwards, ultrathin sections were cut, mounted on copper grids and stained for final examination by transmission electron microscope.
The present investigation revealed the following results:
- Statistical analysis of percentage increase in body weights among the investigated animals revealed non significant difference among control and experimental groups (II and III).
- Histological examination of submandibular salivary gland of group II revealed mild degenerative changes as loss of acinar architecture and intracytoplasmic vacuolization. Also the ducts showed stagnant secretion in their lumen. Few red corpuscles were dispersed between the acini and the ducts.
- In group III more marked atrophic changes were demonstrated as there were extensive intracytoplasmic vacuolization, also some secretory cells were degenerated and their nuclei lost completely leaving homogenous eosinophilic degenerative material.
- The granular convoluted tubules showed decrease in their eosinophilic granules. The striated and excretory ducts demonstrated rupture in their cell membrane with subtotal loss of their cytoplasmic content.
- The blood vessels were markedly dilated and sometimes ruptured with extravasations of red blood corpuscles within the gland.
- The examination of semithin sections of submandibular salivary gland of group III showed widening of intercellular spaces, intracytoplasmic vacuolization and clear loss of zymogen granules. Also the nuclei were darkly stained.
- Electron microscopic findings in submandibular salivary gland of group III added to the structural results. There was loss of cellular contacts accompanied by different degree of cellular degeneration. Karyorrhexis (clumping of chromatin), margination and segregation of chromatin were found. Loss of continuity of nuclear membrane and widening of the nuclear pores as well as intracytoplasmic and intranuclear vacuoles were also observed. The secretory granules appeared to be fused into large vacuoles.
- Dilated rough endoplasmic reticulum and swollen degenerated mitochondria were also frequently detected in the submandibular salivary gland of group III as well as lysosomes and myelin figures.
- The outstanding ultrastructural feature in experimental group III was the presence of dilated blood vessel studded with blood in dissociated connective tissue. Other blood vessels appeared with ruptured endothelium and other showed sinusoidal endothelial lining causing collapse of their lumen.
In the present study the following conclusions could be reached:
1- Orlistat administration for a period of 15 days leads to mild changes in the submandibular salivary gland.
2- Administration of orlistat for longer period 45 days caused more aggravated changes manifested as degeneration and vacuolization of cells leading to possible deterioration of the functions of submandibular salivary gland.
3- Accordingly, we recommend that patients undergoing orlistat therapy should be aware of their oral hygiene and should be subjected to periodic oral and dental care.
4- We recommend those patients to take fat soluble vitamins supplements to overcome the impairment in their absorption that induced by orlistat, as malabsorption of those vitamins leads to negative effects on both the tissues and health

Outcome of immediate implant placement with bone augmentation in the anterior maxillary supporting partial denture
Background: Immediate implantation was suggested to reduce the number of surgical interventions and to preserve the alveolar ridge. Implant placement demands a thorough understanding of anatomic, biologic, surgical, and prosthetic principles. Purpose: The aim of the present study was to evaluate the 18 months survival rate of osseointegration, on the basis of clinical and radiographic examinations, for simultaneous bone grafting with immediate implants supporting partial denture with locator attachment in the maxillary esthetic area. Materials and Methods: A total of twenty patients randomly assigned received 45 immediate implants with bone graft supporting maxillary partial overdentures. The clinical analysis of probing depth, bleeding index, plaque index, and radiographic analysis of crestal bone level at a follow-up interval of 6, 12, and 18 months was evaluated. Results: All implants achieved successful osseointegration. The soft tissue architecture remained stable throughout the healing period of the implants as well as after final prostheses delivery, contributing to esthetically pleasing and biologically sound results. Conclusions: Immediate implants with locator attachment supporting maxillary partial denture can be safe, reliable, and a predictable option for the replacement of teeth in the anterior maxillary zone, providing stability to the peri-implant soft tissue
Validated spectrophotometric methods for simultaneous determination of Omeprazole, Tinidazole and Doxycycline in their ternary mixture
A comparative study of smart spectrophotometric techniques for the simultaneous determination of Omeprazole (OMP), Tinidazole (TIN) and Doxycycline (DOX) without prior separation steps is developed. These techniques consist of several consecutive steps utilizing zero/or ratio/or derivative spectra. The proposed techniques adopt nine simple different methods, namely direct spectrophotometry, dual wavelength, first derivative-zero crossing, amplitude factor, spectrum subtraction, ratio subtraction, derivative ratio-zero crossing, constant center, and successive derivative ratio method. The calibration graphs are linear over the concentration range of 1–20 μg/mL, 5–40 μg/mL and 2–30 μg/mL for OMP, TIN and DOX, respectively. These methods are tested by analyzing synthetic mixtures of the above drugs and successfully applied to commercial pharmaceutical preparation. The methods that are validated according to the ICH guidelines, accuracy, precision, and repeatability, were found to be within the acceptable limits
The Ameliorative Role of Grape Seed Oil on Irradiated Rat Fungiform Papillae
This study was held to clarify changes in the epithelium of the fungiform papillae after irradiation at the microscopic level as well as the effect of grape seed oil (GSO) on these changes and the changes in the innervation of these papillae. Thirty five male Swiss Albino rats were included in this study. The animals were divided into, Grape seed irradiated group (GSI gp): GSO was administrated orally in accompanied with radiation. Irradiated group (IR gp): The same protocol was applied to this group except that the animals received distilled water instead. Control group(C gp): only distilled water was administrated orally. The right tongue halves were prepared for Ultrastructural studies. The left halves were used for detection of the innervations using S100. Degeneration of the cell nuclei and organelles in stratum basale and spinosum of the fungiform papillae in IR gp was obvious. In the GSI gp the nuclei had prominent nucleoli and granulated chromatin. The regeneration of the epithelium in the IR gp was delayed compared with GSI gp. Regeneration of the cytoskeleton was advanced in the GSI gp. Significant obvious decrease in the innervations was indicated in the IR gp at the third day post irradiation compared with both GSI gp and C gp. Toward the end of the experiment, increase in the innervations was more detectable in GSI gp. For these reasons, grape seed oil is recommended to be consumed as a dietary supplement and could be useful in synergizing the hazardous of radiation on the tongue papillae
