Etodolac transdermal cubosomes for the treatment of rheumatoid arthritis: ex vivo permeation and in vivo pharmacokinetic studies
Journal Drug Delivery Volume 24, 2017 - Issue 1In this study, transdermal etodolac-loaded cubosomes were developed in order to relieve patient pain and joints stiffness by providing stable etodolac concentration at the targeting sites through controlled drug delivery via the noninvasive skin route with more sustaining and less frequent dosing. Different ratios and percentages of poloxamer 407 and monoolein were used to formulate the cubosomes using emulsification and homogenization processes. The etodolac-loaded cubosomes showed particle size values ranging from 135.95 to 288.35 nm and zeta potential values ranging from −18.40 to −36.10 mV. All the cubosomes offered an encapsulation efficiency value of about 100% and showed drug loading capacity ranging from 1.28 to 6.09%. The in vitro drug release studies revealed a controlled drug release profile with a drug release rate up to 15.08%/h. Increasing poloxamer concentration in etodolac-loaded cubosomes resulted in nanoparticles with less particle size and faster drug release. The particles exhibited cubic and hexagonal shapes. The DSC and X-ray analysis demonstrated that the drug was encapsulated in the cubosomes bicontinuous structures in amorphous form. In addition, investigated cubosomes exhibited fast drug penetration through excited mice skin followed by slower drug penetration for up to 24 h. The pharmacokinetic study in human volunteers showed that the selected etodolac-loaded cubosomes enhanced the bioavailability of etodolac as compared to the oral capsules (266.11%) with evidence of longer half-life and higher MRT that reached 18.86 and 29.55 h, respectively. The etodolac-loaded cubosomes propose a promising system for treatment of arthritis simply through skin application

Statement of problem: During heat-pressing of lithium dislilicate glass-ceramics, It is more economical to press several restorations from one ingot at the same time. However, this is often not possible and may result in a considerable amount of leftover material. It has been reported that the leftover materials after heat-pressing are reused in some dental laboratories. Sufficient knowledge about the consequences of such procedure is not available. The issue is thereby raised whether the leftover material should be discarded or reused.

Possible correlation between levansucrase production and probiotic activity of Bacillus sp. isolated from honey and honey bee
Five bacterial isolates from honey and bee gut were selected based on their high levansucrase activity and levan yield which were strongly positively correlated. All isolates showed good tolerance to temperature up to 70 °C, to NaCl up to 3 M and to 0.1% H2O2. They maintained over 59 and 64% survival at pH 9.0 and 2.0 respectively, but showed varying tolerance to 0.1% bile salts and pancreatic enzymes. Most isolates were susceptible to widely used antibiotics, but demonstrated diverse antimicrobial activity. Non hemolytic isolates were identified on the basis of 16S rRNA sequencing as Bacillus subtilis HMNig-2 and B. subtilis MENO2 with 97% homology. They exhibited promising probiotic characteristics and achieved highest levansucrase activity of 94.1 and 81.5 U/mL respectively. Both exhibited highest biofilm formation ability in static microtiter plate assay. Also, they achieved 34 and 26% adhesion respectively to Caco-2cells and had highest free radical scavenging activity of 30.8 and 26.2% respectively. The levans of the two isolates showed good antimicrobial activity against some pathogens and exhibited positive prebiotic effect (prebiotic index >1) with Lactobacillus casei and Lactobacillus reuteri. Results suggest a correlation between levansucrase production, levan yield and pre-probiotic activities of the studied strains

Improving tadalafil dissolution via surfactant-enriched tablets approach: Statistical optimization, characterization, and pharmacokinetic assessment
Tadalafil suffers from poor aqueous solubility that could lead to fluctuating blood levels and unreproducible
effect. Thus, this work aimed at improving tadalafil dissolution utilizing the approach of
surfactant-enriched tablets. The feasibility of minimizing various surfactants quantities was investigated
by establishing the ratio of the surfactant to drug that is required for drug solubilization in micellar
solutions. Based on the computed ratios, Tween was precluded from formulation studies due to its poor
solubilizing capacity towards the drug. 23 factorial design was employed to assess the impact of
formulation attributes on tablets' characteristics. Based on the statistical analysis and the desirability
function approach, tablet formulation F6 prepared using CTAB, Avicel PH 102, and 5% Ac-Di-Sol was
selected as the optimum formulation. The selected formulation showed adequate stability after storage at
40 C and 75% R.H. for twelve weeks. Pharmacokinetic study revealed that the selected surfactantenriched
tablet formulation F6 showed enhanced bioavailability compared to the market product Cialis®
Optimization of Cellulase Production by Halobacillus sp. QLS 31 Isolated from Lake Qarun, Egypt.
A halophilic cellulase-producing bacterium was isolated from a sediment sample collected from Lake Qarun (Fayoum Province, Egypt). Molecular identification based on 16S rDNA amplification and sequencing revealed 99% homology with Halobacillus sp. and hence was designated as Halobacillus sp. QLS 31. Medium composition and culture conditions were optimized for enhancing the production of cellulase enzyme using the Plackett-Burman statistical design. Ten variables were evaluated for their influence on cellulase production. Carboxymethyl cellulose (CMC), zinc sulfate (ZnSO4), and inoculum size were found to exert a significant effect on cellulase productivity by Halobacillus sp. QLS 31. The maximum specific activity of cellulase enzyme was 48.08 U/mg. Following the predicted conditions, a 7.5-fold increase in cellulase specific activity (175.47 U/mg) was achieved compared to the basal medium (23.19 U/mg) under the following optimized conditions: temperature (30 °C), fermentation time (2 days ), pH value (9), CMC concentration (1%), inoculum size (1%), yeast extract concentration (0.1%), ammonium sulfate ((NH3)2SO4) concentration (0.1%), sodium chloride (NaCl) concentration (20%), and metal inducers: ZnSO4 (0.1%) and Ca/Mg ratio (0.01%). Thus, the results of this study provide an important basis for more efficient, cheap industrial cellulase production from halophilic Halobacillus sp. QLS 31
Design, synthesis and anticancer evaluation of novel pyrazole, pyrazolo[3,4-d]pyrimidine and their glycoside derivatives
The chalcone derivatives 3a,b were cyclized upon reaction with
thiourea to give the pyrazolo[3,4-d]pyrimidine derivatives 5a,b.
Condensation of 5a,b and their hydrazide derivatives 8a,b with
cyclic and acyclic glucose gave the condensed S- andN-glycosides
7a,b and 9a,b, respectively. Reaction of 3b with ethyl cyanoacetate
followed by reaction with cyclic glucose afforded a mixture
of the O- and/or N-glycoside isomers 12 and 13, respectively.
The pyrazolo[3,4-c]pyrazole derivative 14 was also obtained from
the reaction of 3b with hydrazine hydrate. A number of the synthesized

Among which, the Session Initiation Protocol (SIP) is a signaling protocol that allows the establishment of multimedia sessions and calls between different parties. SIP functionality totally depends on a centralized infrastructure, and complexity arises when deploying such a protocol in a special type of wireless networks, namely Mobile Ad-hoc Networks (MANET), due to the absence of a centralized infrastructure, and the dynamic topology of the later. This paper highlights the the challenges arising when deploying SIP in MANETs and presents an overview of different proposed approaches to overcome these challenge
