A real-time PCR primer/probe set for reliable identification and quantification of Heterodera glycines
The soybean cyst nematode Heterodera glycines is a major pest in soybean cultivation. Identifying and quantifying nematodes can be achieved through probe-based real-time PCR, and two sets of primers/probes have been developed to target H. glycines. However, we found that one set showed low species specificity, and the other showed significant variations in Ct values among H. glycines populations, potentially leading to inaccurate quantification. Therefore, for more reliable identification and quantification, we developed a novel primer/probe set based on the H. glycines mitochondrial cytochrome c oxidase subunit I gene. The novel primer/probe set amplified DNA of all tested H. glycines populations but not that of the other tested species. In addition, there were no significant differences in Ct values among the tested H. glycines populations, indicating accurate quantification across populations using a fixed standard curve. The detection sensitivity of the novel primer/probe set was higher than or comparable to that of the existing primer/probe sets. The novel primer/probe set thus facilitates reliable identification and quantification of H. glycines, supporting decision-making for the appropriate control of H. glycines.This is a post-peer-review, pre-copyedit version of an article published by Brill in Nematology.

Collaborative Exploration and Collection of Vegetable Genetic Resources in Kyrgyz in 2023 and 2024
キルギス共和国水資源・農業・加工産業省と国立研究開発法人農業・食品産業技術研究機構(NARO)は, PGRAsia(Plant Genetic Resources in Asia)プロジェクトの一環として,キルギス国内における野菜遺伝資源の 共同研究を 2019 年から開始した.この共同研究の一環として,2023 年はビシュケク市 , チュイ州,イシク・ クル州,ナルン州およびジャララバード州において野菜類種子を収集するために 2 回の探索を実施した.探 索の結果,各地の自生地,マーケットおよび農家等において 10 科 23 属 37 種に属する合計 199 点の遺伝資源 を収集することができた.また,2024 年にはチュイ州において短期間の探索を 1 回実施し,野生ニンジン 12 点を収集した.収集した種子は 2 等分し,半分はキルギスジーンバンクで保存した.残りの半分は SMTA(標 準材料移転契約)により NARO 遺伝資源研究センターに移送する予定である.A collaborative research between the Department on Expertise and Horticulture of Agricultural Crops, Ministry of Water Resources, Agriculture and Processing Industry of the Kyrgyz Republic, and the National Agriculture and Food Research Organization (NARO) was started in 2019 under the Plant Genetic Resources in Asia Project to survey vegetable genetic resources in Kyrgyz. In 2023, two expeditions to collect vegetable seeds in the city of Bishkek and in the city of Bishkek and Chuy, Issyk-Kul, Naryn, and Jalal-Abad regions were conducted as part of this collaborative research. A total of 199 samples of 37 species from 23 genera and 10 families were collected in the natural habitats, local markets and farmer gardens, etc. In 2024, one short expedition in the Chuy region was conducted, and total 12 samples of wild carrot were collected from the natural habitats. The collected seeds were equally divided into two sets, one of which was stored in the Kyrgyz Genebank, and one of which will be transferred to the Research Center of Genetic Resources, NARO, under a standard material transfer agreement.Table 2点articl
