Heat inactivation of SARS-CoV 2 enabled the measurement of salivary cortisol during COVID-19 pandemic
Background and aim: Salivary cortisol has become an essential tool in the management of cortisol-related disease. In 2020 the sudden outbreak of COVID-19 pandemic caused several concerns about the use of saliva, due to the risk of contamination, and an European consensus further discourage using salivary cortisol. To decrease infectious risk, we handled specimens by applying a heat treatment to inactivate viral particles, further evaluating the impact of the COVID-19 pandemic on the use of salivary cortisol in clinical practice.
Materials and methods: Saliva samples were exposed for 10 min at 70°C, then cortisol was measured using LC-MS/MS. The number of salivary cortisol examinations from 2013 to 2022 was extracted from the local electronic database: those performed in 2019, 2020, and 2021 were analyzed and compared with the historical data.
Results: During 2020 we observed a decrease of 408 (-20%) examinations (p=0.05) compared to 2019; especially in salivary cortisol daily rhythm and salivary cortisol/cortisone ratio (respectively reduction of 47% and 88%, p=0.003 and p=0.001). Analyzing year 2021 compared with 2020 we reported an increase of 420 examinations (+20%, p=0.01), with a complete recovery of salivary cortisol measurement (considering 2019: p=0.71). Major differences were observed between morning salivary cortisol (-20%, p=0.017), LNSC (-21%, p=0.012) and salivary cortisol rhythm (-22%, p=0.056).). No Sars-Cov2 infections related to working exposure were reported among laboratory’s employers.
Conclusions: We speculate that the adoption of an appropriate technique to inactivate viral particles in saliva specimens allowed the safety maintenance of salivary collections, also during the Sars-CoV-2 outbreak

A total of 48 Siboney de Cuba cattle blood samples (35 females and 13 males, with age between 24 and 30 months) were collected.
All animals were genotyped with the GGP Bovine 100k BeadChip (Bos taurus x Bos indicus) containing approximately 100,000 SNPs (Illumina, San Diego, CA, USA)

The way we were, the way we will be. Open science: a change in the scientific paradigm
If we imagine a scientist sleeping for 15 years and waking up today we can just guess his/her surprise: open access journals, papers that are published along with accessible repositories where authors make available the data, the codes that they used to analyze the data and all the relevant digital materials. Even peer reviews are available to the reader, not to mention pre-registration documents that reveal step by step the origin of scientific path followed by the author! All of this is happening because of a double reason. First, we realised that science – often obscured behind paywalls – needed to be accessible by everyone. The second reason for this increase in transparency is perhaps is less noble: we realized that the science we were making needed some adjustments as many of the results reported in the literature were likely false positive. Altogether, this abrupt increment in research transparency and accessibility has been referred to as “open science”. In my talk I will review the major ideas that are emerging nowadays in science to increase accessibility, trustiness and transparency
CT Texture Analysis of Adrenal Pheochromocytomas: a Pilot Study
Abstract: Since radiomics is a promising field of research, we tried to study the application of CT texture analysis in pheochromocytomas (PCCs) and the correlation between the features extracted and the secretory and histopathological data. 17 patients affected by surgically removed PCCs were retrospectively enrolled. Before surgery, all patients underwent contrast-enhanced CT. Patients had catecholamine secretion data available and Adrenal gland Scaled Score (PASS) of the histopathological examination. After a resampling of all CT images, the PCCs were delineated using the LifeX software in all three phases (unenhanced, arterial, and venous) and 58 texture parameters were extracted for each volume of interest. Through the Mann-Whitney test, the correlation between the hormonal hypersecretion, the malignancy score of the lesion (PASS>4), and texture parameters were studied. In the comparison between secreting and non-secreting catecholamine tumors the parameters DISCRETIZED_HUpeak e GLZLM_GLNU in the unenhanced phase and GLZLM_SZE, CONVENTIONAL_HUmean, CONVENTIONAL_HUQ3, DISCRETIZED_HUmean, DISCRETIZED_AUC_CSH, GLRLM_HGRE and GLZLM_SZHGE in the venous phase showed a statistically significant difference (p<0.01) while in the comparison between tumours with low and high PASS the parameters GLZLM_GLNU in unenhanced phase and GLRLM_GLNU e GLRLM_RLNU in the venous phase showed a statistically significant difference. CT texture analysis of pheochromocytomas can be a useful tool for the identification of malignant lesions and lesions that present hormonal hypersecretion
