EnglishThis methodological paper explores different approaches for classifying and clustering longitudinal trajectories in prevention trials, illustrated through data from the MAPT study on Alzheimer’s disease. The aim is to identify homogeneous subgroups of participants and to discuss the relevance of data-driven exploratory methods in inconclusive trials. The manuscript was submitted to Prevention Science and underwent three rounds of peer review. Although the methodological contribution was acknowledged, the paper was not accepted for publication, mainly due to the reviewers’ focus on treatment efficacy rather than methodological innovation. The authors chose to make the manuscript publicly available to promote open science and transparency in methodological research on prevention trials.© 2025 Céline Bougel et al.Distributed under the terms of the Creative Commons Attribution–NonCommercial–NoDerivatives 4.0 International License (CC BY-NC-ND 4.0).This work may be shared and cited for non-commercial purposes, provided that it is not modified and that proper credit is given to the authors. Background: Clinical trials are designed to prove the efficacy of an intervention by means of model-based approaches involving parametric hypothesis testing. Issues arise when no effect is observed in the study population. Indeed, an effect may be present in a subgroup and the statistical test cannot detect it. To investigate this possibility, we proposed to change the paradigm to a data-driven approach. We selected exploratory methods to provide another perspective on the data and to identify particular homogeneous subgroups of subjects within which an effect might be detected. In the setting of prevention trials, the endpoint is a trajectory of repeated measures. In the settings of prevention trials, the endpoint is a trajectory of repeated measures, which requires the use of methods that can take data autocorrelation into account. The primary aim of this work was to explore the applicability of different methods for clustering and classifying trajectories. Methods: The Multidomain Alzheimer Preventive Trial (MAPT) was a three-year randomized controlled trial with four parallel arms (NCT00672685). The primary outcome was a composite Z-score combining four cognitive tests. The data were analyzed by quadratic mixed effects model. This study was inconclusive. Exploratory analysis is therefore relevant to investigate the use of data-driven methods for trajectory classification. The methods used were unsupervised: k-means for longitudinal data, Hierarchical Cluster Analysis (HCA), graphic semiology, and supervised analysis with dichotomous classification according to responder status. Results: Using k-means for longitudinal data, three groups were obtained and one of these groups showed cognitive decline over the three years of follow-up. This method could be applied directly to the primary outcome, the composite Z-score with repeated observations over time. With the two others unsupervised methods, we were unable to process longitudinal data directly. It was therefore necessary to choose an indicator of change in trajectories and to consider the rate of change between two measurements. For the HCA method, Ward's aggregation was performed. The Euclidean distance and rates of change were applied for the graphic semiology method. Lastly, as there were no objective criteria to define responder status, we defined our responders based on clinical criteria. Discussion: In the princeps study, the prevention trial was found to be inconclusive, likely due to the heterogeneity of the population, which may have masked a treatment effect later identified in a refined subgroup of high Beta Amyloid subjects. So, we have adopted an alternative unsupervised approach to subject stratification based on their trajectories. We could then identify patterns of similar trajectories of cognitive decline and also highlight the potential problem of a large heterogeneity of the profiles, maybe due to the final endpoint considered.

Online Matching via Reinforcement Learning: An Expert Policy Orchestration Strategy

    No full text
Online matching problems arise in many complex systems, from cloud services and online marketplaces to organ exchange networks, where timely, principled decisions are critical for maintaining high system performance. Traditional heuristics in these settings are simple and interpretable but typically tailored to specific operating regimes, which can lead to inefficiencies when conditions change. We propose a reinforcement learning (RL) approach that learns to orchestrate a set of such expert policies, leveraging their complementary strengths in a data-driven, adaptive manner. Building on the Adv2 framework (Jonckheere et al., 2024), our method combines expert decisions through advantage-based weight updates and extends naturally to settings where only estimated value functions are available. We establish both expectation and high-probability regret guarantees and derive a novel finite-time bias bound for temporal-difference learning, enabling reliable advantage estimation even under constant step size and non-stationary dynamics. To support scalability, we introduce a neural actor-critic architecture that generalizes across large state spaces while preserving interpretability. Simulations on stochastic matching models, including an organ exchange scenario, show that the orchestrated policy converges faster and yields higher system level efficiency than both individual experts and conventional RL baselines. Our results highlight how structured, adaptive learning can improve the modeling and management of complex resource allocation and decision-making processes

Effect of top metallic contacts on energy conversion performances for near-field thermophotovoltaics

    No full text
International audienceThe design of metallic contact grids on the front side of thermophotovoltaic cells is critical since it can cause significant optical and electrical resistive losses, particularly in the near field. However, from the theoretical point of view, this effect has been either discarded or studied by means of extremely simplified models like the shadowing methods, that consist in simply ignoring the fraction of the semiconductor surface covered by metal. Our study, based on a rigorous three-body theoretical framework and implemented using the scattering matrix approach with the Fourier modal method augmented with adaptive spatial resolution, provides deeper insight into the influence of the front metal contact grid. This approach allows direct access to the radiative power absorbed by the semiconductor, enabling the proposal of an alternative definition for the thermophotovoltaic cell efficiency. By modeling this grid as a metallic grating, we demonstrate its significant impact on the net radiative power absorbed by the cell and, consequently, on the generated electrical power. Our analysis reveals behaviors differing substantially from those predicted by previous simplistic approaches

Terrestrial Education and Marine Structural Design: A Post-Sustainability Framework for Ocean Engineering Innovation

    No full text
International audienceThis paper argues that in an era of accelerating climate change and rising sea levels, what truly matters is not merely the marine structure itself, but the foresight and adaptability embedded in its conception. As global urbanization intensifies along coastlines, and as storms grow stronger and ecosystems degrade, there is an urgent need for transformative approaches to ocean engineering and design. We introduce Terrestrial Education, an interdisciplinary framework that moves beyond conventional sustainability models by integrating ecological intelligence, regenerative systems thinking, and advanced technologies. Unlike traditional approaches that focus on minimizing harm, Terrestrial Education emphasizes active ecological restoration, socio-technical evolution, and planetary stewardship. Drawing on lessons from space exploration, such as closed-loop life support systems, autonomous environmental management, and modular habitat design, we demonstrate how these principles can inspire resilient marine infrastructures, including floating platforms, submerged laboratories, and bio-integrated offshore structures. These designs are envisioned not only for their physical durability but also for their capacity to regenerate ecosystems and foster meaningful human interaction with marine environments. The paper highlights key priorities such as adaptability to climate extremes, energy efficiency through marine renewables, and environmental integration using biomimetic materials. By aligning with the Sustainable Development Goals of the blue economy, Terrestrial Education offers a future-oriented model that harmonizes environmental, technological, and economic objectives. Ultimately, this framework provides a conceptual and operational foundation for reimagining marine structures as catalysts for innovation, educational transformation, and resilient planetary futures in the climate era

An evaluation of Sample Average Approximation applied to the design of impulsive thrust space collision avoidance maneuvers

    No full text
International audienceThis paper investigates how Sample Average Approximation (SAA) techniques can be applied tochance-constrained optimization problems defined for collision avoidance maneuvers design betweenan active satellite and a passive space debris. The objective is to compute impulsive evasive maneuversthat minimize the fuel consumption of the active asset and simultaneously maintaining the collisionprobability below a given threshold. A particular SAA algorithm is then proposed and compared againsta direct optimization algorithm using both synthetic and real conjunction data
