    Human action recognition (HAR) has emerged as a critical research area due to its wide range of real-world applications, including human-computer interaction, intelligent healthcare, virtual reality, surveillance, UAV control, and autonomous systems. Over the past few decades, numerous approaches have been developed to recognize human actions from monocular RGB video sequences. More recently, the advent of depth sensors has fueled the growth of 3D activity analysis and 3D gesture recognition, leading to significant advancements in the field. Among the various techniques proposed, manifold-based approaches have gained prominence due to their ability to effectively model the temporal evolution of 3D skeletal data through manifold-invariant representations. These methods have demonstrated remarkable performance in addressing the challenging task of action recognition.In this work, we explore the properties of the Symmetric Positive Definite (SPD) manifold, one of the most widely used manifolds in action and gesture recognition. We propose a novel recognition framework that integrates both a detector and a classifier, leveraging an SPD-based neural network known as the SPD Siamese Neural Network. We validate its performance through extensive experiments on both segmented and continuous action sequences across multiple datasets. Our results demonstrate that this approach outperforms state-of-the-art methods in various scenarios.Despite these advancements, significant challenges persist, particularly in complex environments such as UAV-based human action recognition. To overcome these limitations, we introduce an improved model, SPDAGG-TransNet, which enhances the baseline SPD Siamese network by refining its temporal-spatial feature extraction and integrating a Transformer module. This enhancement strengthens the model's ability to capture long-range dependencies, enrich feature representations, and maintain the intrinsic geometric properties of SPD representations. By incorporating Transformer encoders, our approach further improves recognition accuracy by effectively modeling both local and global motion dynamics. Extensive evaluations on benchmark datasets, including DHG-14, UAV-Human, and UAV-Gesture, demonstrate that SPDAGG-TransNet achieves state-of-the-art performance.Beyond SPD-based approaches, we also explore hyperbolic space as an alternative geometric framework for motion recognition. Hyperbolic neural networks (HNNs) offer a promising direction for modeling hierarchical and structured relationships in motion data. Unlike conventional Euclidean-based deep learning models, hyperbolic architectures leverage Lorentz transformations and novel optimization techniques, such as the Riemannian Adam optimizer, to stabilize embeddings and enhance scalability. These advancements enable more effective hierarchical motion modeling, making hyperbolic learning particularly suitable for action recognition tasks.Extensive experiments conducted across multiple benchmarks—ranging from hand gesture recognition to full-body action recognition and UAV-based datasets demonstrate the effectiveness of both SPD-based and hyperbolic-based approaches in challenging scenarios. Our findings highlight the superiority of geometric learning frameworks in accurately modeling human motion, ensuring real-time adaptability, and overcoming the limitations of traditional Euclidean methods.La reconnaissance des actions humaines (HAR) est devenue un domaine de recherche essentiel en raison de ses nombreuses applications dans le monde réel, notamment l'interaction homme-machine, la santé intelligente, la réalité virtuelle, la surveillance, le contrôle des drones (UAV) et les systèmes autonomes. Au cours des dernières décennies, de nombreuses approches ont été développées pour reconnaître les actions humaines à partir de séquences vidéo RGB monoculaires. Plus récemment, l'émergence des capteurs de profondeur a favorisé le développement de l'analyse des activités en 3D et de la reconnaissance des gestes en 3D, entraînant des avancées significatives dans le domaine. Parmi les différentes techniques proposées, les approches basées sur les variétés ont gagné en importance en raison de leur capacité à modéliser efficacement l'évolution temporelle des données squelettiques 3D grâce à des représentations invariantes aux variétés. Ces méthodes ont démontré des performances remarquables dans la résolution du défi de la reconnaissance des actions.Dans ce travail, nous explorons les propriétés de la variété des matrices Symmetric Positive Definite (SPD), l'une des plus utilisées en reconnaissance des actions et des gestes. Nous proposons un cadre de reconnaissance innovant intégrant un détecteur et un classificateur, en exploitant un réseau de neurones basé sur SPD, connu sous le nom de SPD Siamese Neural Network. Nous validons ses performances par le biais d'expériences approfondies sur des séquences d'actions segmentées et continues à travers plusieurs ensembles de données. Nos résultats montrent que cette approche surpasse les méthodes de l'état de l'art dans divers scénarios.Malgré ces avancées, des défis majeurs subsistent, en particulier dans des environnements complexes tels que la reconnaissance des actions humaines par drone (UAV). Pour pallier ces limitations, nous introduisons un modèle amélioré, SPDAGG-TransNet, qui optimise le réseau SPD Siamese en affinant l'extraction des caractéristiques spatio-temporelles et en intégrant un module Transformer. Cette amélioration renforce la capacité du modèle à capturer les dépendances à long terme, enrichir les représentations des caractéristiques et préserver les propriétés géométriques intrinsèques des représentations SPD. L'intégration d'encodeurs Transformer améliore encore la précision de la reconnaissance en modélisant efficacement les dynamiques locales et globales du mouvement. Des évaluations approfondies sur des ensembles de données de référence, notamment DHG-14, UAV-Human et UAV-Gesture, démontrent que SPDAGG-TransNet atteint des performances de pointe.Au-delà des approches basées sur SPD, nous explorons également l'espace hyperbolique comme cadre géométrique alternatif pour la reconnaissance des mouvements. Les réseaux de neurones hyperboliques (HNNs) constituent une voie prometteuse pour modéliser les relations hiérarchiques et structurées des données de mouvement. Contrairement aux modèles d'apprentissage profond conventionnels basés sur l'espace euclidien, les architectures hyperboliques exploitent les transformations de Lorentz et des techniques d'optimisation avancées, telles que Riemannian Adam optimizer, pour stabiliser les embeddings et améliorer l'évolutivité.     An Emergent Biosynthetic Pathway to Essential Amino Acids by Metabolic Metathesis

    International audienceAn experimental approach to implanting foreign chemical reactions into living cells is to select for the catalytic production of a vital building block such as an amino acid. Alkene metathesis is unknown in extant biochemistry, but it is emerging as a new type of reaction to be catalyzed by protein enzymes. Here, we show how the alkenic amino acid vinylglycine can be generated in a biocompatible reaction from 5-allyloxy-2-amino-pent-3-enoate (APE) by ring-closing metathesis catalyzed by a standard Hoveyda–Grubbs catalyst. The vinylglycine produced in situ is then used as a precursor of isoleucine and methionine, thus allowing the growth of strains of Escherichia coli requiring these essential amino acids. The robust nutritional screen we have developed paves the way for the directed evolution of genetically encoded metathesis enzymes and the chemical elaboration of metathesis coenzymes

    Méthodes d'analyse comparée des pangénomes procaryotes : explorer la diversité génomique inter-espèces pour une meilleure compréhension du métabolisme

    The boom in sequencing projects has generated over a million prokaryotic genomes in public databases, requiring new approaches to analyze this mass of data. The PPanGGOLiN software suite has been developed to structure this information in the form of pangenome graphs, enabling data compression while preserving gene colocalization information. It also integrates pangenome analysis methods : panRGP, which identifies regions of genomic plasticity, and panModule, which characterizes these variable regions into functional submodules. Despite these advances, there was no method for comparing pangenomes. The aim of this thesis was to develop new approaches to fill this gap. Firstly, PPanGGOLiN was enriched by integrating new methods, such as genomic context search, and by improving its software environment. Secondly, the PANORAMA method, based on PPanGGOLiN graphs, has been designed to annotate macromolecular systems, combining criteria of presence/absence of functions and genomic colocalization, and to compare pangenomes. Applied to bacterial phage defense systems, PANORAMA identified systems and insertion sites conserved between different species. Finally, a first prototype of a graph-oriented database was developed to integrate data from several pangenomes in order to make the most of their information. This approach has made it possible to analyze and compare thousands of bacterial genomes, and to dentify antibiotic resistance modules common to several species, highlighting shared evolutionary mechanisms. This work paves the way for comparative pangenomics, offering a novel framework for exploring the adaptive potential of prokaryotes and better understanding their evolutionary dynamics. By facilitating the comparison of pangenomes and the identification of conserved genomic contexts, these developments contribute to the study of interactions between bacteria and the characterization of biological systems of interest.L'essor des projets de séquençage a généré plus d'un million de génomes procaryotes dans les bases publiques, nécessitant de nouvelles approches pour analyser cette masse de données. La suite logicielle PPanGGOLiN a été développée pour structurer ces informations sous forme de graphes de pangénome, permettant de compresser les données tout en conservant l'information de colocalisation des gènes. Elle intègre également des méthodes d'analyse de pangénome, panRGP, qui identifie les régions de plasticité génomique, et panModule, qui caractérise ces régions variables en sous-modules fonctionnels. Malgré ces avancées, aucune méthode ne permettait de comparer des pangénomes. Les travaux de cette thèse ont consisté à développer de nouvelles approches pour combler cette lacune. Tout d'abord, PPanGGOLiN a été enrichie par l'intégration de nouvelles méthodes, comme la recherche de contextes génomiques, et par une amélioration de son environnement logiciel. Ensuite, la méthode PANORAMA, qui se base sur les graphes de PPanGGOLiN, a été conçue pour annoter des systèmes macromoléculaires, en combinant des critères de présence/absence de fonctions et de colocalisation génomique, et pour comparer des pangénomes. Appliqué aux systèmes de défense bactériens contre les phages, PANORAMA a permis d'identifier des systèmes et des sites d'insertions conservés entre différentes espèces. Finalement, un premier prototype de base de données orientée graphe a été développé pour intégrer les données de plusieurs pangénomes afin d'exploiter au mieux leur information. Cette approche a permis d'analyser et de comparer des milliers de génomes bactériens et d'identifier des modules d'antibiorésistance communs à plusieurs espèces, mettant en lumière des mécanismes évolutifs partagés. Ces travaux ouvrent la voie à la pangénomique comparée, offrant un cadre inédit pour explorer le potentiel adaptatif des procaryotes et mieux comprendre leur dynamique évolutive.     Patterns and drivers of diatom diversity and abundance in the global ocean

Here, we utilize Tara Oceans molecular and morphological data, spanning pole to pole, to describe marine diatom diversity, abundance, and environmental adaptation and acclimation strategies. The dominance of diatoms among phytoplankton in terms of relative abundance and diversity is confirmed, and the most prevalent genera are Chaetoceros , Thalassiosira , Actinocyclus and Pseudo-nitzschia . We define 25 distinct diatom communities with varying environmental preferences illustrative of different life strategies. The Arctic Ocean stands out as a diatom hotspot with 6 of the diatom communities being exclusive to it. Light harvesting and photoprotection are among the cellular functions in which natural diatom populations invest the bulk of their transcriptional efforts. This comprehensive study sheds light on marine diatom distributions, offering insights to assess impacts of global change and oceanic anthropogenic impacts

    The PP2CH- and PBL27-mediated phosphorylation switch of aluminium ion receptor PSKR1/ALR1 controls plant aluminum sensing ability

    International audienceThe ability of plants to sense toxic and nutrient ions is critical for their growth and survival, yet how this ability is regulated remains largely unknown. We previously identified the receptor-like kinase PSKR1/ALR1 (ALR1) in Arabidopsis as a receptor that senses phytotoxic aluminium (Al) ions, which cause severe crop yield loss and forest decline on acidic soils widely distributed over the world. Here we further show that the phosphorylation status of specific Ser residues in ALR1(Ser696/698) controls plant Al-sensing ability. ALR1(Ser696/698) phosphorylation levels are rapidly reduced by Al ions, and the dephosphorylation promotes the interaction and inter-phosphorylation of ALR1 and the BAK1 coreceptor, thereby activating STOP1-dependent Al signalling and resistance. We next identify a clade of PP2C-type phosphatases (PP2CH1 and PP2CH2) that mediate the dephosphorylation of ALR1(Ser696/698). We show that Al ions rapidly increase the protein accumulation of PP2CH1/2 and promote their interaction with ALR1. The lack of both PP2CHs notably increases the phosphorylation levels of ALR1(Ser696/698), therefore reducing the strength of Al signalling. Additionally, we found a receptor-like cytoplasmic kinase, PBL27, responsible for phosphorylating ALR1(Ser696/698) and playing a negative role in the regulation of ALR1-mediated Al signalling. These findings uncover a phosphatase/kinase-mediated phosphorylation switching mechanism of ALR1 that controls plant Al-sensing ability, providing insights into ion-sensing mechanisms in living organisms

    International audienceObjectives: The aim of this study is to assess the value of our gait quality measure [1], based on a multidimensional elastic distance, for quantifying the therapeutic effect on gait of botulinum toxin in rehabilitation.Research question: Does the use of several representatives of normal gait, combined with an elastic distance measure to quantify gait deviation, improve the evaluation of the therapeutic protocol? Method: This retrospective study was carried out on 52 control subjects and 21 patients with hemiparesis who received botulinum toxin injections during rehabilitation. We quantified the therapeutic effect by calculating the deviations from an elastic distance of the patients' cycles at three representatives of normal walking, before and after treatment. We compare this multidimensional measure with the Gait Deviation Index (GDI) [2] and the Global Variable Score (GVS) [3], assessing their respective ability to identify patients' paretic limb and monitor progress in rehabilitation.Results: Unlike the GDI and GVS, our measure enables us to detect the affected limb in patients before treatment. Significant decreases in deviations from norm representatives were observed after treatment with our measure in patients who had made great progress, while GDI and GPS varied little (Figure 1).Conclusions: The study shows that the use of several normal gait references, combined with an elastic distance measurement, enables accurate spatio-temporal gait analysis.     Government decisions for relief materials reserve under supply uncertainty within government-enterprise agreement

    International audiencePre-positioned inventory of relief materials is crucial for the rapid response to potential disasters. Storing relief materials near disaster-prone areas facilitates quick delivery after a disaster. Consequently, governments often collaborate with local enterprises and stores to store materials. However, the proximity to disaster sites can result in the damage of relief materials, exacerbating shortages, particularly during major disasters. Despite this, the literature has not thoroughly addressed how governments pre-position materials considering damage risk. Therefore, this paper constructs a model for the relief supply chains, including a local government, a local supplier and a non-local supplier to determine the government's optimal reserve strategy and quantity. The demand and damage to materials depend on disaster intensity. We derive the conditions under which a specific reserve strategy (no-external reserve, local reserve, non-local reserve, bi-site reserve) is optimal and corresponding reserve quantity. The impacts of supplier characteristics, government-owned reserves, emergency procurement price, disaster intensity distribution, demand-intensity and supply-intensity relationships on optimal prepositioned reserves are identified.     Towards Universal and Black-Box Query-Response Only Attack on LLMs with QROA

    The rapid adoption of Large Language Models (LLMs) has exposed critical security and ethical vulnerabilities, particularly their susceptibility to adversarial manipulations. This paper introduces QROA, a novel black-box jailbreak method designed to identify adversarial suffixes that can bypass LLM alignment safeguards when appended to a malicious instruction. Unlike existing suffix-based jailbreak approaches, QROA does not require access to the model’s logit information or any other internal information. It also eliminates reliance on human-crafted templates, operating solely through the standard query-response interface of LLMs. By framing the attack as an optimization bandit problem, QROA employs a surrogate model and token-level optimization to efficiently explore suffix variations. Furthermore, we propose QROA-UNV, an extension that identifies universal adversarial suffixes for individual models, enabling one-query jailbreaks across a wide range of instructions. Testing on multiple LLMs—including Vicuna, LLama2-chat, Mistral-Large, GPT-3.5, and GPT-4—demonstrates Attack Success Rates (ASR) exceeding 80\% within practical query budgets. These findings highlight critical vulnerabilities, emphasize the need for advanced defenses, and contribute to the development of more robust safety evaluations for secure AI deployment.
