Hydroclimate and Extreme Rainfall Variability over the Past 7500 years in Southeastern and Southern Brazil
Southern and Southeastern Brazil are strongly affected by extreme rainfall events (ERE), which frequently trigger landslides and flash floods, causing severe socioeconomic impacts that is proportionately affect vulnerable populations. Current climate projections indicate that the frequency and magnitude of ERE are likely to increase due to enhanced atmospheric moisture and evaporative demand under global warming. In addition, El Niño–Southern Oscillation (ENSO) events are projected to intensify and occur more frequently, further amplifying ENSO-related extremes through Pacific–South American teleconnections. Despite these projections, the lack of reliable ERE records extending beyond the period of anthropogenic climate change constrains our understanding of the natural variability and forcing mechanisms behind such events, particularly over longer time-scales. In this context, speleothems collected from caves subjected to flood in response to ERE offer a valuable archive for reconstructing past ERE occurrences. The integration of cave hydrological monitoring with paleoclimate reconstructions provides a robust framework for investigating the natural variability of ERE. Precise 230Th–U dating combined with detailed microstratigraphic analyses provide a robust framework for the reconstruction of ERE time series and allows us to assess how their frequency have changed through time. Furthermore, speleothems offer the potential for multi-proxy analyses, allowing the reconstruction of hydroclimate and environmental conditions across multiple temporal scales along with the ERE. By combining ERE reconstructions with a multi-proxy approach, we can generate a comprehensive record of past ERE variability and hydroclimatic changes, allowing us to evaluate the how forcings and changes in the hydroclimate impact the ERE occurrence. In this study, we use multiple speleothems from Malfazido Cave to reconstruct extreme rainfall variability over the past 7,500 years. A multi-year cave monitoring program has evidenced Malfazido Cave response to extreme rainfall events. Stable isotope (δ18O and δ13C) and trace element records reveal persistent interannual periodicities consistent with ENSO variability, as well as multi-centennial cycles evidencing a pattern of shifting wet/dry phases over Southeastern Brazil. The wet anomalies are synchronous with events of North Atlantic cooling, such as the Little Ice Age and Bond events, and are associated with a weakening of the Atlantic Meridional Overturning Circulation (AMOC) and large-scale atmospheric reorganization related to shifts in the Intertropical Convergence Zone (ITCZ). Our results also evidenced a multi-centennial rainfall dipole pattern between Southern (SB) and Southeastern Brazil (SEBRA), resembling the intraseasonal South American Dipole. Switchable valence states in dinuclear cobalt cmplexes : the role of halogenated catecholates and counterions
We present the synthesis and comprehensive characterization of a series of eight dinuclear cobalt complexes, C1–C8, with the general formula [Co2(L)(X4cat)2]2+(A)1–2, wherein X = Br or Cl, A = SO42–, ClO4–, PF6–, or B(Ph)4–, and L denotes a redox inactive bis-tetradentate bridging ligand. Single-crystal X-ray diffraction at 120 K confirms a low-spin Co(III) configuration in all compounds. SQUID magnetometry also shows that the complexes remain diamagnetic below room temperature. However, the complexes bearing sulfate anions, C1 and C5, exhibit a distinct thermally induced valence tautomeric transition above room temperature, marked by an increase in magnetic moment. The temperature of this transition is strongly influenced by the electronic properties of the catecholate (cat) ligands, with electron-deficient tetrahalogenated catecholates stabilizing the low-spin state. In addition, counterions and solvent molecules are found to modulate intermolecular interactions in the solid state. Comparative cyclic voltammetry with previously reported ditert-butyl catecholate (dbucat) complex C9 highlights the influence of ligand electronics on redox potentials, with electron-deficient catecholates shifting redox processes to higher potentials. Interleukin-1 signaling in the blood-brain barrier influences the behavioral response to chronic social stress
Chronic stress is a major risk factor for depression, leading to systemic immune activation and neuroinflammation, both of which contribute to the development of depressive symptoms. The blood-brain barrier (BBB), with endothelial cells as a central component, plays a crucial role in maintaining central nervous system (CNS) homeostasis by regulating the exchange of molecules and immune cells between the blood and CNS. However, the precise impact of chronic stress on BBB function and whether BBB components contribute to the establishment of depression-like behavior following stress remain unclear. Interleukin-1 (IL-1), a key pro-inflammatory cytokine, plays a central role in the development of stress-induced behavioral changes. Its receptor, IL-1R1, is primarily expressed by BBB endothelial cells (BECs) in the CNS, but its expression is restricted to a small subset of these cells under steady-state conditions. Chronic social defeat (CSD) stress has been shown to upregulate IL-1R1 expression on BECs, potentially driving BBB activation during stress. Although systemic endothelial IL 1R1 deletion has been shown to alleviate stress-induced behavioral symptoms, it remains unclear whether IL-1 signaling specifically in BECs mediates these effects.
Using the Slco1c1-CreERT2 mouse strain to delete IL-1R1 selectively in BECs, this study aimed to investigate the role of IL-1 signaling in BECs during CSD and its contribution to depression-like behavior. Here, we show that IL-1 signaling in BECs is critical for mediating stress-induced behavioral dysfunction, as conditional deletion of IL-1R1 in BECs increased stress resilience and ameliorated behavioral changes following CSD. We found that IL-1R1⁺ BECs constitute a small, distinctly activated BEC subset under steady-state conditions. CSD stress further increased IL-1R1 expression on BECs, which was essential for their activation during stress. Notably, IL-1 signaling in BECs influenced behavior independently of peripheral immune cell infiltration or neuroinflammation, suggesting that alternative mechanisms are involved.
While IL-1 signaling is a well-established key regulator of chronic stress responses, our results demonstrate that this effect is specifically mediated through BECs, highlighting the importance of the BBB in the chronic stress response. Taken together, our findings suggest that targeting IL-1 signaling in BECs could be a promising strategy for mitigating stress-induced neurovascular dysfunction and its associated behavioral consequences.Chronischer Stress ist ein wichtiger Risikofaktor für die Entwicklung von Depressionen. Er bewirkt eine anhaltende Aktivierung des Immunsystems sowie Neuroinflammation, wodurch die Entstehung depressiver Symptome begünstigt wird. Die Blut-Hirn-Schranke (BHS) mit ihren Endothelzellen als zentralem Bestandteil steuert den Austausch von Molekülen und Immunzellen zwischen Blut und Zentralnervensystem (ZNS) und trägt somit maßgeblich zur Aufrechterhaltung des Gleichgewichts im ZNS bei. Allerdings ist bislang ungeklärt, wie chronischer Stress die Funktion der BHS beeinflusst und inwieweit ihre Bestandteile zur Entstehung depressiven Verhaltens nach Stress beitragen. Interleukin-1 (IL-1) ist ein wichtiges pro-entzündliches Zytokin mit einer zentralen Rolle in der Entstehung stressinduzierter Verhaltensstörungen. Sein Rezeptor, IL-1R1, wird im ZNS unter homöostatischen Bedingungen hauptsächlich von Endothelzellen der BHS exprimiert, jedoch nur von einem kleinen Anteil dieser Zellen. Es wurde gezeigt, dass „chronischer sozialer Niederlage“ (CSD)-Stress die IL-1R1-Expression in den BHS-Endothelzellen erhöht, was zur Aktivierung der BHS während Stress beitragen könnte. Die Deletion des IL-1R1 in Endothelzellen des gesamten Körpers konnte stressinduzierte Verhaltensstörungen teilweise verhindern. Allerdings ist bislang unklar, ob dieser Effekt durch IL-1-Signale speziell in Endothelzellen der BHS vermittelt wird.
Zur Klärung dieser Frage verwendeten wir das Slco1c1-CreERT2-Mausmodell, das die gezielte Deletion von IL-1R1 in BHS-Endothelzellen ermöglicht. Unsere Ergebnisse zeigen, dass IL-1-Signale in den Endothelzellen der BHS eine zentrale Rolle in der Entwicklung stressinduzierter Verhaltensstörungen spielen, denn die Deletion von IL-1R1 in diesen Zellen verringerte die CSD-induzierten Verhaltensstörungen signifikant. IL-1R1⁺ Endothelzellen bildeten unter homöostatischen Bedingungen eine kleine Zellpopulation mit aktiviertem Phänotyp. CSD führte zu einer verstärkten IL-1R1-Expression in diesen Zellen, die essenziell für ihre Aktivierung während CSD-Stress war. Interessanterweise beeinflusste der IL-1-Signalweg in BHS-Endothelzellen das Verhalten unabhängig von der Infiltration peripherer Immunzellen oder einer generellen Neuroinflammation, was auf andere involvierte Mechanismen hindeutet. Während die Rolle des IL-1-Signalwegs in chronischem Stress bereits etabliert war, zeigen unsere Ergebnisse, dass diese Effekte unter anderem über BHS-Endothelzellen vermittelt werden und unterstreichen die Bedeutung der BHS in der Regulation der Stressantwort. Unsere Ergebnisse legen nahe, dass die gezielte Modulation von IL-1-Signalen in BHS-Endothelzellen eine vielversprechende Strategie zur Verhinderung stressinduzierter BHS-Dysfunktionen und der damit verbundenen Verhaltensveränderungen darstellen könnte.102 Seiten ; Illustrationen, Diagramm
Retrospektive Auswertung und Vergleich von assistierenden Techniken bei stabilisierenden Operationen an der Wirbelsäule am Beispiel von 3D-Navigation und Roboter-assistierten Verfahren
Diese Dissertation untersucht die Genauigkeit der Pedikelschraubenlage von assistierenden Techniken bei stabilisierenden Operationen an der Wirbelsäule, insbesondere durch den Vergleich von 3D-Navigation und Roboter-assistierten Verfahren. Die operative Stabilisierung der Wirbelsäule mit einem Schrauben-Stab-System ist ein anerkanntes Verfahren zur Behandlung von Erkrankungen oder Verletzungen, die zu einer Instabilität der Wirbelsäule führen.
Die korrekte Lage der Pedikelschrauben ist entscheidend für die Stabilität des implantierten Systems und die Vermeidung von Komplikationen wie Verletzungen der Nervenwurzeln. Verschiedene Studien und Metaanalysen haben gezeigt, dass die Unterstützung durch 3D-Navigation oder Operationsroboter zu einer genaueren Schraubenlage und geringeren Komplikationsraten führt. Weiterhin zeigten besagte Untersuchungen, dass Navigationssysteme, welche auf 3D-flouriskopischen Daten, also einem intraoperativen CBCT beruhen, genauere Pedikelschraubenlagen zu liefern scheinen, als solche Systeme, welche auf einem präoperativen CT-Bild beruhen. Der Renaissance-Roboter stellt einen Vertreter dieser CT-basierten Systeme dar, das Brainlab-System bietet beide Technologien an. Beim 3D-Scan wird eine intraoperative 3D-fluoroskopie genutzt, beim Surface-Matching präoperative CT-Daten.
An insgesamt 268 Operationen und 1718 Pedikelschrauben, welche in der Zeit zwischen 2018 und 2022 an der neurochirurgischen Klinik der Universtitätsmedizin Mainz mit Hilfe des Renaissance-Systems, sowie dem Brainlab-System implantiert wurden, wurde retrospektiv eine Auswertung der Pedikelschraubenlagen anhand von CT-Bildern durchgeführt. Hierfür wurde jeder Pedikelschraube ein Grad zwischen 0 und 4 zugewiesen. Weiterhin wurden die OP-Zeit, emittierte Röntgenstrahlung, demographische Daten, Größe der Pedikelschrauben, Lokalisation der Pedikelschrauben, Indikationen, sowie intraoperative Komplikationen oder notwendige Revisionseingriffe erfasst.
Von den 1718 Pedikelschrauben entfielen 728 auf den Roboter, 990 auf das Brainlab-System. Von letzteren wurden 579 Pedikelschrauben hierbei nach Referenzierung mit eine 3D-Fluoroskopie implantiert, 411 nach Referenzierung über ein Surface-Matching mit präoperativen CT-Daten. Durchschnittlich erzielte der OP-Roboter hierbei eine Pedikelschraubenlage von 0.971 Grad, das Brainlab-System insgesamt eine durchschnittliche Lage von 0.866 Grad (-0.105 Grad). Unterteilt in die Subgruppen nach Referenzierung ergab sich bei der 3D-Fluoroskopie eine durchschnittliche Pedikelschraubenlage von 0.808 Grad, beim CT-basierten Surface-Matching von 0.949 Grad. Die CT-basierten Navigationen Roboter und Surface-Matching divergierten also um lediglich 0.022 Grad. Die Ergebnisse dieser Untersuchung decken sich also mit den bisherigen Metaanalysen und zeigen eine durchschnittlich bessere Pedikelschraubenlage bei der Nutzung von intraoperativer 3D-Fluoroskopie bei der navigierten Implantation von Pedikelschrauben. Auch hinsichtlich der definierten Fehllagen im Grad 3 und Grad 4 zeigte das Brainlab-System mit 9.39 % der Pedikelschrauben eine um 2.29 % geringere Fehllagenrate als der Roboter mit 11.68 %. Eine medial perforierende Schraubenfehllage trat bei beiden Systemen jedoch lediglich in 1.65 % der Pedikelschrauben (Roboter) und 0.6 % der Pedikelschrauben (Brainlab) auf. Beide Systeme und ihre jeweiligen Referenzierungsverfahren stellen also eine sichere und valide Technik zur navigierten Pedikelschraubenimplantation dar.
Den statistisch signifikantesten Einfluss auf die Pedikelschrauben stellte der Wirbelsäulenabschnitt dar. Die thorakale Wirbelsäule bietet die anatomisch kleinsten und engsten Pedikel der untersuchten Abschnitte, entsprechend fanden sich hier die häufigste Anzahl an Fehllagen (64.05 % aller Fehllagen) und die schlechtesten durchschnittlichen Pedikelschraubenlagen mit 1.232 Grad. Rein lumbosakral durchgeführte Eingriffe erzielten im Vergleich eine durchschnittliche Schraubenlage von 0.339 Grad (-0.893 Grad). Gerade im rein thorakalen Abschnitt erzielte das Brainlab-System (1.147 Grad) eine um 0.272 Grad bessere durchschnittliche Schraubenlage als der Roboter (1.419 Grad). In der Subgruppenbetrachtung des Brainlab zeigte sich dann, dass das Surface-Matching in diesem Wirbelsäulenabschnitt um 0.132 Grad bessere durchschnittliche Pedikelschraubenlage erzielte als die Referenzierung über eine 3D-Fluoroskopie. Auch die Anteile der Fehllagen im Vergleich zur Gruppengröße waren hier signifikante niedriger. Bei Operationen an der lumbosakralen Wirbelsäule (also dem Segment LW 5/ SW 1) erzielte das Surface-Matching sogar eine durchschnittliche Schraubenlage von 0 Grad, es kam zu keiner Fehllage. Divergierend hierzu kam es jedoch im thorakolumbalen Übergang beim Surface-Matching zu der schlechtesten durchschnittlichen Pedikelschraubenlage des Brainlab-Systems mit 1.246 Grad, sowie den anteilig meisten Fehllagen mit 25.71 % aller im Grad 3 und Grad 4 erfassten Pedikelschrauben bei lediglich 6.78 % der Operationen.
Es bleibt also festzustellen, dass innerhalb des Brainlab-Systems das Surface-Matching für die thorakale Wirbelsäule, sowie den lumbosakralen Übergang eine der 3D-Fluoroskopie überlegene Referenzierungsmethode darstellt und der klinische Einsatz hier zu bevorzugen wäre. In den übrigen Wirbelsäulenabschnitten zeigte sich die 3D-Fluoroskopie hinsichtlich durchschnittlicher Pedikelschraubenlage und Rate an Fehllagen überlegen.
Einen weiteren Einflussfaktor auf die Systeme stellen Größe und Gewicht des Patienten, sowie der daraus resultierende BMI dar. Hinsichtlich der Größe bestand ein statistisch valider Zusammenhang – eine größere Körperlänge führte zu durchschnittlich besseren Pedikelschraubenlagen. Dieser Zusammenhang konnte statistisch valide für den BMI bei der durchschnittlichen Pedikelschraubenlage nicht gezeigt werden, bei übergewichtigen Patienten (Präadipositas, sowie Adipositas Grad 1) kam es jedoch zu vermehrten Fehllagen als bei normalgewichtigen Patienten. Vor allem bei Adipositas Grad 1 kam es beim 3D-Scan zu vermehrten Fehllagen, das Surface-Matching zeigte diesen Effekt nicht.
Die Indikation zur dorsalen Spondylodese hat keinen Einfluss auf die Pedikelschraubenlage. Es konnte gezeigt werden, dass auch hier der Wirbelsäulenabschnitt, in der die jeweilige Operation durchgeführt wird, den größten Einfluss auf die Pedikelschraubenlage hat.
Ein weiterer statistisch signifikanter Einflussfaktor auf die durchschnittliche Pedikelschraubenlage stellt hingegen die Länge der dorsalen Spondylodese dar. Mit zunehmender Länge kam es auch zu einer Zunahme der durchschnittlichen Graduierungen der Pedikelschrauben. Hierbei stellten sich besonders die Spondylodesen über 3, 4 und 5 Segmente als anfällig für Fehllagen und damit eine schlechtere durchschnittliche Pedikelschraubenlage heraus. Systemspezifisch zeigte der Roboter bei der Spondylodese über 4 Segmente eine deutlich erhöhte Anzahl an Fehllagen (45.88 % aller Fehllagen), das Brainlab-System bei Spondylodesen über 3 Segmente, sowie über 5 Segmente mit kumuliert 45.16 % der Fehllagen des Systems. Diese Auffälligkeit trat beim Brainlab-System jedoch ausschließlich bei der Registrierung über die 3D-Fluoroskopie auf. Eine Ursache für dieses Phänomen ließ sich anhand der Daten nicht ableiten. Eine Ungenauigkeit durch manuelles Umsetzen des Roboters, sowie eine möglicherweise ausgebliebene neue Referenzierung beim Brainlab-System mit zunehmender Entfernung zum Referenzstern könnten eine Ursache darstellen. Spezielle Workflows für die Nutzung von navigierenden Techniken an der Wirbelsäule sind von immanenter Wichtigkeit für den Erhalt der Präzision, gerade bei langstreckigen Spondylodesen.
Hinsichtlich der benötigen Zeit/Schraube ergaben sich keine relevanten Unterschiede zwischen den Systemen.
Die erfasste intraoperative Durchleuchtung zeigte beim OP-Roboter die niedrigsten durchschnittlichen Dosisflächenprodukte mit 508.9 cGycm². Ähnliche Werte waren a priori auch für das Surface-Matching zu erwarten. Mit durchschnittlich 1084 cGycm² waren die Dosisflächenprodukte jedoch nahezu doppelt so hoch, was durch mangelnde Erfahrungswerte mit dem System und deshalb durchgeführte Röntgenkontrollen zu erklären wäre. Mit durchschnittlich 1467 cGycm² waren die intraoperativen Dosisflächenprodukte bei der 3D-Fluoroskopie am höchsten, eine präoperative CT-Bildgebung entfällt hier bei der Betrachtung der Gesamtdosis jedoch.XIII, 150 Seiten ; Illustrationen, Diagramm
Ensuring news quality in platformized news ecosystems : shortcomings and recommendations for an epistemic governance
Social media platforms are fundamentally disrupting public communication in two ways. First, non-journalistic actors, such as social media influencers, now have easier access to audiences, increasing the range of epistemic authorities. Second, established news outlets are increasingly reliant on platforms, pressuring them to adapt to the demands of social media. This platformization threatens the quality of public communication and citizens’ realization of their epistemic rights. Drawing on these concerns, this article offers a holistic approach to systematically analyze and practically govern epistemic threats to news quality in the platform-driven hybrid news ecosystem by developing a comprehensive three-stage framework that distinguishes between the production, distribution, and reception of news quality. News quality needs not only to be produced (input) but also to be made visible on platforms (throughput) and processed by audiences (output) in order to contribute to an informed public discourse. Focusing on the EU, the article then discusses current regulatory shortcomings and the need for additional measures to safeguard news quality along its three dimensions
Surgical management of ruptured intracranial arachnoid cysts in young adults : the role of subduroperitoneal shunt placement
Introduction:
Intracranial arachnoid cysts (IAC) are a common incidental finding on brain MRIs in adults. While IAC rupture, leading to severe intracranial hypertension, is well-documented in children, it is rarely reported in adults. As a result, there is limited guidance on managing ruptured IACs in young adults.
In particular, there is no data on the benefits of subduroperitoneal (SDP) shunt placement without interposed pressure valves in young adult patients, a treatment often used in pediatric patients.
Methods:
We present two cases of young adult patients presenting with ruptured IAC with subsequent subdural hematoma/collection and consecutive neurological decline. Based on these sample clinical cases, we will discuss the above-mentioned management strategy and provide an overview of the current available literature.
Results:
This case series demonstrates the successful management of ruptured IAC in young adults through drainage of subdural fluid collections via burr hole trepanation, combined with SDP shunt placement without interconnected pressure valves.
Conclusion:
While there is general consensus that surgery is necessary for symptomatic cases following IAC rupture, the optimal approach remains debated. We provide additional evidence that burr hole trepanation combined with SDP shunt placement without interconnected pressure valves, a method commonly used in children, is a safe treatment option for young adults with ruptured IAC
Unveiling the evolutionary and ecological effects of transgenerational plasticity in monoclonal giant duckweeds
Recent studies have demonstrated that stress experienced by previous generations can
influence the phenotype of their descendants, yet its impact on fitness and adaptability remains
unclear. To investigate this, we exposed multiple genotypes of the clonal giant duckweed
(Spirodela polyrhiza) to copper excess and herbivory by the waterlily aphid (Rhopalosiphum
nymphaeae) over five generations. After another five generations without stress, we
reintroduced both abiotic and biotic stressors under laboratory and natural conditions. Through
ancestral exposure to copper excess in 56 globally distributed genotypes grown under axenic
conditions and minimized natural selection, we found consistent evidence that
transgenerational plasticity is heritable, reproducible, and dependent on both genotype and
environment. The trade-offs of this plasticity could be predicted by the initial resistance of the
plants to stress, likely due to resource allocation. Physiological analysis revealed that ancestral
copper excess increased cyanidin concentrations, likely protecting photosystem II efficiency.
These findings suggest that transgenerational plasticity is regulated by genes involved in plant
reproduction and defence. Additionally, plants exposed to ancestral copper excess boosted
aphid fitness. Unexpectedly, copper-induced transgenerational jasmonates, when
supplemented to plants, benefitted the waterlily aphid, demonstrating that copper-induced
transgenerational plasticity can affect interacting species. Similarly, ancestral aphid herbivory
led to maladaptive effects under recurring herbivory, further increasing aphid reproduction.
Metabolomic and transcriptomic analyses revealed that, in addition to increased jasmonates,
aphid herbivory transgenerationally induced the accumulation of tyramine through regulation
of its biosynthetic machinery. Supplementation of tyramine to plants and aphids confirmed its
adaptive role against herbivory. Under natural conditions, tyramine, but not jasmonates,
exhibited transgenerational plasticity over two consecutive years, and accordingly, aphid
fitness tended to decline outdoors, highlighting the relevance of executing experiments under
both laboratory and natural conditions. Together, these findings demonstrate that
transgenerational plasticity is heritable and plays a crucial role in organismal and community
evolutionary change, as it not only shapes the fitness and phenotype of the affected organisms

Plastizität vererbbar, reproduzierbar und sowohl vom Genotyp als auch von der Umwelt
abhängig ist. Die Kompromisse dieser Plastizität ließen sich anhand der anfänglichen
Widerstandsfähigkeit der Pflanzen gegenüber Stress vorhersagen, was wahrscheinlich auf die
Ressourcenallokation zurückzuführen ist. Physiologische Analysen zeigten, dass der
Kupferüberschuss der Vorfahren die Cyanidin-Konzentration erhöhte, was wahrscheinlich die
Effizienz des Photosystems II schützte. Diese Ergebnisse deuten darauf hin, dass
transgenerationale Plastizität durch Gene reguliert wird, die an der Fortpflanzung und Abwehr
von Pflanzen beteiligt sind. Zudem steigerten Pflanzen, die einem Kupferüberschuss der
Vorfahren ausgesetzt waren, die Fitness der Blattläuse. Unerwarteterweise hatten
kupferinduzierte transgenerationale Jasmonate, die den Pflanzen zugesetzt wurden, einen
positiven Effekt auf die Wasserlilienblattlaus, was zeigt, dass kupferinduzierte
transgenerationale Plastizität auch interagierende Arten beeinflussen kann. In ähnlicher Weise
führte die Herbivorie durch die Vorfahren bei wiederholter Herbivorie zu Fehlanpassungen,
was die Reproduktion der Blattläuse weiter steigerte. Metabolomische und transkriptomische
Analysen zeigten, dass die Herbivorie durch Blattläuse, neben erhöhten Jasmonaten, auch
die Ansammlung von Tyramin transgenerational induzierte, indem sie den
Biosynthesemechanismus regulierte. Die Gabe von Tyramin an Pflanzen und Blattläuse
bestätigte dessen adaptive Rolle im Kampf gegen Herbivorie. Unter natürlichen Bedingungen
zeigte Tyramin, im Gegensatz zu Jasmonaten, über zwei aufeinanderfolgende Jahre
transgenerationale Plastizität, während die Fitness der Blattläuse im Freien tendenziell
abnahm. Dies unterstreicht die Relevanz der Durchführung von Experimenten sowohl unter
Labor- als auch unter natürlichen Bedingungen. Zusammengefasst zeigen diese Ergebnisse,
dass transgenerationale Plastizität vererbbar ist und eine entscheidende Rolle bei der
evolutionären Veränderung von Organismen und Lebensgemeinschaften spielt, da sie nicht
nur die Fitness und den Phänotyp der betroffenen Organismen prägt, sondern auch

Pre-analytical pitfalls : how blood collection tubes influence exercise-induced cell-free DNA concentrations
Circulating cell-free DNA (cfDNA) is a promising biomarker for physiological stress, including exercise-induced responses. However, the lack of standardization in blood collection tubes (BCTs) for quantification of cfDNA hampers inter-study comparisons. In this study, we assessed the impact of different BCTs on exercise-induced cfDNA dynamics. Eleven participants [25 (SD 2.3) years of age] performed three different treadmill exercise protocols, including an all-out test and combinations of constant and interval load. Blood samples were collected before, 5 min and 30 min post-exercise using EDTA, lithium–heparin (LH) and serum BCTs. Concentrations of cfDNA were quantified using quantitative PCR. The cfDNA increased significantly across all protocols and BCTs. A significant effect of BCT on cfDNA concentrations (P = 0.034) was found, with serum showing higher concentrations than EDTA and LH. Although absolute differences from pre- to post-exercise were comparable across BCTs (P = 0.476), fold changes differed significantly (P = 0.012), with the highest observed in EDTA and the lowest in serum. Bland–Altman analyses demonstrated better agreement between EDTA and LH compared with serum. Significant correlations of cfDNA with energy expenditure and peak oxygen uptake were found. These correlations were stronger in EDTA and LH than in serum. Our findings highlight the crucial influence of BCT choice on cfDNA measurements in exercise settings. Given that EDTA and LH reflected exercise load better, they could be preferred for exercise physiology research. This work underscores the need to account for the choice of BCT to improve data comparability across studies. Additionally, these findings might have broader implications for clinical settings where cfDNA is used as a biomarker
Investigating ice formation pathways using a novel two-moment multi-class cloud microphysics scheme
For pure ice clouds in the cold-temperature regime (T<235 K), two major formation pathways are possible. Liquid origin ice clouds stem from droplets that freeze close to water saturation. In-situ-formed ice clouds form directly from the vapor phase below water saturation. For a better investigation of these pathways, we developed a novel microphysics scheme. The new two-moment scheme distinguishes between five ice classes (“ice modes”) each with their own unique formation mechanism: homogeneous freezing of solution droplets, deposition nucleation, homogeneous freezing of cloud droplets and raindrops, immersion freezing, and secondary ice from rime splintering. The ice modes interact with each other, e.g., in competition for growth by deposition of water vapor and aggregation, but also with the other cloud particle classes, i.e., cloud droplets, rain, snow, graupel, and hail.
This scheme was employed to investigate the liquid origin vs. in situ formation in the fully glaciated parts of an idealized convective cloud. The majority of the cloud ice in the deep convection cloud consisted of frozen droplets (liquid origin). This was caused by the high number concentration of cloud droplets available for freezing. In-situ-formed ice was only relevant for the overshoot where ice from both formation pathways mixed.
The new scheme is also useful for investigation of the ice formation in the mixed-phase parts of the convective cloud. We find a vertical layering of ice modes in the cloud. The lowermost layer consists of secondary ice from rime splintering and occurs near the updraft core at temperatures around the Hallett–Mossop zone. At altitudes between 6 and 9 km, ice mostly stems from immersion freezing. We find a correlation between the abundance of ice from immersion freezing and snow. The majority of ice crystals above 9 km stems from homogeneously frozen cloud droplets since ice-nucleating particles (INPs) required for immersion freezing were quickly depleted
