Intermetallische Al9-xFexMo3-Phasen und Eisenmolybdate: Struktur, Magnetis-mus und katalytische Funktionalität
In der vorliegenden Arbeit werden die Reaktivitäten und die Kristallstrukturen von ternären, intermetallischen Phasen Al9-xFexMo3 durch eine Kombination aus ex situ- und in situ-Röntgendiffraktion untersucht. Durch eine Variation des Eisengehaltes werden explizit der Einfluss des Eisens auf die strukturellen und physikalischen Eigenschaften der inter-metallischen Phasen untersucht. Der nominelle Eisengehalt xFe, nom der synthetisierten Proben wurde zwischen 1 und 2.5 gewählt, um die maximal mögliche FeAl Substitution zu bestimmen. Die Proben wurden in der Lichtbogenschmelze hergestellt und bei 600 °C unter Inertgas-Atmosphäre nachbehandelt. Dabei konnte gezeigt werden, dass die τ1-Phase (Al8.5Fe0.5Mo3) eine tetragonale Struktur aufweist, die als Substitutionsvariante des Al3Ti-Strukturtyps zu beschreiben ist. Eine orthorhombisch verzerrte Variante wird für höhere Eisengehalte (xFe) zwischen 1 und 1.8 gefunden. Eine reversible Phasenumwandlung zwischen beiden Strukturtypen findet bei ca. 300 °C statt. Neben der τ1-Phase wird auch die nichtstöchiometrische τ2-Phase erhalten. Diese metastabile Verbindung zerfällt oberhalb von etwa 425 °C. 57Fe-Mössbauerspketroskopische und magnetische Untersuchungen zeigen charakteristische Merkmale eines ungeordneten magnetischen Zustands, der mit einem Spin-Glas-Verhalten bei tiefen Temperaturen einhergeht.
Die Oxidation der intermetallischen Phasen führt u. a. zu Fe2(MoO4)3. Diese Verbindung wurde im Rahmen der folgenden Arbeit im Hinblick auf die erforderliche Aktivierung eines Katalysators für die oxidative Dehydrogenierung (ODH) von Ethanol zu Acetaldehyd eingehend untersucht. Der Fokus dabei liegt auf der Syntheseroute und dem kontrollierten Einbringen struktureller Defekte mittels mechanochemischer Verfahren (Kugelmühle). Diese werden im Rahmen der vorliegenden Arbeit eingehend mit Hilfe der Röntgendiffraktion untersucht. Die Untersuchungen der thermodynamischen Eigenschaften und die Ergebnisse der 57Fe-Mössbauer-Spektroskopie liefern ein tieferes Verständnis der optimierten Katalysatoren mit kontrollierter „Subsurface“ Defektkonzentration. Der optimale Temperaturbereich der ODH-Katalyse, bezogen auf sehr hohe Selektivität und Konversion (> 80%), liegt bei ca. 280 °C und entspricht anwendungstechnischen Anforderungen. Bei Temperaturen oberhalb von 300 °C treten vermehrt Rekristallisationsprozesse (Defektheilungsprozesse) auf.In the present work, the crystal structures and physical properties of ternary intermetallic phases
Al9-xFexMo3 are investigated using a combination of ex situ and in situ X-Ray diffraction. Varying the iron content, the influence of iron on the structural and physical properties of the intermetallic phases is explicitly investigated. The nominal xFe, nom content of the synthesized samples was chosen between 1 and 2.5 to determine the maximum FeAl substitution. The samples were synthesized in an arc furnace and post-treated at 600 °C in an inert gas atmosphere. It was shown that the τ1-phase (Al8.5Fe0.5Mo3) has a tetragonal structure which can be described as a substitution variant of the Al3Ti structure type. In case of higher iron contents (xFe), an orthorhombic distorted variant was found between 1 and 1.8. At approx. 300 °C, a reversible phase transformation between the two structure types takes place. In addition to the τ1-phase the non-stochiometric τ2-phase is also obtained. This metastable compound decomposes above approx. 425 °C. 57Fe-Mössbauer spectroscopic and magnetic investigations show characteristic features of a disordered magnetic state associated with spin glass behavior at low temperatures.
The oxidation of intermetallic phases leads, among side phases, to Fe2(MoO4)3. In this thesis, this compound was investigated in detail regarding the required activation of a catalyst for the oxidative dehydrogenation (ODH) of ethanol to acetaldehyde. Focus was set on the synthesis route and the controlled introduction of structural defects by mechanical methods (ball milling). The influence of these defects on the crystal structure are investigated in detail using X-ray diffraction. The investigations of the thermodynamic properties and the results obtained by using 57Fe-Mössbauer spectroscopy provide a deeper understanding of the optimized catalysts with controlled subsurface defect concentration. The optimal temperature range for ODH catalysis, in terms of very high selectivity and conversion (> 80%), is around 280 °C, and meets application requirements. Application of small angle X-ray scattering for the characterization and quality control of lipid mRNA delivery systems
mRNA basierte Arzneimittel repräsentieren eine neue Generation therapeutischer Ansätze mit vielseitigem Anwendungsgebiet. Die COVID-19 Pandemie im Jahr 2020 zeigte, dass ein großer Vorteil dieser Plattform ihre flexible Anpassungsfähigkeit an verschiedene Krankheiten ist. Neben der mRNA, die als Wirkstoff für die gewünschten Proteine codiert, stellt das Trägersystem einen wichtigen Bestandteil der Formulierung dar. Lipidbasierte Trägersysteme, wie zum Beispiel Lipid Nanopartikel, haben sich dabei als Goldstandard etabliert. Anders als bei klassischen Arzneiformen, bei denen Wirk- und Hilfsstoffe klar unterschieden werden können, fällt diese Differenzierung bei mRNA-basierten Arzneimitteln schwerer, da deren komplexe Formulierung nicht nur die Nukleinsäuren schützt und zur Zelle befördert, sondern auch die Wirksamkeit beeinflusst. Ein tiefgehendes Verständnis der strukturellen Organisation der Systeme ist daher sowohl für die Formulierungsentwicklung als auch für die Qualitätsbewertung von zentraler Bedeutung.
In den vergangenen Jahren wurde erheblicher Aufwand in die Entwicklung neuer Lipide und Herstellungsmethoden investiert, um eine Produktion verbesserter Lipidvehikel in skalierbaren Prozessen zu ermöglichen. Da bislang nur wenige rationale Ansätze zur Formulierungsentwicklung existieren, gestaltet sich das Screening neuer Lipide hinsichtlich ihrer Aktivität als zeit- und ressourcenintensiv. Dabei besteht die Gefahr, dass vielversprechende Kandidaten in frühen Entwicklungsphasen ausgeschlossen werden. Struktur-Wirkungs-Beziehungen sowie die Identifikation struktureller Merkmale mit prognostischer Aussagekraft zur Wirksamkeit können hier einen entscheidenden Beitrag zur Effizienzsteigerung beitragen. Mit der Zulassung der ersten mRNA-basierten Arzneimittel rücken auch regulatorische Anforderungen an Qualität, Wirksamkeit und Unbedenklichkeit zunehmend in den Fokus. Erste Leitlinien von Zulassungsbehörden definieren mittlerweile zentrale Qualitätsmerkmale und schaffen damit den Rahmen für die Bewertung entsprechender Formulierungen.
In dieser Arbeit wurde vor allem die Kleinwinkelröntgenstreuung- ergänzt durch weitere analytische Methoden- genutzt, um lipidbasierte mRNA-Trägersysteme im Detail zu charakterisieren und deren strukturelle Eigenschaften mit der Potenz in in vitro und in vivo Modellen zu korrelieren. Strukturaufklärung durch Kleinwinkelröntgenstreuung liefert dabei wertvolle Einblicke in die innere Struktur und allgemeine Morphologie der Nanopartikel. Dies ermöglicht die Detektion qualitätsrelevanter Merkmale sowie wirksamkeitsvoraussagender Struktureinheiten. Zusammengefasst ermöglichen die Ergebnisse dieser Arbeit, ein tieferes Verständnis von Struktur-Wirkungs-Beziehungen bei lipidbasierten mRNA-Trägersystemen, die in Zukunft sowohl für die Entwicklung verbesserter Plattformen als auch für die Bewertung der Qualität dieser Systeme herangezogen werden können.mRNA-based drugs represent a novel class of therapeutics applicable in various treatments. As the COVID-19 pandemic in 2020 demonstrated, this platform enables the rapid development of drugs against emerging infectious diseases due to its flexible adaptability. Besides the mRNA, which encodes the desired proteins, the carrier system that delivers the mRNA to target cells is a crucial component of the formulation. Lipid-based carrier systems, such as lipid nanoparticles (LNPs), have established themselves as the gold standard for delivery. Unlike traditional drug formulations, where active pharmaceutical ingredients and excipients can be clearly distinguished, this differentiation is more challenging for mRNA-based drugs. The complex lipid composition not only protects and delivers the nucleic acids to cells but also influences efficacy. A deep understanding of the intermolecular interactions within these systems and their structural organization is therefore of great interest not only for formulation development but also for addressing quality issues.
In recent years, significant efforts have been made to develop new carrier systems, including novel lipids and manufacturing methods, to enable the production of improved lipid vehicles through scalable processes. Since only a few paradigms for rational lipid synthesis and formulation development exist, screening new lipids for their activity using in vitro and in vivo models remains a time-consuming process. Additionally, there is a risk that highly potent candidates may be excluded from further development at early stages. Here, structure-activity relationships and the identification of structural fingerprints that can predict high efficacy can help conserve resources and facilitate more efficient formulation development.
With the approval of the first mRNA-based drugs, the quality and safety requirements for these drugs must also be addressed. During the rapid approval process of the pandemic, not all quality standards were established; however, in the years that followed, initial guidelines from regulatory authorities emerged, defining the key quality attributes and setting the framework for the quality control of such formulations.
In this work, small angle X-ray scattering (SAXS) was used in conjunction with other characterization methods to elucidate the properties of lipid-based mRNA carrier systems and correlate their structural properties with potency in in vitro and in vivo models. Structural elucidation through SAXS can provide valuable information about the internal structure of nanoparticles and their overall morphology. This enables the detection of quality-relevant features and structural units that are indicative of high potency. In summary, the results of this work provide a deeper understanding of structure-activity relationships in lipid-based mRNA carrier systems, which can be used for both the development of improved platforms and the quality assessment of these systems in the future.XVI, 177 Seiten ; Illutrationen, Diagramm
The historical climate trend resulted in changed vertical transport patterns in climate model simulations
Convective transport leads to a rapid vertical redistribution of tracers. This has a major influence on the composition of the upper troposphere (UT), a highly climate-sensitive region. It is not yet clear how the convective transport is affected by climate change. In this study, we applied a new tool, the so-called convective exchange matrix, in historical simulations with the chemistry–climate model EMAC (ECHAM/MESSy Atmospheric Chemistry) to investigate the trends in convective transport. The simulated deep convection is penetrating higher but occurs less frequently from 2011 to 2020 than from 1980 to 1989. The increase in the vertical extent of convection is highly correlated with a rise in the tropopause height. The upward transport of air mass increased on average to height levels of 130 hPa and above, but convection transported material less efficiently to the upper troposphere in general from 2011 to 2020 in comparison to the 1980s. These findings give rise to new opportunities to investigate long-term simulations performed by EMAC with regard to the effects of convective transport. Furthermore, they might provide a first insight into the trends in atmospheric convective transport due to changing atmospheric conditions and might serve as an estimate for the convective feedback to climate change
Physical activity decreases somatic symptom distress in the affect and symptom paradigm
Objective:
According to recent empirical taxonomies (eg, the Hierarchical Taxonomy of Psychopathology), somatic symptom distress represents a transdiagnostically relevant dimension of psychopathology. To better understand the mechanisms, somatic symptoms can be experimentally provoked by inducing negative affect (in the Affect and Symptom Paradigm, ASP, formerly known as the Affective Picture Paradigm). Potential moderators of this relation include cardiorespiratory activation and body-focused attention.
Methods:
In this preregistered, cross-sectional study (https://osf.io/sc57z/), we tested whether cardiorespiratory activation and body-focused attention modulate somatic symptoms in the ASP. Participants (N = 144) completed 3 ASPs, interlaced with cardiorespiratory activation (cycling) and rest. Participants were randomized to a body-attention or distraction condition. We measured heart rate, respiratory rate, and end-tidal CO2 during these tasks. The data were analyzed using AN(C)OVAs, t tests, and multilevel models.
Results:
Exercise significantly increased, and rest significantly decreased immediatesomatic symptoms (F (81.86, 262.68) = 53,80, p < .001, η2part = 0.28). High levels of somatic symptoms were significantly related to a sustained greater decrease in somatic symptoms in the ASP after cycling (vs. rest, F(1,136) = 8.061, p = .005, η2part = 0.056). No significant effect of the attention manipulation was observed (Fatt (1, 141) = 0.52, p = .47, η2part = 0.004).
Conclusions:
Rest and exercise temporarily modulate somatic symptom reports, and people with higher baseline somatic symptoms show the strongest beneficial effect of physical activity on symptom reports in the ASP. The findings are compatible with recent models of predictive processing and active inference
Elucidating substituent effects in magnetic properties of redox active cobalt complexes and testing them as potential catalysts for HER
We describe the synthesis of three cobalt complexes, C1–C3, featuring redox-active catechol ligands and a tetradentate salen ligand (Lsal). Structural characterization using single-crystal X-ray diffraction at 120 K, along with UV-vis, infrared spectroscopy, and SQUID magnetometry, provided detailed insights into their electronic and geometric properties. Magnetic measurements revealed that the two tetrahalogenated catechol complexes (C1 and C2) remain diamagnetic up to 400 K, whereas the complex with 3,5-ditert butyl catechol (C3) shows radical characteristics and stays in semiquinone form strongly coupled with a Co(III) centre. This radical behaviour was further confirmed by EPR spectroscopy in MeCN solution at room temperature. Cyclic voltammetry studies demonstrated the influence of catechol substituents on the electronic properties of these complexes, as reflected in shifts in oxidation potential. Preliminary investigations into their electrocatalytic activity for hydrogen production indicate that none of the complexes function effectively as catalysts under the tested conditions
Vergleich der funktionellen und biochemischen Auswirkungen von inhalierten Nano- und Mikropartikeln im Mausmodell mit Schwerpunkt auf die räumliche Verteilung und Unterschiede in der Toxizität je nach Partikelgröße
Die schädlichen Auswirkungen von Feinstaub auf die Gesundheit sind bereits lange
bekannt und seit den 80-er Jahren werden in der EU kontinuierlich Richtlinien für eine
Verbesserung der Luftqualität erarbeitet und überwacht. Dies geschah auf der Basis
von epidemiologischen Studien, die zunächst die Einflüsse von PM10 untersuchten,
dann jedoch erkannt wurde, dass von kleineren Partikel PM2.5 noch schädlichere
Wirkungen ausgehen könnten. Somit werden aktuell Messungen der
Feinstaubbelastung und große epidemiologische Studien in Bezug zu PM2.5
durchgeführt. Durch einige präklinische Studien und wenige epidemiologische
Erkenntnisse erhärtet sich jedoch der Verdacht, dass die schädlichsten systemischen
Auswirkungen durch die kleinsten Partikel, PM0.1 ausgelöst werden, die durch die Luft-
Blut-Schranke in den Kreislauf gelangen und so auch weitere Organe erreichen
können. In dieser Arbeit werden Mikropartikel der Größen 2.16μm und 4.13μm mit
Nanopartikeln der Größe 0.25μm im Hinblick auf ihre räumliche Verteilung nach der
Inhalation im Mausmodell untersucht und molekulare Funktions- und
Inflammationsmarker betrachtet, die Aufschluss auf die schädlichen Auswirkungen
der unterschiedlich großen Partikel liefern können. Die Ergebnisse zeigen, dass
Mikropartikel vorwiegen in der Lunge akkumulieren und dort zu einer lokalen
Aktivierung des Immunsystems mit Entzündungsreaktion führen, die sich jedoch auch
systemisch ausbreiten kann. Nanopartikel dagegen verweilen kürzer in der Lunge und
zeigen dort eine weitaus geringere Immunaktivierung und Inflammation. Jedoch
überwinden sie die Luft-Blut-Schranke und gelangen über den Kreislauf zu weiteren
Organen, wie beispielsweise Herz und sogar in das Gehirn, wo sie zu Inflammation
und Funktionseinschränkungen führen können. Somit verdeutlicht diese Arbeit, dass
bei der Betrachtung von gesundheitlichen Folgen der Feinstaubbelastung eine weitere
Differenzierung zwischen PM2.5 und PM0.1 erfolgen sollte. Sie zeigt die Notwendigkeit
auf, eine standardisierte, flächendeckende Messung von PM0.1 zu etablieren und so
größere epidemiologische Studien zu ermöglichen, die weiteren Aufschluss über den

Temperature-driven transition from Meron-type spin textures to stripe domains and revealing the nanosecond dynamics of antiferromagnetic quasiparticles through sublattice-resolved x-ray microscopy
Magnetic topologically non-trivial quasi-particles, such as skyrmions, hopfions, merons, and bimerons that are stabilized by the Dzyaloshinskii–Moriya interaction, have garnered significant interest in recent years. Here, we demonstrate a temperature-driven transition from meron-type textures to out-of-plane stripe domains by continuously tuning the effective anisotropy across the spin reorientation transition. Combined magnetic force microscopy and SQUID magnetometry reveal the temperature-induced evolution of spin structure and anisotropy. Our results establish temperature as a key control parameter for reconfigurable topological states in SyAFM multilayers
Posttranslationale Regulation der Transportaktivität humaner kationischer Aminosäuretransporter (human cationic aminoacid transporter: hCATs)
Cells depend on transporters for communication, interaction, and the selective uptake of nutrients and other molecules. Despite their importance, the intricate regulatory mecha-nisms that govern transporter activity and surface expression are not fully understood for many of these vital membrane proteins. This work investigated the post-translational regulation of human cationic amino acid transporters (hCATs) by protein kinase C (PKC) involving Rho-GTPases (Cdc42, Rac1, and RhoA) and other potentially involved proteins.
The central finding of the work is that both PKC and Rho-GTPases, especially Cdc42, play a crucial role in modulating the transport activity of all four hCAT isoforms. Activation of PKC leads to a significant reduction in arginine uptake by the hCATs. This effect was demonstrated in Xenopus laevis oocytes as a model system and various human cell lines. Interestingly, the reduced arginine uptake is caused by the altered membrane localization of the transporters (induced endocytosis).
An important aspect of this work is the investigation of the interplay of Rho-GTPases in regulating hCAT transporters showing the involvement of these GTPases in adapting transporter activity to meet cellular arginine demand. The results suggest that the simul-taneous activation of PKC and Rho-GTPases could lead to complex regulation of hCAT transporters, involving a reduced localization of the transporters in the plasma mem-brane.
The importance of membrane localization for the function of hCAT transporters was par-ticularly emphasized in this work. Understanding the mechanisms that regulate mem-brane proteins, especially transporter proteins, is of central importance, as they represent both targets and sensors for the cell's environment.
In summary, this research makes an important contribution to understanding the com-plex regulation of amino acid transporters, especially hCATs. The results show that PKC and Rho-GTPases play a crucial role in modulating transport activity and that membrane localization of the transporters is an important factor for their function. The interaction between PKC and Rho-GTPases could represent a finely tuned system that controls amino acid uptake as needed.In dieser Arbeit wurde die posttranslationale Regulation der kationischen Aminosäuret-ransporter (hCATs) durch Proteinkinase C (PKC) unter Beteiligung der Rho-GTPasen (Cdc42, Rac1 und RhoA) und anderen potenziellen beteiligten Proteinen untersucht.
Zentrale Erkenntnis der Arbeit ist, dass sowohl PKC als auch Rho-GTPasen, insbesondere Cdc42, eine entscheidende Rolle bei der Modulation der Transportaktivität aller vier hCAT-Isoformen spielen. Die Aktivierung von PKC führt zu einer signifikanten Reduktion der Argininaufnahme durch die hCATs. Dieser Effekt konnte sowohl im Modellsystem der Xenopus-Oocyten als auch in verschiedenen humanen Zelllinien nachgewiesen werden. Interessanterweise wird die verminderte Argininaufnahme durch die veränderte Memb-ranlokalisation der Transporter (wahrscheinlich induzierte Endozytose) verursacht.
Ein weiterer wichtiger Aspekt der Arbeit ist die Untersuchung des Zusammenspiels von Rho-GTPasen bei der Regulation der hCAT-Transporter. Die Aktivierung von Rho-GTPasen z.B. durch EGF, deutet auf eine Beteiligung dieser GTPasen bei der Regulation der Trans-portaktivität hin. Die Ergebnisse legen nahe, dass die gleichzeitige Aktivierung von PKC und Rho-GTPasen zu einer komplexen Regulation der hCAT-Transporter führt, die sich in einer reduzierten Oberflächenexpression der Transporter äußert.
Die Bedeutung der Membranlokalisation für die Funktion der hCAT-Transporter wurde in dieser Arbeit besonders hervorgehoben. Es ist von zentraler Bedeutung, die Mechanismen zu verstehen, die die Membranproteine, insbesondere die Transporterproteine, regulie-ren, da sie sowohl Angriffspunkte als auch Sensoren für die Umgebung der Zelle darstel-len.
Zusammenfassend lässt sich sagen, dass diese Forschungsarbeit einen wichtigen Beitrag zum Verständnis der komplexen Regulation von Aminosäuretransportern, insbesondere der hCATs, leistet. Die Ergebnisse zeigen, dass PKC und Rho-GTPasen eine entscheidende Rolle bei der Modulation der Transportaktivität spielen und dass die Membranlokalisati-on der Transporter ein wichtiger Faktor für ihre Funktion ist. Die Interaktion zwischen PKC und Rho-GTPasen könnte ein fein abgestimmtes System darstellen, das die Aminosäu-reaufnahme bedarfsgerecht steuert.XIX, 162 Seiten ; Illustrationen, Diagramm
