Isomerization of poly(ethylene glycol) : a strategy for the evasion of anti-PEG antibody recognition
PEGylation, the conjugation of poly(ethylene glycol) (PEG) to nanocarriers or protein-based active pharmaceutical ingredients (APIs), is a key strategy in nanomedicine to extend the circulation time of therapeutics in the bloodstream based on the stealth effect of PEG. However, the growing prevalence of anti-PEG antibodies in the population can lead to pronounced immune responses upon drug administration and accelerated blood clearance of PEGylated drugs, resulting in the loss of the stealth effect. We introduce the randomized PEG (rPEG) technology designed to strongly reduce the antigenicity of PEG while preserving its core benefits. This conceptually novel approach is based on an introduction of hydrophilic side chains along the PEG backbone. The synthesis is performed via anionic ring-opening copolymerization of ethylene oxide (EO) and glycidyl methyl ether (GME), resulting in constitutional isomers of PEG. By optimization of the reaction conditions, an ideally random distribution of the side chains in the polymer backbone could be achieved. Since previous studies show a relation between polymer chain regularity and immune system response, our approach specifically aims at introducing an irregular comonomer sequence via copolymerization, while translating the hydrophilicity and low toxicity of PEG to rPEG. Biocompatibility was evaluated using peripheral blood mononuclear cells (PBMC). Increasing the GME content in the copolymers did not decrease cell viability. Furthermore, all rPEG samples did not show complement activation in vitro at all tested concentrations. Enzyme-linked immunosorbent assays (ELISA) utilizing backbone- and end group-selective anti-PEG antibodies showed drastically reduced recognition and antibody binding for the constitutional isomers of PEG

Electrosynthesis of carboxylic acid derivatives, sulfonic acid derivatives, and electrochemical allylic oxidation reaction
Im Rahmen dieser Dissertation wurde die elektrochemische Synthese von Cabonsäurederivaten, Sulfonsäurederivaten, sowie die elektrochemische allylische Oxidation erforscht. Hierfür wurden Forschungsarbeiten in vielfältigen Bereichen der Elektrosynthese durchgeführt, z. B. elektrochemische Dehydratisierungsreaktionen und elektrochemische Multikomponentenreaktionen mit Schwefeldioxid. Das Zusammenspiel von Elektrosynthese mit anderen aufstrebenden Technologien wie Flow Chemie und Laborautomatisierung wurde untersucht und weiterentwickelt. Carbonsäureanhydride wurden synthetisiert durch elektrochemische Dehydratisierung der entsprechenden Carbonsäuren. Sulfonsäureamide und Sulfonate wurden durch elektrochemische Multikomponentenreaktion von Arenen mit Aminen bzw. Alkoholen und SO2 synthetisiert, sowie auch durch elektrochemische Dehydratisierung von Sulfonsäuren zu den entsprechenden Sulfonsäureanhydriden, welche nachfolgend mit Alkoholen oder Aminen umgesetzt wurden. Der Fortschritt im Gebiet der elektrochemischen Dehydratisierungsreaktionen wurde in einem Review-Artikel zusammenfassend erörtert. Um eine open-source Plattform für automatisierte Elektrosynthese zu entwerfen wurde das LABS-System (Laboratory Automation and Batch Scheduling) hervorgebracht, welches unter Verwendung einer Durchflusszelle mithilfe einer elektrochemischen Phenol-Kreuzkupplungsreaktion sowie der elektrochemischen Synthese von Sulfonaten als Modellreaktionen entwickelt wurde. Abschließend wurde ein vereinfachter Ansatz zur elektrochemischen allylischen Oxidation entwickelt, was eine wichtige Methode zur Funktionalisierung von Naturstoffen zu Hochwertprodukten darstellt.This dissertation explored the electrochemical synthesis of carboxylic acid derivatives, sulfonic acid derivatives, and electrochemical allylic oxidation reaction. To achieve this, research in various areas of electrosynthesis was carried out, such as electrochemical dehydration reactions and electrochemical multicomponent reactions using sulfur dioxide. The cooperation of electrosynthesis with other evolving technologies such as flow chemistry and laboratory automation was investigated and further developed. Carboxylic anhydrides were synthesized by electrochemical dehydration of their corresponding carboxylic acids. Sulfonamides and sulfonates were synthesized via electrochemical multicomponent reactions of arenes with amines or alcohols and SO2, and also by electrochemical dehydration of sulfonic acids to their sulfonic anhydrides via subsequent treatment of the sulfonic anhydrides with an excess of amines or alcohols. The recent progress in electrochemical dehydration reactions was summarized in a review article. To establish an open-source platform for automated electrosynthesis, the LABS system (Laboratory Automation and Batch Scheduling) was developed, which was benchmarked by using a flow electrolysis cell with electrochemical phenol cross coupling and electrochemical multicomponent synthesis of sulfonates as test reactions. Evolution of plant specialized metabolites : beyond ecological drivers
Plants produce a highly diverse array of specialized metabolites. Traditionally, the evolution of these metabolites has been studied primarily through the lens of plants’ ecological interactions with herbivores, pathogens, and pollinators, as many of them exhibit defense and/or attraction functions. However, increasing evidence suggests that many specialized metabolites, along with their precursors, also act as cellular signals that regulate cell growth and differentiation. We propose that these intrinsic functions are at least equally important factors in shaping the evolution of plant chemical defenses. We further discuss how future research that combines modern single-cell techniques and evolutionary genomics will provide novel insights into the evolutionary process of specialized metabolism diversification
Vom Konzept zur Evaluation: Herausforderungen und Erkenntnisse bei der patient:innenorientierten Entwicklung und Rekrutierung für eine psychoonkologische Online-Selbsthilfe in einer randomisiert kontrollierten Studie
Fast die Hälfte aller Menschen erkrankt im Laufe ihres Lebens an Krebs und die Erkrankung geht häufig mit vielen psychosozialen Belastungen für die Betroffenen einher. Dennoch erhalten nicht alle Betroffenen die notwendige psychische Unterstützung. Eine Möglichkeit, diese Versorgungslücke zu schließen, besteht in E-Mental Health Angeboten, welche die traditionelle face-to-face Versorgung ergänzen können. Jedoch stößt die Implementierung dieser Interventionen oft auf Barrieren, die dafür verantwortlich sein können, dass die Intervention entweder gar nicht erst bei den Patient:innen ankommt oder nicht in dem Ausmaß wie vorgesehen genutzt wird. Diese Dissertation fokussiert mögliche Ansatzpunkte, diese Herausforderung einer geringen Inanspruchnahme und Adhärenz zu überwinden und konzentriert sich dabei auf zwei spezielle Barrieren: Zum einen besteht aus Patient:innensicht häufig eine unzureichende Passung zwischen den eigenen Bedürfnissen und Wünschen hinsichtlich einer psychoonkologischen Online-Intervention und den tatsächlichen Themen und Inhalten, die in der Online-Intervention behandelt werden. Ein möglicher Lösungsansatz, diese Barriere zu überwinden, liegt in einem patient:innenorientierten Entwicklungsprozess, welcher Krebspatient:innen an verschiedenen Stellen in die Entwicklung einbezieht. Zudem wird die Herausforderung thematisiert, eine ausreichende Anzahl an Patient:innen für Studien zur Evaluation psychoonkologischer Online-Interventionen zu gewinnen und somit sicherzustellen, dass Patient:innen auch tatsächlich mit der Intervention erreicht werden. Dieser Barriere wird mit einer diversifizierten Rekrutierungsstrategie, bestehend aus sowohl traditionellen als auch Online-Rekrutierungsmethoden, entgegnet.
Beide Barrieren werden in dieser Dissertation ausführlich beleuchtet und Einblicke in die angewendeten potentiellen Lösungsansätze sowie daraus resultierenden Erkenntnisse und Herausforderungen aus methodischer und praxisorientierter Perspektive gegeben. Die Ergebnisse zeigen vor allem die Bedeutsamkeit eines angemessenen Erwartungsmanagements auf, welches bei der gesamten Kommunikation während der Entwicklungs- und Evaluationsphase berücksichtigt werden sollte. Bei der Entwicklung der Intervention und der Planung der Studie ist es zentral, Patient:innen mit einzubeziehen. Hierbei sollte individuell entschieden werden, welches Ausmaß der Partizipation unter den gegebenen Rahmenbedingungen (z.B. der entsprechenden Studienpopulation) am geeignetsten ist. Bei der Anwendung einer diversifizierten Rekrutierungsstrategie, welche auch Social Media einschließt, muss vor allem berücksichtigt werden, dass die Gefahr besteht, dass sich das Problem einer nicht repräsentativen Stichprobe verstärken kann. Es empfiehlt sich daher, bereits bei der Entwicklung sowie der Planung der Rekrutierungsstrategie schwer erreichbare Gruppen, z.B. Männer und ältere Personen, einzubeziehen. Zudem gibt es beim Einschluss von Social Media als Rekrutierungskanal wichtige in der Praxis zu beachtende Punkte, um keine methodischen oder ethischen Probleme zu erzeugen. Werden diese bei der Planung der diversifizierten Rekrutierungsstrategie berücksichtigt, kann Social Media einen wichtigen ergänzenden Rekrutierungskanal für klinische Studien darstellen.
Alles in allem zeigt sich, dass es von großem Nutzen ist, während des gesamten Entwicklungs- und Evaluationsprozesses einer Online-Intervention die Bedürfnisse der Zielgruppe zu berücksichtigen, ihre Erreichbarkeit zu planen und ihre Einbeziehung in den Forschungs- und Entwicklungsprozess zu ermöglichen. Diese Dissertation beleuchtet wichtige Aspekte für Forschende, die zukünftige Online-Interventionsstudien im psychoonkologischen Bereich konzipieren und bietet wertvolle Hinweise für deren Umsetzung.II, 53 Seiten ; Illustrationen, Diagramm
Experimental induction of state rumination in youth soccer players on the pitch : how can we evaluate an effect of rumination on soccer-specific performance?
Based on the goal progress theory, we examined whether cueing an unresolved goal triggers state rumination in athletes and consequently leads to performance deterioration. However, experimental approaches are usually conducted in the laboratory and only rarely in the field. Therefore, the aim of this proof-of-concept study was to verify whether the findings obtained in previous experimental studies could be transferred and confirmed in the field. For this purpose, the study was applied during regular training sessions. Fifty soccer players (f = 17, m = 33, age range: 14–18 years; Mage = 16.38) were divided into three different conditions—one experimental condition in which a goal-related rumination was induced and two control conditions in which a comparable goal-related induction or a neutral task was performed. Before and after the inductions during athletic practice, soccer players performed a sport-specific test to examine potential changes in athletes' performance as a function of condition and answered questions related to their state rumination. Different mixed ANOVAs were performed to examine (1) whether we could significantly elicit rumination in soccer players and (2) whether the application of the experimental procedure had an impact on performance in the subsequent sport-specific test. Two main findings were identified: First, the experimental procedure generally transfers well to the field. Second, however, the expected effects of state rumination on performance in the sport-specific test were not obtained. Reasons for the lack of clear effects and approaches for future research are discussed
Deep Learning Reaction Framework (DLRN) for kinetic modeling of time-resolved data
Model-based analysis is essential for extracting information about chemical reaction kinetics in full detail from time-resolved data sets. This approach combines experimental hypotheses with mathematical and physical models, enabling a concise description of complex system dynamics and the extraction of kinetic parameters like kinetic pathways, time constants, and species amplitudes. However, building the final kinetic model requires several intermediate steps, including testing various assumptions and models across multiple experiments. In complex cases, some intermediate states may be unknown and are often simplified. This approach requires expertise in modeling and data comprehension, as poor decisions at any stage during data analysis can lead to an incorrect kinetic model, resulting in inaccurate results. Here, we introduce DLRN, a new deep learning-based framework, designed to rapidly provide a kinetic reaction network, time constants, and amplitude for the system, with comparable performance and, in part, even better than a classical fitting analysis. We demonstrate DLRN’s utility in analyzing multiple timescales datasets with complex kinetics, different 2D systems such as time-resolved spectra and agarose gel electrophoresis data, experimental datasets as nitrogen vacancy and strand displacement circuit (using photoluminescence and transient absorption techniques), even in scenarios where the initial state is a hidden, non-emitting dark state
