    There is a continuously increasing pressure associated with the appearance of Salmonella enterica Serovar typhimurium (S. typhimurium) and Shigella sonnei (S. sonnei) that have developed pathogenic multiple antibiotic resistance and the cost of cure and control of these enterobacteriaceae infections increases annually. The current report for first time demonstrated the distinguished antimicrobial action of camel lactoferrin (cLf) obtained from the milk of different clans of camel in Saudi Arabia against S. typhimurium and S. sonnei. These cLf subtypes showed comparable antimicrobial potential when tested against the two bacterial strains but were superior to either bovine (bLf) or human lactoferrin (hLf). The synergism between lactoferrins and antibiotics concerning their antibacterial efficacies against the two bacterial strains was evident. Exploring mechanisms by which camel lactoferrin can kill S. typhimurium and S. sonnei revealed that cLf affects bacterial protein profile. Besides, it interacts with bacterial lipopolysaccharides (LPS) and numerous membrane proteins of S. typhimurium and S. sonnei, with each bacterial strain possessing distinctive binding membrane proteins for lactoferrin. Furthermore, as evidenced by electron microscopy analysis, cLf induces extracellular and intracellular morphological changes in the test bacterial strains when used alone or in combination treatment with antibiotics. Lactoferrin and antibiotics combination strongly disrupts the integrity of the bacterial cells and their membranes. Therefore, cLf can kill S. typhimurium and S. sonnei by four different mechanisms, such as iron chelation, affecting some bacterial proteins, binding to bacterial LPS and membrane proteins, and impairing the integrity of the bacterial cells and their membranes

    Liquid–liquid phase separation (LLPS) is responsible for the formation of so-called membrane-less organelles (MLOs) that are essential for the spatio-temporal organization of the cell. Intrinsically disordered proteins (IDPs) or regions (IDRs), either alone or in conjunction with nucleic acids, are involved in the formation of these intracellular condensates. Notably, viruses exploit LLPS at their own benefit to form viral replication compartments. Beyond giving rise to biomolecular condensates, viral proteins are also known to partition into cellular MLOs, thus raising the question as to whether these cellular phase-separating proteins are drivers of LLPS or behave as clients/regulators. Here, we focus on a set of eukaryotic proteins that are either sequestered in viral factories or colocalize with viral proteins within cellular MLOs, with the primary goal of gathering organized, predicted, and experimental information on these proteins, which constitute promising targets for innovative antiviral strategies. Using various computational approaches, we thoroughly investigated their disorder content and inherent propensity to undergo LLPS, along with their biological functions and interactivity networks. Results show that these proteins are on average, though to varying degrees, enriched in disorder, with their propensity for phase separation being correlated, as expected, with their disorder content. A trend, which awaits further validation, tends to emerge whereby the most disordered proteins serve as drivers, while more ordered cellular proteins tend instead to be clients of viral factories. In light of their high disorder content and their annotated LLPS behavior, most proteins in our data set are drivers or co-drivers of molecular condensation, foreshadowing a key role of these cellular proteins in the scaffolding of viral infection-related MLOs

    This study describes what the graduates of a teacher educator program continue to apply in their classrooms and schools up to fifteen years after they began teaching. The data for this study included focus group notes, interview notes, and questionnaire responses. Teacher educators analyzed the data in order to explore their inquiry about how the educator preparation program had impacted and continued to impact graduates. The voices of the graduates highlighted ways they continued to apply foundational ideas from the program in their classrooms

    Practitioner Inquiry for Turbulent Times: Learning to Take an Inquiry Stance Toward Teaching Difficult Topics Through a Teacher Inquiry Community

    Amid turbulent times and politically polarized communities, many teachers require support if they are to teach or engage with difficult topics in their curricula or professional practices, yet few teachers actually receive any formalized support for addressing such topics. This article responds by describing the work of an inquiry community of inservice educators that was designed to assist teachers in learning to address difficult topics by integrating practitioner inquiry and student inquiry with asset-based and trauma-informed lenses. The article outlines the community’s conceptual foundations then describes how a team of university-based teacher educators facilitated the community’s work. A participating teacher’s reflection illustrates how the support of the community transformed her teaching of one potentially difficult topic—the American Civil War—as she shifted her pedagogy from being a provider of knowledge to facilitating students’ inquiries. The article highlights implications for future scholarship about professional learning, student inquiry, and teacher retention
