    The phytosiderophore analogue proline-2′-deoxymugineic acid is more efficient than conventional chelators for improving iron nutrition in maize

    Iron (Fe) is an essential nutrient but has poor bioavailability because of its low solubility under conditions of high pH, such as calcareous soils. Previously, we synthesized an analogue of the natural Fe(III) chelator 2′-deoxymugineic acid secreted by graminaceous plants for efficient Fe uptake, designated as proline-2′-deoxymugineic acid (PDMA). Soil application of Fe(III)-PDMA ameliorated symptoms of Fe deficiency in rice. In the present study, we explored the potential of PDMA as Fe nutrition supplement in maize, a major graminaceous crop, and rice for comparison. In calcareous soil pots, Fe deficiency chlorosis of maize was efficiently recovered by a single application of Fe(III)-PDMA. In contrast, maize plants treated with conventional Fe(III)-chelator complexes, such as Fe(III)-ethylenediaminetetraacetic acid (EDTA) or Fe(III)-N,N′-di(2-hydroxybenzyl)ethylenediamine-N,N′-diacetic acid monohydrochloride (HBED), showed little or no recovery. Similarly, rice Fe chlorosis in calcareous soil pots was efficiently recovered by a single application of Fe(III)-PDMA, but not by other conventional Fe(III)-chelator complexes. Application of Fe(III)-PDMA was also effective in ameliorating Fe deficiency chlorosis in hydroponically grown maize seedlings; other Fe(III)-chelator complexes showed minimal or no efficacy. Addition of low concentrations of metal-free PDMA recovered Fe chlorosis of maize or rice in the presence of Fe(III)-EDTA, suggesting possible ligand exchange from EDTA to PDMA for subsequent Fe(III)-PDMA uptake by ZmYS1 and OsYSL15 transporters. Similar effects were observed with Fe(III)-HBED, which has a higher stability constant, but to a lesser extent than Fe(III)-EDTA, suggesting that ligand exchange from HBED to PDMA might be less prone to occur than for chelators with moderate stability constants, such as EDTA. Ferric-chelate reductase assays of maize roots showed substantial reduction of Fe(III)-PDMA, but this reduction activity was not increased under Fe-deficient condition. These results suggested that PDMA is an efficient reagent for improvement of Fe nutrition in graminaceous crops, including maize, because of suitability of Fe(III)-PDMA as a substrate for chelation-based Fe uptake systems.journal articl

    Blue light irradiation inhibits the M2 polarization of the cancer-associated macrophages in colon cancer

    Recent studies have shown that blue light-emitting diode (LED) light has anti-tumor effects, suggesting the possibility of using visible light in cancer therapy. However, the effects of blue light irradiation on cells in the tumor microenvironment, including tumor-associated macrophages (TAMs), are unknown. Here, THP-1 cells were cultured in the conditioned medium (CM) of HCT-116 cells to prepare TAMs. TAMs were divided into LED-irradiated and control groups. Then, the effects of blue LED irradiation on TAM activation were examined. Expression levels of M2 macrophage markers CD163 and CD206 expression were significantly decreased in LED-irradiated TAMs compared with the control group. While control TAM-CM could induce HCT-116 cell migration, these effects were not observed in cells cultured in TAM-CM with LED irradiation. Vascular endothelial growth factor (VEGF) secretion was significantly suppressed in LED-exposed TAMs. PD-L1 expression was upregulated in HCT-116 cells cultured with TAM-CM but attenuated in cells cultured with LED-irradiated TAM-CM. In an in vivo model, protein expression levels of F4/80 and CD163, which are TAM markers, were reduced in the LED-exposed group. These results indicate that blue LED light may have an inhibitory effect on TAMs, as well as anti-tumor effects on colon cancer cells.journal articl

    Association between motivation scale score and oral condition on receiving basic periodontal therapy

    The purpose of this study was to evaluate the usefulness of the motivation scale score (MSS) for Japanese adults and investigate motivation related to the oral condition on receiving basic periodontal therapy using MSS. The participants enrolled in this study were 221 patients with periodontal disease aged 20-64 years who visited a dental clinic in Shizuoka City, Japan for the first time. MSS was calculated using a 14-itemquestionnaire concerning the patients’ motivation assessment scale, and principal component analysis of MSS was performed. The associations among MSS, items of oral health behavior, periodontal conditions [Bleeding on probing (BOP) rate, Periodontal inflamed surface area (PISA)], and oral hygiene status [O’Leary’s Plaque control record (PCR)] were evaluated at the baseline and after basic periodontal treatment. MSS was shown to be useful in terms of reliability and validity and was classified into five factors. MSS and MSS-Factor 1 (oral hygiene practices) of participants with BOP of less than 10% were significantly higher than in of participants with BOP of more than 10% at the baseline. In addition, MSS and MSS-F1 were significantly higher among participants who received regular dental check-ups within a year and used interdental cleaning tools. MSS-F1 was significantly higher in non-smokers. By binominal logistic regression analysis, the use of interdental cleaning tools was significantly correlated with BOP of less than 10% at the baseline. All items related to oral conditions and oral health behavior improved after basic periodontal therapy, except for current smoking status. After basic periodontal therapy, MSS was significantly higher in participants with PCR of less than 20%, and MSS-F1 was significantly higher in participants who used interdental cleaning tools or were non-smokers. It was revealed that MSS used in this study was useful and associated with the periodontal condition and oral health behavior at the baseline. The values of MMS and MSS-F1 significantly increased after basic periodontal therapy. The use of interdental cleaning tools was associated with a good oral condition and higher MSS. Therefore, high motivation might lead to a good oral condition in patients with periodontal disease through the use of interdental cleaning tools by influencing patients’ compliance to improve their oral health behaviors.本研究の目的は,日本の成人に対するモチベーションスケールスコア(MSS)の有用性を評価すること,および歯周基本治療における口腔状態に関連するモチベーションを,MSS を用いて検討することである.  対象者は静岡市の歯科診療所に来院した20~64 歳の初診歯周病患者221 名である.MSS は患者のモチベーション評価尺度に関する 14 項目の質問紙を用いて算出し,MSS の主成分分析を行った.また,ベースライン時および歯周基本治療後の MSS と口腔保健行動項目,歯周状態(Bleeding on probing (BOP) 率,Periodontal inflamed surface area(PISA))および口腔衛生状態(O’Leary のPlaque control record (PCR))との関係を評価した.  MSS は信頼性と妥当性の点からその有用性が示され,5 つの要素に分類された.ベースライン時BOP10% 未満の者はBOP10% 以上の者と比較してMSS およびMSS-F1(口腔衛生行動)が有意に高かった.また,年1 回の歯科健診受診,歯間部清掃用具の使用の者はMSS およびMSS-F1 が有意に高く,非喫煙者でMSS-F1 が有意に高かった.二項ロジスティック回帰分析において,ベースライン時BOP10% 以上と統計学的に有意な関連を認めた因子は歯間部清掃用具の使用であった.一方,歯周基本治療後,喫煙状況を除くすべての項目で良好な改善が認められた.このうち,歯周基本治療後のPCR20% 未満の者はMSS が有意に高く,歯間部清掃用具の使用および非喫煙者においてMSS-F1 が有意に高かった.  本研究で用いたMSS は有用であり,ベースライン時の歯周組織状態および口腔保健行動と関連した.歯周基本治療介入後,MSS およびMSS-F1 の値は有意に上昇した.歯間部清掃用具の使用は良好な歯周組織状態と関連し,MSS の高値と関連した.それ故,高いモチベーションが,歯間部清掃用具の使用によって口腔保健行動の改善に対する患者のコンプライアンスに影響を与え,歯周病患者の良好な口腔状態を導いた可能性がある.doctoral thesi

    電気炉酸化スラグ細骨材と膨張材を使用した短繊維補強コンクリートの急速凍結融解抵抗性の検討

    鋼材の使用量を削減するための短繊維補強コンクリートの普及・高性能化を最終目的として,短繊維を混入することで著しく損なわれてしまう流動性を向上させるために電気炉酸化スラグ細骨材を使用した。さらに,収縮特性やひび割れ抑制に効果的な膨張材を用いた短繊維補強コンクリートのスケーリングに着目した急速凍結融解に関する実験的検討を行った。短繊維としては,鋼繊維とビニロン繊維の2種類とし,短繊維容積混入率を最大4Vol.%,膨張材量は最大30kg/m3とした。実験的検討の結果,単位水量200kg/m3の短繊維補強コンクリートは膨張材の有無にかかわらず高い急速凍結融解抵抗性を有することが明らかになった。journal articl

    Study on prediction of early adverse events by CapeOX therapy in patients with colorectal cancer

    CapeOX is a regimen used as postoperative adjuvant chemotherapy for the treatment of advanced recurrent colorectal cancer. If early adverse events occur, treatment may not progress as planned and further dose reduction may be necessary. In this study, we investigated whether pre-treatment medical records could be used to predict adverse events in order to prevent adverse events caused by CapeOX treatment. The 178 patients were classified into two groups (97 in the adverse event positive group and 81 in the adverse event-negative group) based on withdrawal or postponement of four or fewer courses. In univariate analysis, age, height, weight, body surface area (BSA), creatinine clearance, muscle mass, and lean body mass were associated with early adverse events (P < 0.05). The area under the receiver operating characteristic curve obtained by Stepwise logistic regression analysis using the Akaike information criterion method was 0.832. For nested k-fold cross validation, the accuracy rates of the support vector machine, random forest, and logistic regression algorithms were 0.71, 0.70, and 0.75, respectively. The results of the present study suggest that a logistic regression prediction model may be useful in predicting early adverse events caused by CapeOX therapy in patients with colorectal cancer.journal articl

    【Introduction】Familial Mediterranean fever(FMF) is an autoinflammatory disease that is characterized by repeated periodic fever and serositis. FMF is mainly inherited in an autosomal recessive manner. While the prognosis of FMF is good with oral colchicine which is recommended to take for avoiding amyloidosis, FMF is associated with many medical and social issues, such as stigma against genetic diseases and infertility due to oral colchicine. 【Case】From year X, a female teenager repeatedly developed fever of 38℃ that continued for 2-3 days and unilateral chest and back pain once a month. She visited a nearby clinic, however, her symptoms spontaneously disappeared. She was kept under observation without definite diagnosis. In May of X+4, she developed fever and right chest pain and referred to our hospital. By the time she presented to our hospital, symptoms had disappeared and there were no abnormalities in a peripheral blood examination or on whole-body CT or cardiac ultrasound. Based on her clinical course, we suspected an autoinflammatory disease ; therefore, we performed a genetic examination with informed consent from the patient and her guardian. The results obtained revealed that heterogenous pathogenic variant in the MEFV gene(M694I), and, thus, she was diagnosed with FMF. The oral administration of colchicine was initiated in November of the same year, which resolved fever and chest and back pain. In X+7, she started a job, which affected her daily life and the regularity with which she took oral colchicine. Therefore, chest and back pain relapsed. She expressed concerns about infertility and teratogenicity caused by oral colchicine and received a second opinion and genetic counseling, which improved compliance with oral colchicine. 【Discussion】Adolescent and young adult generation patients have strong phycological stress and anxiety about the future, such as educational plans, employment, marriage, and pregnancy, which need to be considered in their treatment. Further improvement in awareness of FMF and the establishment of a medical care support system through inter-professional work, including internal medicine, pediatrics, obstetrics and gynecology, clinical genetic staff and clinical psychologists, are needed.journal articl

    アベマシクリブ投与中に重篤な間質性肺疾患をきたしたが救命しえた1例

    Abemaciclib, Selective cyclin-dependent kinase 4 and 6 inhibitor has become available for use in hormone receptor-positive and HER2 - negative unresectable or recurrent breast cancer, significantly extending progression-free survival and improving prognosis. However, caution is necessary as in some cases it may cause drug-induced interstitial lung disease, occasionally leading to severe exacerbations. The case involves a 75-year-old female who was emergency transported to our hospital due to dyspnea occurring 11 months after initiating abemaciclib + letrozole therapy for hormone receptor-positive, HER2 - negative recurrent breast cancer. Upon arrival, she presented with typeⅠrespiratory failure with PCO2 : 24.5 mmHg, PO2 : 61.2 mmHg, and CT revealing ground-glass opacities in both lung fields. Diagnosed with drug-induced interstitial lung disease, she underwent endotracheal intubation followed by steroid pulse therapy. After three rounds of steroid pulses, symptoms gradually improved, allowing for discharge home with oxygen supplementation. Despite the development of severe interstitial lung disease in this case, prompt initiation of treatment facilitated successful life-saving intervention.journal articl

    Orbital apex syndrome secondary to acute invasive fungal rhinosinusitis diagnosed by transnasal endoscopic biopsy of the optic canal : A case report

    Orbital apex syndrome (OAS) is a complex condition characterized by visual loss, diplopia, and eye pain that occurs secondary to several pathological processes involving the orbital apex. We report a case of acute invasive fungal rhinosinusitis (AIFRS) associated with OAS. A 76-year-old man with left-sided visual loss, diplopia, palpebral ptosis, and headache was diagnosed with OAS secondary to Tolosa-Hunt syndrome and received systemic corticosteroid therapy from his neurologist. Owing to persistent symptoms, we opened the optic canal using a transnasal endoscopic approach for a surgical biopsy of the orbital apex lesions. Histopathological evaluation revealed numerous Aspergillus organisms in the biopsied granuloma. After surgical debridement, he received a 12-month course of voriconazole, and no recurrence of AIFRS occurred during 8-year follow-up. Patients with OAS may occasionally be prescribed corticosteroids because the clinical manifestations of AIFRS-induced OAS are similar to those observed in OAS secondary to Tolosa-Hunt syndrome, especially no nasal symptoms which is known to respond to corticosteroid therapy. Because both AIFRS induced OAS and OAS secondary to Tolosa-Hunt syndrome induce ophthalmoplegia, proptosis, eye pain, it is sometimes difficult to differentiate these two diseases in early stage. However, corticosteroid therapy causes exacerbation of fungal infection in patients with AIFRS-induced OAS resulting in delayed accurate diagnosis and poor prognosis. AIFRS is associated with a high mortality rate ; therefore, transnasal endoscopic biopsy of orbital apex lesions before corticosteroid administration is recommended in patients with OAS.journal articl

    Acute myeloid leukemia developed through myeloproliferative features during immunosuppressive therapy for juvenile idiopathic arthritis

    A 17-year-old male with thrombocytosis and exacerbation of arthralgia during intensified immunosuppressive therapy with tocilizumab, prednisolone, and methotrexate for juvenile idiopathic arthritis (JIA) was referred to our department. Bone marrow examination revealed myelodysplastic syndrome/myeloproliferative neoplasm, unclassifiable (MDS/MPN-U). Peripheral myeloblasts disappeared temporarily after discontinuation of tocilizumab but progressed to acute myeloid leukemia six months after the development of MDS/MPN-U. The patient sustained complete remission after unrelated bone marrow stem cell transplantation, followed by chemotherapy. The arthralgia also improved after chemotherapy. The possibility of developing malignancies during immunosuppressive therapy in patients with JIA should be considered.journal articl
