Symptome einer Depression bei Diagnosestellung einer Demenz oder eines Mild Cognitive Impairments

Dementielle Erkrankungen nehmen mit der alternden Allgemeinbevölkerung in Deutschland einen wichtigen Stellenwert ein. Prognosen gehen davon aus, dass sich die Zahl der Erkrankten bis 2050 verdoppeln wird.
Ziel dieser Studie ist es zu untersuchen, wie viele der Personen mit einer erstmalig diagnostizierten neurodegenerativen Erkrankung zu diesem Zeitpunkt auch die Kriterien einer bestehenden Depression erfüllen und ob sich hierbei Unterschiede zwischen den verschiedenen Entitäten zeigen. Folgende neurodegenerative Erkrankungen wurden berücksichtigt: Mild Cognitive Impairment (MCI), Demenz vom Alzheimertyp (AD), Parkinson Demenz (PDD), Lewy-Body-Demenz (LBD), Frontotemporale Demenz (FTD), vaskuläre Demenz (VaD) und solche mit einer gemischten oder unklaren Genese.
Material und Methoden
Die Datenerhebung erfolgte mittels einer retrospektiven Analyse der Patientenakten der Klinik für Psychiatrie und Psychotherapie der Universitätskliniken des Saarlandes.
Berücksichtigt wurden Patient*innen, die in dem Zeitraum vom 01.01.2018 bis zum 31.12.2019 in einer der zugehörigen Institutionen vorstellig wurden.
Ausgehend von der so gebildeten Grundgesamtheit wurde eine Selektion mit folgenden ICD-10-Schlüsseln als Einflusskriterien durchgeführt: F00.-, F01.-, F02.-, F03.-, F06.7, G20.-, G31.-, U51.-. Die so gebildete Studienpopulation umfasst N=330 Personen.
Die ursprünglich voneinander getrennten Gruppen wurden nach Aktenanalyse in die folgenden Kategorien eingeteilt: AD (1), MCI (2) und andere Demenzen (3) mit PDD, LBD, FTD, VaD und Demenzen gemischter oder unklarer Genese.
Mittels Chi-Quadrat-Test wurde überprüft, ob zwischen den so gebildeten Untergruppen ein Unterschied in der Verteilung (Depression/keine Depression) besteht.
Ergebnisse
Es zeigt sich eine statistisch signifikante Abhängigkeit zwischen dem Bestehen von Symptomen einer Depression und der jeweilig diagnostizierten neurodegenerativen Erkrankung.
Für die AD (1) erfüllen 43,7% die Kriterien einer bestehenden Depression, bei dem MCI (2) sind dies 61,2% und in der Gruppe der anderen Demenzen (3) 63,8% der Personen.
Für 263 Patient*innen wurde zu dem Diagnosezeitpunkt ein Minimental-Status-Test (MMST) erhoben. In der Gruppe mit unauffälligem MMST-Ergebnis erfüllen zu 57,4 % die Kriterien einer bestehenden Depression, 62,1% bei der leichten Demenz sowie 41% für die moderate und 20 % einer schweren. Der Zusammenhang zwischen der erreichten Punktzahl im MMST und dem Bestehen von Symptomen einer Depression ist statistisch signifikant.
Schlussfolgerung
Die Ergebnisse der Studie bestätigen den Zusammenhang zwischen dem Bestehen (von Symptomen) einer Depression und einer neurodegenerativen Erkrankung zum Zeitpunkt ihrer Erstdiagnose. Es können Unterschiede zwischen den Diagnosegruppen festgestellt werden. Weiterhin zeigen die Ergebnisse, dass bei der Diagnosestellung in einem frühen Stadium der neurodegenerativen Erkrankung (gemessen an der Punktzahl im MMST) das Bestehen von Symptomen einer Depression wahrscheinlicher ist als zu einem Zeitpunkt, zu dem die Erkrankung bereits weiter fortgeschritten ist.Abstract
Objective
Dementia is becoming increasingly important in Germany as the general population ages. Forecasts assume that the number of sufferers will double by 2050. The aim of this study is to investigate how many of the people diagnosed with a neurodegenerative disease for the first time also meet the criteria for existing depression at this time and whether there are dif-ferences between the various entities. The following neurodegenerative diseases were taken into account: Mild Cognitive Impairment (MCI), Alzheimer's dementia (AD), Parkinson's de-mentia (PDD), Lewy body dementia (LBD), frontotemporal dementia (FTD), vascular demen-tia (VaD) and those with a mixed or unclear genesis.
Material and Method
The data were collected by means of a retrospective analysis of the patient files of the Klinik für Psychiatrie und Psychotherapie des Universitätsklinikum des Saarlandes. Patients who presented themselves at one of the associated institutions between January 1, 2018 and December 31, 2019 were taken into account. Based on the population thus formed, a selec-tion was carried out using the following ICD-10 codes as influencing criteria: F00.-, F01.-, F02.-, F03.-, F06.7, G20.-, G31.-, U51.-. The study population thus formed comprises N=330 people. The groups that were originally separate from one another were divided into the fol-lowing categories after file analysis: AD (1), MCI (2) and other dementias (3) with PDD, LBD, FTD, VaD and dementias of mixed or unclear origin. The chi-square test was used to check whether there was a difference in the distribution (depression/no depression) between the subgroups thus formed.

There is a statistically significant relationship between the presence of symptoms of depres-sion and the neurodegenerative disease diagnosed. For AD (1), 43.7% meet the criteria for existing depression, for MCI (2) this is 61.2% and in the group of other dementias (3) 63.8% of people. A mini-mental status examination (MMSE) was administered to 263 patients at the time of diagnosis. In the group with an unremarkable MMSE result, 57.4% met the criteria for existing depression, 62.1% for mild dementia, 41% for moderate dementia and 20% for se-vere dementia. The relationship between the score achieved in the MMSE and the presence of symptoms of depression is statistically significant.
Conclusion
The results of the study confirm the link between the presence (of symptoms) of depression and a neurodegenerative disease at the time of its initial diagnosis. Differences can be identi-fied between the diagnosis groups. Muscle atrophy in polytraumatized patients - a longitudinal observational pilot study
In polytraumatized patients, muscle atrophy appears more pronounced than in
immobilized healthy study participants. However, rates and trajectories of the
acute muscle atrophy and associated parameters have not been reported. In a
prospective longitudinal pilot study with 10 patients (Injury Severity Score (ISS)
≥16), hand grip strength and inflammatory blood parameters were assessed.
Skeletal muscle thickness of the rectus femoris (RF), vastus lateralis (VL), and
tibialis anterior (TA), and subcutaneous tissue thickness over these muscles were
measured via ultrasound. Muscle oxygen saturation (SO2
), relative haemoglobin
content (rHb), blood flow (BF), and blood flow velocity (BFV) were captured
by laser-Doppler and white-light spectroscopy. Three women and seven men
were included (age 43.2 ± 22.5 years; height 176.4 ± 5.7 cm; body weight 83.0
± 14.5 kg; ISS 24.5 ± 4.6 points). Hand grip strength increased (p < 0.001) at a
rate of 0.85%/d. Muscle thickness decreased (p < 0.001) at rates of −0.47%/d
(RF), −0.39%/d (VL), and −0.38%/d (TA); no difference in the rate of decline
between muscles (p = 0.908). Recovery of VL thickness was observed between
the third and fourth week (p = 0.016). There were no changes in subcutaneous
tissue thickness. Muscle perfusion parameters SO2
, rHb, BF and BFV showed
high variability with significant time effects only in the rHb of the TA (p =
0.003). CRP and leukocyte count decreased (both p < 0.001). Unexpectedly,
grip strength increased despite a reduction in muscle thickness, likely after
decreasing compared with pre-injury. Possible reasons are discussed
Heterologous expression and genetic engineering of natural product biosynthetic pathways
The misuse and overuse of antibiotics, combined with the ability of bacteria to adapt and develop resistance, have led to more and more cases of infections that are very hard or even impossible to treat. Multi-resistant pathogens are already a serious threat to human health today, and their growing numbers worldwide are alarming. To address this crisis, the discovery of new antibacterial compounds and their optimisation into clinical candidates are of crucial importance. This thesis focuses on the antibiotic darobactin, which represents a promising novel class of antibiotics, effective against Gram-negative pathogens. The work describes the generation of novel darobactin derivatives, some of which demonstrate improved bioactivity, such as, the lead candidate D22 showing promising efficacy in in vitro and in vivo models. In addition, structural limits of the biosynthetic production method were explored. However, a key challenge remains: overcoming production limitations in the heterologous host Escherichia coli. To address this, the thesis investigates alternative production systems, including Gram-positive and an eukaryotic host, as potential solutions. Additionally, efforts to identify the previously unknown peptidase involved in darobactin's maturation process are described, which could pave the way for enhancing its production efficiency.Der unsachgemäße und übermäßige Einsatz von Antibiotika sowie die Fähigkeit von Bakterien, sich anzupassen und Resistenzen zu entwickeln, haben zu immer mehr Infektionen geführt, die schwer oder sogar unmöglich zu behandeln sind. Multiresistente Erreger stellen bereits heute eine ernsthafte Bedrohung für die menschliche Gesundheit dar und ihre weltweite Zunahme ist alarmierend. Um diese Krise zu bewältigen, ist die Entdeckung neuer antibakterieller Wirkstoffe und deren Entwicklung zu klinischen Kandidaten entscheidend. Im Mittelpunkt dieser Arbeit steht das Antibiotikum Darobactin, eine vielversprechende neue Wirkstoffklasse gegen Gram-negative Erreger. Es wird die Herstellung neuartiger Darobactin-Derivate beschrieben, von denen einige verbesserte Bioaktivität zeigen, darunter der Spitzenkandidat D22, der vielversprechende Wirksamkeit in in vitro- und in vivo-Modellen zeigt. Zudem wurden die strukturellen Grenzen der biosynthetischen Produktionsmethode untersucht. Eine zentrale Herausforderung bleibt jedoch die Überwindung der Produktionslimitierungen im heterologen Wirt Escherichia coli. Zur Lösung dieses Problems werden alternative Produktionssysteme, darunter Gram-positive und ein eukaryotischer Wirt, als potenzielle Ansätze untersucht. Zusätzlich werden Strategien zur Identifizierung der bislang unbekannten Peptidase beschrieben, die an der Reifung von Darobactin beteiligt ist und deren Entschlüsselung zur Produktionsoptimierung beitragen könnte
Combating antimicrobial resistance through underexplored drug targets and drug-delivery systems
Finding new ways to fight infections is more important than ever, particularly due to the global threat of antimicrobial resistance, which has rendered many current treatments ineffective, leading to prolonged hospital stays and increasing healthcare costs. This thesis tackles this challenge by investigating underexplored drug targets, discovering new antitubercular agents, and improving antibiotic efficacy through advanced drug-delivery systems. First, we focus on an underexplored drug target, energy-coupling factor transporters, which are crucial for nutrient uptake in Gram-positive bacteria but are absent in humans, making them an ideal target for selective inhibition. In parallel, we explore the development of new antitubercular agents targeting Mycobacterium tuberculosis. Both projects involve compound library screening, hit identification, and lead optimization to discover new inhibitors that serve as potent anti-infective agents against these pathogens. Second, we explore biodynamers, dynamic biopolymers with pH-responsive reversible covalent bonds, for the delivery and controlled release of potent drugs. These biocompatible and biodegradable materials offer unique drug-delivery advantages such as infection and endo/lysosome-responsive behavior. This research focuses on designing a novel fluorescent biodynamer to study polymer behavior and fate in situ after cellular uptake, with the potential to answer unresolved questions and advance drug-delivery applications.Die Entwicklung neuer Strategien zur Bekämpfung von Infektionen ist angesichts der weltweiten Bedrohung durch antimikrobielle Resistenzen, die viele aktuelle Behandlungen unwirksam gemacht haben, dringender denn je. Diese Doktorarbeit befasst sich mit diesem Problem, indem sie wenig erforschte Zielproteine untersucht, neue antituberkulöse Wirkstoffe entdeckt und die Wirksamkeit von Antibiotika durch fortschrittliche Wirkstofftransportsysteme verbessert. Im ersten Teil konzentrieren wir uns auf den Energy Coupling Factor (ECF)- Transporter, der für die Nährstoffaufnahme Gram-positiver Bakterien unerlässlich ist, beim Menschen jedoch nicht vorkommt und daher ein ideales Ziel für selektive Hemmstoffe darstellt. Parallel dazu erforschen wir die Entwicklung neuer Wirkstoffe gegen Mycobacterium tuberculosis. Beide Projekte umfassen das Screening von Substanzbibliotheken, die Identifizierung von hits und die Optimierung von Leitstrukturen, um neue potente Hemmstoffe zu entdecken. Im zweiten Teil werden Biodynamere, dynamische und pH-sensitive Biopolymere mit reversiblen kovalenten Bindungen, für die Abgabe und kontrollierte Freisetzung wirksamer Arzneimittel untersucht. Diese biokompatiblen und biodegradierbaren Materialien zeichnen sich durch ein infektions sowie endo-/lysosomenresponsives Verhalten aus. Ein neuartiges fluoreszierendes Biodynamer wurde entwickelt, um das intrazelluläre Verhalten des Polymers und den Verbleib in situ nach der zellulären Aufnahme zu untersuchen, um die Anwendung der Arzneimitteltransport voranzutreiben
Vergleich der Effekte von Delaire-Maske und skelettal verankerter Klasse III-Mechanik
Ziel: Das Ziel dieser Arbeit war der Vergleich von Therapieeffekten von Delaire-Maske und skelettal verankerter Klasse III-Mechanik in Anlehnung an die Methode nach De Clerck. Patienten und Methode: Untersucht wurden Patienten, die aufgrund einer frühen ausgeprägten maxillären Mikro- beziehungsweise Retrognathie mittels Delaire-Maske (Gruppe 1) oder skelettal verankerter Klasse III-Mechanik (Gruppe 2) therapiert wurden. Zur Beurteilung der Therapieeffekte wurden extraorale Fotoaufnahmen, Modellpaare und Fernröntgenseitenbilder vor Therapiebeginn und nach frühestens einem Jahr nach Insertion der Apparaturen ausgewertet und verglichen. Ergebnisse: In Gruppe 2 konnte eine höchst signifikante (p<0,001) Verlagerung des Oberkiefers nach anterior beobachtet werden. Die sagittale Kieferrelation veränderte sich in beiden Gruppen hoch signifikant (p<0,01). Die vertikale Kieferrelation vergrößerte sich in Gruppe 1 hoch signifikant (p<0,01). In Gruppe 1 war eine hoch signifikante (p<0,01) Proklination der Oberkiefer-Schneidezähne zu beobachten. Zudem zeigte sich in Gruppe 1 eine hoch signifikante (p<0,01) Retroinklination der Unterkiefer-Schneidezähne. Die Oberlippe der Patienten aus Gruppe 1 verlagerte sich signifikant (p<0,05) nach anterior. In Gruppe 2 verlagerte sich die Unterlippe signifikant (p<0,05) nach posterior. Die anteriore und posteriore Zahnbogenbreite vergrößerten sich in Gruppe 1 höchst signifikant (p<0,001). Die sagittale Frontzahnstufe vergrößerte sich in Gruppe 1 höchst signifikant (p<0,001) und in Gruppe 2 signifikant (p<0,05). Veränderungen der Lippenstufe waren in Gruppe 1 signifikant (p<0,05). Schlussfolgerung: Beide Behandlungsmethoden erzielten eine Protraktion der Maxilla. Bei Verwendung der Delaire-Maske ist mit dentalen Nebenwirkungen und einer Bissöffnung durch posteriore Rotation der Mandibula zu rechnen. Diese Nebenwirkungen sind bei der skelettal verankerten Klasse III-Mechanik nicht zu beobachten.Aim: The aim of this study was to compare the effectiveness of the treatment with Delaire type facemask versus bone anchored Class III treatment. Method: Patients who were treated early due to maxillary micrognathia or retrognathia using a Delaire type facemask (group 1) or bone anchored Class III treatment (group 2) were investigated. To
assess the treatment effects, extraoral photographs, dental casts and cephalometric radiographs were analysed and compared before the treatment and at earliest one year after insertion of the appliances. Results: In group 2, anterior displacement of the maxilla was observed with the highest significance (p<0.001). The sagittal jaw relation changed highly significant in both groups (p<0.01). The vertical jaw relation increased highly significant in group 1 (p<0.01). A highly significant (p<0.01) proclination of the maxillary incisors was observed in group 1. In addition, a highly significant (p<0.01) retroinclination of the mandibular incisors was observed in group 1. The upper lip protruded significantly (p<0.05) in group 1. The lower lip retruded significantly in group 2 (p<0.05). The anterior and posterior dental arch width and the sagittal anterior tooth step increased with the highest significance in group 1 (p<0.001). The overjet increased significantly in group 2
(p<0.05). The changes in the lip level were significant in group 1 (p<0.05).
Conclusion: Both methods achieved protraction of the maxilla. When using the Delaire type facemask, dental side effects and bite opening due to posterior rotation of the mandible are to be expected. These side effects are less expected with the bone anchored Class III treatment
Development of an adverse outcome pathway for 5⍺-reductase inhibition: Environmental implications for endocrine disruption
With the rising use of hair loss treatments, concerns have increased about environmental exposure to 5αreductase (SRD5A) inhibitors such as finasteride and dutasteride, classified as endocrine-disrupting chemicals (EDCs). To assess their risks, this study applied the Adverse Outcome Pathway (AOP) framework in vertebrate and invertebrate models. A sensitive LC-MS/MS assay was developed to evaluate SRD5A inhibition in human and fish cell lines, providing species-specific insights. In zebrafish embryos, inhibition caused hormonal disruptions, including reductions in dihydrotestosterone (DHT) and 17βestradiol, underscoring DHT’s role in estrogenic signaling. In H295R cells, GC-MS/MS-based profiling of 17 steroid hormones revealed systemic disruptions across progestins, glucocorticoids, mineralocorticoids, and androgens, extending beyond OECD TG 456 assays. To enhance ecological relevance, Daphnia magna was studied; genomic data suggested an SRD5A-like enzyme involved in ecdysteroidogenesis. Finasteride exposure impaired reproduction, affected ecdysteroid-related gene expression, and altered lipid metabolism. These findings connect molecular initiating events to adverse outcomes across species, advancing robust AOPs for regulatory toxicology. This work highlights the systemic and environmental risks of SRD5A inhibitors and supports sustainable chemical management to protect ecosystems and human health.Mit der zunehmenden Nutzung von Haarwuchsmitteln wächst die Sorge über Umweltbelastungen durch 5α-Reduktase-(SRD5A)-Inhibitoren wie Finasterid und Dutasterid, die als endokrine Disruptoren (EDCs) gelten. Zur Risikobewertung wurde das Adverse Outcome Pathway (AOP)-Konzept in Wirbeltier- und Wirbellosenmodellen angewendet. Ein sensitiver LC-MS/MS-Assay ermöglichte die Analyse der SRD5A-Hemmung in menschlichen und Fischzelllinien und lieferte artspezifische Einblicke. In Zebrafischembryonen führte die Hemmung zu Hormonstörungen, darunter verringerte Spiegel von Dihydrotestosteron (DHT) und 17β-Östradiol, was die Rolle von DHT in östrogener Signalgebung verdeutlichte. In H295R-Zellen zeigte das GC-MS/MS-Profiling von 17 Steroidhormonen systemische Störungen über Gestagene, Glukokortikoide, Mineralokortikoide und Androgene hinaus und erweiterte damit bestehende OECD-TG-456-Ansätze. Zur ökologischen Relevanz wurde Daphnia magna untersucht; genomische Daten deuten auf ein SRD5A-ähnliches Enzym in der Ecdysteroidogenese hin. Eine Exposition gegenüber Finasterid beeinträchtigte die Reproduktion, veränderte den Lipidstoffwechsel und beeinflusste ecdysteroidassoziierte Genexpression. Die Ergebnisse verknüpfen molekulare Initiierungsereignisse mit nachteiligen Wirkungen über Spezies hinweg und fördern robuste AOPs für die regulatorische Toxikologie. Damit werden die systemischen und ökologischen Risiken von SRD5AInhibitoren hervorgehoben und Grundlagen für nachhaltiges Chemikalienmanagement geschaffen
Reciprocal effects of conditioned medium on gene and protein expression of limbal epithelial cells and limbal fibroblasts in congenital aniridia
Congenital aniridia is marked by substantial inflammatory changes to the ocular surface. However the exact mechanisms of epithelial-stromal interaction are not fully understood. The purpose of this study was to investigate inflammatory cytokine expression in limbal epithelial cells and fibroblasts following exposure to each other’s conditioned medium (CM). Healthy primary limbal epithelial cells (pLECs) and healthy (LFC) or aniridia primary limbal fibroblasts (AN-LFC) were isolated. A PAX6-deficient limbal epithelial cell line (mut-LSCs) modeled aniridia. pLECs underwent siRNA-mediated PAX6 knockdown (siPAX6 pLECs) with control cells transfected with non-specific siRNA (siCtrl pLECs). siCtrl and siPAX6 pLECs were treated with LFC-CM and AN-LFC-CM for 24 hours, while LFC and AN-LFC were treated with pLECs-CM and mut-LSCs-CM for 48 hours. Gene and protein expression of IL-1β, IL-6, IL-8, TNF-α and VEGF-A were measured using qPCR and ELISA. Except for an increased IL-8 protein expression in siPAX6 pLECs treated with LFC-CM, gene and protein levels of inflammatory biomarkers remained unchanged in siCtrl and siPAX6 pLECs, regardless of treatment with LFC-CM or AN-LFC-CM. In LFCs, pLECs-CM decreased TNF-α mRNA and IL-8 protein, while increasing IL-1β, IL-6, IL-8 mRNA and IL-6 protein. LFCs treated with mut-LSCs-CM showed decreased TNF-α mRNA and increased IL-6 protein. AN-LFCs treated with pLECs-CM showed increased IL-6, IL-8 and VEGF-A mRNA and IL-6 protein. mut-LSCs-CM did not alter AN-LFC expression. Limbal fibroblasts’ secretome minimally inflames limbal epithelial cells, suggesting a supportive niche role. In contrast, pLECs-CM induces a stronger fibroblast response, indicating abnormal interactions in congenital aniridia
Undifferenzierte Oli-neu Zellen als in vitro-Modell für Multiple Sklerose : Eine Cuprizon-induzierte Reduktion der Connexin29- und PLP-1-Expression ist umkehrbar durch den Pannexin-1-Inhibitor Probenecid
Multiple Sklerose (MS) ist eine neurodegenerative Erkrankung, welche durch Symptome wie progrediente Lähmungszustände, vegetative Störungen oder kognitive und psychomotorische Veränderungen gekennzeichnet ist. Diese sind auf nervale Schädigungen im Rahmen von autoimmunbedingten Inflammationsreaktionen und Demyelinisierungen zurückzuführen. Hieran maßgeblich beteiligt sind die Oligodendrozyten, welche im differenzierten Stadium das Myelin bilden. Aber auch nicht-myelinisierende Oligodendrozyten-Vorläuferzellen werden durch die MS beeinträchtigt. Ein in vivo-Modell zur Untersuchung des Pathomechanismus der MS ist das Cuprizon-(CPZ)-Modell, anhand dessen die toxische Komponente der Erkrankung abgebildet werden kann. Durch die Applikation des Kupferchelators Cuprizon konnte ein Oligodendrozytenuntergang und folglich eine Demyelinisierung induziert werden.
Oligodendrozyten können im panglialen Synzytium mit anderen Gliazellen interagieren. Dabei erfolgt ein Informationsaustausch maßgeblich über Gap Junction-Kanäle. Diese bestehen aus Connexinen (Cx), einer Familie von Transmembranproteinen mit zelltypspezifischer Expression der Isoformen. Neben Connexinen zeichnen sich Oligodendrozyten durch die spezifische Expression weiterer Proteine, wie beispielsweise dem Proteolipid-Protein (PLP1) und dem Myelin-Basic Protein (MBP), aus. Oligodendrozyten exprimieren zudem weitere Membranproteine, die Pannexine, welche ebenfalls Membrankanäle ausbilden können. Diese Kanäle können durch den Wirkstoff Probenecid, einem Pannexin-1-Inhibitor, blockiert werden. Hierdurch konnten im Tiermodell präventive Effekte auf eine CPZ-induzierte Demyelinisierung dargestellt werden.
Das Ziel dieser Arbeit bestand darin, die oben genannten Proteine quantitativ und qualitativ in immortalisierten Oligodendrozyten-Vorläuferzellen der Zelllinie Oli-neu im undifferenzierten Zustand darzustellen. Weiterführend bestand die Überlegung, dass die Auswirkungen des CPZ auf Oligodendrozyten durch eine Dysfunktion von Connexinen oder anderen oligodendrozytenspezifischen Proteinen bedingt sein könnten, weshalb dies in vitro analysiert werden sollte. Außerdem konnte Pannexin-1 im Rahmen dieser Arbeit in Oli-neu Zellen nachgewiesen werden. Unter der Vorstellung, dass Oli-neu Zellen somit eine Zugangsmöglichkeit für PBN bieten könnten, um den CPZ-bedingten Effekten entgegenzuwirken, erfolgte die Untersuchung dieser Effekte in vitro. Hierdurch könnte eine Remyelinisierung durch Oligodendrozyten-Vorläuferzellen begünstigt werden, was so einen möglichen Ansatz zur Therapie der MS darstellen könnte.
Mittels RNA-Analyse konnten oligodendrozytäre Connexine und spezifische Proteine quantitativ und qualitativ in Oli-neu Zellen nachgewiesen werden. Auf Proteinebene erfolgte der Nachweis mittels einer Immunfluoreszenzfärbung. Da sich unter den Connexinen Cx29 als sehr stark exprimiert erwies, wurden die Oli-neu Zellen hinsichtlich dieses Connexins mit der Gefrierbruchtechnik (FRIL, Freeze-fracture replica immunogold labeling, „Gefrierbruch-Replika Immunogoldmarkierung“) im Elektronenmikroskop untersucht. Hierbei konnte eine ultrastrukturelle Ähnlichkeit zu Cx29 im peripheren Nervensystem festgestellt werden, da sich dieses Connexin sowohl in Oli-neu Zellen als auch in Schwann-Zellen des peripheren Nervensystems in hexamerer Formation, den sogenannten Rosetten, anordnete. Unter der Auffassung, dass hemihexamere Connexin-Anordnungen sich später zu ebensolchen Rosetten formieren könnten, wurden solche prägnanten Formationen als präformierende Komplexe benannt.
Durch die weiterführende CPZ-Applikation zeigte sich in Oli-neu Zellen hinsichtlich der Proteinexpression in der Immunfluoreszenzfärbung für Cx29, MBP und PLP1 ein signifikanter Abfall der immunpositiven Fläche. Durch die Applikation des PBN konnten diese Effekte für Cx29 und PLP1 umgekehrt werden.
Zusammengefasst bleibt zu unterstreichen, dass im Rahmen dieser Arbeit erstmals die ultrastrukturelle Darstellung von Cx29 in Oli-neu Zellen gezeigt werden konnte. Hieraus ergeben sich vielfältige Möglichkeiten zur weiterführenden Untersuchung der Oli-neu Zellen, z. B. hinsichtlich der Connexin-Funktion und der Connexin-Kopplung in Co-Kultivierung mit anderen Zellen. Darüber hinaus erfolgte erstmals der Nachweis von Panx1 in Oli-neu Zellen. Durch die anschließende Applikation des Pannexin-1-Inhibitoren PBN, konnten die CPZ-bedingten Effekte auf die Proteinsynthese von Cx29 und PLP1 umgekehrt werden. Somit stellt der Wirkstoff PBN einen möglichen Ansatz zur Therapie der MS dar, was es weiter zu untersuchen gilt.Immature Oli-neu cells as an in vitro model for multiple sclerosis: cuprizone-induced reduction of Connexin29- and PLP-1-expression can be reversed by the pannexin-1-inhibitor probenecid
Multiple sclerosis (MS) is a neurodegenerative disease characterized by symptoms like progressive paralysis, vegetative disorders or cognitive changes. These symptoms are based on axonal damage in the context of autoimmune-related inflammatory reactions and demyelination. Mature oligodendrocytes are involved in this, since they are responsible for the formation of myelin in the central nervous system. In addition, non-myelinating oligodendrocyte precursor cells are also affected by MS. There are several models for the investigation of the pathomechanism of MS. The cuprizone (CPZ) model can be used to demonstrate the toxic component of MS. The application of the copper chelator cuprizone causes oligodendrocyte death and consequently demyelination.
Oligodendrocytes interact with other glial cells in the panglial syncytium by exchange of information, for example via gap junction channels. These channels consist of connexins (Cx), a family of transmembrane proteins with cell type-specific expression. In addition to this, oligodendrocytes specifically express other proteins, such as the proteolipid protein (PLP1) and the myelin basic protein (MBP). Furthermore, oligodendrocytes express another group of transmembrane proteins, the pannexins, which can form channels connecting the cytoplasm to the extracellular space. These channels can be blocked specifically by Probenecid (PBN). There are hints that PBN could reverse the effects caused by CPZ.
In this work, it was the aim to visualize the mentioned proteins quantitatively and qualitatively in immortalized and immature oligodendrocyte precursor cells of the cell line Oli-neu. Moreover, it was hypothesized that the CPZ-induced effects might be caused by a dysfunction of connexins or other oligodendrocyte-specific proteins. With regard to this, the CPZ effects were analyzed in immature Oli-neu cells. In this study, pannexin-1 was detected in Oli-neu cells for the first time. Thus, the effects of CPZ and a possible counteraction by PBN should be investigated in Oli-neu cells.
Via analysis of mRNA-expression, oligodendrocytic connexins and specific proteins were detected quantitatively and qualitatively in Oli-neu cells. For protein analysis, immunofluorescence staining was used. Cx29 was found to be very prominent. For examination of this connexin at the ultrastructural level, the freeze-fracture technique (FRIL, freeze-fracture replica immunogold labeling) was used. In both Oli-neu cells and Schwann-cells of the peripheral nervous system, Cx29 is frequently observed in hexameric formations, the so-called rosettes. Under the assumption that hemi-hexameric connexin arrangements could later form into these rosettes, they were called „präformierende Komplexe”.
Additionally, a CPZ-application to Oli-neu cells showed a significant decrease in the immunopositive areas of Cx29, MBP and PLP1. The application of PBN reversed these effects for Cx29 and PLP1.
To summarize, it remains to point out that the ultrastructural representation of Cx29 in Oli-neu cells was shown for the first time in this work. This opens many possibilities for further investigation of Oli-neu cells like the function of this connexin and potential coupling partners in co-cultivation with other cells. Furthermore, Panx1 was detected in Oli-neu cells for the first time. By the application of the Panx-1 inhibitor PBN, the CPZ-related effects on the protein synthesis of Cx29 and PLP1 could be reversed. This is why PBN represents a possible approach for the treatment of MS and should be investigated in more detail
Repeatability of Corneal Astigmatism and Equivalent Power with the MS-39 Tomographer Derived from Model Surface Fitting in a Cataractous Population
We investigated the repeatability of the MS-39 in determining power vector components—the spherical equivalent (SEQ) and astigmatic powers (C0 and C45) and asphericity (Q)—of corneal epithelium, stroma, and endothelium in a large patient cohort. In this retrospective cross-sectional single-centre study, we evaluated a dataset containing 600 MS-39 anterior segment tomography measurements from 200 eyes (three repeat measurements each) taken prior to cataract surgery. The exported measurements included height map data for the epithelium, stroma, and endothelium surface. Model surfaces (spherocylinder (SphCyl), cylindrical conoid (CylConoid), and biconic (Biconic), all in the 3/6 mm zone) were fitted using nonlinear iterative optimisation, minimising the height difference between the measurement and model. The mean (MEAN) and standard deviation (SD) for each sequence of measurements were derived and analysed. In the 3 mm and 6 mm zone, the MEAN SEQ was 53.47/53.56/53.57 and 53.21/53.54/53.54 D for SphCyl/CylConoid/Biconic for the epithelium, −4.47/−4.51/−4.51 and −4.45/−4.50/−4.50 D for the stroma, and −6.23/−6.26/−6.26 and −6.18/−6.29/−6.30 D for the endothelium. With the three surface models and the 3/6 mm zone, the SD for SEQ/C0/C45 was in the range of 0.04 to 0.11/0.05 to 0.13/0.04 to 0.11 D for epithelium; 0.01 to 0.02/0.01 to 0.05/0.01 to 0.06 D for stroma; and 0.01 to 0.02/0.02 to 0.07/0.03 to 0.07 D for endothelium. Fitting floating model surfaces with astigmatism to map data of the corneal epithelium, stroma, and endothelium seems to be a robust and reliable method for extracting equivalent power and astigmatism using all the datapoints within a region of interest

The 2025 conference of the research section “Comparative Politics” of the German Political Science Association (DVPW) was dedicated to the topic: “Citizen’s
Trust and Societal Polarization in Times of Transformation: Comparative Perspectives from Different World Regions.” Around 100 scholars from across Europe gathered from March 27 to 28 at Saarland University in Saarbrücken for the largest
political science event to date hosted by the two political science chairs of the
university’s still young department for European Social Research.
The conference is the result of two international research projects that are part of
the Horizon Europe program: “Activating European Citizens Trust in Times of Crises
and Polarization” (ActEU, led by the University of Duisburg-Essen and Saarland
University) and PROTEMO (“Emotional dynamics of protective policies in an
age of insecurity”, led by Saarland University). The aim of the conference was to
address and discuss the most pressing challenges facing representative democracy
across 20 panels: citizens’ trust—or distrust—in “their” political system, institutions,
political representatives, and the EU as well as the interrelationships between trust,
emotions and societal polarization. These overall topics are linked to long-term
transformations related to migration and integration, climate change, technological
change, gender inequalities, Europeanisation or region-specific challenges. Each
of the 20 panels offered valuable perspectives on the concepts, determinants and
consequences of political trust, polarization and participation
