    Uncertainty-Aware Predictive Process Monitoring in Healthcare: Explainable Insights into Probability Calibration for Conformal Prediction

    In high-stakes decision-making environments, predictive models must deliver not only high accuracy but also reliable uncertainty estimations and transparent explanations. This study explores the integration of probability calibration techniques with Conformal Predic tion (CP) within a predictive process monitoring (PPM) framework tailored to healthcare analytics. CP is renowned for its distribution-free prediction regions and formal coverage guarantees under minimal assumptions; however, its practical utility critically depends on well-calibrated probability estimates. We compare a range of post-hoc calibration meth ods—including parametric approaches like Platt scaling and Beta calibration, as well as non-parametric techniques such as Isotonic Regression and Spline calibration—to assess their impact on aligning raw model outputs with observed outcomes. By incorporating these calibrated probabilities into the CP framework, our multilayer analysis evaluates improvements in prediction region validity, including tighter coverage gaps and reduced minority error contributions. Furthermore, we employ SHAP-based explainability to explain how calibration influences feature attribution for both high-confidence and ambigu ous predictions. Experimental results on process-driven healthcare data indicate that the integration of calibration with CP not only enhances the statistical robustness of uncertainty estimates but also improves the interpretability of predictions, thereby supporting safer and robust clinical decision-making

    Arbeiten zu einer neuen Strategie zur Synthese von Cavicularin

    Das makrocyclische Bisbibenzyl Cavicularin wurde 1996 aus dem Lebermoos Cavicularia densa isoliert. Die einzigartige Struktur, die axiale und planare Chiralität kombiniert macht Cavicularin zu einer ebenso interessanten wie herausfordernden Zielverbindung in der Naturstoffsynthese. Bisher konnten einige sowohl racemische als auch atropselektive Synthesen von Cavicularin realisiert werden. Im Rahmen dieser Arbeit wurde versucht eine neue Synthesestrategie zur atropselektiven Synthese von Cavicularin zu entwickeln. Diese Strategie basiert auf einer Makrocyclisierung durch Auxiliar-gesteuerte atropdiastereoselektive HECK-Reaktion. Als Auxiliar findet eine zur Biarylachse ortho-ständige p-Tolylsulfinylgruppe Verwendung.The macrocyclic Bisbibenzyl Cavicularin was isolated from the liverwort Cavicularia densa in 1996. Its unique structure combining axial and chiral chirality makes it an interesting and challenging target molecule in natural product synthesis.     The proteomic landscape of trophoblasts unravels calcium-dependent syncytialization processes and beta-chorionic gonadotropin (ß-hCG) production

    Background The syncytiotrophoblast (STB) layer of the placenta is formed by cell fusion of cytotrophoblasts, acts as a feto-maternal barrier, is required for the production of pregnancy hormones such as chorionic gonadotropin, estradiol and progesterone and is also responsible for feto-maternal mineral exchange such as calcium. Adequate mineral supply and placental hormone production are essential for the maintenance of pregnancy, and disturbances in trophoblast integrity are associated with pregnancy complications. Since knowledge about the identity and expression levels of proteins in trophoblast and syncytiotrophoblast cells is limited so far, we analyzed the proteomes of trophoblast-like and syncytiotrophoblast-like BeWo cells under different calcium conditions. The investigation of protein expression profiles in combination with hormone assays can provide a better understanding of calcium dependent cellular processes in trophoblasts and syncytiotrophoblasts. Methods Here, we combine human trophoblast model cell cultures, hormone assays, antibody-based detection methods and high-resolution mass spectrometry analyzes to assess changes in cellular processes during syncytialization. Results We monitored the changes in protein expression profiles during forskolin induced syncytialization of trophoblast-like cells in an unbiased manner and show that the expression of numerous proteins is strongly altered. Among them are enzymes of the glucocorticoid and sex hormones synthesis pathways such as cytochrome P450 (CYP) 19A1, CYP11A1, adrenodoxin (FDX1), hydroxysteroid dehydrogenase (HSD) 11β2 and HSD17β1, whose expression is strongly induced by syncytialization. The production of beta human chorionic gonadotropin (ß-hCG), progesterone and estradiol increase during syncytialization, while the secretion and synthesis of ß-hCG and the expression of several protein syncytiotrophoblast markers show a clear calcium dependence. Conclusion The broad applicability of semi-quantitative proteome profiling of cytotrophoblast- and syncytiotrophoblast-like cells provides new insights into signaling processes that occur in cytotrophoblasts / syncytiotrophoblasts during pregnancy

    Tolerability of PSMA radioligand therapy in metastatic prostate cancer patients with baseline mild to moderate leukopenia

    Background Aim of this study was to analyze the safety of prostate-specifc membrane antigen radioligand therapy (PSMA-RLT) in patients with metastatic castration-resistant prostate cancer (mCRPC) with preexisting mild to moder‑ ate leukopenia (CTCAE≥1). Results Thirty-seven mCRPC patients with preexisting leukopenia (leukocyte count<3.8× 109 /L) were included in this study. Patients received a median of 3 cycles of [ 177Lu]Lu-PSMA-617 (range 1–9). No signifcant diference in leu‑ kocyte counts was observed between baseline and follow-up after each PSMA-RLT cycle: frst cycle (3.0±0.5 at base‑ line vs. 3.4±1.4 at follow up [in× 109 /L], p=0.0921), second cycle (3.1±0.4 vs. 3.8±1.7, p=0. 0.0509), and third cycle (3.1±0.4 vs. 3.2±2.0, p=0.2929), respectively. Similarly, baseline and end of treatment values, irrespective of the num‑ ber of administered cycles, did not reveal a signifcant diference (3.0±0.5 vs. 3.5±1.4, p=0.0684). After the end of therapy, irrespective of the number of administered cycles, 27% patients remained stable in terms of CTCAE scor‑ ing, 46% changed to a higher score and 27% improved to a lower score. Conclusion Although marked preexisting leukopenia is often considered a relative contraindication for PSMA-RLT, our fndings indicate that PSMA-RLT is feasible in patients with leukopenia of CTCAE grade ≥1. In our cohort, leuko‑ cyte counts remained stable without signifcant RLT-induced deterioration. Therefore, patients with leukopenia should not be categorically excluded from receiving PSMA-RLT.     Langzeitergebnisse nach Mitralklappenrekonstruktion mit Papillarmuskelreposition bei ischämischer Kardiomyopathie

    Klinischer Hintergrund: Die ischämische Mitralklappeninsuffizienz (MI) kann als typische Komplikation eines Myokardinfarktes auftreten und ist mit einer schlechten Prognose assoziiert. Eine aggressive Ringraffung mit einem geschlossenen starren Anuloplastie- Ringimplantat (RING) kann zu einem zufriedenstellenden Operationsergebnis führen, solange keine erhebliche LV-Dilatation oder eine schwere restriktive Segelbeweglichkeit vorliegen. Am Universitätsklinikum des Saarlandes Homburg/ Saar wurde eine spezifische Rekonstruktionstechnik entwickelt, welche den posterioren Papillarmuskel mithilfe einer transventrikulären Polytetrafluorethylen-Naht (STRING) in Richtung des aortomitralen Übergangs repositioniert. Methodik: Zwischen 9/2004 und 7/2013 wurde die RING+STRING-OP-Technik bei 104 Patienten (73 männlich, Alter 68±10 Jahre, MPAP 32,85±11,14mmHg, EF 33,76±11,73%, LVEDD 60,91±7,61mm) angewandt. 20 dieser Patienten hatten sich zuvor einer Koronararterien-Bypass-OP unterzogen – bei drei Patienten war bereits eine Mitralklappenrekonstruktion durchgeführt worden. Alle Patienten erhielten eine moderate Ringanuloplastie (partieller oder kompletter Ring, mittlere Größe 30 mm) kombiniert mit einer STRING-Naht. Begleitende Eingriffe waren Koronarbypass (n=87), Trikuspidalklappenrekonstruktion (n=62) und Vorhofablation (n=32). Die Perfusionszeit an der Herz-Lungen-Maschine betrug 110±31 min, die myokardiale Ischämiezeit betrug 71±20 min, eine intraaortale Ballonpumpe wurde bei neun Patienten notwendig. Ergebnisse: Die 30-Tage-Mortalität lag bei 6 %, die 5-Jahres-Überlebensrate bei 71 %. Nach fünf Jahren waren 89 % der Patienten frei von einer MI ≥ II und 95 % frei von einer Reoperation. Die Dimensionen des linken Ventrikels nahmen enddiastolisch (von 61±8 auf 57±8 mm, p<0.001) und endsystolisch (von 49±9 auf 46±8 mm, p <0.001) ab. Eine Subgruppenanalyse bezogen auf den präoperativen LVEDD ( 65 mm: n=29) zeigte keinen Unterschied in Bezug auf Überleben (p=0.27) oder die Freiheit von einer Reoperation nach 5 Jahren (p=0.1). Fünf Patienten mussten sich einer Reoperation bei MIRezidiv (partieller Ring: n=5, präoperativer LVEDD > 65 mm: n=3) unterziehen. Eine erneute Rekonstruktion erfolgte bei einem dieser 5 Patienten - die übrigen 4 erhielten einen Mitralklappenersatz. Schlussfolgerung: Die Kombination aus moderater Ringraffung und einer zusätzlichen Papillarmuskelreposition führt zu einer geringeren Rate von Rezidiv-Mitralklappeninsuffizienz bei Hochrisiko-Patienten mit schwerer restriktiver Segelbeweglichkeit. Stabile Rekonstruktionsergebnisse und ein Reverse Remodeling des linken Ventrikels konnten sogar mit partiellen Ringimplantaten und trotz erheblicher präoperativer Dilatation des linken Ventrikels erreicht werden. Die Korrektur der pathologischen Veränderung des subvalvulären Apparates ist bei der ischämischen Mitralklappeninsuffizienz somit von entscheidender Bedeutung.Objective: Ischemic mitral regurgitation (MR) is a typical complication of myocardial infarction and is associated with a poor prognosis. Undersized ring annuloplasty with complete rigid annuloplasty devices (RING) can lead to satisfactory results unless marked left ventricular dilatation and severe leaflet tethering are present. The Department of Thoracic and Cardiovascular Surgery of the Saarland University Medical Center has developed a disease-specific repair technique, which relocates the posterior papillary muscle towards the aorto-mitral continuity using a transventricular polytetrafluorethylene-suture (STRING). Methods: Between 9/2004 and 7/2013 this principle (RING+STRING) was applied in 104 patients with a tenting height > 10 mm (73 male, age 68±10 years, MPAP 36±10 mmHg, EF 34±12 %, LVEDD 61±8 mm). Twenty of those patients had previous CABG – three patients already had previous mitral valve repair. All patients underwent only moderately undersized annuloplasty (partial or complete ring devices, median size 30 mm) combined with STRING. Concomitant procedures were CABG (n=87), tricuspid valve repair (n=62) and atrial ablation (n=32). Cardiopulmonary bypass time was 110±31 min, myocardial ischemic time was 71±20 min, an intraaortic balloon pump was required in 9 patients. Results: 30-day mortality was 6 %, 5-year survival 71 %. Freedom from MR > II at 5 years was 89 %, freedom from reoperation at 5 years 95 %. Left ventricular dimensions (enddiastolic: 61±8 to 57±8 mm, p<0.001; endsystolic: 49±9 to 46±8 mm, p<0.001) decreased. Subgroup analyses for preoperative LVEDD ( 65 mm: n=29) revealed no differences between groups for survival (p=0,27) and freedom from reoperation (p=0,1) at 5 years. Five patients underwent reoperation for recurrent mitral regurgitation (partial ring: n=5, preoperative LVEDD > 65 mm: n=3). Successful redo repair could be performed in one patient – all other underwent mitral valve replacement. Conclusions: The combination of moderate annular downsizing and adjunctive papillary muscle repositioning leads to low rate of recurrent mitral regurgitation in high-risk-patients with severe leaflet tethering. Durable results and LV reverse remodeling could even be achieved with the use of partial annuloplasty rings and in the presence of significant preoperative LV-dilatation.     Application of Protein Structure Encodings and Sequence Embeddings for Transporter Substrate Prediction

    Membrane transporters play a crucial role in any cell. Identifying the substrates they translo cate across membranes is important for many fields of research, such as metabolomics, pharmacology, and biotechnology. In this study, we leverage recent advances in deep learning, such as amino acid sequence embeddings with protein language models (pLMs), highly accurate 3D structure predictions with AlphaFold 2, and structure-encoding 3Di sequences from FoldSeek, for predicting substrates of membrane transporters. We test new deep learning features derived from both sequence and structure, and compare them to the previously best-performing protein encodings, which were made up of amino acid k-mer frequencies and evolutionary information from PSSMs. Furthermore, we compare the performance of these features either using a previously developed SVM model, or with a regularized feedforward neural network (FNN). When evaluating these models on sugar and amino acid carriers in A. thaliana, as well as on three types of ion channels in human, we found that both the DL-based features and the FNN model led to a better and more consistent classification performance compared to previous methods. Direct encodings of 3D structures with Foldseek, as well as structural embeddings with ProstT5, matched the performance of state-of-the-art amino acid sequence embeddings calculated with the ProtT5-XL model when used as input for the FNN classifier

    Hämodynamische Veränderungen durch mechanische Thrombektomie nach akuter Lungenembolie mit intermediärem und hohem Risiko

    Hintergrund: Zur Behandlung der akuten Lungenarterienembolien (LAE) mit intermediärem und hohem Risiko standen zusätzlich zur therapeutischen Antikoagulation nur die systemische Thrombolyse und die chirurgische pulmonale Embolektomie zur Verfügung. Da die systemische Thrombolyse aber mit einem erhöhten Risiko für potenziell lebensbedrohliche Blutungskomplikationen einhergeht, werden perkutane kathetergestützte Therapien als Alternativen untersucht. Die kathetergestützte mechanische Thrombektomie ließ sich in ersten klinischen Studien bei Patienten mit intermediärem und hohem Risiko sicher durchführen und konnte akut die Hämodynamik verbessern. Die Fragestellung der vorliegenden Arbeit ist es, die Sicherheit, klinischen Ergebnisse sowie die unmittelbaren und mittel- bis langfristigen hämodynamischen Effekte der mechanischen Thrombektomie bei akuter LAE mit Rechtsherzbelastung zu untersuchen. Methoden: In dieser prospektiven open-label-Studie wurden insgesamt 60 Patienten mit akut symptomatischer, computertomographisch dokumentierter LAE mit Rechtsherzbelastungszeichen einer mechanischen Thrombektomie mit dem FlowTriever System unterzogen. Eine Rechtsherzkatheteruntersuchung wurde unmittelbar vor und nach der Thrombektomie sowie erneut nach drei Monaten durchgeführt. Zudem wurde eine transthorakale Echokardiographie vor der Thrombektomie, zum Zeitpunkt der Entlassung sowie nach drei Monaten durchgeführt. Ergebnisse: Sechzig Patienten (33 % weiblich) unterzogen sich einer mechanischen Thrombektomie, von denen 35 auch die Nachuntersuchung drei Monate nach dem Eingriff abschlossen. Die meisten Patienten wiesen eine Hochrisiko-LAE auf (17 % der Patienten), definiert durch einen PESI-Score IV–V, einen sPESI-Score ≥ 1, hämodynamische Instabilität, Rechtsherzbelastungszeichen und positive kardiale Biomarker. 79 % der Patienten hatten ein intermediär-hohes Risiko, basierend auf einem PESI-Score III–IV, einem sPESI-Score ≥ 1, Rechtsherzbelastungszeichen, positiven kardialen Biomarker und keine Anzeichen von einer hämodynamischen Instabilität. Vor der Thrombektomie war der systolische pulmonalarterielle Druck (PAPs) stark erhöht (Mittelwert 54,3 ± 13,4 mmHg). Der mittlere PAPs verringerte sich unmittelbar nach dem Eingriff um 13,7 mmHg (95 %-Konfidenzintervall (KI): -16 bis -11,3; p < 0,001) und drei Monate nach der Thrombektomie um weitere 9,9 mmHg (95 % -KI: -13,9 bis -5,9 p < 0,001). Das Verhältnis vom rechtsventrikulären und linksventrikulären Diameter (RV/LV) nahm bis zum Zeitpunkt der Entlassung um 0,3 (95 %-KI: -0,36 bis -0,25; p < 0,001) ab. Der Anteil der Patienten mit einem TAPSE/PAPs von < 0,31 mm/mmHg, als Hinweis auf eine eingeschränkte rechtsventrikuläre-pulmonalarterielle Kopplung, konnte von anfänglich 44 % der Patienten vor der Entlassung vollständig aufgehoben werden. Kein Patient starb innerhalb von 48 Stunden nach dem Eingriff oder während der 30-tägigen Nachbeobachtungszeit. Schlussfolgerung: Bei Patienten mit intermediärem und Hochrisiko-LAE mit Rechtsherzbelastungszeichen erwies sich die mechanische Thrombektomie als effektive Methode mit niedrigem Komplikationsrisiko. Sie führte zu einer Reduktion des pulmonalarteriellen Drucks und einer Verbesserung der rechtsventrikulären Funktion sowie der rechtsventrikulär-pulmonalarteriellen Kopplung. Diese positiven hämodynamischen Effekte persistierten über einen Zeitraum von mindestens drei Monaten nach dem Eingriff. Randomisierte, kontrollierte Studien sind erforderlich, um die Rolle der mechanischen Thrombektomie bei der Behandlung von akuter LAE weiter zu evaluieren. Background: High-risk and intermediate-risk pulmonary embolism (PE) have been treated until today mostly with systemic thrombolytics, which can have serious bleeding side effects. Novel interventional therapies are increasingly being used because of beneficial acute hemodynamic effects and a good short-term safety profile. However, middle to long-term hemodynamic outcomes have not been examined thoroughly yet. The aim of the present study is to investigate the safety, clinical outcomes and immediate and middle-long-term haemodynamic effects of mechanical thrombectomy in acute LAE with right heart overload. Methods: In this prospective, open-label, single-centre study, a total of 60 patients with acute symptomatic pulmonary embolism as confirmed by computed tomography showing signs of right heart overload underwent mechanical thrombectomy with the FlowTriever aspiration system. A right heart catheterisation was performed immediately before and after the thrombectomy and three months after the patients were discharged. Transthoracic echocardiography was conducted before thrombectomy, at discharge, and after three months. Results: Sixty Patients (33% female) underwent mechanical thrombectomy, of which 35 completed the three-month follow-up examination with right heart catheterization. Most patients presented a high-risk acute pulmonary embolism (17% of patients), defined by a PESI score IV–V, an sPESI score ≥1, hemodynamic instability, signs of right heart overload, and positive cardiac biomarkers. 79% of the patients had an intermediate-high risk, based on a PESI score III–IV, an sPESI score ≥1, no sign of haemodynamic instability, signs of right heart overload, and positive cardiac biomarkers. Before thrombectomy, mean systolic PAP (PAPs) was severely elevated (54.3 ± 13.4mmHg). Mean PAPs dropped by 13.7mmHg (95% confidence interval (CI): -16 to -11.3; p < 0.001) immediately after the procedure and continued to decrease from postthrombectomy to three months (-9.9mmHg, 95% CI: -13.9 to -5.9 p < 0.001. RV/left ventricular (LV) ratio decreased before discharge by 0.3 (95% CI: -0.36 to -0.25; p < 0.001). The proportion of patients with a right ventricular arterial coupling (TAPSE/PAPs) <0.31mm/mmHg decreased from 44% at baseline to none before discharge, same results were seen at three months control. No patient died within 48 hours post-procedure or during the 30 days of follow-up. Zusammenfassung 4 Conclusion: Mechanical thrombectomy for acute intermediate- and high-risk PE with right heart overload seems to be effective and with a low risk of complications. It quickly reduced PAP, improved right heart function and enhanced RV-arterial coupling. The reduction in PAP was maintained for at least three months. Randomized, controlled trials are required to further assess the role of mechanical thrombectomy in the management of acute PE

    Identifikation robuster Merkmale für die technische Diagnostik von netzgespeisten Asynchronmotoren und der angetriebenen Arbeitsmaschinen

    Technische Diagnostik ist eine Disziplin, die lange Zeit nur für große Anlagen mit hohen Schadenssummen angewendet wurde, jedoch verschieben jüngste Entwicklungen die Anwendungsgrenze auf kleine Anlagen und Aggregate. Sensorarme Ansätze, die dies ermöglichen, profitieren aktuell von zwei Entwicklungen, dem Data-Mining und der Beschreibung spezieller Maschinenmerkmale. Probleme bei der Anwendung von technischer Diagnostik bleiben Falsch-Positiv-Bewertungen und die Erreichung der notwendigen Fehlerraten bei leicht veränderten Randbedingungen. Grund dafür ist, dass es derzeit kein Verfahren gibt, das die leistungsstarken Expertenmerkmale im Kontext einer systematischen Wissensgewinnung bewertet und dadurch oft nur partikulare Lösungen vorhanden sind. In der vorliegenden Arbeit wird das Knowledge Discovery from Experimental Data als Erweiterung des Knowledge Discovery in Database erarbeitet. In der Praxis erworbene Daten zeigen, dass mit den richtigen Merkmalen eine Klassifizierung der Fehlerzustände auch bei auftretenden Störgrößen möglich ist. Überprüft wurde dies an den exemplarisch betrachteten Fehlern Kavitation, Rotorstabbruch und Fehlausrichtung. Mit dem vorgestellten Verfahren wird es somit möglich sein, bei weiteren Kombinationen von Fehlern und Aggregaten effizient nach geeigneten Merkmalssätzen für die technische Diagnostik zu suchen. Gleichzeitig stehen für die genannten Fehler leistungsfähige Merkmalsvektoren zur Verfügung.Technical diagnostics is a discipline that for a long time was only used for large systems with high damage sums, but recent developments are shifting the application limit to small systems and aggregates. Approaches with few sensors that make this possible are currently benefiting from two developments: data mining and the description of special machine features. Problems with the application of technical diagnostics remain false-positive evaluations and achieving the necessary error rates under slightly different conditions. The reason for this is that there is currently no method that evaluates the powerful expert characteristics in the context of systematic knowledge acquisition, which means that often only partial solutions are available. In this thesis, Knowledge Discovery from Experimental Data is developed as an extension of Knowledge Discovery in Database. Data acquired in practice show that, with the right features, it is possible to classify fault states even when disturbance variables occur. This was tested using the faults cavitation, broken rotor bar and misalignment as examples. With the method presented, it will therefore be possible to efficiently search for suitable feature sets for technical diagnostics for further combinations of faults and aggregates.     Learning and exploiting temporal dependencies in the synthesis and analysis of video signals

    The acquisition, reproduction, analysis and modification of visual information are important in all parts of human life - even more so since the advent of sufficiently capable computers. Especially the computational treatment of the temporal dimension is challenging, but also beneficial for many applications. This thesis explores the temporal dimension in three different contexts: For the detection of semantically relevant manipulations, it demonstrates that previous detection methods can be fooled by the same improvements to the manipulation technique that would fool human observers. New methods are presented to nevertheless achieve high detection accuracy, and especially temporal dependencies are shown to help generalise to unseen manipulation methods. For the synthesis of new video signals, previous work has constructed models that entangle spatial and temporal features. This thesis separates these features, reducing memory demand and computation time, as well as the amount of data necessary for training. For the reconstruction of video signals from event data, a data modality for which training data is scarce, the thesis presents a method to turn event data into watchable signals, without using any training data at all, but outperforming previous methods that do so. In each of these contexts, the thesis highlights the degree to which solutions depend on training sets of different sizes, and the impact this has on performance and computational cost.Erfassung, Reproduktion, Analyse und Modifikation visueller Informationen sind wichtig für alle Bereiche menschlichen Lebens -- insbesondere seit der Verfügbarkeit leistungsfähiger Rechner. Die vorliegende Arbeit untersucht diese Dimension in drei verschiedenen Kontexten: Für die Erkennung semantisch relevanter Manipulationen wird gezeigt, dass Manipulationen, die menschliche Betrachter zuverlässig täuschen, auch die bisherigen maschinellen Erkenner in die Irre führen. Neue Erkenner werden eingeführt, denen die Modellierung zeitlicher Abhängigkeiten zu erhöhter Robustheit gegenüber ungesehenen Manipulationen verhilft. Bei der Synthese neuer Videosignale haben vorherige Arbeiten räumliche und zeitliche Zusammenhänge ineinander verwoben modelliert. Die vorliegende Arbeit trennt diese Dimensionen und reduziert so Speicherbedarf, Rechenzeit und Bedarf an Trainingsdaten. Für die Rekonstruktion von Videosignalen aus Event-Daten sind Trainingsdaten nur schwer zu beschaffen. Die Arbeit rekonstruiert Videosignale aus Event-Daten besser als vorherige Methoden, ohne Trainingsdaten zu benötigen. Für alle drei Aufgaben beleuchtet die Arbeit den Bedarf an Trainings-Datensätzen verschiedener Größen, sowie den daraus resultierenden Einfluss auf Ausgabequalität und Ressourcenverbrauch

    Harnessing stochastic forces: Exploring noise-induced phenomena in physarum-inspired network design models

    In this dissertation, we study models for the design of transport networks inspired by the paradigmatic organism Physarum polycephalum. Beyond characterizing the networks generated in a deterministic setting, we analyze how adding stochastic forces influences the topological properties of the emerging networks as well as their adaptability to dynamically changing environmental conditions. As key results, we report on noise-induced resonances, optimizing the adaptability to external factors as well as the robustness, efficiency, and cost of the resulting networks for finite noise amplitudes. Further, we shed light on the role of the interplay of nonlinear dynamics and stochastic forces in the emergence of these noise-induced phenomena, providing crucial insights for optimization algorithms in general. Our results challenge the traditional notion of noise being detrimental, demonstrating that it can even be used as a resource.In dieser Dissertation untersuchen wir Modelle zur Entwicklung von Transportnetzwerken, die von dem paradigmatischen Organismus Physarum polycephalum inspiriert sind. Neben der Charakterisierung der erzeugten Netzwerke im deterministischen Fall analysieren wir, wie das Hinzufügen stochastischer Kräfte die topologischen Eigenschaften der entstehenden Netzwerke sowie deren Anpassungsfähigkeit an sich dynamisch ändernde Umweltbedingungen beeinflusst. Als zentrale Ergebnisse berichten wir von rauschinduzierten Resonanzen, die die Anpassungsfähigkeit an externe Faktoren sowie die Robustheit, Effizienz und Kosten der generierten Netzwerke bei endlicher Rauschamplitude optimieren. Darüber hinaus beleuchten wir die Rolle des Zusammenspiels von nichtlinearer Dynamik und stochastischen Kräften bei der Entstehung dieser rauschinduzierten Phänomene, was entscheidende Einblicke in Optimierungsalgorithmen im Allgemeinen bietet. Unsere Ergebnisse hinterfragen die traditionelle Betrachtung von Rauschen als Störfaktor und zeigen vielmehr, dass es sogar von Vorteil sein kann
