A Data-Driven Approach to Lean and Digital Process Re-Modeling for Sustainable Textile Production: A Case Study
This study presents a data-driven framework that integrates lean management and digital business process modelling to enhance sustainability in textile manufacturing. Conducted in a company producing industrial safety textiles from Peru, this research applies lean tools within a digital BPM structure supported by real-time data tracking. The integrated approach led to increased production efficiency (from 79% to 86%), reduced setup times, and improved operational agility. The digital infrastructure empowered operators and supported informed decision-making. This work contributes to Industrial Engineering, Business Administration, and MIS by offering a holistic model that bridges lean principles with Industry 4.0 technologies. The findings, though context-specific, provide actionable insights for manufacturers aiming for smart and sustainable operations. Future research should validate the proposed framework across diverse industrial contexts and assess its longitudinal impact on lean performance outcomes
Selektion und Charakterisierung eines HLA-C*07:02 restringierten, CMV-spezifischen Antikörpers mittels Phagen-Display
Eine Zytomegalievirus (CMV)-Reaktivierung zählt zu den häufigsten Komplikationen bei Menschen nach allogener Stammzelltransplantation (HSCT) oder solider Organtransplantation (SOT). Trotz antiviraler Prophylaxe ist die CMV-Reaktivierung immer noch mit einer deutlich erhöhten Morbidi-tät und Mortalität bei Transplantationspatienten vergesellschaftet. Bei Gesunden wird die Infektion durch T-Zellen kontrolliert, bei Immunsupprimierten ist die T-Zellantwort für eine CMV-Kontrolle nicht mehr ausreichend.
Die T-Zell-abhängige Immunantwort stützt sich auf die intrazelluläre Prozessierung und Präsentati-on von Viruspeptiden durch MHC-Klasse-Ⅰ-Moleküle der virusbefallenen Zellen.
MHC-Klasse-Ⅰ-Moleküle werden durch die drei Gene HLA-A, HLA-B und HLA-C kodiert. T-Zellen erkennen mit ihrem T-Zell Rezeptor (TCR) den MHC/Peptid-Komplex, der von den infizierten Zel-len präsentiert wird. Aktuelle Studien konnten zeigen, dass HLA-C*07:02 kodierte MHC-Ⅰ-Rezeptoren kaum von immunevasiven Mechanismen des CMV betroffen sind.
TRC-like Antikörper können, im Gegensatz zu herkömmlichen Antikörpern, ähnlich dem TCR der T- Zellen den MHC/Peptid-Komplex binden und bieten sich daher als mögliche Therapieoption in Situationen mit T-Zell Defekt an.
Mit Hilfe der Phagen-Display-Methode haben wir HLA-C*07:02 restringierte, CMV-spezifische An-tikörper identifiziert. Hierzu wurden zunächst HLA-C*07:02 Komplexe synthetisiert, um diese mit dem CMV-Peptid CRVLCCYVL zu beladen.
Durch Inkubation von Fab-Antikörper-präsentierenden Phagen mit diesen MHC-Ⅰ-Komplexen konn-ten mehrere HLA-C*07:02/CMV-Peptid-bindende Antikörperfragmente (Fab) identifiziert werden. Eine weitere Selektion der Phagen erfolgte an biotinylierten HLA-C*07:02/CRVLCCYVL Komple-xen. TG1 E. coli Bakterien wurden mit den selektierten Phagen infiziert und lösliche Fab produziert.
Die Bindungsfähigkeit der Fab wurde an HLA-C*07:02-exprimierenden, lymphoblastoiden Zelllinien (LCL), die mit dem CMV-Peptid beladen wurden, getestet. Die Fab zeigten keine Bindung an mit einem Kontrollpeptid beladene HLA-C*07:02-Komplexe, sowie an andere getestete HLA-C Allele.
Insgesamt konnten drei Fab-Klone mit spezifischer Bindungsfähigkeit gegenüber den HLA-C*07:02/CRVLCCYVL Komplexen identifiziert werden.A Cytomegalovirus (CMV) reactivation in humans after allogeneic stem cell transplantation (HSCT) or solid organ transplantation (SOT) is one of the most common complications. Despite antiviral prophylaxis, CMV reactivation is still associated with significantly increased morbidity and mortality in transplant patients. In healthy individuals, infection is controlled by T-cells, while in immuno-suppressed individuals the T-cell response is no longer adequate.
The T-cell immune response depends on the intracellular processing and presentation of viral pep-tides by MHC-class-Ⅰ-molecules of virus-infected cells.
The MHC-class-Ⅰ-molecules are encoded by the three genes HLA-A, HLA-B and HLA-C. T-cells recognize the MHC/CMV-peptide complex presented by infected cells with their T-cell receptor (TCR). Recent studies have shown that HLA-C*07:02-encoded MHC-Ⅰ receptors are hardly affected by the immune evasion mechanisms of CMV.
TRC-like antibodies, in contrast to conventional antibodies, can bind the MHC/peptide complex similar to the TCR of T-cells and therefore offer a potential therapeutic option for patients with T-cell defects.
Using the phage display method, we identified HLA-C*07:02 restricted CMV-specific antibodies.
For this purpose, we first synthesized HLA-C C*07:02 receptors to load them with the CMV peptide CRVLCCYVL.
By incubating phages with these MHC-Ⅰ/peptide-complexes, several specific Fab-antibody presenting phages were identified. To ensure a binding ability, phages were further selected on biotinylated HLA-C*07:02/CRVLCCYVL complexes. TG1 E. coli bacteria were infected with the selected phages and soluble Fab were produced.
The binding of the Fab was tested on HLA-C*07:02 expressing lymphoblastoid cell lines (LCL) loaded with the CMV peptide. Selected Fab showed no binding to control peptide-loaded HLA-C*07:02 receptors, as well as to LCLs expressing MHC complexes of different HLA-C alleles.
In total, three Fab clones were identified that showed specific binding to the HLA-C*07:02/CRVLCCYVL complex
PSMA PET/CT in biochemical recurrence of prostate cancer with PSA levels ≤ 0.2 ng/mL: a German multicenter analysis of conventional PSMA tracers, including [68Ga]Ga-PSMA-11, [68Ga]Ga-PSMA I&T, and [18F]PSMA-1007
Background
Prostate-specific membrane antigen (PSMA)-directed positron emission tomography/computed tomography (PET/CT) has emerged as a highly accurate imaging modality for detecting tumor lesions in patients with biochemical recurrence (BCR) of prostate cancer (PC). While detection rates of lesions suspicious for PC relapse are known to increase with rising prostate-specific antigen (PSA) levels, data on the efficacy of PSMA PET/CT at very low PSA values (≤ 0.2 ng/mL) remain limited.
Methods
In this multicenter study, we analyzed 321 patients with BCR and a PSA value ≤ 0.2 ng/mL across five German academic centers, using three different PSMA-targeted radiotracers: [68Ga]Ga-PSMA-11, [68Ga]Ga-PSMA I&T, and [18F]PSMA-1007 and analyzed the detection rates and potential predictive parameters.
Results
The overall pooled detection rate was 29.6%. No statistically significant differences in detection rates were observed between the three radiotracers ([68Ga]Ga-PSMA-11 29.4% vs. [68Ga]Ga-PSMA I&T, 22.5% vs. [18F]PSMA-1007 32.4%, p ≥ 0.314). Detection rates were significantly higher in patients with a PSA level > 0.15 ng/mL (p = 0.029, φ = 0.122), in those with an initial Gleason score > 7 (p = 0.018, φ = 0.141) and in those receiving androgen deprivation therapy (p = 0.031, φ = 0.120).
Conclusion
All three radiotracers demonstrated comparable diagnostic performance, with no significant superiority observed between the 68Ga- and 18F-labeled tracers in the patient sample investigated (overall pooled detection rate: 29.6%). This positivity rate can serve as an expectation horizon for both the attending physician and the patient in the case of low PSA values. Further studies with larger cohorts, preferably conducted in a prospective setting, are needed to confirm and expand upon our findings
Investigating Calcium Dependent Regulation of IL-1α Release: Unravelling the Significant Role of Purinergic Signaling with Implications in Chronic Kidney Disease.
Chronic kidney disease (CKD) is characterized by irreversible changes in kidney function persisting for more than three months. Lethal cardiovascular events in CKD patients account for 7.6% of cardiovascular disease-related deaths. Elevated levels of IL-1α have been observed in monocytes of CKD patients. In this study, we aimed to investigate the importance of Ca²⁺ signals in IL-1α biogenesis and release, as well as the channels mediating these signals and their alterations in monocytes from CKD donors. Moreover, IL-1α localization in monocytes and transcriptional alterations associated with CKD were investigated.
To investigate the importance of Ca²⁺ in IL-1α release, concentrations of IL-1α and IL-1β released in the medium upon chelating Ca²⁺ with EGTA or BAPTA-AM were measured using ELISA, showing a significant reduction in both cytokines when intracellular Ca²⁺ was chelated. Inhibition of calpain, a Ca²⁺-dependent protease known to cleave IL-1α, showed a significant reduction in IL-1α in the medium, but had no effect on IL-1β. Moreover, Ca²⁺ signaling through three main Ca²⁺ pathways, including TRPM2, store-operated calcium entry (SOCE), and the purinergic family, were investigated using live-cell Ca²⁺ imaging. Although no significant alterations were observed in expression levels or Ca²⁺ signals via SOCE or TRPM2 in CKD monocytes, purinergic signaling was significantly increased compared to control cells. Expression analysis using qPCR showed higher expression levels of P2Y11, P2X4, and P2X7 in monocytes from CKD patients. Our results showed that the activation of P2X7 is crucial for IL-1α release, and inhibition of P2X7 and to a lesser extent P2X4 reduced IL-1α and IL-1β release from monocytes. Analysis of monocyte surface markers showed alterations of the fraction of CD16⁺ monocytes following priming and treatment with ATP in CKD monocytes.
The current work also studied the effects of several kinases activated downstream of the P2 family on IL-1α release and surface expression. Release of IL-1α but not IL-1ß was significantly inhibited by PKCβ2 inhibition. Moreover, surface expression of IL-1α and the CD16 marker were increased upon NFκB inhibition. RNA sequencing results identified 143 up-regulated and 79 downregulated genes in CKD monocytes. Among these, MMP25 and MMP9 were significantly up-regulated. Incubating LPS-primed cells with an MMP pan-inhibitor led to a significant decrease in IL-1α release while an increased surface expression of IL-1α was observed only if the inhibitor was included during priming with LPS.

To explore intracellular localization, IL-1α was overexpressed with a GFP tag at the N-terminus and mCherry at the C-terminus in HEK293 cell line or primary monocytes. We observed nuclear localization of IL-1α in mature monocytes with a bean-shaped nucleus and cytoplasmic localization in immature monocytes with a round nucleus.
In conclusion, our findings highlight the pivotal role of P2X4, P2X7, PKCβ2, and MMPs in IL-1α release and surface expression. Additionally, we observed a strong correlation between CD16 and IL-1α surface expression.Zusammenfassung: Die chronische Nierenerkrankung (CKD) ist durch irreversible Veränderungen der Nierenfunktion gekennzeichnet, die länger als drei Monate andauern. Tödliche kardiovaskuläre Ereignisse bei CKD-Patienten machen 7.6 % der an Herz-Kreislauf-Erkrankungen bedingten Todesfälle aus. Bei den Monozyten von CKD-Patienten wurden erhöhte IL‑1α Spiegel beobachtet. In dieser Studie wollten wir die Bedeutung von Ca²⁺ Signalen für die Biogenese und Freisetzung von IL‑1α untersuchen, ebenso wie die Kanäle, die diese Signale vermitteln, und deren Veränderungen in Monozyten von CKD Spendern. Darüber hinaus wurden die Lokalisation von IL‑1α in Monozyten und die mit CKD assoziierten transkriptionellen Veränderungen analysiert.
Um die Bedeutung von Ca²⁺ bei der Freisetzung von IL‑1α zu untersuchen, wurden mittels ELISA die Konzentrationen von IL‑1α und IL‑1β gemessen, die nach der Chelatierung von Ca²⁺ mit EGTA oder BAPTA‑AM in das Medium freigesetzt wurden. Dabei zeigte sich, dass beide Zytokine signifikant reduziert wurden, wenn das intrazelluläre Ca²⁺ gecheliert wurde. Die Hemmung von Calpain, einer Ca²⁺ abhängigen Protease, die dafür bekannt ist, IL‑1α zu spalten, führte zu einer signifikanten Verringerung von IL‑1α im Medium, während IL‑1β unbeeinflusst blieb. Außerdem wurde die Ca²⁺ Signalübertragung über drei Hauptwege: TRPM2, store-operated calcium entry (SOCE) und die purinerge Familie, mittels Live-Cell-Ca²⁺-Imaging untersucht. Obwohl in CKD Monozyten weder signifikante Veränderungen der Expressionsniveaus noch der Ca²⁺ Signale über SOCE oder TRPM2 festgestellt wurden, war die purinerge Signalübertragung im Vergleich zu Kontrollzellen signifikant erhöht. Eine Expressionsanalyse mittels qPCR ergab zudem höhere Expressionsniveaus von P2Y11, P2X4 und P2X7 in den Monozyten von CKD-Patienten. Unsere Ergebnisse zeigten, dass die Aktivierung von P2X7 entscheidend für die Freisetzung von IL‑1α ist und dass die Hemmung von P2X7 – und in geringerem Maße von P2X4 , die Freisetzung von IL‑1α und IL‑1β aus den Monozyten reduziert. Die Analyse der Oberflächenmarker der Monozyten ergab Veränderungen im Anteil der CD16⁺ Monozyten nach dem Priming und der Behandlung mit ATP in CKD-Monozyten.
Die vorliegende Arbeit untersuchte zudem die Effekte mehrerer Kinasen, die nachgeschaltet der P2 Familie aktiviert werden, auf die Freisetzung und Oberflächenexpression von IL‑1α. So wurde die Freisetzung von IL‑1α, nicht aber die von IL‑1β, durch die Hemmung von PKCβ2 signifikant reduziert. Darüber hinaus führte die Hemmung von NFκB zu einer erhöhten Oberflächenexpression von IL‑1α und des CD16‑Markers. RNA Sequenzierungsergebnisse

identifizierten 143 hochregulierte und 79 herunterregulierte Gene in CKD Monozyten. Unter diesen waren MMP25 und MMP9 signifikant hochreguliert. Die Inkubation von LPS geprimten Zellen mit einem MMP‑Pan‑Inhibitor führte zu einer signifikanten Abnahme der IL‑1α Freisetzung, während eine erhöhte Oberflächenexpression von IL‑1α nur beobachtet wurde, wenn der Inhibitor bereits während des Primings mit LPS hinzugefügt wurde.
Um die intrazelluläre Lokalisation zu untersuchen, wurde IL‑1α in der HEK293‑Zelllinie oder in primären Monozyten überexprimiert, wobei ein GFP Tag am N‑Terminus und mCherry am C‑Terminus angefügt wurde. Dabei beobachteten wir in reifen Monozyten mit bohnenförmigem Zellkern eine nukleäre Lokalisation von IL‑1α, während in unreifen Monozyten mit rundem Zellkern eine zytoplasmatische Lokalisation vorherrschte.
Zusammenfassend heben unsere Ergebnisse die zentrale Rolle von P2X4, P2X7, PKCβ2 und MMPs bei der Freisetzung und Oberflächenexpression von IL‑1α hervor. Zudem wurde eine starke Korrelation zwischen der Oberflächenexpression von CD16 und IL‑1α festgestellt
Relating space and time in cryptography
This thesis studies the interplay between space and time in cryptography. It explores trade-offs between these two resources for honest parties and shows how fine-grained constraints on an adversary's capabilities can unlock new cryptographic functionalities. Adopting this perspective, we present results in four subfields of cryptography: 2PC: We prove tight lower bounds on the cost of oblivious transfer protocols, a central primitive for two-party computation, revealing a fundamental trade-off between communication and public-key operations. SNARG: We construct designated-verifier SNARGs with very short proofs, highlighting a novel trade-off between proof size and verifier efficiency. Incompressible Encryption: We construct a new incompressible encryption scheme. Incompressible encryption remains secure even after key exposure, assuming adversaries had limited space when the ciphertext was transmitted. This offers a lightweight alternative to forward secrecy for long messages. Space-Hard Functions: We define and construct verifiable space-hard functions and space-lock puzzles, space-based analogues of verifiable delay functions and time-lock puzzles. These enable new applications such as deniable proofs and leverage space limitations of the adversary. Together, these results demonstrate how a careful study of space-time trade-offs yields both foundational insights and practical cryptographic tools.Diese Arbeit untersucht das Zusammenspiel von Raum und Zeit in der Kryptographie. Sie analysiert Kompromisse zwischen diesen Ressourcen für ehrliche Parteien und zeigt, wie gezielte Beschränkungen der Fähigkeiten eines Angreifers neue Möglichkeiten eröffnen. Aus dieser Sicht liefern wir Beiträge in vier Bereichen: 2PC: Wir beweisen untere Schranken für die Kosten von Oblivious-Transfer-Protokollen, einem zentralen Primitive für Zwei-Parteien-Berechnung, und zeigen einen grundlegenden Trade-off zwischen Kommunikation und Public-Key-Operationen. SNARG: Wir konstruieren Designated-Verifier-SNARGs mit sehr kurzen Beweisen und demonstrieren einen neuen Trade-off zwischen Beweisgröße und Verifizierer-Effizienz. Inkomprimierbare Verschlüsselung: Wir stellen ein neues Schema vor, das auch nach Schlüsselkompromittierung sicher bleibt, solange der Angreifer beim Empfang nur begrenzten Raum hatte. Es bietet eine leichte Alternative zur Vorwärtsgeheimhaltung bei langen Nachrichten. Raum-Harte Funktionen: Wir definieren und konstruieren die ersten verifizierbaren raum-harte Funktionen und Raum-Lock-Rätsel, raumbasierte Gegenstücke zu Time-Lock-Rätseln. Diese ermöglichen neue Anwendungen wie ableugbare Beweise und nutzen Raumbeschränkung des Angreifers. Diese Resultate zeigen, dass eine gezielte Analyse Kompromissen zwischen Raum und Zeit sowohl theoretische Einsichten als auch praxisnahe Werkzeuge liefert
Parental Adverse Childhood Experiences (ACEs) in an Early Childhood Mental Health Outpatient Clinic in Germany: Prevalence and Associations with Child Psychiatric Diagnoses
Parental adverse childhood experiences (ACEs) are linked to negative outcomes in children, including emotional and behavioral problems, developmental delays, and higher risk for psychopathology. Most research focuses on school-aged children or community samples, with few studies examining preschool-aged children in child psychiatric care. Understanding parental ACEs in this population is crucial, as early childhood is a sensitive developmental period, and intergenerational effects may be particularly pronounced in children already presenting with psychiatric symptoms. Background/Objectives: The goal of this study was to analyze how parents of patients in an early childhood (0–5.9 yrs) mental health outpatient clinic differ from the general population in terms of the frequency of ACEs. In addition, we investigated the connection between mental health disorders in young children and the specific ACE scores of their parents. Methods: A total of 116 caregivers (34.45 years (SD = 5.28)) and their children (71.6% boys, 28.4% girls) at an average age of 3.99 years (SD = 1.35, range = 0.31–5.95) were included in the analysis. The legal guardians completed the 10-item ACE questionnaire. The young children were diagnosed as part of outpatient treatment using the DC:0–5 classification system. We analyzed the ACE scores and diagnoses descriptively and in comparison to a community sample. Results: An average value of 2.38 parental ACEs was reported by our sample, and 68.1% (n = 79) reported at least one ACE. The high-risk group with four or more ACEs comprised 30.2% (n = 35). The most common diagnosis in young children was the Disorder of Dysregulated Anger and Aggression of Early Childhood, followed by global developmental delay. Adjustment disorder was third in terms of frequency. Among the examined child psychiatric diagnoses, adjustment disorder showed a significant correlation with parents being affected by the ACE category of neglect (OR = 2.54; 95% CI: 1.012–6.369; p = 0.047). Conclusions: Parents who presented their children at an early childhood mental health outpatient clinic reported significantly more ACEs as compared to representative data on ACEs in adulthood. These results highlight the need for further studies with larger samples to enable a more in-depth analysis of the general intergenerational transmission processes and the differential transmission of specific ACEs to specific diagnoses in preschool-aged children
Understanding and supporting software model evolution through edit operation mining and AI-based software model completion
Model-based Systems Engineering has become increasingly important in managing the complexity of modern software systems. However, the evolution of software models in large-scale, real-world projects remains a significant challenge due to the lack of effective and automated methods. This thesis addresses this problem by providing a comprehensive theoretical foundation for software model evolution, and, based on this theory, presents practical approaches to understand and support model evolution, which will be evaluated and validated by empirical studies. The first part of the thesis develops a theory to software model evolution and introduces novel techniques---backing the theory---based on graph mining, large language models, and graph neural networks. These methods allow to automatically define model transformations, and support software model completion, solely based on model histories from model version control systems. The second part motivates and shows the relevance and usefulness of the research behind the thesis by providing insights into the complexity of a real-world industrial model-driven product line at our industry partner. In the third part, controlled experiments provide evidence for the theory developed in the first part of the thesis. The findings demonstrate the feasibility and that there is a potential for data-driven intelligent modeling assistants to support model-based engineering---not only boosting productivity of engineers, but also accuracy and overall quality of software models. This research contributes to advancing Model-based Systems Engineering by proposing automated solutions that support the continuous evolution of software models, ultimately leading to more robust and maintainable systems.Modellbasiertes Systems Engineering hat sich bei der Bewältigung der Komplexität moderner Softwaresysteme als zunehmend wichtig erwiesen. Die Evolution von Softwaremodellen in großen, realen Projekten bleibt jedoch aufgrund des Mangels an effektiven und automatisierten Methoden eine erhebliche Herausforderung. Diese Arbeit befasst sich mit diesem Problem, indem sie eine umfassende theoretische Grundlage für die Evolution von Softwaremodellen bereitstellt und darauf aufbauend praktische Ansätze zum Verständnis und zur Unterstützung der Modellevolution vorstellt, die durch empirische Studien evaluiert und validiert werden. Der erste Teil der Arbeit entwickelt eine Theorie zur Evolution von Softwaremodellen und führt neuartige Techniken—zur Untermauerung der Theorie -– ein, die auf Graph Mining, großen Sprachmodellen und Graph Neural Networks basieren. Diese Methoden ermöglichen es, Modelltransformationen automatisch zu definieren und die Vervollständigung von Softwaremodellen zu unterstützen, ausschließlich basierend auf Modellhistorien aus Modellversionskontrollsystemen. Der zweite Teil motiviert und zeigt die Relevanz und Nützlichkeit der Forschung hinter dieser Arbeit, indem er Einblicke in die Komplexität einer realen, modellgetriebenen Produktlinie bei unserem Industriepartner gibt. Im dritten Teil liefern kontrollierte Experimente Belege für die im ersten Teil der Arbeit entwickelte Theorie. Die Ergebnisse demonstrieren die Machbarkeit und das Potenzial datengestützter intelligenter Modellierungsassistenten zur Unterstützung des modellbasierten Engineerings – nicht nur zur Steigerung der Produktivität der Ingenieure, sondern auch der Genauigkeit und der Gesamtqualität von Softwaremodellen. Diese Forschung trägt dazu bei, das modellbasierte Systems Engineering voranzutreiben, indem sie automatisierte Lösungen vorschlägt, die die kontinuierliche Evolution von Softwaremodellen unterstützen und letztendlich zu robusteren und wartbareren Systemen führen
Risikofaktoren für das Auftreten von Rezidiven nach endovaskulärer Behandlung intrakranieller Aneurysmen
Intrakranielle Aneurysmen treten bei etwa zwei bis fünf Prozent der erwachsenen Bevölkerung auf. Nach der Diagnosestellung steht die Entscheidung im Vordergrund, ob und mit welcher Methode eine Behandlung erfolgen soll. Neben der chirurgischen Methode des Clippings hat sich in den letzten Jahren die endovaskuläre Methode des Coilings als etablierte Alternative entwickelt. Trotz intensiver Forschung besteht weiterhin Uneinigkeit darüber, welche der beiden Verfahren langfristig überlegen ist. Eine zentrale Rolle spielt dabei das Risiko eines Rezidivs nach erfolgter Behandlung.
Ziel dieser Arbeit war es, Risikofaktoren zu identifizieren, die ein Rezidiv nach endovaskulärer Behandlung intrakranieller Aneurysmen begünstigen. Hierzu wurden retrospektiv Merkmale von 100 Aneurysmen erfasst und statistisch ausgewertet. Bei 29 Aneurysmen trat ein Rezidiv auf, von denen 24 erneut behandelt wurden. Rezidivierte Aneurysmen zeigten größere Dom-, Hals- und Höhenmaße sowie eine niedrigere Packdichte, eine höhere Coilanzahl und häufiger eine unvollständige Okklusion. Eine vorbestehende Subarachnoidalblutung oder atherosklerotische Gefäßveränderungen erhöhten ebenfalls das Rezidivrisiko. Keine signifikanten Zusammenhänge ergaben sich für Stentverwendung, Schlaganfall, weitere Aneurysmen oder die anatomische Lage.
Die Ergebnisse ermöglichen eine genauere Einschätzung des Rezidivrisikos und können helfen, die individuelle Behandlungsentscheidung bei intrakraniellen Aneurysmen zu verbessern.Intracranial aneurysms can be detected in approximately two to five percent of the adult population. Once an aneurysm has been diagnosed, the treating physician and patient must decide whether treatment is necessary and, if so, which therapeutic approach should be chosen. In addition to the surgical method of clipping, the endovascular technique of coiling has become an established alternative in recent years. Despite extensive research, it remains a matter of debate which method provides the better long-term results. The risk of recurrence after treatment plays a central role in this context.
The aim of this study was to identify risk factors associated with recurrence after endovascular treatment of intracranial aneurysms. For this purpose, the characteristics of 100 aneurysms were retrospectively analyzed. A recurrence occurred in 29 aneurysms, of which 24 required retreatment. Recurrent aneurysms showed larger dome, height, and neck dimensions, lower packing density, a higher number of coils used, and more frequent incomplete occlusion. Patients with subarachnoid hemorrhage or atherosclerotic vessel changes also had a higher risk of recurrence. No significant associations were found for stent use, stroke, additional aneurysms, or the anatomical location of the lesion.
These findings contribute to a better understanding of the recurrence risk after endovascular treatment of intracranial aneurysms and may support improved individualized treatment decisions in the future
Nomogram for Deep Vein Thrombosis Prediction Post-Endovascular Thrombectomy in Acute Ischemic Stroke: A Retrospective Multicenter Observational Study
Background
Deep vein thrombosis (DVT) is a frequent complication following endovascular thrombectomy (EVT) in patients with acute ischaemic stroke (AIS), potentially leading to fatal pulmonary embolism (PE). Identifying patients early at high risk for DVT is clinically important. This study developed and validated a nomogram combining laboratory findings and clinical characteristics to predict the risk of lower-extremity DVT after EVT in patients with AIS.
Methods
This retrospective multicentre observational study was conducted in two tertiary hospitals in China, enrolling 640 patients who underwent ultrasonography for DVT diagnosis within 10 days following EVT. Data on medical history, examination and laboratory results were collected for logistic regression analyses to develop a DVT risk nomogram.
Results
Logistic regression analyses identified critical predictors of DVT: lower limb National Institutes of Health Stroke Scale (NIHSS) score ≥ 2, elevated D-dimer levels (≥ 1.62 mg/L) and prolonged puncture-to-recanalization time (PRT ≥ 66 min). The nomogram demonstrated good discriminative ability (AUC 0.741–0.822) and clinical utility across internal and external validation cohorts. Additionally, the presence of DVT was significantly associated with reduced functional independence at 90 days post-EVT, highlighting the negative impact of DVT on patient recovery (OR = 3.85; 95% CI: 2.18–6.78; p < 0.001).
Conclusion
The study provides a practical clinical tool for early detection and intervention in patients with AIS at high risk for DVT following EVT. Early identification and intervention may help improve outcomes in patients with AIS undergoing EVT.
Relevance to Clinical Practice
This nomogram helps in the early detection and proactive management of DVT in AIS patients, which can reduce severe complications and improve patient recovery outcomes.

From Native Glycosaminoglycans to Mimetics: Design, Mechanisms, and Biomedical Applications
Glycosaminoglycans (GAGs) are essential regulators of numerous biological processes
through their interactions with growth factors, chemokines, cytokines, and enzymes. Their
structural diversity and heterogeneity, however, limit reproducibility and translational
use, as native GAGs are typically obtained from animal-derived sources with notable
batch-to-batch variability. To overcome these challenges, a wide range of GAG mimetics
has been developed with the aim of replicating or modulating the biological functions
of native GAGs while offering improved structural definition, accessibility, and thera peutic potential. Polysaccharide-based GAG mimetics, including derivatives of heparan
sulfate, hyaluronan, dextran, and other natural glycans, represent one major strategy,
whereas non-saccharide-based mimetics provide alternative scaffolds with enhanced stabil ity and selectivity. Both approaches have yielded compounds that serve as valuable tools
for dissecting GAG/protein interactions and as candidates for therapeutic development.
Biomedical applications of GAG mimetics span diverse areas such as cancer, cardiovascular
and inflammatory diseases, bone and cartilage regeneration, wound healing, and infectious
diseases. This mini-review summarizes key developments in the design and synthesis of
GAG mimetics, highlights their potential biomedical applications, and discusses current
challenges and future perspectives in advancing them toward clinical translation
