Cannabidiol and Hydroxypropyl-β-Cyclodextrin for the Development of Deflated Spherical-Shaped Inhalable Powder.

peer reviewedIn addition to the known therapeutic indications for cannabidiol, its administration by inhalation appears to be of great interest. Indeed, there is evidence of cannabidiol's efficacy in several physiological pathways, suggesting its potential for a wide range of applications for both local and systemic pulmonary administration like cancers. Significant advances in pulmonary drug delivery have led to innovative strategies to address the challenges of increasing the respirable fraction of drugs and standardizing inhalable products. Among different devices, dry powder inhalers offer significant advantages including high stability and ease of use. Particle engineering using techniques such as spray drying is now the focus of research and is expected to improve upon, rather than completely replace, traditional carrier-based formulations. The development of carrier-free powders (without lactose-carrier) is mainly used for medicines with low active ingredient doses, which limits the technology. Previously, we demonstrated the benefits of using a cyclodextrin to obtain deflated spherical-shaped powders by spray drying. In this study the potential of this excipient with a very poorly water-soluble active molecule was investigated. Inhalable cannabidiol powders were developed by spray drying, using the solubility enhancers hydroxypropyl-beta-cyclodextrin and ethanol to optimize cannabidiol water-solubility. Electron microscopy images revealed consistent deflated spherical shapes, while particle size analysis showed low polydispersity and suitable sizes for deep lung deposition (2 µm). The selected engineered powders (without ethanol) had very high fine particle fractions (> 60%) due to their deflated surface. Finally, the powder was instantly solubilized leading to drug dissolution, which is important for therapeutic efficacy. In conclusion, this study successfully develops a cannabidiol inhalation powder by particle engineering having suitable aerosolization behavior. Due to the speed of the process and the performance of the finished product, this work opens the door for future studies. It has been shown that active molecules that are only slightly soluble in water can be formulated effectively as a powder for inhalation. Other molecules could be tested and subsequent in vivo studies conducted to demonstrate correlation with these in vitro results

Hydroelectric power plant and upstream fish migration: evaluation of the efficiency of a behavioural barrier

peer reviewedHydropower plant (HPP) weirs disrupt river ecosystems by fragmenting habitats. Moreover, the strong water currents from HPP discharges can mislead fish, causing them to enter nonviable or dangerous output channels. This study investigates a submerged behavioural barrier installed downstream of an HPP in the Our River in Belgium. The barrier’s goal is to repel fish from the turbine output channel and guide them towards a safer fishway passage, thereby aiding their upstream migration. We used radio telemetry to track the movements of brown trout (Salmo trutta L.) and evaluate the barrier’s effectiveness in preventing misdirected migration. The results showed significant attraction for the output channel but 50% of the fish were successfully deterred by the behavioural barrier where the success was clearly influenced by changes in water flow conditions. The fishway had a moderate success rate, with 58% of fish using it effectively. Fish that passed through the barrier were less likely touse the fishway. These results highlight the interest of equipping certain sites with behaviouralbarriers during upstream migration and the need to improve their design to promotea balance between hydropower production and the conservation of aquatic biodiversity.14. Politiques de la littérature. Fortunes et infortunes d’une formule critique

    No full text

Black coral patches and reef fish diversity: An integrative multidisciplinary approach

    No full text
peer reviewedThe black coral Antipathes galapagensis is a habitat engineer that may play critical roles for marine biodiversity but is still largely underexplored because typically found in the mesophotic zone. However, A. galapagensis occasionally colonizes shallow waters providing an excellent opportunity to assess its ecological role. Here, we compared the reef fish assemblages between rocky reefs with and without black coral in the Bay of La Paz (Mexico). To achieve a comprehensive understanding of the diversity in each habitat, we adopted an integrative approach that combined different methodologies tailored for specific groups, i.e., Underwater Visual Census for conspicuous, Passive Acoustic Monitoring for nocturnal soniferous, and environmental DNA for cryptobenthic species. Black coral habitats were found in isolated patches rather than extensive forests and demersal fish assemblages were largely similar between the habitats, except for Lutjanidae, with higher densities in the black coral habitat. However, small benthic fish species (5–20 cm) showed higher species richness, density, and distinct compositions in black coral. Historical fish surveys in reefs of the region confirmed these findings. Acoustic analyses revealed differences between the sites, although no relationship with black coral was detected. Although environmental DNA supported the role of black coral for Lutjanidae, it failed to capture the diversity of cryptobenthic fish, pointing to methodological challenges discussed in our study. Overall, our findings underscore the ecological significance of black coral for reef fishes, even in isolated patches, and highlight the need for further extensive studies on this underexplored and imperiled habitat.14. Ultrasound- and clinical-defined disease activities are associated with altered bone microarchitecture and lower bone mineral density in patients with rheumatoid arthritis.

peer reviewed[en] BACKGROUND/AIMS: We investigate if rheumatoid arthritis (RA) activity, defined clinically and with ultrasound, is associated with bone macro- and micro-architecture impairment on high-resolution peripheral quantitative computed tomography (HR-pQCT) and dual X-ray densitometry (DEXA). METHODS: Disease activity was evaluated in 61 RA patients, with clinical indices and ultrasound (hands and wrists). Bone mineral density (BMD) and architecture were analyzed with HR-pQCT and DEXA. RESULTS: Ultrasound RA disease activity parameters [synovitis, power doppler (PD)-positive joints, sum of positive power doppler signals and tenosynovitis] were associated with altered HR-pQCT bone density and structure at tibia or radius (trabecular volumetric BMD, trabecular bone volume fraction, trabecular thickness and cortical porosity). In addition, wrist ultrasound activity was specifically locally associated with impaired local bone microarchitecture at distal ipsilateral radius. Clinical and functional RA disease activity parameters (number of swollen joints, Health Assessment Questionnaire and disease activity score DAS28-CRP) were also correlated with HR-pQCT parameters (total and trabecular volumetric BMD, trabecular thickness and cortical thickness). At the hip, BMD correlated with VAS-fatigue and DAS28-ESR. The number of synovitis detected by ultrasound was higher when total hip T-score was lower than - 1. DISCUSSION: Ultrasound and clinical disease activity parameters were associated with impaired HR-pQCT parameters (distal radius and tibia), with lower trabecular and cortical bone densities and impaired bone microarchitecture (organization of spans and cortical porosity). In addition to systemic contribution to bone impairment, a local correlation between wrist US activity and HR-pQCT at distal radius was observed. CONCLUSION: Patients with active RA, especially with US evaluation, are at higher risk for altered bone density and structure
