Osteogenic CpG Oligodeoxynucleotide, iSN40, Inhibits Osteoclastogenesis in a TLR9-Dependent Manner
A CpG oligodeoxynucleotide (CpG-ODN), iSN40, was originally identified as promoting the mineralization and differentiation of osteoblasts, independent of Toll-like receptor 9 (TLR9). Since CpG ODNs are often recognized by TLR9 and inhibit osteoclastogenesis, this study investigated the TLR9 dependence and anti-osteoclastogenic effect of iSN40 to validate its potential as an osteoporosis drug. The murine monocyte/macrophage cell line RAW264.7 was treated with the receptor activator of nuclear factor-κB ligand (RANKL) to induce osteoclast differentiation, then the effect of iSN40 on was quantified by tartrate-resistant acid phosphatase (TRAP) staining and real-time RT-PCR. iSN40 completely inhibited RANKL-induced differentiation into TRAP+ multinucleated osteoclasts by suppressing osteoclastogenic genes and inducing anti-/non-osteoclastogenic genes. Treatment with a TLR9 inhibitor, E6446, or a mutation in the CpG motif of iSN40 abolished the intracellular uptake and anti-osteoclastogenic effect of iSN40. These results demonstrate that iSN40 is subcellularly internalized and is recognized by TLR9 via its CpG motif, modulates RANKL-dependent osteoclastogenic gene expression, and ultimately inhibits osteoclastogenesis. Finally, iSN40 was confirmed to inhibit the osteoclastogenesis of RAW264.7 cells cocultured with the murine osteoblast cell line MC3T3-E1, presenting a model of bone remodeling. This study demonstrates that iSN40, which exerts both pro-osteogenic and anti-osteoclastogenic effects, may be a promising nucleic acid drug for osteoporosis.ArticleLife 2024, 14(12), 1572journal articl
Reductions of Parvalbumin Positive Interneurons and Adult Hippocampal Neurogenesis were Observed in the Genetically Mimicked Mouse Models for IQSEC2-related Disorders
Background : IQSEC2-related disorders are a genetic syndrome characterized by intellectual disability and various neurodevelopmental disorders. We previously generated Iqsec2 knockout mice and conducted electrophysiological and behavioral assays to study their phenotypic manifestations. However, histological features observed in other mouse models of neurodevelopmental disorders have never been examined. We focused on the adult neurogenesis and interneuropathy as features of neurodevelopmental disorders and investigated them with the Iqsec2 knockout mouse. Methods : Four-month-old Iqsec2 knockout male mice were injected with bromodeoxyuridine (BrdU) to label newly born hippocampal neurons. We estimated the number of the adult-born neurons in the hippocampus by immunohistochemistry with antibodies against BrdU and NeuN, a neuronal marker. We also quantified parvalbuminpositive neurons, a dominant subtype of GABAergic interneurons by immunohistochemistry. Results : We observed that the number of parvalbumin interneurons decreased in the medial prefrontal cortex and the dentate gyrus of the ventral part of the hippocampus. The number of the BrdU positive neurons in the dentate gyrus of the hippocampus decreased in Iqsec2 knockout mice. The reduction of BrdU positive neurons was observed both in the dorsal and ventral parts of the hippocampus. Conclusion : A decrease in parvalbumin-positive neurons, occurred in the medial prefrontal cortex and the hippocampus, which indicates that a mechanism of IQSEC2-related disorders may involve a deficit of interneurons. Considering that adult neurogenesis seems important for cognitive brain functions, a reduction of adult-born neurons in the hippocampus may be related to some of the phenotypes of IQSEC2-related disorders, such as intellectual disability.信州医学雑誌 72(6) : 385-396, (2024)journal articl

Recent trends and concerns on age-specific folate intake among women in Nagano prefecture and in Japan
目的:女性における年齢階層別葉酸摂取量の最近の動向を示し、若い女性の葉酸欠乏のリスクについて述べる。また、葉酸過剰摂取に伴う懸念について、文献的に考察する。方法:全国女性の年齢階層別葉酸摂取量および食品摂取量の年次推移は、厚生労働省の発表データ、長野県女性の葉酸摂取量は、長野県発表データを用いた。結果:2001 年から2019 年における全国女性の年齢階層別葉酸摂取量は、全年齢層で減少しており、20歳代から40 歳代ではそれ以上の年代に比べて摂取量が少なく、20 歳代では2009 年以降その平均値が、推奨量240 μg/ 日を下回るようになった。長野県の2019 年における18-29 歳女性の葉酸摂取量中央値は、204 μg/ 日と推定平均必要量の200 μg/ 日に近似していた。全国女性の野菜類、果実類、藻類、豆類の摂取量の年次推移では、全年齢層で豆類以外減少傾向であった。考察:若い女性の葉酸摂取量の減少は明らかであり、20 歳代では半数近くが推定平均必要量を満たさない可能性が示唆された。葉酸過剰摂取の懸念も指摘されており、若い世代の女性に対し、葉酸摂取に焦点を当てた食育が必要と考える。Objective: This study aims to exhibit recent trends in folate intake among women across different age groups and discuss the risk of folate deficiency in young women. Additionally, concerns related to excessive folate intake are explored through a literature review. Methods: National trends in age-specific folate intake and food consumption were analyzed using data from the Ministry of Health, Labour and Welfare, while folate intake among women in Nagano Prefecture was assessed using data from the prefectural office. Results: The age-specific folate intake among women nationwide from 2001 to 2019 exhibited a decreasing trend across all age groups. In the 20s to 40s age range, the intake was notably lower compared to older age groups, and in the 20s, the average intake has fallen below the recommended dietary allowance level of 240 μg/day since 2009. Among women aged 18 to 29 in Nagano Prefecture, the median folate intake in 2019 was 204 μg/day, approximating the estimated average requirement level of 200 μg/day. The annual trends in vegetable, fruit, seaweed, and legume consumption among women of all age groups, except for legumes, showed a decreasing pattern. Discussion: The noticeable decline in folate intake among young women, with nearly half of those in their 20s potentially not meeting the estimated average requirement, raises significant concerns. While caution may also be necessary for excessive folate intake, dietary education focusing on folate intake would be required for the younger generation of women.journal articl
Temporal Phosphorus Dynamics in Shallow Eutrophic Lake Suwa, Japan
It can be difficult to decrease the water phosphorus (P) concentration in eutrophic shallow lakes, even if the external P loading is reduced, owing to a high level of internal P loading to surface water from sediment. However, in shallow Lake Suwa, Japan, lake water P concentration has largely decreased in recent years owing to low levels of internal P loading, as well as declining external P loading. We measured water/sediment P and iron (Fe) concentrations and the P release rate from sediment in Lake Suwa, and then compared it with data from the 1970s. In the 1970s, the P concentration throughout the lake water was high during the hypoxic period. Recently, however, the P concentration has increased only in the hypolimnion during the hypoxic period. This suggests that internal P loading from sediment to surface water has largely been suppressed during the hypoxic period in recent years. This may be due to (i) stronger water stratification from global warming, (ii) a greater decrease in the P release rate from the sediment owing to a decline in sediment P concentration from the 1970s to 2020, and (iii) stronger formation of the Fe–P cycle in Lake Suwa recently, compared with that in the 1970s.
