Myogenic Anti-Nucleolin Aptamer iSN04 Inhibits Proliferation and Promotes Differentiation of Vascular Smooth Muscle Cells
De-differentiation and subsequent increased proliferation and inflammation of vascular smooth muscle cells (VSMCs) is one of the mechanisms of atherogenesis. Maintaining VSMCs in a contractile differentiated state is therefore a promising therapeutic strategy for atherosclerosis. We have reported the 18-base myogenetic oligodeoxynucleotide, iSN04, which serves as an anti-nucleolin aptamer and promotes skeletal and myocardial differentiation. The present study investigated the effect of iSN04 on VSMCs because nucleolin has been reported to contribute to VSMC de-differentiation under pathophysiological conditions. Nucleolin is localized in the nucleoplasm and nucleoli of both rat and human VSMCs. iSN04 without a carrier was spontaneously incorporated into VSMCs, indicating that iSN04 would serve as an anti-nucleolin aptamer. iSN04 treatment decreased the ratio of 5-ethynyl-2′-deoxyuridine (EdU)-positive proliferating VSMCs and increased the expression of α-smooth muscle actin, a contractile marker of VSMCs. iSN04 also suppressed angiogenesis of mouse aortic rings ex vivo, which is a model of pathological angiogenesis involved in plaque formation, growth, and rupture. These results demonstrate that antagonizing nucleolin with iSN04 preserves VSMC differentiation, providing a nucleic acid drug candidate for the treatment of vascular disease.ArticleBiomolecules 2024, 14(6), 709journal articl

Staggered structural dynamic-mediated selective adsorption of H2O/D2O on flexible graphene oxide nanosheets
Graphene oxide (GO) is the one of the most promising family of materials as atomically thin membranes for water-related molecular separation technologies due to its amphipathic nature and layered structure. Here, we show important aspects of GO on water adsorption from molecular dynamics (MD) simulations, in-situ X-ray diffraction (XRD) measurements, and ex-situ nuclear magnetic resonance (NMR) measurements. Although the MD simulations for GO and the reduced GO models revealed that the flexibility of the interlayer spacing could be attributed to the oxygen-functional groups of GO, the ultra-large GO model cannot well explain the observed swelling of GO from XRD experiments. Our MD simulations propose a realistic GO interlayer structure constructed by staggered stacking of flexible GO sheets, which can explain very well the swelling nature upon water adsorption. The transmission electron microscopic (TEM) observation also supports the non-regular staggered stacking structure of GO. Furthermore, we demonstrate the existence of the two distinct types of adsorbed water molecules in the staggered stacking: water bonded with hydrophilic functional groups and “free” mobile water. Finally, we show that the staggered stacking of GO plays a crucial role in H/D isotopic recognition in water adsorption, as well as the high mobility of water molecules.journal articl
Factors Associated With Health-Related Quality of Life Among Community-Dwelling Older Adults Without Social Participation in Japan: A Cross-Sectional Study
This study aimed to identify the characteristics of community-dwelling older adults who exhibited high health-related quality of life (HRQOL) without social participation. This cross-sectional observational study collected data using a mailed questionnaire. This study included 1,183 community-dwelling older adults aged ≥65 years who lived in Japan. HRQOL was assessed via the EuroQol 5-Dimensions 5-Levels (EQ-5D-5L). Multivariate analysis was performed; full health status (EQ-5D-5L = 1) was used as the objective variable, and socio-demographic, lifestyle, and health-related variables were used as the explanatory variables. Driving (OR = 3.846), economic situation (1.938), purpose in life (2.211), primary illness (0.398), low physical function (0.051), and depressive symptoms (0.288) were associated with higher HRQOL among community-dwelling older adults without social participation. This exploratory study suggests the possibility of promoting and improving the HRQOL of community-dwelling older adults without social participation by strengthening specific factors among them.journal articl
