    Microstructure and mechanical behavior of cross-linked biopolymer networks

    Fiber-achtige netwerken zijn alom tegenwoordig. Vilt en papier zijn voorbeelden waar we de netwerk structuur eenvoudig herkennen; rubber is een netwerk op moleculaire schaal, maar in ons eigen lichaam vinden we biopolymere netwerken in zacht weefsel alsook in het cytoskelet binnenin cellen. Het mechanisch gedrag van al deze materialen hangt uiteraard af van de eigenschappen van de fibers of filamenten in het netwerk maar ook van de microstructuur van het netwerk, vooral bij grote vervormingen. Van een elastiekje weet de lezer dat het steeds moeilijker wordt om het verder te vervormen; in-vitro experimenten hebben laten zien dat sommige biofysische netwerken een nog sterkere mate van versteviging vertonen. De versteviging van verknoopte biopolymere netwerken wordt niet alleen bepaald door de eigenschappen van de filamenten en van de moleculen die de filamenten met elkaar verknopen tot een netwerk (de zgn. cross links), maar ook door de microstructuur van het netwerk. Ondanks veel theoretisch en experimenteel onderzoek in het recente verleden, bevatte het verstevigingsgedrag van biopolymere netwerken nog vele geheimen. De kernboodschap van dit proefschrift is dat deze geheimen vooral verstopt zaten in de microstructuur. Uitgebreide computersimulaties hebben een diep inzicht verschaft in de relatie tussen de microstructuur en de mechanische eigenschappen van biofysische netwerken. Hiermee is het verstevigingsgedrag van een groot scala aan biofysische netwerkmaterialen geünificeerd en kan nu met een betrekkelijk eenvoudig model worden verklaard. A random fibre network is an ubiquitous microstructure in many materials, ranging from metallic open cell foams through felts, paper and rubbers to the scaffolds of biopolymer filaments in soft tissue and the cell cytoskeleton. While the mechanical behavior of these materials evidently depends on the constituents, the underlying network microstructure is particularly important at large strains. Numerous rheological experiments have shown that random fibre networks of in vitro reconstituted biopolymer networks exhibit strong nonlinear elastic strain-stiffening accompanied with an increase of the shear modulus up to three orders of magnitude. Several synthetic polymer network systems show a similar phenomenology as biopolymer networks, albeit with a much weaker increase of the modulus up to a factor of ~2. The strain-stiffening in biopolymer networks is, generally, attributed to the properties of the fibres, the cross-links that mediate the inter-fibre force transmission and the network microstructure. Despite much research in the recent past, the mechanical properties of biopolymer networks still hold many mysteries. The conjecture in this thesis is that they are mostly hidden in the network microstructure. Extensive computer simulations have provided a deep insight in the relationship between microstructure and mechanical properties of biophysical networks. This understanding has provided a unified explanation of stiffening in a wide range of biophysical network materials.

    Making coke a more efficient catalyst in the oxidative dehydrogenation of ethylbenzene using wide-pore transitional aluminas

    The thermal activation of a silica-stabilized γ-alumina impacts positively on the oxidative dehydrogenation of ethylbenzene (EB) to styrene (ST). A systematic thermal study reveals that the transition from γ-alumina into transitional phases at 1050 °C leads to an optimal enhancement of both conversion and selectivity under pseudo-steady state conditions; where active and selective coke have been deposited. The effect is observed in the reaction temperature range of 450–475 °C at given operation conditions resulting in the highest ST yield, while at 425 °C this effect is lost due to incomplete O2 conversion. The conversion increase is ascribed to the ST selectivity improvement that makes more O2 available for the main ODH reaction. The fresh aluminas and catalytically active carbon deposits on the spent catalysts were characterized by gas adsorption (N2 and Ar), acidity evaluation by NH3-TPD and pyridine adsorption monitored by FTIR, thermal and elemental analyses, solubility in CH2Cl2 and MALDI-TOF to correlate the properties of both phases with the ST selectivity enhancement. Such an increase in selectivity was interpreted by the lower reactivity of the carbon deposits that diminished the COx formation. The site requirements of the optimal catalyst to create the more selective coke is related to the higher density of Lewis sites per surface area, no mixed Si–Al Brønsted sites are formed while the acid strength of the formed Lewis sites is relatively weaker than those of the bare alumina.

    How superdiffusion gets arrested: ecological encounters explain shift from Lévy to Brownian movement

    Ecological theory uses Brownian motion as a default template for describing ecological movement, despite limited mechanistic underpinning. The generality of Brownian motion has recently been challenged by empirical studies that highlight alternative movement patterns of animals, especially when foraging in resource-poor environments. Yet, empirical studies reveal animals moving in a Brownian fashion when resources are abundant. We demonstrate that Einstein’s original theory of collision-induced Brownian motion in physics provides a parsimonious, mechanistic explanation for these observations. Here, Brownian motion results from frequent encounters between organisms in dense environments. In density-controlled experiments, movement patterns of mussels shifted from Lévy towards Brownian motion with increasing density. When the analysis was restricted to moves not truncated by encounters, this shift did not occur. Using a theoretical argument, we explain that any movement pattern approximates Brownian motion at high-resource densities, provided that movement is interrupted upon encounters. Hence, the observed shift to Brownian motion does not indicate a density-dependent change in movement strategy but rather results from frequent collisions. Our results emphasize the need for a more mechanistic use of Brownian motion in ecology, highlighting that especially in rich environments, Brownian motion emerges from ecological interactions, rather than being a default movement pattern.

    Binnen het Nederlandse circusveld bestaat de opvatting dat de professionele Nederlandse circusartiesten moeite hebben met het ontwikkelen van een succesvolle carrière. Dit onderzoek zet de carrières van professionele Nederlandse circusartiesten uiteen aan de hand van verschillende sociologische theorieën om te onderzoeken hoe Nederlandse circusartiesten hun kansen op de arbeidsmarkt kunnen vergroten, aan de hand van de onderzoeksvraag: Hoe kunnen de kansen op een succesvolle carrière voor professionele Nederlandse circusartiesten vergroot worden? Het onderzoek bestaat uit 2 delen: een theoretisch en empirisch deel. In het theoretische deel betreft een uiteenzetting over circus(artiesten) en de sociologische onderzoeksvelden betreffende kunstenaarsprofielen (van Winkel, Gielen en Zwaan) en de artistieke biotoop (Gielen en Laermans). Voor het empirische deel zijn 12 kwalitatieve diepte-interviews afgenomen met professionele Nederlandse circusartiesten. De respondenten worden op basis van de empirie ingedeeld in de geformuleerde circusartiestprofielen. In combinatie met de artistieke biotoop kan worden afgelezen hoe succesvol succesvol de carrières van de artiesten zijn. Vervolgens wordt deze biotoop vergeleken met de 'ideale' en wordt antwoord gegeven op de onderzoeksvraag.

    Strategies for gastrin releasing peptide recepror targeted imaging in prostate cancer

    Prostaatkanker wordt het vaakst vastgesteld bij mannen ouder dan vijftig, waarbij een positieve familieanamnese predisponeert voor het ontstaan van kanker. Als de ziekte in een vroeg stadium wordt gediagnosticeerd, is de vijfjaarsoverleving bijna 100%. Positron emission tomography (PET) en single photon emission computed tomography (SPECT) kunnen middels hoge sensitiviteit, specificiteit en resolutie helpen bij het (re)stageren van prostaatkanker en daarbij op moleculair niveau tumor informatie verkrijgen. De gastrin releasing peptide receptor (GRPR) kan gebruikt worden voor moleculair beeldvorming bij prostaatkanker. GRPR komt in hoge expressie voor in humane kankercellijnen en kan gebruikt worden als doel voor kankerdetectie. Het doel van deze thesis was nieuwe bombesine tracers te ontwikkelen voor de GRPR, die in hoge expressie voorkomt in prostaatkanker. Betrekkelijk eenvoudig te maken, hoogkwalitatieve probes zouden breed ingezet kunnen worden voor prostaatkankerdetectie en follow up, waarbij meer invasieve technieken (zoals de rectale echografie en PSA-bepalingen) dan wel beeldvormende technieken met wisselende sensitiviteit en specificiteit mogelijk minder gebruikt hoeven te worden. Diverse bombesine-peptiden, gelabeld met verschillende SPECT en PET radionucliden, werden in vivo en in vitro getest. Deze nieuwe probes toonden goede binding en werden met succes getest in diermodellen. Voor de in deze thesis geteste probes zullen extra studies verricht moeten worden voor translatie naar de kliniek. Prostate cancer (PCa) is most commonly diagnosed in men over the age of 50, but the genetic history of their family are already an indication of a possible predisposition. When diagnosed at an early stage, the 5-year survival rate of PCa is almost 100%. Positron emission tomography (PET) and single photon computed tomography (SPECT) with high sensitivity, specificity and resolution may help to investigating the staging and restaging of prostate cancer and obtaining molecular tumor information. Gastrin releasing peptide receptor (GRPR) may be a valid target for nuclear imaging of PCa. GRPR is highly expressed in several human cancer cell lines and could be used as a potential target for cancer detection. The aim of this thesis was to develop novel bombesin tracers to target Gastrin Releasing Peptide Receptor (GRPR), which is overexpressed in Prostate Cancer (PCa). Relatively easy to make, high quality probes could be widely used for prostate cancer detection and follow up, thus rendering more invasive techniques (like trans rectal ultrasound and PSA blood tests) or imaging techniques with varying sensitivity and specificity less important. Different bombesin peptides have been radio-labeled with different SPECT and PET radionuclides and evaluated in vitro and in vivo. These novel probes showed good targeting abilities and were successfully tested in animal models. For clinical translation of the probes studied in this dissertation additional studies should be conducted.

    State space and graphical models for estimating networks dynamics

    Aanvankelijk werd de sequencing van het genoom werd door velen leken gezien alsof alles over de genetische werkelijkheid bekend geworden was. Echter, sequencing stelde ons alleen maar voor aan de spelers van een zeer complex genomisch spel, waarvan we de regels nu pas beginnen te begrijpen. Een manier om dit spel te begrijpen is door het uitvinden van hoe de spelers met elkaar omgaan: genomische netwerk reconstructie is een aantrekkelijk paradigma van genomische wetenschap, maar inhomogene monsters en complexe tijdsdynamiek betekent dat de regels van de genomische interactie spel verschillend kunnen zijn voor verschillende subgroepen of zelfs veranderen in de tijd. Dit proefschrift ontwikkelt nieuwe methoden om te gaan met deze twee scenario's. Initially, the sequencing of the genome in 2000 was seen by many laymen as if everything about the genetic reality had become known. In fact, sequencing merely introduced us to the players of a very complex genomic game, whose rules we only begin to understand. One way to understand the genomic game is by finding out how the players interact: genomic network reconstruction is an attractive paradigm of genomic science, but inhomogeneous samples and complex time dynamics, means that the rules of the genomic interaction game may be different for different subgroups or even change over time. This thesis develops new methods to deal with these two scenarios.

    The foreign body reaction

    Biomaterialen worden wereldwijd veel gebruikt, bijvoorbeeld voor het repareren, herstellen en beschadigen van beschadigd of ziek weefsel. Het lichaam reageert op deze biomaterialen met een ontstekingsreactie die de ‘foreign body reaction’ (FBR) wordt genoemd. Wanneer het lichaam biomaterialen niet verdraagt, kan dat grote gevolgen hebben. Daarom wordt veel onderzoek verricht naar de FBR. Van Putten stelt dat veel onderzoek gestuurd wordt door aannames en hypothesen die nog bewezen moeten worden. Hij pleit daarom voor meer fundamenteel onderzoek naar de cellulaire en moleculaire processen die de FBR sturen, en levert daaraan zelf in zijn proefschrift een bijdrage. Een van zijn conclusies is dat de aanwezigheid van endotoxines, celwandbestanddelen van gram-negatieve bacteriën, in biomaterialen de lichaamsreactie kan beïnvloeden. Ook concludeert hij dat de FBR gemoduleerd kan worden door gebruik te maken van de menselijke cytokine (een molecuul die een rol speelt in de immuunafweer) Interleukine-10. Biomaterials are widely investigated and applied in tissue engineering, for example for the repair, restoration and augmentation of damaged or diseased tissue. Introducing these biomaterials into the body induces a biomaterial-dependent inflammatory response called the foreign body reaction (FBR). This inflammatory response is critically important for the functionality of the biomaterial. Adverse reactions to introduced biomaterials, ranging from tissue engineered constructs to orthopedic implants, have been reported. These examples indicate that our current understanding of the FBR is not sufficient to completely predict or prevent adverse foreign body reactions. Although the FBR has been studied for decades, many of the fundamental cellular and molecular mechanisms, that are crucial to the course of the FBR, are poorly understood at best. Several extensive reviews provide a good general overview of the processes that are involved in the FBR. Due to a shortage of fundamental research on the cellular and molecular mechanisms driving the FBR these reviews are full of assumptions and hypotheses that remain to be proven. In this thesis we describe our efforts to contribute to the general understanding of the FBR.

    The testing effect applied to procedural skills using an echocardiogram simulator

    This study focuses on long-term retention of knowledge and skills required for making transthoracic echoes. Declarative knowledge and procedural skills are interconnected for many skills. Before making an echocardiogram, the practitioner needs to collect from declarative memory what echocardiogram is suitable to visualize what he is looking for. Before being able to diagnose a patient based on the echocardiogram, the practitioner needs the skill to make the appropriate echocardiogram. The aim of this study was to find the best way to achieve long-term retention of declarative knowledge and procedural skills by applying the testing effect and spacing effect. The testing effect explains that declarative memory can be improved when subjects retrieve information rather than restudying the material an equal amount of time; i.e. retesting is more effective than restudying (e.g. Abbott, 1909; Gates, 1917; Agarwal, Karpicke, Kang, Roediger III, & McDermott, 2008; Zaromb & Roediger III, 2010). The spacing effect explains that a repetition will be most beneficial ‘if the material had been in storage long enough as to be just on the verge of being forgotten’ (e.g. McGeoch, 1943; Banaji & Crowder, 1989; Pashler, Rohrer, Cepeda & Carpenter, 2006). The testing effect and spacing effect have been repeatedly shown on declarative knowledge; both effects have not been studied often in combination with procedural skills. Medical students have been trained on the basic anatomy and function of the heart (declarative knowledge) and received a theoretical introduction to transthoracic echocardiography. Additionally subjects were trained on making heart echoes (procedural skills). Groups with or without an interim test between the training and the final test have been compared on both their declarative performance and their procedural performance. As expected, interim testing has a beneficial effect on the long-term retention of declarative knowledge. This effect goes beyond declarative performance, as the groups that took an interim procedural test outperformed the groups that did not take an interim test. It seems the testing effect can be generalized to procedural skills.
