    Wearable magnetoencephalography in a lightly shielded environment

    Wearable magnetoencephalography based on optically pumped magnetometers (OPM-MEG) offers non-invasive and high-fidelity measurement of human brain electrophysiology. The flexibility of OPM-MEG also means it can be deployed in participants of all ages and permits scanning during movement. However, the magnetic fields generated by neuronal currents – which form the basis of the OPM-MEG signal – are much smaller than environmental fields, and this means measurements are highly sensitive to interference. Further, OPMs have a low dynamic range, and should be operated in near-zero background field. Scanners must therefore be housed in specialised magnetically shielded rooms (MSRs), formed from multiple layers of shielding material. The MSR is a critical component, and current OPM-optimised shields are large (>3 m in height), heavy (>10,000 kg) and expensive (with up to 5 layers of material). This restricts the uptake of OPM-MEG technology. Here, we show that the application of the Maxwell filtering techniques signal space separation (SSS) and its spatiotemporal extension (tSSS) to OPM-MEG data can isolate small signals of interest measured in the presence of large interference. We compare phantom recordings and MEG data from a participant performing a motor task in a state-of-the-art 5-layer MSR, to similar data collected in a lightly shielded room: application of tSSS to data recorded in the lightly shielded room allowed accurate localisation of a dipole source in the phantom and neuronal sources in the brain. Our results point to future deployment of OPM-MEG in lighter, cheaper and easier-to-site MSRs which could catalyse widespread adoption of the technology

    Employee Organisational Commitment and Corporate Environmental Sustainability Practices: Mediating Role of Organisation Innovation Culture

    The growing concerns about climate change have seen global leaders and the international community launch diverse initiatives, deals and reforms in an attempt to combat its negative impact. In response to these initiatives, corporates are increasingly prioritising environmental sustainability practices, such as reducing resource use, recycling and redesigning products and services to transition to sustainable operations, as a means of promoting trust and credibility, increasing their reputation and protecting the planet. However, the extant literature does not provide a clear understanding of the determinants of promoting corporate environmental sustainability practices (CESPs). Thus, drawing upon both resource-based views (RBVs) of the firm and the economic views of neo-institutional theory (NIT), this study examined the role of employee organisational commitment (EOC) and organisation innovation culture (IC) in promoting CESP. This quantitative study collected the required data through a questionnaire survey of senior to mid-level managers of a total of 201 Dhaka Stock Exchange (DSE)-listed and non-listed firms. A two-step structural equation modelling (SEM) technique, employing SmartPLS, was followed to test the study's hypothesised associations. The findings provide evidence of a direct association between EOC and CESP and an indirect association through IC. Our findings are robust as alternative models were developed and tested based on different control and instrumental variables relating to organisational characteristics. The findings of the study contribute to the environmental sustainability literature by providing empirical evidence of the importance of EOC and IC in promoting CESP. Further, the findings inform managers, governments, foreign investors and other stakeholders on the importance of building EOC and developing suitable cultural practices within an organisation that promote CESP

    Data Checking of Asymmetric Catalysis Literature Using a Graph Neural Network Approach

    The range of chemical databases available has dramatically increased in recent years, but the reliability and quality of their data are often negatively affected by human-error fidelity. The size of chemical databases can make manual data curation/checking of such sets time consuming; thus, automated tools to help this process are highly desirable. Herein, we propose the use of Graph Neural Networks (GNNs) to identifying potential stereochemical misassignments in the primary asymmetric catalysis literature. Our method relies on the use of an ensemble of GNN models to predict the expected stereoselectivity of exemplars for a particular asymmetric reaction. When the majority of these models do not correlate to the reported outcome, the point is labeled as a possible stereochemical misassignment. Such identified cases are few in number and more easily investigated for their cause. We demonstrate the use of this approach to spot potential literature stereochemical misassignments in the ketone products resulting from catalytic asymmetric 1,4-addition of organoboron nucleophiles to Michael acceptors in two different databases, each one using a different family of chiral ligands (bisphosphine and diene ligands). Our results demonstrate that this methodology is useful for curation of medium-sized databases, speeding this process significantly compared to complete manual curation/checking. In the datasets investigated, human expert checking was reduced to 2.2% and 3.5% of the total data exemplars

    Protective efficacy of a bivalent equine influenza H3N8 virus-like particle vaccine in horses

    Equine influenza (EI) is a highly contagious acute respiratory disease of wild and domesticated horses, donkeys, mules, and other Equidae. EI is caused by the Equine Influenza virus (EIV), is endemic in many countries and outbreaks still have a severe impact on the equine industry globally. Conventional EI vaccines are widely used, but a need exists for a platform that facilitates prompt manufacturing of a highly immunogenic, antigenically matched, updated vaccine product. Here we developed a plant-produced bivalent EI virus-like particle (VLP) vaccine candidate which lacks the viral genome and are therefore non-infectious. We conducted a pilot safety/dose response study of a plant produced bivalent VLP vaccine expressing the HA proteins of Florida clade (FC) 1 and FC2 EIV in 1:1 ratio. Groups of three EIV seronegative horses were vaccinated using four antigen levels (0 sham control, 250, 500, 1000 HAU/dose component). Two doses of vaccines were administered one month apart, and horses were observed for adverse reactions, which were minimal. Sera were collected for hemagglutination inhibition (HI) testing using FC1 and FC2 viruses. One month after the second dose, all horses were challenged with the aerosolized FC1 virus. Horses were observed daily for clinical signs, and nasopharyngeal swabs were collected to quantify viral RNA using qPCR and infectious virus by titration in embryonated hens' eggs. Results showed that all vaccinated groups seroconverted prior to challenge. Post-challenge, both clinical scores and virus shedding were much reduced in all vaccinates compared to the sham-vaccinated controls. We conclude that the VLP vaccines were safe and effective in this natural host challenge model. A safe, efficacious, new-generation bivalent EI VLP vaccine produced in plants, which can promptly and regularly be antigenically matched to ensure optimal protection, will pave the way to highly competitive commercially viable vaccine products for all economic environments globally

    Assessing Public Morality and the Necessity of Restrictions on ECHR Rights

    One does not have to be an animal lover to consider that unnecessary suffering during the ritual slaughter of animals is morally unjustified. The moral objection can arguably be met by legally requiring that animals be stunned before being subjected to ritual slaughter. However, such requirements have been challenged by religious believers as being an unjustified limitation on their manifestation of their religious beliefs. This article analyses how such legal challenges have been adjudicated in the legal orders of the European Union and the European Convention on Human Rights. After outlining the ECtHR’s approach to restrictions based on public morality, this article focuses on the compatibility with Article 9 ECHR of restrictions on animal slaughter for religious reasons. It then considers (i) how the ECtHR assesses the necessity of such restrictions, particularly its approach to general measures; (ii) the requirement that restrictions be the least restrictive measures; (iii) restrictions being applied to only part of a State’s territory; and (iv) the wider implications of justifying such restrictions on the basis of public morality. The article concludes by briefly reflecting on (i) consistency and coherence between the two European Legal Orders, and (ii) analogous issues under the International Covenant on Civil and Political Rights (1966)

    The “special relationship,” and the overseas Chinese: the Information Research Department (IRD) and the United States Information Agency (USIA) cold war partnership in East Asia, 1950s-1970s

    From the 1950s to the 1970s, Britain’s strategic role in the Cold War in East and Southeast Asia shaped the post-WWII contours and ramifications of what Winston Churchill famously dubbed the Anglo-American ‘special relationship’. Through its clandestine Information Research Department (IRD), Britain targeted anti-communist propaganda, focusing on neutral nations and the overseas Chinese communities. The IRD assessed communist influence among emigrant Chinese communities along with their views of the United States. The IRD served to support American goals while bolstering Britain’s regional influence. Despite occasional divergences, the IRD and the United States Information Agency (USIA) coordinated efforts to counter communist expansion, reflecting the adaptability of their ‘special relationship’

    Cordycepin generally inhibits growth factor signal transduction in a systems pharmacology study

    Cordycepin (3′ deoxyadenosine) has been widely researched as a potential cancer therapy, but many diverse mechanisms of action have been proposed. Here, we confirm that cordycepin triphosphate is likely to be the active metabolite of cordycepin and that it consistently represses growth factor-induced gene expression. Bioinformatic analysis, quantitative PCR and western blotting confirmed that cordycepin blocks the PI3K/AKT/mTOR and/or MEK/ERK pathways in six cell lines and that AMPK activation is not required. The effects of cordycepin on translation through mTOR pathway repression were detectable within 30 min, indicating a rapid process. These data therefore indicate that cordycepin has a universal mechanism of action, acting as cordycepin triphosphate on an as yet unknown target molecule involved in growth factor signalling

    Magnetic Resonance Imaging Reveals Novel Insights into the Dual Mode of Action of Bisacodyl: A Randomized, Placebo-controlled Trial in Constipation

    Bisacodyl is a widely used laxative that stimulates both motility and secretion. Our aim was to exploit the unique capabilities of MRI to define bisacodyl's mode of action. Two placebo-controlled cross-over trials were performed, one using a single dose of Bisacodyl 5 mg while the second dosed daily for 3 consecutive days. Serial MRI was performed every 75 minutes. Primary endpoint: ascending colon water content as assessed by T1AC AUC300–450 minutes. Secondary endpoints included: small bowel water content, whole gut transit time (WGTT), colonic volumes, stool frequency, and consistency using Bristol Stool Form Score (BSFS). Exploratory endpoints: changes in the serial segmental volumes were quantified from the number of “mass movements” defined as episodes when segmental volume change from the previous scan was > 20% of baseline volume. We also measure the time to defecate after dosing. After 3 days of bisacodyl, ascending colon water content (T1) was 62% greater than after placebo, mean difference T1 AUC300–450 minutes 50.2 (61.0) sec.min, 95% CI (9.2, 91.2), P = 0.02, while after a single dose difference was only 11% (P = 0.58). Both single and repeated doses shortened WGTT (P < 0.049) and time to defecate (P 0.01). Only repeated doses significantly increased small bowel water content (P < 0.03), the number of “mass movements” (P = 0.048), bowel frequency (P = 0.006), and BSFS (P = 0.03). Repeated, compared to single dosing of Bisacodyl, additionally increases small bowel and colon water content and increases the number of “mass movements” thereby increasing its laxative effect. MRI is a non-invasive, patient-acceptable technique for evaluating drugs which alter secretion and/or motility

    A Protracted War Against Counterfeits: Navigating Tension Management Under External Pressures on an E-Commerce Platform

    This study examines how external pressures drive a platform leader to fight counterfeiting over time. Acknowledging the tension between the illegality and legitimacy of counterfeiting, we find that the fighting effort involves an enduring balance between the two poles of the tension until counterfeits have been sharply reduced. We demonstrate how the management of the tension entails an interactive process in which a platform leader decouples counterfeit fighting actions to pursue legitimacy. We suggest that pressures from external forces predict a process of decoupling and legitimation through which the platform leader promotes ecosystem acceptance and builds a shared understanding of ecosystem purposes, and the endeavours to balance platform quality and activity prompt the platform leader to pursue legitimation which demonstrates the viability of an ecosystem. We show that these processes are accompanied by activating and stabilising the tension, which predicts varied strength and scope of measures and the reduction of counterfeits over time
