    Einfluss der Vorbehandlungsmethoden und Befestigungsmaterialien auf die Scherhaftfestigkeit von gedrucktem und gefrästem Zirkonoxid

    Im klinischen Alltag haben sich verschieden konventionelle, adhäsive und selbstadhäsive Befestigungskomposite für die definitive Befestigung von Kronen und Brücken aus Zirkonoxidkeramik (3Y-, 4Y-, 5Y-TZP), Lithiumdisilikatkeramik und zirkonoxidverstärktem Lithiumsilikat bewährt. Es soll untersucht werden ob diese Befestigungskomposite auch für die Befestigung von additiv gefertigter 3Y-TZP Zirkonoxidkeramik geeignet sind und ob sich Empfehlungen für ein Befestigungsprotokoll ableiten lassen. Es wurden je 105 Prüfkörper aus gedrucktem Zirkonoxid (Lithacon Y210, Lithoz), gefrästem Zirkonoxid (Katana Zirconia HTML plus, Kuraray Noritake) und gefrästem CAD/CAM Komposit (Grandio disc, VOCO) hergestellt. Alle Prüfkörper wurden zunächst poliert (Körnung P2000) und korundgestrahlt (50µm, 1bar). Anschließend wurden die Prüfkörper pro Material zu 7 verschiedenen Befestigungsprotokollen zugeteilt: Panavia SA Cemement Universal (Kuraray Noritake), Ceramic Primer plus + Panavia SA Cemement Universal (Kuraray Noritake), SpeedCem plus (Ivoclar), Monobond plus + SpeedCem plus (Ivoclar), Ceramic primer plus + Panavia V5 (Kuraray Noritake), Monobond plus + Variolink esthetic (Ivoclar), Panavia 21 (Kuraray Noritake) Nach Herstellung des Klebeverbundes wurden die Proben im Wasserbad (30°C, 30 Tage) und im Thermolastwechsler (7.500 Zyklen, 5°C/55°C) künstlich gealtert. Anschließend wurde die Scherhaftfestigkeit gemessen (MPa) und eine Bruchflächenanalyse durchgeführt. Die Mittelwerte der Scherhaftfestigkeitsmessung liegen zwischen 0,56MPa und 19,91MPa. Die Ergebnisse der Scherprüfung und der Bruchflächenanalyse zeigen eine signifikante Abhängigkeit von Restaurationsmaterial, Befestigungsmaterial und der Verwendung eines Haftvermittlers. Die in der Studie getesteten Befestigungskomposite weisen für additiv gefertigter Zirkonoxidkeramik vergleichbare oder bessere Scherhaftfestigkeitswerte im Vergleich zu gefräster Zirkonoxidkeramik auf. Die Ergebnisse lassen annehmen, dass die Befestigungsprotokolle für gefrästes Zirkonoxid auch bei additiv gefertigtem Zirkonoxid anwendbar sind

    High-mass star formation across the Large Magellanic Cloud

    Star formation is a fundamental process that shapes the evolution of galaxies, with high-mass (M > 8 M⊙) star formation playing a critical role as a major driver of energy input and chemical enrichment. Despite significant progress, the mechanisms governing (high-mass) star formation remain incompletely understood, particularly within the unique extra galactic environments of the Large Magellanic Cloud (LMC). The LMC provides a rare laboratory for studying high-mass star formation under distinct environmental conditions compared to our Galaxy, the Milky Way, including lower metallicity, enhanced ultraviolet (UV) radiation, reduced cosmic ionization rates, and large-scale dynamical interactions. These factors might influence key star- formation processes such as fragmentation, core mass distribution, clustering, chemical composition, and feedback mechanisms. Thus, studying these processes in the LMC offers critical insights into how environmental factors shape star formation across different galactic contexts. This thesis presents the first systematic survey of 20 massive young stellar objects in the LMC, selected from Spitzer and Herschel catalogs. The sample spans diverse physical and chemical conditions, evolutionary stages, and locations across the LMC, where metallicity gradients and varying levels of external feedback provide a statistically significant dataset for understanding star formation. Using high-resolution observations from ALMA, complemented by ATCA data and ancillary datasets from Herschel, Spitzer, and VISTA, this study explores the physical and chemical properties of star-forming regions. Among the main findings of this work, the high-angular resolution reveal diverse fragmentation and clustering patterns. Chemical properties exhibit a complex interplay with metallicity, deviating from linear scaling, and signatures of large-scale interactions and metallicity mixing emerge. The detection of filaments, outflows, and HII regions further confirms the dynamic feedback processes shaping gas morphology. This multiwavelength characterization of 20 star-forming regions advances our understanding of high-mass star formation in low-metallicity environments, emphasizing the influence of environmental diversity. By combining physical, chemical, and kinematic insights, this study provides a statistically robust foundation for future extra- galactic star formation research

    THE REGULATION OF TNF SIGNALING IN METABOLISM-RELATED DISEASES

    Tumor necrosis factor (TNF) is a proinflammatory cytokine with a well-established role in diabetes pathogenesis. In addition to driving inflammation, TNF can induce cell death; yet the contribution of TNF-induced cell death to β-cell loss in diabetes remains unclear. Receptor- interacting protein kinase 1 (RIPK1) is a key regulator of TNF-induced cell death. The kinase function of RIPK1 is essential for TNF-induced necroptosis and under certain circumstances, apoptosis. In contrast, the scaffolding function of RIPK1 is essential to limit cell death. We studied the role of TNF-induced cell death in the pathogenesis of diabetes by dissecting the kinase versus scaffold function of RIPK1. To study the role of this important checkpoint in TNF signaling in β-cells, we used two models that induce hyperglycemia by either β-cell toxicity or insulin resistance. These are the Multiple Low Dose Streptozotocin (MLDSTZ) and the high-fat diet models, respectively. Using Ripk1D138N mice, which express a kinase-inactive version of RIPK1, we demonstrated that RIPK1 kinase activity does not drive the pathogenesis of type 1 diabetes (T1D), at least in the Multiple Low Dose Streptozotocin (MLDSTZ) model. We then generated Ripk1β-KO mice, which lack RIPK1 specifically in pancreatic β-cells. Notably, these mice are viable and have normal glycemic control over their lifespan. Moreover, Ripk1β-KO mice were not sensitized to MLDSTZ or high-fat diet (HFD). Hence, although RIPK1 is vital for the survival of many cell types, including keratinocytes and immune cells, its deletion in β-cells did not result in any spontaneous phenotype. Islets isolated from Ripk1β-KO mice were resistant to TNF-induced cell death and were not sensitized to cytokine cocktail-induced cell death. While RIPK1 is crucial for the survival of many cell types, its deletion in β-cells did not sensitize them to cell death. Interestingly, we found that β-cells express high levels of cFLIP relative to Caspase-8, likely conferring resistance to TNF-induced apoptosis. Additionally, β-cells exhibited low RIPK3 expression, explaining their resistance to necroptosis. These findings suggest that β-cells fine-tune the expression of prosurvival and pro-death proteins to limit TNF-induced cell death, thereby preserving β-cell mass and maintaining normoglycemia. In a second project, we investigated the role of LUBAC, another important checkpoint of TNF signaling in glycogen metabolism. LUBAC, an E3 ubiquitin ligase, acts downstream of various immune and cytokine receptors and is crucial for limiting TNF-induced cell death. In humans, LUBAC deficiency leads to autoinflammation and immunodeficiency. Unexpectedly, many patients also develop cardiomyopathy and myopathy due to toxic polyglucosan (a less- branched form of glycogen) accumulation in cardiac and skeletal muscles. 9 To explore LUBAC's role in glycogen metabolism, we generated HoipMuscle-KO mice, lacking HOIP, the main catalytic component of LUBAC, in cardiac and skeletal muscle. These mice were viable and displayed no overt phenotypes. However, histological analysis revealed polyglucosan accumulation in the heart, reinforcing the hypothesis that LUBAC regulates glycogen metabolism through an unknown mechanism. Our preliminary results suggest that LUBAC may regulate autophagy, as ATG16L1, a key scaffold protein for LC3 lipidation, was significantly downregulated in HoipMuscle-KO hearts. The precise mechanism by which LUBAC controls glycogen metabolism warrants further investigation. In summary, my work revealed that β-cells regulate TNF-mediated cell death pathways in an alternative way, independent of classical checkpoints. While elevated cFLIP levels may play a key role in this resistance, other molecular mechanisms are likely involved and warrant further investigation. Additionally, LUBAC, a critical checkpoint in TNF signaling, appears to regulate glycogen metabolism, potentially by facilitating autophagy-mediated clearance of polyglucosan bodies

    Charakterisierung der Rolle der NADPH-Oxidase Duox2 bei der Darm-Homöostase und der Immunabwehr von intestinalen Pathogenen

    Duox2 ist eine von insgesamt sieben humanen und sechs murinen Isoformen der Noxe, deren Funktion darin besteht Sauerstoff (O2) zu Reaktiven Sauerstoffspezies (reactive oxygen species; ROS) zu katalysieren. Das Endprodukt von Duox2, Wasserstoffperoxid (H2O2), kann einerseits oxidativen Schaden verursachen und andererseits als Signalmolekül fungieren. Im Darm wird eine Über- oder Unterexpression von Duox2 mit entzündlichen Darmerkrankungen assoziiert. Welche genau Rolle Duox2 in der Darm-Homöostase und der Immunantwort gegen intestinale Pathogene hat, ist noch nicht bekannt. Um dieser Frage nachzugehen, wurde in dieser Arbeit anhand von Mäusen mit einer spezifischen Defizienz für Duox2 in intestinalen Epithelzellen (Duox2IEC-KO) die Darm- Homöostase sowie die Immunantwort gegen eine bakterielle Darminfektion mit C. rodentium untersucht. Die durch den Intestinalen-Epithelzell-spezifischen-Knockout (IEC-KO) entstandene Duox2- Defizienz wurde dabei nicht durch die Hochregulierung anderer Nox-Isoformen kompensiert. Es ergaben sich keine strukturellen Veränderungen der einzelnen Darmabschnitte in Länge, Gewicht oder Beschaffenheit der Mucosa. Einzig der Darminhalt des Cecums der Duox2IECKO- Mäuse war schwerer als der der WT-Mäuse. Bei der Untersuchung der Auswirkung der Duox2-Defizienz auf ausgewählte Entzündungsparameter zeigte sich, dass das Gewicht der mLN in den Duox2IEC-KO-Mäuse geringer war als in den WT-Mäusen. Auch die Expressionen des antimikrobiellen Peptids Reg3γ und des mit chronischen Entzündungen assoziierten iNos wiesen eine verminderte Expression im Ileum bzw. im Ileum und Colon auf. Bei der Untersuchung des Mucus zeigte sich ein verringerter Anteil der Becherzellen an der Gesamtzellzahl im Colon der Duox2IEC-KO-Mäuse. Zusammen mit der niedrigeren Expression von CLCA1 in allen drei Darmabschnitten der Duox2IEC-KO-Mäuse, welches an der Schichtung des Mucus mitwirkt, wiesen diese Ergebnisse auf eine veränderte mucosale Struktur in dem Duox2-defizienten Gewebe hin. Von den Mikrobiota, als im Mucus lebenden Organismen, wiesen die γProteobakterien ein vermehrtes Vorliegen im Cecum und Colon der Duox2IEC-KOMäuse auf. Nach Infektion der Duox2IEC-KO-Mäuse mit C. rodentium ließ sich kein Unterschied bezüglich der Schwere der Infektion feststellen. Darüber hinaus waren die ohne Infektion festgestellten Unterschiede in der Expression von Reg3γ und iNos nach Infektion mit C. rodentium nicht mehr vorhanden. Auch CLCA1 als Marker für eine mucosale Strukturveränderung wies nach Infektion nicht länger eine niedrigere Expression in den Darmabschnitten der Duox2IEC-KO10 Mäuse auf. Die bei nicht infizierten Duox2IEC-KO-Mäusen festgestellten Veränderungen führten also nicht zu einer veränderten antibakteriellen Immunantwort. Zusammenfassend konnte in dieser Arbeit gezeigt werden, dass eine Defizienz von Duox2 im intestinalen Gewebe sich auf mucosale Einflussfaktoren wie CLCA1 und die Becherzellzahl, auf mit Entzündungen im Zusammenhang stehende Einflussfaktoren wie iNos und Reg3γ sowie auf die Mikrobiota im Darm auswirkt. Auf den Verlauf der Infektion mit C. rodentium hatte die Duox2-Defizienz hingegen keine Auswirkung. Die genaue Rolle von Duox2 innerhalb dieser komplexen Vorgänge und die funktionellen Auswirkungen der Veränderungen müssen weiter untersucht werden

    Lifelong Learning through Workplace Learning: A University Perspective

    This study aims to identify key dimensions in the implementation of workplace learning in higher education. Utilizing an integrative literature review, 38 papers meeting the inclusion criteria were analyzed from an initial pool of 227 articles retrieved from various databases. The objective was to provide an overview of how workplace learning is implemented in higher education contexts. Additionally, a semi-structured interview was employed as the primary data collection instrument, involving 12 faculty members from a Laotian university as key informants. The collected data were analyzed using a deductive category assignment approach. By collating various perspectives, the findings propose ten critical dimensions that should be considered when integrating workplace learning in higher education institutions. These dimensions include personal interest, self-confidence, individual engagement, group learning, trust, management, expertise, collaborations, policy, and culture. Future research may further validate these findings through a quantitative research design

    Importance of dendritic cells in pathogenesis and therapy of conjunctival melanoma

    Conjunctival melanoma is a very rare malignant tumor, which occurs in less than 5 % of ocular melanomas. Because of the high recurrence rate of CM, current therapy options are insufficient and, for advanced CM, no standardized therapy is available. The immunological principles, especially the composition of dendritic cell subsets and their functions, are almost completely unknown in CM. DCs are antigen-presenting cells and link the innate and the adaptive immune system. In this dissertation, we investigated the immune cell composition in the physiological conjunctiva, CM and in the draining lymph nodes, the lymph- and hemangiogenic profile and the effect of local injection of non-activated classical DCs on survival, metastases and tumor progression in vivo in immunocompetent C57BL/6N mice using a HGF-Cdk4R24C CM model. DCs can be subclassified into cDC₁, cDC₂, plasmacytoid and monocyte derived DCs. Subclassification showed that CD103- cDC₂ are the most common DC subset in healthy conjunctiva and that CD103- cDC₂, CD103+ cDC₁, moDCs and pDCs are significantly increased in CM. A local injection of non-activated cDC₂ showed neither positive nor negative effects. Furthermore, the angiogenic factors expressed by cDCs and tumor cells were verified in vitro and identified Osteopontin as the most relevant cytokine expressed by the tumor cells, which actually interacts with DCs. We evaluated a combined therapy strategy for CM by subconjunctival OPN blockade and activated cDC vaccination. The combined therapy slowed down tumor progression and inhibited tumor growth completely in 35 % of cases. To better understand the mode of action of the OPN blockade, tumor cells and cDCs, the effect on blood- and lymphatic endothelial cells was studied in co-culture with the supernatant of DCs and tumor cells in combination with an OPN blocking antibody or additional OPN protein in vitro. The results of this thesis will help to get a better understanding of the immunological principles in the development of CM and the influence of the tumor microenvironment on the activity of DCs and on blood- and lymhangiogenesis

    Paving the Path toward Smart Sustainable Mobility; Data-Driven Operations Management for Emerging Mobility Systems: Sharing, Automation, and Electrification

    Despite significant advances in mobility systems, they continue to perform inefficiently, contributing to a range of global challenges. To address these issues, many researchers predict that the future of transportation will be characterized by shared, autonomous, and electric vehicles (SAEVs). However, this transformation poses several managerial and societal challenges that must be addressed to fully realize its potential benefits. This thesis highlights the potential of research at the intersection of information systems and operations management to improve the performance of next-generation mobility systems (Chapter 1). By integrating data-driven decision models four research projects underscore this potential. Chapter 2 studies the cooperative charging management of SAEVs to maximize profits and service quality while accommodating uncertain demand, limited charging infrastructure, and time-varying electricity prices. A distributed approach using cooperative multi-agent reinforcement learning is proposed that outperforms centralized static charging strategies and provides insights to improve the performance of SAEVs. Chapter 3 explores the impact of emerging technologies on the physical world and their interaction with user adoption behavior. Specifically, it evaluates a hybrid system that combines autonomous and human-operated ride-hailing services. The study first identifies mobility user preferences toward autonomous services, and using an agent-based model predict and analyze the future of such hybrid service platforms, and evaluate potential changes. The agent-based model enables analysis of the end-to-end impact of key factors, such as trust in the technology. Chapter 4 proposes a method for leveraging data-driven digital twin frameworks to design large-scale charging hubs. Such problem classes are difficult to solve with traditional mathematical programming optimization. This study shows how high-fidelity, data-driven simulation environments coupled with reinforcement learning can achieve arbitrary scalability and high modeling flexibility. The benchmark experiments show that the proposed model designs result in superior cost and service-level performance under real-world operating conditions. Chapter 5 supports the operational management of EVCHs through dynamic pricing. Drawing on cutting-edge deep reinforcement learning algorithms, a model-free solution is provided to find optimal pricing policies. The proposed pricing policy is a time-dependent function of the service rate, called dynamic capacity-based pricing. Benchmark analysis shows that the proposed model not only ensures high profits for EVCHs, but also successfully reshapes aggregated demand as desired, even in environments with high variability in supply and demand

    Einfluss des HPV-Status auf die ekto- und endozervikale Zytologie gemäß den Kriterien der Münchner Nomenklatur III

    Das Ziel der Dissertationsarbeit ist die Bestimmung der Häufigkeit der ekto- und synchron erhobenen endozervikalen zytologischen Befunde und deren Abhängigkeit von HP-Viren. In Vorarbeiten der Arbeitsgruppe an der Klinik und Poliklinik für Frauenheilkunde und Geburtshilfe konnte bereits gezeigt werden, dass die ekto- und endozervikale zytologische Verteilung von den zugrunde liegenden Pap-Klassifikationen abhängig ist. Zum damaligen Zeitpunkt lagen keine Ergebnisse zum HPV-Status vor. Es wurde vermutet, dass die unterschiedliche Antwort des ekto- und endozervikalen Epithels unmittelbare Folge einer HPV-Infektion ist. Zur Abklärung dieser Hypothese wurden die Ergebnisse von 1133 Patientinnen analysiert, die in der Klinik für Frauenheilkunde und Geburtshilfe an der Universität zu Köln eine ekto- und endozervikale zytologische Untersuchung und zusätzlich eine HPV-PCR-Untersuchung erhielten. Die Analyse bezieht sich ausschließlich auf plattenepitheliale Befunde, die sowohl in der Ekto- als auch in der Endozervix zu finden sind. Die Datensammlung erstreckte sich über einen zehnjährigen Zeitraum von 2011 bis 2021. Hierbei wurden die demographischen Befunde, die getrennt erhobenen ekto- und endozervikalen zytologischen Befunde und die in der HPV-PCR ermittelten Genotypen tabellarisch erfasst und ausgewertet. Zunächst wurde die Verteilung der Altersklassen untersucht. Hier zeigte sich eine signifikante Häufigkeit in zwei Altersgruppen, und zwar von 26 bis 30 Jahren (N=264) und von 31 bis 35 Jahren (N=245). Die Zahl der Patientinnen über 50 Jahre war mit einer Zahl von 91 gering. Bei Analyse der Daten fiel auf, dass HPV eine differente Auswirkung auf die ekto- und endozervikale Zytologie hatte. Hierbei zeigte sich in der Pap-Gruppe I/ II-a, d.h. in der Gruppe ohne Dysplasien ein Überwiegen der ektozervikalen Befunde gegenüber den endozervikalen Befunden bei definierter HPV-Konstellation. Im Bereich der mittel- bis schwergradigen Dysplasien zeigte sich ein reziprokes Verhalten, insbesondere in der Gruppe der Pap IVa-p. Hierbei war die endozervikale Zytologie gegenüber der ektozervikalen Zytologie signifikant erhöht. Um diese Befunde weiter einschätzen zu können, wurde die Abhängigkeit des Verhältnisses der ekto- zu endozervikalen Befunden zu den einzelnen HP-Viren untersucht. Ist der ektozervikale zytologische Befund schwergradiger als der endozervikale wurde der Begriff des „ektozervikalen Shifts“ eingeführt. Im umgekehrten Verhältnis spricht man von einem endozervikalen Shift. In allen Gruppen zeigen sich zunächst qualitativ ähnliche Befunde bezüglich des ekto- und endozervikalen Shifts. Bei zytologisch unauffälligen Befunden beziehungsweise geringgradige Dysplasien ist der ektozervikale Shift vorherrschend, wohingegen bei mittleren- bis schwergradigen Dysplasien der endozervikale Shift überwiegt. Bei der Analyse des Verhältnisses von HPV16-Monobefunden zu HPV16+-Kombinationsbefunden zeigte sich in der Kombination eine signifikante Steigerung sowohl des endozervikalen Shifts als auch des ektozervikalen Shifts. Die Kombination von HPV 16 mit anderen HR-Viren potenziert den ekto- und endozervikalen Shift. Bei allen HR-HP-Viren der Gruppe der Gardasil®9-HR-HPV-Monobefunde zeigte sich qualitativ ein Überwiegen der endozervikalen Zytologie zu ektozervikalen Zytologie. Zusätzlich wurde die Gruppe der Nicht- Gardasil®9-HR-HPV untersucht. Hier zeigte sich gegenüber der Gardasil®9-Gruppe eine deutliche Erhöhung des endozervikalen Shifts. Der Befall mit HP-Viren insbesondere der Befall mit HR-HPV oder einer Kombination von HR-HPV führt zu einer Akkumulation mäßig- beziehungsweise schwergradiger Dysplasien in der Endozervix. Diese Zellen sind in der Routinediagnostik mittels Kolposkopie nicht zu erfassen und aufgrund der klinischen Relevanz von besonderer Bedeutung
