Caracterización de la vulnerabilidad sísmica de las edificaciones residenciales empleando análisis de conglomerados y redes neuronales : aplicación a escenarios de riesgo en Murcia (España) y Valdivia (Chile)
En esta tesis se estudia la vulnerabilidad sísmica de edificios de uso residencial teniendo en cuenta no solo factores que determinan su comportamiento estructural y el sistema resistente a cargas laterales, sino también modificadores edificatorios o urbanos, relacionados con la configuración geométrica del edificio en el espacio tridimensional o en relación a los edificios colindantes.
Se aplican diferentes métodos de análisis estadístico para mejorar la caracterización de las tipologías constructivas observadas. Así, el método de análisis de conglomerados permite encontrar los agrupamientos de variables (atributos del edificio, incluyendo sus modificadores edificatorios) más comunes y característicos de cada zona de estudio. Además, se aplican técnicas de redes neuronales (perceptrón multicapa) para validar los resultados obtenidos por el análisis de conglomerados y determinar la variabilidad (o representatividad) con la que se puede encontrar un modificador asociado a una tipología constructiva. Como resultado de estos análisis estadísticos se obtiene un catálogo de conglomerados de tipologías edificatorias (CTE) para cada zona de estudio. Seguidamente, se utiliza el método del índice de vulnerabilidad para cuantificar la vulnerabilidad sísmica de esos CTEs y se establecen los rangos de variación más probable y posible.
La metodología desarrollada se aplica a dos ciudades seleccionadas por sus diferencias en cuanto a características sísmicas, climáticas y urbanísticas: Murcia (España) y Valdivia (Chile). Si bien ambas destacan por su alta peligrosidad sísmica en un contexto nacional, las características sismotectónicas de Valdivia le confieren una peligrosidad mucho mayor que Murcia en una comparativa global. Por contra, la vulnerabilidad sísmica de Murcia es mucho mayor que la de Valdivia, encontrándose gran cantidad de edificios que han sido erigidos son tener en cuenta ninguna consideración sismorresistente. Por el contrario, la sucesión de eventos sísmicos importantes en Valdivia ha conducido a la destrucción de los edificios antiguos y la predominancia de las edificaciones con un diseño sismorresistente notable. Por último, Valdivia presenta una trama urbana más regular que Murcia, con predominio de construcciones bajas de madera. Por su parte, en Murcia la trama urbana es más irregular (especialmente en los barrios antiguos) y abundan las construcciones de mampostería y hormigón armado.
Se plantean diversos escenarios de riesgo en ambas localidades. Para ello, se identifican las principales fuentes sísmicas, incluyendo fallas activas y sectores de la zona de subducción, se seleccionan los modelos de movimiento fuerte más adecuados para cada zona de estudio y se calcula el movimiento sísmico esperado en cada ciudad considerando el efecto de sitio. Para cada localidad, se identifican y asignan las curvas de fragilidad y las curvas de vulnerabilidad a los diferentes CTE para calcular la distribución de daño esperado y la proporción de pérdida esperada, respectivamente.
Finalmente se comparan y discuten los resultados obtenidos en cada zona y se proponen unas conclusiones y líneas futuras de investigación identificadas a raíz de este estudio.
ABSTRACT
In this PhD thesis, I study the seismic vulnerability of residential buildings takin into account not only the factors that control their structural response and lateral load-resisting system, but also urban modifiers linked to the 3D geometric configuration of the building and its relation with the adjoining buildings.
I apply several statistical methods to improve the characterization of observed building typologies: I use the cluster analysis to identify the more common groups of variables (building attributes, including urban modifiers) that characterize each study area. In addition, I apply artificial neural networks (multilayer perceptron) to validate the results given by the cluster analysis and to determine the variability (or representativeness) of finding a certain urban modifier associated to a building typology. As a result of these statistical analyses, I obtain a catalogue of building typology clusters (BTC) for each study area. Subsequently, I use the vulnerability index method to quantify the seismic vulnerability of these BTCs for each study area, including their most likely and possible range of variation.
The method developed in this thesis is applied in two cities that are chosen because of their different characteristics regarding seismic activity, climatic conditions and urban patterns: Murcia (Spain) and Valdivia (Chile). Both cities rank as high seismic hazard in a national context, although the seismotectonic characteristics around Valdivia reflect a clearly higher seismic hazard than Murcia in a global context. By contrast, the seismic vulnerability of Murcia is much higher than the vulnerability of Valdivia. In Murcia, there are many relatively old, non-engineered (without earthquake-resistant design) buildings still erected. In Valdivia, the succession of significant seismic events has led to the destruction of old buildings and the prevalence of buildings with a notable level of earthquake-resistant design. In addition, Finally, Valdivia presents a more regular urban pattern than Murcia, with predominance of low-rise timber structures in Valdivia and irregular urban pattern in Murcia (especially in the oldest districts), where masonry and reinforced concrete buildings are abundant.
Several seismic risk scenarios are proposed for both study areas. This requires identifying the main seismic sources, including active faults and patches of subduction zones, selecting appropriate ground motion prediction models for each tectonic region, and computing the expected seismic strong motion including site effects for each study area. For each city, I identify and assign fragility curves and vulnerability curves to the different CTEs to calculate the expected damage distribution and expected loss ratios, respectively.
Finally, I compare and discuss the results obtained for each study area and propose son conclusions and future research lines
Concierto cromático y diagrama de flujos : Carlos Raúl Villanueva : Ciudad Universitaria de Caracas
El color aparece como un tema accesorio en los trabajos de investigación y las publicaciones sobre la arquitectura de la Ciudad Universitaria de Caracas de Carlos Raúl Villanueva. Aspecto sorpresivo dada la evidencia cromática que presentan las obras del campus universitario y por el sólido sustento de una teoría cromática elaborada en sus escritos, Notas Docentes y Cuadernos de Notas que se constituyen en parte central de un discurso arquitectónico de lo humano. En la experimentación espacial de las mismas se reconoce el diagrama de flujos de Villanueva -explicación esquemática y funcional del espaciotemporal del Conjunto Central- como un instrumento que permite analizar el fenómeno cromático como un elemento esencial de su proceso proyectual.
El color se conceptualiza como un componente vital y existencial de relaciones espaciales lumínico-cromáticas orientadas a la socialización del arte y a la Síntesis de las Artes Mayores. Villanueva acentúa cromáticamente los elementos plásticos tradicionales -invariantes locales que hunden sus raíces en la arquitectura colonial y popular- como partes integrantes del constructo mental del ensayo de integración de los colores en atmósferas creadas por superficies arquitectónicas que vibran por efecto del acceso de la luz natural al espacio fluido.
El diagrama de flujos es la representación gráfica de las relaciones que establece el color entre distintos elementos constructivos, plásticos y el medio físico, así como sus efectos cromáticos sobre la unidad del marco arquitectónico considerando el contexto medioambiental y cultural. Totalidad construida sobre el desplazamiento de un sujeto sometido a la acción simultánea del espacio y el tiempo que se pone en movimiento impulsado cromáticamente. El color es una materia sensorial del diseño de las relaciones espaciotemporales entre cuerpos arquitectónicos, piezas artísticas y entorno medioambiental. En una primera fase, y por medio de seis mecanismos arquitectónicos de control cromático, se estudia la estructura espacial de la aparición del color desde un análisis posicional que toma valores plásticos y constructivos. Los grados de interacción plástica y funciones espaciales asignadas a los colores diseñan atmósferas cromáticas para ser percibidas por un observador que deviene en actor del espacio. En una segunda etapa, y aplicando la metodología de la síntesis, los flujos cromáticos introducen el tiempo en el espacio bajo esquemas que integran las Artes Mayores en la arquitectura.
El observador interpreta los flujos cromáticos en tiempos diseñados como movimientos de un pentagrama musical donde los colores están compuestos espacialmente como sonidos. El ordenamiento cromático del marco espacial vincula al color con el movimiento del observador que introduce directamente la dimensión temporal en un espacio sensible, vacío intermedio que es intuido como interacción entre objetos construidos y elementos artísticos abiertos al medio físico circundante bajo una estructura de lógica arquitectónica.
La demostración del diagrama de flujos como método objetivo para proyectar con colores se inicia con un cuerpo teórico donde se describen sus tres conceptos generales: la técnica del movimiento que ordena con flujos cromáticos el espacio, el diseño de una estética de relaciones cromáticas que integra las artes en el medio físico y la proyección de los flujos cromáticos por medio de diagramas según un orden lógico e intuitivo. En el cuerpo empírico se aplica el diagrama de flujos en tres conjuntos edificados de la Ciudad Universitaria para probarlo como herramienta de diseño y análisis del color espacial en el proyecto. Cada caso práctico corresponde a una división orgánica según los usos universitarios que establece Villanueva: el Conjunto Estudiantil (habitar), Conjunto Central (cultivar el espíritu) y Conjunto Facultad de Arquitectura y Urbanismo (estudiar). En 1948 Villanueva empieza un viaje hacia el color, experimentando e innovando con el fenómeno cromático hasta llegar al Pabellón de Montreal de 1968 cuando los colores representan a Venezuela como país.
En definitiva, el objetivo principal de la tesis es verificar que el diagrama de flujos es un método universal para el análisis, la representación y el proyecto cromático del espaciotemporal. Villanueva idealiza cromáticamente en blanco y negro su espacio interior, sin necesidad de dibujar explícitamente los tonos, las saturaciones y los valores de sus colores, con lo que mantiene el grado de abstracción que exige una aplicación metodológica como dispositivo de carácter universal y científico.
ABSTRACT
Colour appears as an accessory theme in Carlos Raúl Villanueva's research work and publications on the architecture of the University City of Caracas. This is a surprising aspect given the chromatic evidence presented by the works on the university campus and the solid support of a chromatic theory elaborated in his writings, Teaching Notes and Notebooks, which constitute a central part of an architectural discourse of the human. In their spatial experimentation, Villanueva's flow diagram - a schematic and functional explanation of the space-time of the Conjunto Central - is recognised as an instrument that allows us to analyse the chromatic phenomenon as an essential element of his design process.
Colour is conceptualised as a vital and existential component of light-chromatic spatial relations oriented towards the socialisation of art and the Synthesis of the Major Arts. Villanueva chromatically accentuates the traditional plastic elements - local stable variants rooted in colonial and popular architecture - as integral parts of the mental construct of the essay on the integration of colours in atmospheres created by architectural surfaces that vibrate as a result of the access of natural light to the fluid space.
The flow diagram is the graphic representation of the relationships that colour establishes between different constructive and plastic elements and the physical environment, as well as its chromatic effects on the unity of the architectural framework considering the environmental and cultural context. Totality built on the displacement of a subject subjected to the simultaneous action of space and time that is set in motion chromatically driven. Colour is a sensorial matter of the design of spatial and temporal relationships between architectural bodies, artistic pieces and environmental surroundings. In a first phase, and by means of six architectural mechanisms of chromatic control, the spatial structure of the appearance of colour is studied from a positional analysis that takes on plastic and constructive values. The degrees of plastic interaction and spatial functions assigned to the colours design chromatic atmospheres to be perceived by an observer who becomes an actor in the space. In a second stage, and applying the methodology of synthesis, the chromatic flows introduce time in space under schemes that integrate the Major Arts in architecture.
The observer interprets the chromatic flows in times designed as movements of a musical stave where the colours are spatially composed as sounds. The chromatic ordering of the spatial framework links colour with the movement of the observer who directly introduces the temporal dimension in a sensitive space, an intermediate void that is intuited as an interaction between built objects and artistic elements open to the surrounding physical environment under a structure of architectural logic.
The demonstration of the flow diagram as an objective method for designing with colour begins with a theoretical body in which its three general concepts are described: the technique of movement that arranges the space with chromatic flows, the design of an aesthetic of chromatic relationships which integrates the arts in the physical environment and the projection of chromatic flows by means of diagrams according to a logical and intuitive order. In the empirical body, the flow diagram is applied in three built-up areas of the University City to test it as a tool for the design and analysis of spatial colour in the project. Each case study corresponds to an organic division according to the university uses established by Villanueva: the Conjunto Estudiantil (to live), Conjunto Central (to cultivate the spirit) and Conjunto Facultad de Arquitectura y Urbanismo (to study). In 1948 Villanueva began a journey towards colour, experimenting and innovating with the chromatic phenomenon until he reached the Montreal Pavilion in 1968, when the colours represented Venezuela as a country.
In short, the main objective of the thesis is to verify that the flow diagram is a universal method for the analysis, representation and chromatic project of space-time. Villanueva chromatically idealises in black and white his interior space, without the need to explicitly draw the tones, saturations and values of its colours, thus maintaining the degree of abstraction required by a methodological application as a universal and scientific device
Improvements in high-speed rail network environmental evaluation and planning: An assessment of accessibility gains and landscape connectivity costs in Spain
High Speed Rail (HSR) in Spain began in 1992 and curently has a network of about 3400 km. In this 25-year period, accessibility has progressively increased, and its spatial distribution has changed throughout the territory. This has had important implications for territorial cohesion, which is a strategic planning objective of transport infrastructures in the European Union. However, this increase in HSR has environmental impacts; among other effects, new transport infrastructures imply a loss of landscape structural connectivity. This paper has two objectives: to study the distribution and development of the changes across the territory produced by the arrival of HSR in Spain between 1990 and 2025, by looking at changes in potential accessibility and structural habitat connectivity at the national scale; and to study – at the same scale – the relationship between both variables through the cost of accessibility improvements (socioeconomic variable) in terms of the loss of structural connectivity (environmental variable). All the analyses were done using GIS-based indicators, reported in the literature to be useful at the Strategic Environmental Assessment (SEA) scale. The methodology is applied in different horizon years, namely 1990, 2007, 2016, and new proposed HSR lines to 2025 in Spain. The results show that throughout the whole period (1990−2025), accessibility improved by 74.7 % due to the extension of HSR. The most important period was 1990−2007, when the main socio-economic centres were connected by HSR, while connectivity, the environmental variable, declined by −3.4 %. The greatest connectivity loss was concentrated in 2007−2016, when the areas with the highest connectivity values in 1990 were the most affected. Our results show that a 1 % increase in accessibility implied a loss of connectivity of 0.03 % in the interval 1990−2007, while in the second period, the same 1 % increase entailed a 0.17 % loss; in the 2016−2025 scenario the value is intermediate (0.08). Our work contributes to adapting connectivity studies to the environmental assessment tiers in the transportation sector (i.e. plans, programmes and projects). Each stage has its limitations due to the lack of specific data or problems of spatial scale which must be publicly reported during environmental assessment processes. We also highlight the need for new research to establish a reliable spatial scale to assess the different effects of HSR corridors at the strategic level
Use of supervised learning to predict suicidal and non-suicidal self harm risk
Suicide is a very prevalent, tragic public health concern. Artificial intelligence can be a helpful tool for predicting suicide attempts and identifying important factors in suicide risk. In the present work, we use four supervised machine learning techniques - random forest, K-nearest neighbors, classification trees, and generalized linear models - to classify individuals with suicide attempts. We use data from University College London’s longitudinal Millennium Cohort Study, which followed a cohort of individuals from age 9 months to age 17 and sampled data over 7 time points. We use variables from wave 6, (age 14) to predict suicidality at wave 7 (age 17). We find that the classification trees we created performed the best, with metrics in the following ranges: accuracy 90.52% – 92.23%; true positive rate 17.00% – 19.43%; true negative rate 95.75% – 97.64%; false positive rate 2.36% – 4.25%; false negative rate 80.57% - 84.64%. Additionally, through modelling, we identified a number of important variables that aligned with the literature, for example, history of self-harm, sex of the participant, and variables implicated in the emotional and behavioral functioning of the participant
Analysis of RNA Sequencing Data from Chicken DF-1 Cells under Acute and Persistent Infection by Infectious Bursal Disease Virus
Immune Bursal Disease Virus (IBDV) causes severe immunosuppression in chickens, causing an immense economic impact for the poultry industry worldwide. Members of the Birnaviridae family, including IBDV, show a dual behavior, producing both rapid acute and long-term persistent infections in the host. However, the mechanisms involved in persistent IBDV infection establishment have been poorly studied. RNA Sequencing emerged as a promising strategy to elucidate the mechanisms underlying persistent IBDV infection.
Transcriptome analysis, particularly differential expression analysis (DEA), has become pivotal in deciphering molecular mechanisms governing biological processes. edgeR, DESeq2, and limma have emerged as prominent approaches for DEA in RNA-Seq data. These tools empower researchers to uncover novel insights into gene regulation, disease mechanisms, and therapeutic targets, opening doors to transformative discoveries. However, the complexity of these programs can be a significant barrier for non-expert users without prior bioinformatic training.
We introduce EasyDEA, an automated and user-friendly tool for Differential Expression Analysis. EasyDEA been developed in close cooperation with active research groups to address real-world problems, and has been expanded as further needs arose, in line with agile development philosophy. EasyDEA allows non-expert users to perform effective DEA and pairwise comparisons in one single command. Additionally, it incorporates several highly demanded functionalities for downstream analysis, such as functional enrichment and clustering analysis, and generates easily interpretable output files and reports Furthermore, to demonstrate its effectiveness, we used EasyDEA to analyze real RNA-Seq data and elucidate the transcriptional mechanisms involved in acute and persistent IBDV infection, obtaining valuable results.
Even though further improvements need to be made in organization and modularity, EasyDEA is a highly effective tool for DEA, that provides robust, comparable, and reproducible results; thereby, promoting scientific advancements in the field of transcriptome research
Molecular and biological characterization of the mycovirome of botrytis cinerea and development of mycoviral reverse genetic systems
Botrytis cinerea es un hongo patógeno de plantas que causa podredumbre gris en más de 3000 especies de plantas, incluyendo más de 200 cultivos. Se encuentra ampliamente distribuido por todo el mundo y causa importantes pérdidas millonarias en cultivos como la vid, tomate, o pimiento, entre otros. Existen diversos factores que lo hacen uno de los principales hongos patógenos del mundo. En primer lugar, además de presentar una amplia gama de huéspedes, posee diversas formas de infección y supervivencia que incluyen esporas, micelio y esclerocios. Estas estructuras pueden infectar distintas partes de la planta en campo, y a los frutos en postcosecha. En segundo lugar, B. cinerea presenta una gran plasticidad genética que le permite generar resistencia a fungicidas en un tiempo corto. La principal medida de control de B. cinerea es la aplicación de fungicidas químicos, pero debemos encontrar alternativas más respetuosas con el medio ambiente, que eviten la generación de resistencias en el hongo, y que puedan ser usadas en estrategias de control integrado. En la búsqueda de soluciones alternativas más sostenibles el uso de micovirus se encuentra en pleno proceso de investigación.
Los micovirus son virus que infectan a hongos de manera natural. A pesar de que la mayoría de micovirus no presentan ningún efecto en el fenotipo observable del hongo, existen cada vez más ejemplos de micovirus que son capaces de reducir la virulencia de los hongos en la planta. Esta tesis tiene como objetivo caracterizar el micoviroma del hongo B. cinerea en aislados de campo procedentes de vid, para así poder proponer nuevas vías de control biológico basadas en micovirus.
En este trabajo se obtuvo una colección de aislados de campo de B. cinerea que se encontraron infectando racimos de uvas de viñedos de diferentes regiones de España (Jerez, La Rioja, Penedés y Ribera del Duero) y de Italia (Piemonte, Lombardia, Véneto, Basilicata y Sicilia). A partir de los aislados obtenidos, se llevó a cabo una caracterización biológica de los aislados de España para determinar su fenotipo y su virulencia, e identificar aislados hipovirulentos para, posteriormente, asociar la disminución de virulencia con la presencia de determinados micovirus. Los datos obtenidos muestran que algunos de los aislados son hipovirulentos, y que esa característica puede estar asociada con la presencia de micovirus. Por otra parte, se determinó el micoviroma de las colecciones de España e Italia mediante secuenciación masiva de ARN total (RNAseq). Los datos obtenidos de RNAseq fueron analizados mediante un pipeline bioinformático y se detectaron un total de 92 micovirus. Los micovirus identificados pertenecían a diversas familias de virus con genomas de ARN de simple cadena y sentido positivo (scARN(+)) y negativo (scARN(-)), de ARN de doble cadena (dcARN) y de ADN de cadena simple circular. Algunos de los virus identificados pertenecían a grupos que no habían sido asociados previamente a B. cinerea tales como narnavirus y micovirus tipo alfavirus, umbravirus y timovirus de scARN(+); virus bisegmentados y tetrasegmentados de dcARN; un virus trisegmentado de scARN (-) y un virus de DNA. Entre los resultados más relevantes obtenidos, se descubrieron los primeros micovirus trisegmentados relacionado filogenéticamente con los narnavirus, Botrytis cinerea binarnavirus 1, 2, 3 y 5 (BcBNV1, 2, 3, y 5), que tienen el gen que codifica para la RNA polimerasa dependiente de RNA (RdRp) dividido en dos segmentos; se identificó el primer virus trisegmentado tipo bunyavirus, Botrytis cinerea bocivirus 1, con segmentos genómicos monocistrónicos que codifican para una RdRp, una proteína de cápsida y una posible proteína de movimiento. Adicionalmente, se identificó el primer micovirus tetrasegmentado de simple cadena de ADN circular perteneciente a la familia Genomoviridae, Botrytis cinerea ssDNA virus 1 (BcssDV1). BcBNV2 y BcssDV1 han sido caracterizados molecularmente en detalle en esta tesis doctoral. Adicionalmente, se construyó un clon de cDNA basado en BcBNV2 para determinar los requerimientos del virus para replicar su genoma usando dos proteínas independientes que deben constituir una RdRp funcional. Esta parte no se ha completado con éxito debido a las dificultades asociadas a la construcción de un clon infecciosos basado en un virus multisegmentado.
El desarrollo de sistemas genéticos reversos basados en micovirus es útil para realizar estudios de genómica funcional del hongo. Para ello se caracterizó molecularmente un virus previamente identificado infectando B. cinerea en distintas partes del mundo, Botrytis virus F (BVF), y se usó para construir un clon infeccioso. La construcción del clon infeccioso fue exitosa y permitió determinar la región promotora de la síntesis del RNA subgenómico para la traducción de su proteína de cápsida, y el efecto de la infección sobre el fenotipo de dos cepas del hongo, una cepa de laboratorio libre de virus, y una cepa de campo infectada con dos virus. El no efecto sobre el fenotipo del hongo, o la disminución de la virulencia de la cepa infectada con otros micovirus, le convierten en un candidato ideal para el desarrollo de un vector de expresión viral o de un vector de inducción de silenciamiento génico, para usarlo en biotecnología o en estrategias de control biológico.
En resumen, los resultados de esta tesis han permitido expandir el conocimiento de la gran diversidad micoviral existente, de la transferencia horizontal de micovirus entre diferentes hongos, pero también de eventos de cruces de virus entre reinos, frecuentemente, hongos y plantas; y generar herramientas que puedan ser usadas en estrategias de control biológico y para el estudio de las interacciones entre micovirus, hongos y plantas.
ABSTRACT
Botrytis cinereais a plant pathogenic fungus that causes gray mold on more than 3000 plant species, including over 200 crops. It is widely distributed worldwide and causes significant economic losses in crops such as grapevine, tomato, and pepper, among others. Several factors contribute to its status as one of the world's leading fungal pathogens. Firstly, it has a wide range of hosts and various infection and survival methods, including spores, mycelium, and sclerotia. These structures can infect different parts of the plant pre-harvest and fruits post-harvest. Secondly, Botrytis cinereaexhibits great genetic plasticity, enabling it to develop resistance to fungicides in a short period. Chemical measures are the main control method for B. cinerea;however, we must seek more environmentally friendly alternatives that prevent the development of resistance in the fungus and can be employed in integrated pest management strategies. In the search for more sustainable alternatives, the use of mycoviruses is currently under investigation.
Mycoviruses are viruses that naturally infect fungi. While most mycoviruses do not affect the observable phenotype of the fungus, there are increasing instances of mycoviruses capable of reducing the virulence of fungi in plants. This thesis aims to characterize the mycovirome of the fungus Botrytis cinereaisolated from grapevine. From these results, the goal is to propose new methods of biological control based on mycoviruses.
In this study, we set a collection of B. cinereaisolates from infected grape bunches of vineyards from different regions of Spain (Jerez, La Rioja, Penedés y Ribera del Duero) and Italy (Piemonte, Lombardia, Veneto, Basilicata y Sicilia). Based on the obtained isolates, a biological characterization of the Spanish isolates was conducted to determine their phenotype and virulenceandidentify hypovirulent isolates in order to subsequently correlate the decrease in virulence with the presence of specific mycovirus. Resulted data showed some isolates were hypovirulent, which may be associated with the infection of mycoviruses. Additionally, the mycovirome of the Spanish and Italian collections was determined by high throughput sequencing total RNA (RNAseq). RNAseq data were analysedfollowing a bioinformatic pipeline, revealing a total of 92 mycoviruses. The identified mycoviruses belonged to diverse families of viruses with positive single-stranded RNA (ssRNA(+)) and negativesingle-strandedRNA (ssRNA(-)) genomes, double-stranded RNA (dsRNA) genomes, and single-stranded circular DNA genomes.
Some of the identified viruses were from clades not previously associated with B. cinerea, such as narnaviruses and alpha-like, umbra-like, and tymo-like mycoviruses of ssRNA (+) genomes; bisegmented and tetrasegmented viruses of dsRNAgenomes; a trisegmented virus of ssRNA(-) and a DNA virus. Among the most relevant results obtained, the first trisegmented mycoviruses phylogenetically related to narnaviruses were discovered. These mycoviruses, Botrytis cinerea binarnavirus 1, 2, 3, and 5 (BcBNV1, 2, 3, and 5), have the gene encoding for RNA-dependent RNA polymerase (RdRp) split into two segments. It was also identified the first trisegmented virus of the bunyavirus type, Botrytis cinerea bocivirus 1, which has monocistronic genomic segments encoding for an RdRp, a capsid protein, and a movement protein. Furthermore, it was described the first tetrasegmented mycovirus with a circular single-stranded DNA genome belonging to the Genomoviridaefamily, Botrytis cinerea ssDNA virus 1 (BcssDV1).
BcBNV2 and BcssDV1 have been molecularly characterized in detail in this doctoral thesis. Additionally, acDNA clone based on BcBNV2was constructed to determine the virus's requirements for replicating its genome using two independent proteins that must constitute a functional RdRp. This part has notbeen successfully completed due to the difficulties associated with constructing an infectious clone based on a multisegmented virus.
The development of reverse genetic systems based on mycoviruses is useful to conduct functional genomics studies of the fungus. To achieve this, a previously identified virus infecting B. cinereafrom different parts of the world, Botrytis virus F (BVF), was molecularly characterized and used to construct an infectious clone. The construction of the infectious clone was successful, allowing to determine the promoter region for the synthesis of subgenomic RNA for the translation of its capsid protein, and the effect of infection on the phenotype of two strains of the fungus: a virus-free laboratory strain and a field strain infected with two viruses. The absence of an effect on the fungal phenotype, or the reduction of virulence in the strain infected with other mycoviruses, makes it an ideal candidate for the development of a viral expression vector or a gene silencing induction vector, for its use in biotechnology or biological control strategies.
In summary, the results of this thesis have expanded our knowledge of the extensive diversity of mycovirusesandhorizontal transfer of mycoviruses between different fungi, as well as cross-kingdom virus events, often involving fungi and plants. Moreover, this research has produced tools that can be utilized in biological control strategies and to study interactions among mycoviruses, fungi, and plants
Diseño de un emulador de la tecnología en simulador de protocolos anestésicos mediante el análisis de calidad del monitor BIS™
Resumen:
La especialidad de la anestesiología se considera hoy día primordial y de alta importancia en el campo de la medicina, no obstante, ha sufrido una evolución desde sus comienzos, de forma que con el avance de los estudios científicos y la tecnología el paciente está menos expuesto a sufrir daños colaterales debido a una intervención quirúrgica cuando se le somete, por ejemplo, a anestesia general. Así mismo, garantizar la integridad del paciente durante y tras la intervención quirúrgica es en la actualidad una tarea rutinaria realizada por el personal médico, anestesista en este caso, que acompaña a cada paciente antes, durante y posterior a su paso por quirófano.
El presente trabajo aborda el estudio que parte del equipamiento tecnológico empleado hoy día en el quirófano para monitorizar la profundidad de hipnosis del paciente, considerada como variable no medible, y que es supervisada por parte del médico anestesista. El equipamiento, monitor BIS™ (Bispectral Index) VISTA es una herramienta que, mediante el procesamiento del electroencefalograma (EEG), es capaz de indicar en una escala de 0 a 100 el grado de hipnosis del paciente.
Aunque los productos puestos en el mercado ya han sido supervisados y depurados por el personal experto, tanto médico como desde el punto de vista de la ingeniería, cabe la posibilidad de que el grado de hipnosis del paciente mostrado al personal médico se vea distorsionado por una serie de artefactos, como por ejemplo el mero hecho de emplear el bisturí para realizar una incisión sobre el paciente. Esta es, por tanto, uno de los puntos que se han analizado en este documento, para proponer una primera solución que, sumado a la experiencia del anestesista, tiene como objetivo que la información del estado del paciente no se vea distorsionada.
Dado que no es posible su implementación directa en el plano quirúrgico para no comprometer la integridad física de los pacientes, se emulará la parte del equipamiento tecnológico sobre un Simulador de Protocolos Anestésicos (SPA) desarrollado por el Grupo de Bioingeniería y Telemedicina (GBT) de la Universidad Politécnica de Madrid (UPM) en colaboración con el Servicio de Anestesiología del Hospital Universitario Infanta Leonor (HUIL). Por último, se dotará a dicho simulador de los mecanismos que permitan paliar las alteraciones promovidas por los artefactos anteriormente citados con el fin de que se disminuyan y faciliten, sin pretensión de suplantarlas, la toma de decisiones del personal clínico, que en este caso corresponden al médico anestesista.
Abstract:
Anaesthesiology specialty is nowadays considered essential and very important in the health field. Nevertheless, has been suffered an evolution from the beginnings, so that with the development of the scientific studies and the technology the patient is less exposed to suffer collateral damages because of a surgical treatment when, for example, general anaesthesia is used. In this term, guarantee the integrity of the patient during and after the surgical treatment is in the actuality a daily task done by the doctors, anaesthetist in this case. This figure supervises any patient before, during and after he passed through the operation theatre.
This Project studies the part of the technology equipment used today in the operation theatre to monitor the depth of the hypnosis of the patient, considered as a variable that cannot measure, and it is supervised by the anaesthetist. The equipment, monitor BIS™ (Bispectral Index) VISTA is a tool capable of indicating on a scale 0 to 100 the depth of the hypnosis of the patient through processing the electroencephalogram (EEG).
Although the products commercialized has been approved and debugged by the experts, also doctors and engineers, does the possibility exists that the Depth of hypnosis of the patient showed to the medical personnel could be distorted by an artifact series. E.G: the use of a scalpel to practise an incision to the patient. This is one of the main points analysed in this document to propose a solution that, added to the experience of the anaesthetist, have the objective that the health of the patient did not be affected.
Instead of implementing it directly in the surgical field to not affect the physical integrity of the patients, the technology equipment will be emulated through the Anaesthetic Protocols Simulator developed by Grupo de Bioingenieria y Telemedicina (GBT) of the Technical University of Madrid in collaboration with the Anaesthesiology Service of the Hospital Universitario Infanta Leonor (HUIL). Finally, the simulator will have diverse mechanisms that allow to solve the alterations of the artefacts mentioned before with the purpose of decrease and facilitate, without the intention of supplant, the decision making of the medical personnel, that in this case corresponds to the anaesthetist
Students’ attitude and teachers’ perspective towards CLIL programs: Spanish secondary school bilingual programs
El estudio investiga la aplicación de programas bilingües en la Educación Secundaria española porque consideramos crucial, en esta etapa avanzada del uso de los programas AICLE en España, analizar y estudiar algunos aspectos de este método con el fin de enfatizar las pautas de buenas prácticas y obtener un conocimiento profundo de lo que se está haciendo en este campo (Coyle, 2007). Uno de los dos objetivos básicos de este estudio era ofrecer un análisis de perspectivas múltiples de esta implementación bilingüe para conocer la actitud de los estudiantes hacia los programas bilingües. El otro objetivo era estudiar las perspectivas de los maestros sobre este programa. Convencidos de que el cambio introducido por CLIL va más allá del concepto “solo hazlo en inglés” (Lasagabaster & Sierra, 2010), esta investigación cuantitativa utilizó cuestionarios para recopilar datos de los docentes para mostrar cómo se percibe la metodología CLIL dentro de las aulas más allá de lo teórico. Este artículo forma parte de una línea de investigación en lingüística aplicada que surgió a partir de dos proyectos anteriores centrados en el estudio de la lingüística de corpus a partir de la producción de los alumnos del master bilingüe. Dichos proyectos son: "Análisis de patrones de errores gramaticales, estilísticos y de léxico en lengua inglesa a partir de producciones escritas de inmigrantes digitales" (Convocatoria 2014-2015, Universidad Internacional de La Rioja) y "Detección y análisis del comportamiento lingüístico de producciones escritas de estudiantes universitarios" (Referencia del proyecto: B0036-1617-104-ETEL. Universidad Internacional de La Rioja, 2016-2018). Los resultados del estudio fueron alentadores para seguir utilizando los programas bilingües en la Educación Secundaria en España porque muestran que los estudiantes tienen una actitud positiva hacia el aprendizaje del inglés. La actitud del profesorado también ha sido positiva. La investigación ofrece conclusiones muy importantes que revelan que las necesidades de los profesores son diversas para enfrentarse a este desafío en sus aulas. En este caso destacamos el resultado inesperado del estudio que es el acuerdo entre los participantes de que los cursos de formación recibidos no son suficientes y que incluso algunos no son muy útiles. También es importante indicar que los resultados indican que CLIL proporciona un ambiente de aprendizaje positivo capaz de mejorar la motivación de los estudiantes para aprender inglés y otras materias, sin embargo, todavía hay mucho por hacer para ayudar al profesorado a sobresalir en su rol y garantizar mejores resultados de aprendizaje para los alumnos. Las implicaciones de estos resultados son muy importantes en el diseño de los cursos de formación para el profesorado que forma parte de los programas CLIL y van más allá de lo que recibe el profesor cuando esté ejerciendo su labor con el fin de mejorar el dominio de la lengua inglesa, y por eso propone que este camino empieza durante la carrera universitaria del profesor en la que hay que incluir asignaturas impartidas completamente en inglés como una forma de prepararle a enfrentarse al reto de impartir sus clases en inglés
Implementación y Desarrollo de un sistema IoT para la monitorización de las condiciones ambientales en un hospital

La evolución exponencial que ha vivido el Internet of Things ha permitido llevar a cabo multitud de proyectos revolucionarios en diversos ámbitos de investigación. Uno de esos ámbitos, el Internet of Medical Things, busca mejorar los procesos de detección, diagnóstico y tratamiento de enfermades. Dotar a los dispositivos que se encargan de estos procesos de la capacidad de conectarse a internet y enviar información en tiempo real, permite apreciar la gran importancia del IoMT para el campo de la medicina en nuestros días.
El propósito general de este Proyecto Fin de Grado titulado “Implementación y Desarrollo de un sistema IoT para la monitorización de las condiciones ambientales en un hospital” es el de diseñar e implementar un sistema capaz de tomar medidas de ciertas condiciones ambientales y de la calidad del aire en hospitales, para su posterior visualización en ThingsBoard y en una aplicación móvil creada desde cero para el sistema operativo Android, abriendo las puertas a un posterior análisis que permita mejorar la recuperación del paciente.
El sistema se ha desarrollado de forma que sea fácilmente escalable y la aplicación móvil visual e intuitiva para ayudar a que haya una rápida visualización de la información. En la presente memoria se puede encontrar la información necesaria para comprender el diseño, implementación y pruebas realizadas en el sistema.

The exponential evolution that the Internet of Things is experiencing has allowed a multitude of revolutionary projects to be carried out in various fields of research. One of these areas, the Internet of Medical Things, seeks to improve the processes of detection, diagnosis, and treatment of diseases. Providing the devices that are responsible for these processes with the ability to connect to the internet and send information in real time allows us to appreciate the great importance of the IoMT for the field of medicine today.
The general purpose of this Final Degree Project entitled "Implementation and Development of an IoT system for monitoring environmental conditions in a hospital." is to design and deploy a system capable of taking measurements of certain environmental conditions and air quality in hospitals, for subsequent visualization on ThingsBoard and in a mobile application created from scratch for the Android operating system, fostering a further analysis that will let improve patients’ recovery.
The system has been developed to be easily scalable and the mobile application visual and intuitive to aid rapid visualization of the information. In this report you can find the necessary information to understand the design, implementation, and testing of the system
Evaluación económica de la restauración hidrológico forestal como herramienta para la gestión de la sedimentación en embalses
Los embalses del mundo están perdiendo su capacidad a una tasa anual de 0,5 - 1% por año debido a los procesos de sedimentación. Las estrategias para revertir esta tendencia son variadas, si bien pueden agruparse básicamente en dos opciones: reducir la cantidad de sedimentos que llegan a los embalses a través de la ejecución de medidas en sus cuencas vertientes o la gestión de dichos sedimentos una vez depositados en el vaso de los embalses mediante las técnicas actualmente disponibles.
La primera opción, se basa en la Restauración Hidrológico Forestal (RHF) entre cuyos principales objetivos está desarrollar coberturas vegetales que protejan el suelo de la cuenca, reduciendo la emisión de sedimentos hacia los embalses. La segunda opción la componen, sobre todo, las técnicas de dragado de sedimentos así como las medidas necesarias para el mantenimiento de las infraestructuras y equipos de los embalses y sus costes asociados.
En esta tesis se lleva a cabo la comparación, desde un punto de vista financiero, de estas dos opciones.
La investigación parte del desarrollo de un modelo empírico y estadístico basado en los datos del CEDEX (Centro de Estudios y Experimentación de Obras Públicas de España) sobre el estudio de 131 embalses durante el período 1967 - 2004, en el que se elaboraron 272 informes sedimentológicos y batimétricos. A partir de este información, se ha desarrollado un modelo de SSY (Specific Sediment Yield o Producción Específica de Sedimentos, en t ha-1 año-1) para cuencas de tamaño medio.
Del conjunto de embalses analizados, se seleccionan cuatro de ellos, Puente Nuevo (Guadalquivir), Guadalmellato (Guadalquivir), Fuensanta (Segura) y San Juan (Tajo) para simular los efectos sobre la sedimentación de las medidas de RHF y de gestión de sedimentos en el embalse, y compararlos mediante técnicas de análisis financiero de proyectos (en este caso se ha seleccionado utilizar como indicador el periodo de payback, se han considerado los tipos de interés del 3%, 5% y 7%, y que los efectos de la RHF evolucionan linealmente hasta alcanzar su objetivo protector pasados 20 años).
El resultado obtenido muestra que las medidas de RHF, en este caso la repoblación forestal, son especialmente indicadas sobre superficies con ninguna o poca protección del suelo (sobre 2.797 ha afectadas por un incendio en la cuenca del embalse de Fuensanta) donde la inversión es compensada en menos de 10 años para los tres tipos de interés por los ahorros en costes.
Cuando la repoblación forestal se extiende sobre una superficie suficientemente grande de la cuenca y su incidencia sobre la protección del terreno es significativa, caso de los embalses de Puente Nuevo y San Juan (donde se repueblan 12.337 ha 15.863 ha respectivamente), de nuevo la RHF es una buena opción desde el punto de vista financiero, alcanzando plazos de pay-back de 32 y 21 años al 3% respectivamente y de 26 años al 5% en San Juan.
Para el resto de situaciones, incluidas todas las simuladas en el embalse de Guadalmellato, el pay-back de la inversión en RHF no se alcanza en un plazo financieramente razonable.
Finalmente destacar que, aunque no sea el objeto de esta tesis el análisis de las externalidades de la RHF, la consideración de unos valores medios de 108 € ha-1 año-1 descritos en la bibliografía consultada como beneficios obtenidos de los servicios de los ecosistemas forestales, permiten casi todas las opciones de RHF analizadas resulten financieramente atractivas frente al coste y efectos de la gestión de sedimentos en embalses.
ABSTRACT
The world's reservoirs are losing their capacity at an annual rate of 0.5 - 1% per year due to sedimentation processes. The strategies to reverse this trend are varied, although they can basically be grouped into two options: the reduction of the amount of sediment that reaches the reservoirs through the execution of measures in their catchment basins or the management of those sediments once deposited in the bed of the reservoirs by means of currently available techniques.
The first option is based on Forest Hydrological Restoration (RHF, by its acronym in Spanish) whose main objectives are to develop vegetation covers that protect the soil of the watershed, reducing the emission of sediments towards the reservoirs. The second option is made up, above all, of sediment dredging techniques as well as the necessary measures for the maintenance of the infrastructure and equipment of the reservoirs and their associated costs.
In this thesis, the comparison is carried out, from a financial point of view, of these two options.
The research is based on the development of an empirical and statistical model based on data from the CEDEX (Center for Studies and Experimentation of Public Works of Spain) on the study of 131 reservoirs during the period 1967 - 2004, in which 272 sedimentological and bathymetric reports were prepared. Based on this information, a SSY model (Specific Sediment Yield, in t ha-1 year-1) has been developed for mediumsized basins.
Four of them, Puente Nuevo (Guadalquivir), Guadalmellato (Guadalquivir), Fuensanta (Segura) and san Juan (Tajo) are selected from the set of reservoirs analysed, to simulate the effects on sedimentation of RHF and sediment management measures in the reservoir, and to compare them through project financial analysis techniques (In this case, the pay-back period has been selected as an indicator, the interest rates of 3%, 5% and 7% have been considered, and that the effects of the RHF evolve linearly until reaching its protective objective after 20 years).
The result obtained shows that RHF measures, in this case reforestation, are especially appropriate on surfaces with little or no soil protection (over 2,797 ha affected by a fire in the Fuensanta reservoir basin) where the investment is compensated. in less than 10 years for the three interest rates due to cost savings.
When the reforestation extends over a sufficiently large area of the basin and its incidence on the protection of the land is significant, in the case of the Puente Nuevo and San Juan reservoirs (where 12,337 ha and 15,863 ha respectively are reforested), the RHF is again a good option from a financial point of view, reaching pay-back terms of 32 and 21 years at 3% respectively and from 26 years to 5% in San Juan.
For the rest of the situations, including all those simulated in the Guadalmellato reservoir, the pay-back of the investment in RHF is not reached within a financially reasonable period.
Finally, it should be noted that, although the analysis of the externalities of the RHF is not the object of this thesis, the consideration of average values of 108 € ha-1 year-1 described in the bibliography consulted as benefits obtained from ecosystem services forestry, allow almost all the RHF options analyzed to be financially attractive compared to the cost and effects of sediment management in reservoirs
