Ulcerative Colitis and Alcohol: Facts and Myths
Ulcerative colitis (UC) is a chronic inflammatory disease of the colon and rectum, characterized by symptoms such as diarrhea, hematochezia, abdominal pain, and distension. The etiology of UC remains unclear, but it is believed to result from a combinationof immune system, environmental, and genetic factors. Among environmental influences, dietary and lifestyle choices, including alcohol consumption, have been scrutinized for their potential impact on UC. Alcohol, a widely consumed beverage, has complex effects on the gastrointestinal system. While moderate alcohol intake is often associated with certain health benefits, its role in inflammatory bowel diseases like UC is contentious. Research indicates that alcohol can exacerbate UC symptoms by altering gutmicrobiota, increasing intestinal permeability, and enhancing inflammatory responses. This review explores the multifaceted relationship between alcohol consumption and UC, highlighting how alcohol can disrupt the gut barrier, modify microbiota composition, and interact with UC medications, potentially diminishing their efficacy. Despite some studies suggesting that certain types of alcohol, such as red wine, may have anti-inflammatory properties, the consensus is that individuals with UC should approach alcohol consumption with caution. The variability in individual responses to alcohol further complicates this issue. This review underscores the need for more research to fully understand the interplay between alcohol and UC and to provide clear guidelines for patients. Until then, it is advisable for UC patients to consult healthcare professionals to make informed decisions regarding alcohol use

Development and characterization of surface solid dispersion Moringa oleifera leaf extract-microcrystalline cellulose by co-grinding method
Context: Moringa oleifera leaf extract is rich in flavonoid compounds and potential antioxidant sources. However, the development of M. oleifera leaf extract into an herbal product is limited by challenges such as inconsistent quality, hygroscopicity, and poor solubility. Surface solid dispersion (SSD) is a versatile method to enhance both the physical properties and solubility of such extracts.
Aims: To enhance the physical and solubility properties of M. oleifera leaf extract using SSD with microcrystalline cellulose as a carrier.
Methods: SSD was prepared using a co-grinding method with extract-to-carrier ratios of 1:2 and 1:4.
Results: This study revealed that the SSD effectively absorbed the extract onto the surface of microcrystalline cellulose. Hence, the SSD powder exhibited improved moisture content and flowability. Thermal and crystallinity analyses revealed a reduction in crystal lattice regularity compared to the pure extract, as evidenced by decreased enthalpy and diminished peak intensity. The total flavonoid content of SSD 1:2 was 14.70 ± 0.35 mg QE/g, whereas SSD 1:4 was 7.00 ± 0.07 mg QE/g. Flavonoid solubility compound from SSD 1:2 was 62.26 ± 0.62 μg/mL, while the solubility of SSD 1:4 was 63.58 ± 0.62 μg/mL. The solubility of the flavonoid compound from SSD preparation increased about 1.16-fold compared to the physical mixture.
Conclusions: SSD preparation of M. oleifera leaf extract improves its physical and chemical properties, making it more suitable for formulation

Genetic Polymorphisms as Treatment Biomarkers for Gynecological Malignancies Treated With Carboplatin and Paclitaxel: A Systematic Review
Purpose
Gynecological tumors, which correspond to the group of neoplasms that affect the female reproductive system, have high incidence and mortality rates. This systematic review aimed to summarize the most recent advances in identifying pharmacogenetic variants associated with the clinical outcomes of carboplatin-paclitaxel chemotherapy in these patients.
Methods
A comprehensive literature search was conducted across eight databases to identify studies published up to July 17, 2024. Two reviewers independently selected the studies and extracted the data; disagreements were resolved by two additional reviewers.
Findings
Out of the 2375 records that were found, only 20 met the eligibility criteria. The main findings were: (1) The three most extensively investigated genes were ATP binding cassette subfamily C member 1 (ABCB1), cytochrome P450 2C8 (CYP2C8), and glutathione S-transferase P1 (GSTP1); (2) three variants, rs1128503 (ABCB1), rs10509681 and rs11572080 (CYPC28), appear to have a significant association with important adverse drug reactions (in particular, neutropenia, thrombocytopenia, and peripheral sensory neuropathy). Others, as is the case with rs1045642 (ABCB1) and rs1695 (GSTP1), have inconsistent results, and the extent to which these results can be extrapolated is still limited; and (c) most of the included studies concerned Asian or European patients.
Implications
Therefore, future research should include more extensive analyses with more inclusive cohorts. As a limitation of the study, a meta-analysis was not possible due to the significant heterogeneity among the studies
Optimization of Silver Nanoparticle Dermal Patch Films for Enhanced Wound Healing: Formulation and Characterization Study
Background: Silver nanoparticles (AgNPs) are known for their potent antibacterial properties, making it suitable for wound healing applications.
Aims: This study focuses on formulating AgNPs into dermal patch films DPFs), leveraging the adhesive properties of the film for the effective delivery of active ingredients.
Methods: AgNPs were synthesized through a green synthesis method using Plantago major L. Leaf extract as a bio-reductant. Five distinct formulations, ranging from AgNPs concentration of 0% (control); 0,005%; 0,01%; 0,05%; and 0,10%, were optimized and denoted as Formula 1 to 5 (F1-F5), respectively. The films were fabricated by solvent casting method employing a manual film applicator. A variety of evaluations were then performed on the films, including assessments of their physical and chemical characteristics. These characteristics included organoleptic properties, film thickness, folding endurance, surface pH, loss on drying (LOD), crystallinity, the interaction between active ingredients and excipients, the morphological characteristics of the films, and a wound healing study.
Results: All formulations resulted in smooth and transparent films. Favorable outcomes were observed in film thickness and surface pH measurements. Formulations F1-F4 demonstrated exceptional folding endurance (>200 times). This is also affirmed by a reduction in the -OH peak in the Fourier transform infrared (FTIR) spectrum. Powder X-Ray diffraction (PXRD) analysis showed that F1-F4 had adopted an amorphous structure, while F5 retained crystalline AgNPs. The drying process revealed that F5 exhibited the lowest moisture loss. Scanning electron microscope (SEM) imaging displayed distinct morphologies among the five formulations. F4 and F5 exhibited the highest percentage of wound healing.
Conclusion: The formulation of AgNPs synthesized through a green synthesis method, utilizing Plantago major L. leaf extract as a bio-reductant, has demonstrated significant improvements in the physical characteristics, particularly in formulations F1-F4. Notably, F4 exhibited the highest wound healing efficacy. Therefore, the findings of this study suggest that F4 (AgNPs 0.05%) represents the most promising dermal patch films formulation for enhanced wound healing applications
