Sparse grid approximation of the Riccati equation
In this article, we study the sparse grid discretization for the numerical solution of the algebraic Riccati equation (ARE). This approach is of particular interest for the solution of large scale AREs. Such AREs arise, for example, from the discretization of the operator Riccati equations associated with the linear quadratic control of systems evolving in a Hilbert space . Following [4, 45], we formulate the ARE as a nonlinear operator equation on the space of Hilbert–Schmidt operators and derive the matrix equation for the sparse grid discretization. Provided that degrees of freedom are used to discretize the space , the sparse grid approximation of the ARE requires degrees of freedom. Especially, we propose an algorithm that evaluates the approximated ARE with operations. This considerably reduces the cost of solving the ARE compared to the memory requirement and complexity of the regular tensor product discretization. Numerical results are presented to validate the approach
Safety assessment of herbal drugs as alternatives to treat non-psychotic mental diseases during pregnancy
Pregnancy is an extraordinary state for the expecting mother. Her body goes through many
changes to bring forth new life. Amidst the joy of bearing a child, up to 20% of pregnant women
will suffer from depression. Other ailments are anxiety, restlessness, stress, and sleeping issues,
which are categorized as non-psychotic mental diseases (NMDs). In order to prevent further
discomfort for the expecting mothers and negative birth outcomes like preterm birth or
preeclampsia, medications are prescribed to mitigate the symptoms. Typical treatments are
selective serotonin reuptake inhibitors (e.g., citalopram) and benzodiazepines (e.g., diazepam)
to treat mild depression and anxiety, respectively. Those synthetic drugs are for instance known
to cross the placental barrier, which might be one of many factors having physicians and
pregnant women discuss possible alternatives. Alternatives to treat NMDs are valerian,
lavender, hops, St. John’s Wort (SJW), and California poppy. Treatment with herbal medication
is as old as the history of medicine itself and remedies of natural origin have ever since been
considered trustworthy. This notion is also reflected in the different handling of synthetic drugs
versus herbal ones. The regulations worldwide differ. In some countries herbal medications are
categorized as dietary supplements. Although the Committee on Herbal Medicinal Products has
regulations in place for herbal medications; data on those products are often missing and most
herbal medication are simply labelled as not recommended during pregnancy and lactation.
The high trust in herbal medications and the lack of safety data at the same time, potentially
expose pregnant women and their unborn child to unknown risks.
To assess the safety of valerian, hops, SJW, and California poppy we first established
bioanalytical methods for the compounds attributed to the therapeutic effect, namely valerenic
acid, humulone, hyperforin, hypericin, and protopine. Lavender and its volatile constituent
linalool have been excluded from the following studies.
In a first step the bioanalytical methods allowed determination of foetal exposure to those single
compounds in an ex vivo placental perfusion model. We could show that protopine and valerenic
acid crossed the placental barrier, while hyperforin only transferred to a small extent into the
foetal circuit. Hypericin on the other hand, accumulated in the placenta.
To make an informed safety assessment, knowledge of potential metabolites is also needed. We
therefore investigated phase I and phase II metabolism for valerenic acid, protopine, and
humulone in human liver microsomes. For valerenic acid, we were able to identify five
dehydrovalerenic acid isomers, valerenic acid glucuronide, and oxoglucuronide. Protopine was
metabolised to dimethylcryptopine, and to a metabolite formed by catechol-O-methylation and
a third metabolite formed by ketone reduction. Finally, for humulone we could identifiy eleven
hydroxyhumulone isomers, three dihydroxyhumulone isomers, and a metabolite formed by
aliphatic hhydroxylation and ketone reduction. Those metabolites have been described in
human liver microsomes for the first time.
The third part of the project was dedicated to screening for potential mitochondria targeted liver
toxicity in HepG2 and HepaRG cells. Hypericin was, in term of toxic effects, the most promising
candidate. Hypericin turned out to be a potent mitochondria toxicant. It showed inhibition of
complexes I, III, and IV in the electron transport chain and a decrease in mitochondrial ATP
production, which leads to mitochondrial reactive oxygen species accumulation. Nrf2 was
activated, which eventually leads to necrosis.
In conclusion we could show that the foetus is directly exposed to valerenic acid and protopine,
while exposure to hyperforin is minimal. Further studies to determine the potential harm for the
unborn are still needed. An investigation for possible influences of the accumulation of
hypericin in the placenta (change in the viability of the placenta and therefore consequences for
the foetus) also needs to be carried out further since we now know the toxicant potential of
hypericin. Additionally, a study of the identified metabolites and their interactions with the
body are necessary. Finally, an assessment of the full extracts in vitro and in vivo is required
since those single compounds are rarely consumed alone, but as part of complex matrix in the
herbal medication.
With the obtained results we cannot conclude that the investigated compounds or the full
extracts are safe
Combination therapy against soil-transmitted helminthiasis: from egg excretion patterns to treatment effects
Soil-transmitted helminths (STHs) are parasitic nematodes dwelling in the intestinal tract of
their hosts. The four major worms infecting humans are Ascaris lumbricoides, Trichuris trichiura, hookworm and Strongyloides stercoralis. Infections with STHs belong to the group of Neglected Tropical Diseases (NTDs), which notoriously affect the most impoverished populations in subtropical and tropical regions. An estimated 1.5 billion people are infected worldwide, causing a considerable disease burden and exacerbating the vicious cycle of socio-economic struggles and disease. The World Health Organization (WHO) promotes global coordinated control efforts with preventive chemotherapy (PC) as the central strategy. This approach primarily aims at curbing heavy infections, however the ultimate goal goes beyond morbidity control to elimination of STH infections as a public health problem. Single-dose albendazole and mebendazole are the drugs of choice to treat STH infections in PC schemes. Both drugs have excellent safety profiles and work well against A. lumbricoides infections, however the efficacy against hookworm is merely moderate and neither of the drugs are sufficiently efficacious in reducing T. trichiura infections. The co-administration of two or more drugs is a promising strategy to increase the efficacy of treatments. Furthermore, re-purposing drugs applied for other pathogens may be a comparatively fast and effective way to broaden the currently available treatment options against STHs.
The core work of this PhD centered on two randomized controlled trials (RCTs) assessing the efficacy and safety of two combination treatments against infections with T. trichiura. One objective was to contribute to a multi-country trial investigating the combination treatment of ivermectin-albendazole in comparison to albendazole monotherapy in children and adults between 6 and 60 years of age in three settings: Pemba Island, Tanzania in East Africa, Côte d’Ivoire in West Africa and Lao PDR in Southeast Asia. The results found the combination treatment to be superior to albendazole on Pemba Island and in Lao PDR, with CRs of 49% vs. 6% and 66% vs. 8%, respectively. In Côte d’Ivoire however, the combination treatment did not result in a superior efficacy to albendazole alone.
A sub-study conducted within the framework of the multi-country trial in Lao PDR had the aim of determining the optimal timepoint at which to collect stool samples for the assessment of anthelminthic drug efficacies. For this, the daily egg excretion patterns of STHs during 28 days following ivermectin-albendazole and albendazole treatment were analyzed. The assessment of samples collected at 14-21 days post-treatment could be confirmed as an adequate timeframe for an accurate representation of the true efficacy.
A second RCT assessed the safety and efficacy of the combination treatment of moxidectin-albendazole in comparison to ivermectin-albendazole in adolescents infected with T. trichiura on Pemba Island, Tanzania. Non-inferiority by two percentage points in terms of egg reduction rate (ERR) could not be shown, with 96.8% (95% CI 95.8 – 97.6) for moxidectin-albendazole in comparison to 99.0% (95% CI 98.7 – 99.3) for ivermectin-albendazole. The moxidectin-albendazole combination treatment resulted in a higher CR compared to moxidectin and albendazole administered in monotherapy, with the latter difference not being statistically significant. Combined ivermectin-albendazole led to significantly higher CRs than the respective ivermectin and albendazole monotherapies.
In conclusion, the successes achieved by PC to date should be expanded in order to meet the WHO 2030 targets for STH infections and to move from morbidity control towards elimination. Improved treatment options should be incorporated into PC schemes delivered via MDA programs. The results presented in this thesis confirmed the ivermectin-albendazole combination therapy to be superior in terms of CRs against T. trichiura compared to albendazole monotherapy in two of the three settings of the multi-country study. Furthermore, even though non-inferiority could not be shown for the moxidectin-albendazole combination treatment compared to ivermectin-albendazole in terms of ERRs among adolescents infected with T. trichiura on Pemba Island, further research in different settings is warranted as adding moxidectin to the albendazole standard treatment might be a valuable option in areas in which ivermectin is not available. The NTD landscape needs to be carefully considered in order to find the optimal treatment option for each setting to potentially target various pathogens and to enhance treatment efficacy
Physiologische und neuromuskuläre Veränderungen aufgrund verschiedener Neigungen (geradeaus, bergauf, bergab) und akut ermüdeter Bauch- und Beinmuskulatur beim Laufen auf dem Laufband
Hintergrund: Trailrunning liegt im Trend. Das Bergauf- und Bergablaufen bewirkt neigungsspezifische Veränderungen der Biomechanik des Laufens wodurch muskuläre Aktivierungsmuster sowie physiologische Anpassungen die Folgen sind (Vernillo et al., 2017). Dabei ist der spezifische Einfluss von Bein- und Rumpfermüdung auf Parameter der sportlichen Performance ein wenig erforschtes Gebiet (Willson et al., 2005).
Methoden: Die vorliegende Studie (FARUN) ist angelegt als eine randomisierte, kontrollierte Cross-Over Studie. 12 ambitionierte Läufer*innen (6 weiblich, 6 männlich) absolvierten jeweils nach einem Gewöhnungstermin zwei Testtermine. Dabei untersucht wurde der Effekt von Laufneigung (geradeaus, bergauf, bergab) sowie ermüdeter Bauch- und Beinmuskulatur auf physiologische Parameter (Herzfrequenz und Borg) sowie neuromuskuläre Veränderungen (Muskelaktivität) der für das Laufen relevanten Bauch- und Beinmuskulatur während dem Lau-fen auf dem Laufband.
Ergebnisse: Die statistische Analyse (Mixed Models) zeigte für die Herzfrequenz, das subjektive Belastungsempfinden (Borg) und die Muskelaktivität des Gastrocnemius signifikante Unterschiede zwischen den Laufneigungen. Sowohl Bauch- als auch Beinermüdung führten zu signifikanten, neigungsunabhängigen Unterschieden der Herzfrequenz und Borg, wobei die Effektstärken bei Beinermüdung tendenziell höher ausfielen. Die Bauchermüdung hingegen führte neigungsunabhängig zu stärkeren Reduktionen der Muskelaktivität in mehreren Muskeln im Vergleich zur Beinermüdung. Akute Bauch- sowie Beinermüdung bewirkten keine signifikante neigungsabhängigen Effekte auf die physiologischen Parameter (HF und Borg) sowie Muskelaktivierung der gemessenen Rumpf- und Beinmuskeln.
Schlussfolgerung: Diese Studie zeigte, dass sich sowohl Laufneigung als auch akute Bauch- und Beinermüdung signifikant und spezifisch auf die Herzfrequenz, die wahrgenommene Anstrengung und die Muskelaktivität beim Laufen auswirken. Die Ergebnisse liefern wertvolle Erkenntnisse für sportwissenschaftliche Anwendungen, insbesondere zur Optimierung von Trainings- und Rehabilitationsprogrammen
Der Einfluss des Manschettendrucks während einer einmaligen Krafttrainingseinheit mit Blutflussrestriktion auf die Hypoalgesie sowie zentrale und periphere Ermüdung: eine Studie zur Erforschung der Dosis-Wirkungs-Beziehung
Hintergrund: Blood Flow Restriction Training (BFRT) kann sowohl neuromuskuläre Ermüdung als auch Hypoalgesie hervorrufen. Obwohl diese Effekte bereits dokumentiert sind, bleiben die zugrunde liegenden Wirkmechanismen weitgehend unklar. Insbesondere ist der Einfluss des arteriellen Okklusionsdrucks (AOP) auf Ermüdung und Schmerzlinderung bisher nicht vollständig verstanden, was die Optimierung von BFRT für klinische und sportliche Anwendungen erschwert.
Methoden: An der Studie nahmen 18 Probanden1 teil, die jeweils an drei Messterminen mit unterschiedlichen Manschettendrücken (0 % AOP, 40 % AOP, 80 % AOP) untersucht wurden. Dabei wurden die Veränderungen des Druckschmerzes mit der Borg-Skala sowie der Voluntary Activation (VA) und des Resting Twitch (RT) jeweils vor der Kraftübung, eine Minute danach und 10 Minuten danach mit der Twitch-Interpolation-Methode gemessen. Zur Auswertung wurde eine Dosis-Wirkungs-Analyse durchgeführt, um die Effekte der unterschiedlichen Druckbedingungen zu vergleichen.
Ergebnisse: Die Ergebnisse zeigen, dass ein höherer Manschettendruck (80 % AOP) sowohl eine signifikante Reduktion der VA als auch des RT bewirkt, was auf eine stärkere zentrale (ZE) und periphere Ermüdung (PE) hinweist. Zudem war die Schmerzreduktion bei 80 % AOP signifikant ausgeprägter als bei niedrigeren Druckbedingungen. Periphere Ermüdung (PE) zeigte dabei einen stärkeren Einfluss auf die Schmerzveränderung als die zentrale Ermüdung (ZE).
Schlussfolgerungen: Die Studie liefert wichtige Erkenntnisse über den Einfluss des Manschettendrucks auf neuromuskuläre Ermüdung und Hypoalgesie beim BFRT. Sie zeigt, dass ein höherer Manschettendruck (80 % AOP) einen grösseren Einfluss auf zentrale und periphere Ermüdung sowie auf die Schmerzlinderung hat als niedrigere Druckstufen, was das Verständnis der zugrunde liegenden Mechanismen vertieft
Schlaf, körperliche Aktivität und kognitive Leistungsfähigkeit bei jugendlichen Psychiatriepatienten: Einfluss eines Bewegungscoachings
Hintergrund: Diese Masterarbeit befasst sich mit dem Zusammenhang zwischen körperlicher Aktivität, Schlafqualität und kognitiver Leistungsfähigkeit. Weiter geht es um die potenziell positiven Effekte einer Bewegungsintervention auf diese drei Variablen. Die Schlafqualität und kognitive Leistungsfähigkeit können bei Jugendlichen mit psychischen Problemen leiden, weshalb körperliche Aktivität potenziell eine Lösung ist.
Methoden: 17 Proband*innen haben an der Studie teilgenommen und wurden in Kontrollgruppe (N=7) und Interventionsgruppe (N =10) aufgeteilt. Nach Eingangsuntersuchungen erfuhr die Interventionsgruppe eine dreimonatige Bewegungsintervention mit Bewegungscoachings. Danach wurden wieder körperliche Aktivität gemessen (Accelerometer), Schlafqualität eruiert (PSQI-Fragebogen) und kognitive Leistungsfähigkeit getestet (Strooptest). Die Resultate wurden mit der Spearman – Rangkorrelationsanalyse auf Zusammenhänge geprüft und den Effekt der Bewegungsintervention wurden mit dem Mann – Whitney – U – Test errechnet.
Resultate: Es gab keinerlei Korrelation zwischen den drei Variablen. Weiter zeigt sich die Bewegungsintervention als nicht effektiv, da keine signifikanten Resultate zustande kamen. Die Effektstärke sprach jeweils für die Kontrollgruppe und die Interventionsgruppe hat eine weniger grosse Verbesserung erfahren.
Diskussion: Die kleine Stichprobe wurde durch das Verwenden des Mann – Whitney – U – Test berücksichtigt, trotzdem hat sie ihre Spuren hinterlassen. Es wurden keine signifikanten Resultate gefunden, was unter anderem an der kleinen Stichprobe liegen kann. In Zukunft sollte weiter Forschung in diese Richtung betrieben werden, aber grössere Probandengruppen sollten angezielt werden
Wirkung von Velocity-Based-Training im Nachwuchsspitzenfussball
Hintergrund: Pro Spiel treten bis zu 1400-mal explosive und schnellkräftige Handlungen wie Sprints, Sprünge und Zweikampfsituationen auf. Es ist somit von grosser Bedeutung, mit Fussball-Junioren die Explosivkraft und Schnelligkeit zu trainieren (Schlumberger, 2006). Das Velocity-Based-Training (VBT) gilt als effektive Methode zur Verbesserung der Muskelkraft, Sprungkraft und Sprintleistung (Baena-Marín et al., 2022; Guerriero et al., 2018; X. Zhang et al., 2022). Diese Arbeit untersucht, ob und wie VBT erfolgreich im Trainingsalltag eines Fußball-Nachwuchsleistungszentrums integriert werden kann, und analysiert die Effekte von Teilnahmefrequenz in VBT-Einheiten und dem Trainingload auf die Entwicklung im CMJ, LCMJ, 10m- und 40m-Sprint zwischen Messzeitpunkt (MZP) 1 & 2 im U19- und U21-Team des FC Basel 1893.
Methoden: Insgesamt wurden 23 männliche Spieler (Alter: 17.87 ± 0.92) in die Datenauswertung eingeschlossen. Zur statistischen Analyse wurde das Programm IBM SPSS (Version 30) genutzt. Die Fragestellung wurde mit einfachen und multiplen linearen Regressionanalysen überprüft. In der Datenauswertung wurden das Bestimmtheitsmass (R2), die p-Werte aus der ANOVA-Tabelle, die Regressionskoeffizienten (B) sowie deren p-Werte miteinbezogen.
Ergebnisse: Die Resultate zeigen, dass die Teilnahmefrequenz und der Trainingload keinen signifikanten Einfluss auf die Leistung in den erfassten Parametern haben. Die Regressionskoeffizienten zeigen jedoch im Mittel einen positiven Trend auf, pro Teilnahme mehr verbessern sich die Spieler in allen Parametern, CMJ (B = 0.357), LCMJ (B = 0.428) und 10m-Sprint (B = -0.009), bis auf den 40m-Sprint, wo die Leistung gleichbleibt (B = 0.000). Schlussfolgerungen: Die Untersuchung zeigt, dass der Zusammenhang zwischen Trainingsfrequenz, Trainingload und der Entwicklung im CMJ, LCMJ, 10m-Sprint und 40m-Sprint nicht signifikant ist. Jedoch weisen die Regressionskoeffizienten im CMJ, LCMJ und 10m-Sprint auf positive Trends hin. Diese Arbeit soll zeigen, dass es im Kontext des Hochleistungssports nicht nur um die Trainingsmethoden, sondern auch um die praktische Umsetzbarkeit geht. Die niedrige Teilnahmefrequenz, bedingt durch Belastungen wie Spiele und Nationalmannschaftseinsätze, könnte die signifikanten Ergebnisse beeinflusst haben. Die praktische Umsetzung von VBT im Hochleistungssport ist herausfordernd und erfordert Maßnahmen wie optimiertes Feedback und Gewichtsanpassungen. Zukünftige Untersuchungen sollten größere Stichproben, Kontrollgruppen und zusätzliche Variablen einbeziehen, um die Ergebnisse zu validieren und die praktische Umsetzung von VBT im Hochleistungssport weiter zu verbessern
Optical spectroscopy of shallow silicon vacancy centers in diamond nanostructures
Color centers in diamond represent the basic building blocks for various quantum technology applications, including sensing [1–5], quantum information processing [6], and quantum communication [7–9]. In quantum sensing, the negatively charged silicon vacancy (SiV−) center is a particularly promising candidate for all-optical single-spin quantum sensing at sub-kelvin temperatures and tesla-range magnetic fields due to its outstanding optical and spin qualities in this regime [10, 11]. The SiV− boasts low optical lifetimes (~ 1.7ns), a high Debye-Waller factor (~ 0.7) and optical coherence protected by its crystallographic inversion symmetry [12, 13], as well as an all-optically addressable spin with long coherence times [10]. What is more, the study of complex physical systems such as skyrmions [14] and other strongly correlated electron systems below 1K and high magnetic fields would benefit immensely from the high spatial resolution and sensitivity offered by single-spin scanning probe magnetometry, such as demonstrated with near-surface nitrogen vacancy (NV) centers in diamond from 4-300K [1, 15] incorporated in this geometry.
At ultra-low temperatures (< 1K), however, charge-state instabilities of near-surface NV centers obstruct their use for such applications. Therefore, the SiV− center presents an excellent candidate in this particular regime, although the combination of an emitter close (< 100 nm) to the surface together with high optical coherence in nanostructures is still difficult to attain, a vital prerequisite for all-optical SiV scanning probe magnetometry.
In this thesis, we address this issue by demonstrating lifetime-limited optical coherence of single shallow (< 50nm) SiV− centers incorporated into diamond parabolic reflectors [16], suitable for scanning probe operation, paving the way towards all-optical ultra-low temperature scanning probe magnetometry at high magnetic fields. Further, the unlocked spectral stability of the SiV− enables a detailed study of its orbital structure using photon correlation measurements under resonant optical driving, shedding light on its photon bunching dynamics.
Finally, we turn to yet another promising defect center, the neutrally charged silicon vacancy (SiV0) center in diamond. While it offers a unique combination of excellent optical properties and long spin coherence times even above 1K [17], its deterministic stabilization in the diamond host is challenging. Here, we present a method for charge-state control of the SiV center, where SiV0 is produced by surface transfer doping enabled by hydrogen termination and subsequent recovery of SiV− using a continuous optical charge-state conversion technique. This enables accessible charge-state control and exciting prospects for SiV-based quantum technologies by incorporating SiV0 into open microcavities [18]. Our results constitute a key step towards the use of near-surface SiV− centers for sensing in extreme conditions and offer powerful approaches for charge-state control of diamond color centers for quantum technology applications
Akuter Einfluss von bewegungsintegrierten Lernmethoden auf die Lernleistung von Primarschulkindern
Heutige Forschungserkenntnisse haben gezeigt, dass körperliche Aktivität einen positiven Einfluss auf die kognitive Leistungsfähigkeit hat und dadurch im Rahmen der Schulbildung eine wichtige Rolle für den Bildungserfolg einnehmen kann. Neben der positiven Beeinflussung der kognitiven Fähigkeiten, kann körperliche Aktivität zudem durch gezielte Integration in den Lernprozess die Aneignung von Wissen erleichtern. In Anbetracht dieser Studienerkenntnisse bedarf es mit der zunehmenden körperlichen Inaktivität in unserem Alltag und der bewegungskargen Gestaltung des westlichen Schulunterrichts neuer Lehrmethoden, die sich diesen positiven Einfluss von körperlicher Aktivität zu Nutze machen.
Die Forschungsrichtung des Embodied Learning beschäftigt sich mit dem Effekt von körperlicher Aktivität auf die Lernleistung und strebt die Etablierung von körperintegrierten Lernmethoden an. Im Rahmen dieses Forschungsfeldes wurde die ExEmbL-Studie durchgeführt, mit dem Ziel, den akuten Einfluss von körperintegrierten Lernmethoden auf die Lernleistung von PrimarschülerInnen zu überprüfen. Basierend auf bisherigen Forschungsergebnissen der Embodied-Learning-Forschung wurde die Hypothese aufgestellt, dass SchülerInnen durch die Integration von körperlicher Bewegung in den Lernprozess bessere Lernerfolge erzielen werden.
Es wurde ein einmaliges Lernprogramm mit drei unterschiedlichen Testgruppen durchgeführt, welche dieselben Lerninhalte mit unterschiedlicher Integration von körperlicher Bewegung erlernen mussten. Die rekrutierten Primarschulkinder im Alter zwischen 8 bis 12 Jahren wurden zufällig aufgeteilt in eine Gruppe, welche den Lerninhalt mit einer hohen Verknüpfung zwischen Lerninhalt und Bewegung lernte, eine Gruppe, welche den Lerninhalt mit einer niedrigen Verknüpfung zwischen Lerninhalt und Bewegung lernte und eine Kontrollgruppe, die den Lerninhalt anhand einer konventionellen Lehrmethode ohne Bewegung erlernte.
Im Anschluss an das durchgeführte Lernprogramm wurde der Lernerfolg anhand eines Cued-Recall-Tests überprüft. Die Resultate zeigen ein besseres Abschneiden der Gruppe mit einer niedrigen Verknüpfung zwischen Lerninhalt und körperlicher Bewegung im Cued-Recall-Test, während die Gruppe mit der hohen Verknüpfung am schlechtesten abschnitt. Diese Resultate indizieren, dass nicht jede Art von körperlicher Bewegung einen positiven akuten Einfluss auf die Lernleistung von Primarschulkindern hat, sondern die Art der Verknüpfung relevant für den Lernerfolg ist
