    Auswirkung einer Sport- und Bewegungsintervention auf das metabolische Syndrom bei Flüchtlingen in einem Flüchtlingslager

    Hintergrund: Das metabolische Syndrom (MetS) ist ein Bündel von Risikofaktoren, welche bei Zwangsvertriebenen vermehrt vorkommen können. Dabei erhöht sich das Risiko für kardiovaskuläre Erkrankungen, Typ-II Diabetes mellitus sowie die allgemeine als auch die kardiovaskulär bedingte Mortalität. Das Ziel dieser Arbeit ist es zu untersuchen, ob mit einer Sport- und Bewegungsintervention in einem Flüchtlingslager, der kontinuierliche Score für das MetS reduziert werden kann. Methoden: Die Daten dieser Arbeit stammen aus einer pragmatischen, randomisierten Kontrollstudie. Von 98 Teilnehmenden absolvierten 49 eine Sport- und Bewegungsintervention, über den Zeitraum von neun Wochen. Die körperliche Aktivität wurde mittels "International Physical Activity Questionnaire" erhoben, während objektiv erfasste Variablen verwendet wurden, um den siMS und das MetS zu berechnen. Der Zusammenhang der körperlichen Aktivität mit den Bestandteilen des MetS sowie mit dem siMS wurde mittels Pearson Korrelation, der Effekt der Intervention mittels ANCOVA geprüft. Ergebnisse: Es wurde wenig Evidenz für einen Zusammenhang der körperlichen Aktivität mit den Bestandteilen des MetS und dem siMS gefunden. Lediglich für den Taillenumfang konnte sowohl bei den Baseline- (r = -0.145, p = 0.046) als auch bei den Post-Messungen (r = -0.178, p = 0.040) ein signifikanter negativer Zusammenhang gefunden werden. Mit der ANCOVA wurde sowohl für Modell 1 (F (1,79) = 0.695, p = 0.407, partielles η² = 0.009) als auch für Modell 2 (F (1,76) = 0.316, p = 0.576, partielles η² = 0.004) keine Evidenz für einen Unterschied der Interventionsgruppe von der Kontrollgruppe gefunden. Schlussfolgerung: Die Intervention hat den erwünschten gesundheitsfördernden Effekt für das MetS nicht erbracht. Diese Untersuchung liefert erste Erkenntnisse zu der mangelnden Forschungslage des hier bearbeiteten Themas. Dabei erkannte Limitationen, wie beispielsweise die unregelmässige Teilnahme an den Interventionseinheiten oder die Ernährung im Flüchtlingslager können in weiteren Untersuchungen berücksichtigt werden

    Der Einfluss von Fitness als Moderator bei physiologischen und psychologischen Reaktionen in schulischen Belastungssituationen

    Hintergrund: Der Einfluss von körperlicher Aktivität und Fitness auf physiologische und psychologische Stressreaktionen wird in der Wissenschaft seit Längerem untersucht. Unter Laborbedingungen konnte dabei teilweise ein abfedernder Effekt der Fitness auf die Stressreaktivität nachgewiesen werden. Untersuchungen zu alltäglichen Stressbelastungen gibt es bis anhin nur im Ansatz. Das Ziel dieser Studie ist es, zu ergründen, ob die körperliche Fitness bei Jugendlichen während eines Real-Life-Stressors in Form einer schulischen Leistungssituation als Moderator der physiologischen und psychologischen Stressreaktivität wirksam ist. Methoden: Die Studie wurde an der Regionalschule Lenzburg mit 79 Lernenden (M = 14.19 ± 0.79 Jahren; 49.4 % Frauen) durchgeführt. Während einer Prüfungssituation und in einer normalen Unterrichtsstunde wurde die Herzfrequenzvariabilität der Lernenden aufgezeichnet. Ergänzend diente ein psychologischer Fragebogen zur subjektiven Beurteilung der aktuellen Stimmung und Angst. Die körperliche Fitness wurde mittels Shuttle-Run- und Griffkrafttest erhoben, während die Lernenden in zwei Fitnessgruppen bezüglich ihrer sportlichen Leistung aufgeteilt wurden. Mit Hilfe des t-Tests erfolgte hernach der Vergleich der Fitnessgruppen in relevanten Variablen. Abschliessend wurde durch eine Varianzanalyse getestet, ob die Einteilung in die Fitnessgruppen einen Einfluss auf die physiologische und psychologische Stressreaktivität zeigt. Ergebnisse: Die kardiovaskuläre Fitness wirkt als Moderator bezüglich der psychologischen Stressreaktivität. Kardiovaskulär fitte Jugendliche zeigen allgemein verbesserte physiologische Werte hinsichtlich der Herzfrequenzvariabilität, allerdings hat die kardiovaskuläre Fitness keinen Moderatoreffekt. Zwischen den Fitnessgruppen der Griffkraft zeigen sich keine Unterschiede. Schlussfolgerungen: Die Ergebnisse verdeutlichen, dass eine gute kardiovaskuläre Fitness zur Verbesserung der Herzfrequenzvariabilität beitragen und als Moderator psychologische Stressreaktionen während Alltagssituationen abfedern kann

    Einleitung: Schlaganfälle gehören weltweit zu den häufigsten Ursachen für Tod, langfristige körperliche Beeinträchtigungen und Einschränkungen der Leistungsfähigkeit und der Mobilität. Die Gefässe des Gehirns und des Auges sind sich sehr ähnlich, weshalb die retinalen Gefässe bei der Einschätzung des Risikos für Schlaganfälle von grossem Nutzen sind. Diese Arbeit untersucht, wie sich die körperliche Aktivität und Mobilität im ersten Jahr nach einem Schlaganfall auf die Gefässgesundheit auswirken. Methoden: Daten, die im Rahmen der Mobitec-Stroke-Studie an 2 Messterminen im Abstand von 6 Monaten innerhalb eines Jahres nach dem Schlaganfall von 34 Probanden (71.7 ± 10 Jahre) erhoben worden waren, wurden ausgewertet. Dabei handelt es sich um Daten zur körperlichen Aktivität (gemessen mittels Accelerometer), zur Mobilität und zum Lebensraum der Probanden (gemessen mittels GPS-Messgerät und Life Space Assessment (LSA) Fragebogen) sowie zur statischen retinalen Gefässanalyse. Mittels multipler linearer Regression wurde untersucht, ob sich die unabhängigen Variablen auf die Änderungen der retinalen Gefässdurchmesser – konkret auf die zentralen retinalen arteriolären und venulären Äquivalente (CRAE und CRVE) sowie die arterio-venöse Ratio (AVR) – auswirken. Resultate: Es konnten keine signifikanten Effekte der untersuchten unabhängigen Variablen auf die Differenz zwischen den beiden Terminen von CRAE, CRVE und AVR gefunden werden. Die AVR verbesserte sich jedoch signifikant vom ersten auf den zweiten Messtermin um 0.02 (95% KI: 0.00, 0.03, P = 0.009). Schlussfolgerung: Trotz nicht signifikanter Resultate der linearen Regression heben die Erkenntnisse dieser Arbeit die Wichtigkeit von körperlicher Aktivität in Bezug auf die Mobilität und die Gefässgesundheit hervor, insbesondere in der frühen Rehabilitationsphase nach einem Schlaganfall

    Interplay of internal and external regulation and their joint impact on student engagement in class

    Student engagement in class is the time during class when students are actively and productively learning in regard to specific scholastic content. Educational scientists agree that student engagement is a substantial predictor of academic performance. Nevertheless, factors influencing student engagement in elementary school have not been investigated systematically. Accordingly, this dissertation examines factors which might affect elementary students’ engagement during class. Based on a synthesis of theoretical models and a systematic literature review of studies that empirically examine observable student engagement in elementary schools, a working model is presented. The model proposes that student engagement is affected by personal, social, and instructional factors relating to internal and external regulatory processes. It is assumed that in a classroom context, internal and external regulatory processes might interact with each other and jointly contribute to student engagement during class. The sample used consists of 34 fifth grade elementary school classes in Switzerland. The data used in this dissertation was collected using three instruments: First, a longer questionnaire about attributes of the students which are conceptualized as more stable. Second, a standardized observation of students’ on-task engagement employing a 15-second time-sampling approach. Third, a short student questionnaire after each observed lesson capturing the student’s self-perception during the lesson. The data analysis strategy follows a quantitative approach, taking into account dependencies between students from the same classroom. Research question 1 investigated a triangulation of student engagement in class. The analysis revealed that a comparison referent effect, similar to the 'Big-Fish-Little-Pond'-Effect, can be observed regarding student engagement. Research question 2 examined the relationship between motivation, self-control, and student engagement. The results indicate that self-control as well as different types of internal and external motivation significantly impacted student engagement at both the individual and class levels. Research question 3 examined the relationship between social connectedness within a class and student engagement. The data suggests that students with slightly lower engagement levels than the class average were more likely to be chosen as best friends by their classmates. Overall, the results of the dissertation support the significance of social factors related to classmates in influencing student engagement in class. Therefore, it is important to consider not only individual and instructional factors, but also social factors when promoting student engagement in the classroom

    Investigation and targeting of early degenerative response in retinal cells in Ischemia-Reperfusion glaucoma mouse model: paving the way for therapeutic neuroprotection strategy

    Glaucoma is a group of neuropathies characterized by progressive degeneration of retinal ganglion cells (RGCs) causing vision loss. It is the leading cause of irreversible blindness with 76 million people affected worldwide. Currently, the only approved treatments aim at lowering intraocular pressure (IOP), the main modifiable risk factor. However, some patients continue losing vision even though they have a well-controlled or nonelevated IOP. Therefore, there is an unmet need to develop new neuroprotective treatments to prevent RGC loss in order to slow down disease progression and ultimately preserve vision. The study aimed at better understanding the mechanisms involved in RGC degeneration in order to develop RGC neuroprotective strategies. To do so, we used the ischemia-reperfusion (I/R) mouse model of glaucoma. We first characterized the I/R mouse model and showed degeneration of the RGCs with different degrees of severity: mild, moderate and severe, 7 days following the injury. Resulting visual dysfunction was observed. Performing single-cell RNA sequencing allowed us to identify initial genes and pathways transcriptionally up- or down-regulated following injury in RGCs as well as other retinal cell types. We explored RGC surrounding cell response and evaluated the implication of cellular senescence in the retina following I/R injury. We identified at early I/R time point glial cells, rather than RGC, expressing senescence-related genes, although RGC showed hallmarks of senescence at the later time point. We investigated further the early RGC stress response following I/R injury and identified the transcriptor factor 3 (ATF3) as a significant factor responding to I/R injury. We confirmed the relevance of this target as we observed increased ATF3 transcript expression in human glaucomatous post-mortem eyes. We decided to inhibit expression of Atf3 and relative genes that were activated in response to I/R injury to assess their effect on RGCs survival. The strategy was to use sh-miR (artificial miRNA), delivered into retinal cells using adeno-associated viral (AAV) vectors, in order to knock down target gene expression. We quantified RGC survival and observed a reduction of the degree of severity of RGC degeneration by knocking down Atf3, Ecel1 and Cdkn1a. Our results demonstrate that targeting ATF3 and related pathways might have a neuroprotective effect on RGCs and could be considered for potential neuroprotection strategies to treat glaucoma patients with the ultimate goal of preserving patient vision

    Regularity analysis for semilinear elliptic PDEs with random data

    In this thesis we consider a class of semilinear elliptic partial differential equations with uncertainty regarding the diffusion coefficient. We derive the analytic and Gevrey smooth dependence of the solution of such equations on the diffusion coefficient, both as an element of a suitable function space and when given by some (possibly nonlinear) parametric expansion. Such smoothness results then enables one to quantify the uncertainty in the solution using state-of-the-art numerical methods. We approach this task in a modular and abstract fashion. We first introduce the necessary combinatorical objects and results. We then review the definitions and behaviour of linear and multilinear maps as well as powers, polynomials and formal power series between (possibly infinite-dimensional) vector spaces. Following this, their corresponding continuous versions between Banach spaces are considered. With this we also review the definitions and behaviour of Peano and (higher-order) Fréchet differentiability, as well as analyticity and Gevrey smoothness. Especially, we prove various qualitative and quantitative results concerning both the composition of maps and implicit map theorems for polynomials and power series, as well as Peano differentiable, Fréchet differentiable, analytic or Gevrey smooth maps. These mathematical tools enable our analysis of the class of semilinear elliptic partial differential equations

    Does the Central Nitrogen Atom Make a Difference? A Comparison of Non‐Coordinating Pyridine and Benzene Spacers in Multitopic Ligands

    Abstract In metal coordination compounds of the divergent ligands 4,2′:6′,4“‐terpyridine and 3,2′:6′,3”‐terpyridine and their 4′‐functionalized derivatives, the central pyridine ring of the 4,2′:6′,4“‐terpyridine and 3,2′:6′,3”‐terpyridine domains is non‐coordinated. We present an overview of data from the Cambridge Structural Database to assess whether there are structural similarities between the coordination compounds of 4,2′:6′,4“‐tpy and 1,3‐di(pyridin‐4‐yl)benzene, and between 3,2′:6′,3”‐tpy and 1,3‐di(pyridin‐3‐yl)benzene. Based upon structurally characterized compounds, it emerges that the coordination chemistry of ligands including one or more 4,2′:6′,4“‐terpyridine or 3,2′:6′,3”‐terpyridine metal‐binding domains is more abundantly exemplified than that of corresponding ligands based upon 1,3‐di(pyridin‐4‐yl)benzene and 1,3‐di(pyridin‐3‐yl)benzene.     Mapping risk preferences: psychometric and empirical properties across measurement approaches and domains

    Risk preference, a fundamental building block of economic theory, plays a significant role in understanding individual differences in risk-taking across the behavioural sciences. Despite employing various measurement approaches and domains for its assessment, inconsistencies persist regarding these operationalizations’ reliability and the inherent characteristics of risk preference. This dissertation consists of three manuscripts that extensively investigate the psychometric properties (i.e., convergent validity and temporal stability) of risk preference and is supplemented by a fourth manuscript that delves into its empirical properties (i.e., selection and socialization effects). The aim is to enhance our comprehension of the individual differences in risk preference and to quantify its development over time. In our analysis of psychometric properties, we employed meta-analytic methods to thoroughly analyse and synthesise data, with the goal of estimating and comparing the convergent validity and temporal stability (i.e., rank-order stability and mean-level change) of risk preference across various measures. Our meta-analyses provided several significant insights. Firstly, we discovered a notable lack of convergence among different measurement approaches and domains. Secondly, we identified substantial differences in rank-order stability across these measures. Thirdly, our results challenge existing theories by demonstrating an absence of significant age-related differences in revealed risk preferences across various task types. However, we consistently found a decline in stated risk preference with age across diverse risk domains, genders, and populations. In our exploration of empirical properties, we applied the propensity score matching (PSM) method to control for confounding variables, meticulously examining the intricate relationship between self-reported risk preferences and life events in family and career domains. Our detailed analysis indicated that selection effects were more pronounced than socialization effects, suggesting that individuals’ self-reported risk preferences exhibited greater predictive ability on life events while being less influenced by those events. In conclusion, this dissertation conducts an in-depth examination of both the psychometric and empirical properties of risk preferences, aiming to elucidate whether, how, and why risk preferences vary among individuals. It reveals the complex nature of risk preferences across various measurement approaches and domains, providing invaluable insights into individual differences in risk preferences for researchers, professionals, and policymakers
