    Transition metal-catalyzed intermolecular C(sp³)-H animation reactions

    Nitrogen is a fundamental element in almost every area of chemistry, playing an essential role in biological systems, pharmaceuticals, agrochemicals, and material science. In organic synthesis, the development of efficient methods to introduce nitrogen-based functionalities in a chemo- regio- and stereoselective manner is a major challenge. In this context, transition metals complexes have been studied and designed to directly functionalize ubiquitous C–H bonds, with a number of attractive and straightforward methods to forge new C–N bonds without the need of pre-functionalized materials. Indeed, C(sp3)–H amination reactions have recently emerged as powerful tools for the synthesis of nitrogen-containing pharmaceutical fragments and natural products. This thesis focuses on the development of novel transition metal-catalyzed intermolecular C(sp3)–H amination reactions. Specifically, we explored the design and synthesis of a safe and highly reactive amidating reagent for the direct and selective synthesis of vicinal aminoalcohols and diamines. The studied reagent, in combination with an iridium-based catalytic system, promotes the C(sp3)–H amination of alcohols and diamines under mild and practical conditions. Furthermore, the high reactivity of iridium nitrenoids was employed for the development of an intermolecular protocol for the C(sp3)–H amination of benzylic and aliphatic substrates. Next, we discovered that catalytic amounts of abundant and benign FeCl2 can promote the site-selective C(sp3)–H amination of pharmaceutically relevant N-heterocycles, in an appealing late stage functionalization logic. The developed method was then applied to the total synthesis of the reported structure of senobtusin, an atypical alkaloid with an amidine moiety. In addition, kinetic studies helped gaining insights in the reaction’s mechanism. Altogether, the research presented in this thesis contributes to the growing field of C–H amination, offering novel, sustainable and atom-economical strategies for the synthesis of amines

    Zusammenhang von körperlicher Fitness und Stress mit Schlafproblemen, Depressionsschwere und Wohlbefinden

    Hintergrund Schlafstörungen und Depressionen sind grosse Probleme der modernen Welt und chronischer Stress wird häufig als mögliche Ursache dieser Krankheiten genannt und vermindert darüber hinaus auch das Wohlbefinden. In der Prävention und Therapie dieser Krankheiten hat die Stressreduktion einen grossen Stellenwert, aber auch körperliche Aktivität wird oft als Methode eingesetzt, um den Stress zu vermindern und psychische Krankheiten zu behandeln. In dieser Arbeit wurden depressive Patienten und gesunde Kontrollen bezüglich des Zusammenhangs von Stress und körperlicher Aktivität auf Schlafstörungen, Depressionsschwere und Wohlbefinden untersucht und verglichen. Methoden Die untersuchten Daten stammen aus der PACINPAT-Studie. Stressbelastung, Schlafstörungen, Depressionsschwere und Wohlbefinden wurden mittels Fragebogen erhoben und die körperliche Fitness mittels eines submaximalen Ergometrietests geschätzt. Resultate Stress ist sowohl bei den gesunden Kontrollen als auch bei den depressiven Patienten mit schlechteren Werten bei Schlafstörungen, Depressionsschwere und psychischem Wohlbefinden assoziiert (p < .001). Beim körperlichen Wohlbefinden konnte keine Signifikanz (p = .778) bei den Gesunden festgestellt werden, bei den Depressiven ist mehr Stress mit einem besseren körperlichen Wohlbefinden assoziiert (p = .023). Für die körperliche Fitness konnte keine Signifikanz mit Schlafproblemen, Depressionsschwere oder dem Wohlbefinden festgestellt werden, zum Teil ist jedoch eine Tendenz für bessere Werte bei höherer körperlicher Fitness erkennbar. Schlussfolgerungen Stress kann einen grossen Einfluss auf Schlafstörungen, Depressionsschwere und Wohlbefinden haben und sollte in Prävention und Therapie behandelt werden. Für körperliche Fitness sind weitere Studien von Nöten, um einen Effekt klar nachweisen zu können

    Chains of Dependency: Changing Importation Practices in the Democratic Republic of Congo

    The reliance on imported products has rendered many sub-Saharan African countries like the DRC vulnerable to global disruptions such as supply chain bottlenecks that have been engendered and made visible by COVID-19 pandemic dislocations. New questions continue to emerge with regards to the dynamics of transnational dimensions of trade, as a result of deepening Sino-African relations. Transnational trade is partly dependent on the geographic mobility of the individuals who travel abroad to select goods to be packed into cargo containers or suitcases to be transported to the DRC. With the COVID-19 pandemic producing global disruptions in supply chains of all manner, countries such as the DRC—where it is individual traders themselves who manage import logistics—are confronting pointed challenges with regards to the way in which geographic immobility has stymied the flow of goods into the country. Through which means do goods continue to flow into the DRC? Who now stands to benefit from import to Congo? Through an examination of the gendered geographies of supply chains—as they relate to transnational trade—as well as local social networks, this chapter’s empirical findings bring together the dialectical oscillations of global and local, and formal and informal into an analytical theoretical framework

    How Uphill and Downhill Running Interfere Posture and Muscle Activity: Descriptive Laboratory Study

    Background: Downhill and uphill running alter running kinematics, force distribution, and muscle activity. While lower limb changes are well studied, research on the back and pelvis remains limited. This study examined how six gradients (-15% to 15%) affect back and pelvic posture and muscle activity. Understanding these changes is important for injury prevention and post-back injury protocols. Methods: Twelve healthy recreational runners (6 women; age 25.0 (SD 1.6) years; body mass 61.1 (3.5) kg; height 167.8 (5.4) cm; and 6 men; age 26.3 (1.2) years; body mass 75.0 (6.9) kg; height 178.0 (8.5) cm) participated. Kinematic changes such as lumbar lordosis, hip drop, pelvic tilt, and trunk inclination were assessed using 3D motion capture. Muscle activity in the lower limbs and trunk was measured via surface electromyography. Results: Compared to level running lumbar lordosis was increased at -15% (mean difference MD: 1.2 (4.9) degrees, Cohen’s d = 0.73) and decreased at +15% (MD: -1 (3.6) degree, d = 0.39). Total hip drop decreased at -15% (MD: 14.2 (11) mm, d = 1.93). Muscular activity of lower limb muscles was higher at steep downhill and uphill gradients. While of the abdominals only obliquus externus showed grade-specific increases, activity of erector spinae was relevantly reduced at -5% and -10%. Discussion: Increased lumbar lordosis during downhill running may increase lumbar spine overload, especially in runners with lower back issues. For injury prevention and rehabilitation, we recommend starting with uphill running, followed by level, and then downhill. Additionally, grade-specific training is vital due to varying demands

    Zusammenhang zwischen der kardiorespiratorischen Fitness und der retinalen Gefässfunktion bei Glaukom-Patienten*innen

    Hintergrund Das Glaukom ist eine chronisch fortschreitende Optikusneuropathie. Sie gilt als weltweit häufigste Ursache für irreversible Erblindung, wobei das POAG „primary open angle glaucoma“ die häufigste Form darstellt. Vaskulärer Risikofaktoren gewinnen zunehmend an Relevanz, da vaskuläre Dysregulationen anhand von abgeschwächten Flimmerreaktionen oder übermässigen arteriellen Konstriktionen aufgezeigt werden. Höhere kardiorespiratorische Fitness (CRF) hängt bei Nicht-Glaukompatienten*innen mit einer verbesserten mikrovaskulären Gesundheit zusammen. Das Ziel dieser Arbeit war es herauszufinden, ob bei POAG-Patienten*innen ein Zusammenhang zwischen der CRF und der retinalen Gefässfunktion besteht. Methoden Die retinale Gefässfunktion wurde bei 18 POAG Probanden*innen (Alter = 61 9 Jahre, 5 Hochdruckglaukom, 13 Normaldruckglaukom) anhand der dynamischen Netzhautanalyse, 3 Flickerphasen à jeweils 20 Sekunden, und die CRF anhand eines individuellen Rampenprotokoll auf dem Fahrradergometer, erfasst. Ergebnisse Die multiple lineare Regression zeigt nach Kontrolle von Alter, BMI, Geschlecht und Blutdruck keine signifikanten Zusammenhänge zwischen der retinalen Gefäßgesundheit (aFID: p = 0.717, vFID: p = 0.096, aCON: p = 0.404) und der CRF. Die Dilatationswerte der POAG-Probanden*innen (aFID = 3.17% , vFID = 5.18%) sind vergleichbar zu Studien mit gesunden Probanden und der VO2peak, variiert stark unter den Probanden*innen (M: 25.83ml/min/kg, SD: ± 6.35ml/min/kg). Schlussfolgerungen Es konnte kein Zusammenhang zwischen der kardiorespiratorischen Fitness und der retinalen Gefässfunktion bei POAG-Probanden*innen festgestellt werden. Die geringe Stichprobengrösse führte zu einer hohen Variabilität retinaler Biomarker, CRF sowie Störvariablen. Zukünftig soll ein anderes Studiendesign, grössere Stichprobe, Längsschnittstudie und Trennung der POAG-Formen, das mögliche Potential von Bewegung auf die Gefässfunktion der Glaukompatienten*innen erfassen und die Relevanz für die Therapie aufzeigen

    Exploring conformationally and state-resolved ionic and neutral reactions

    Understanding the impact of the molecular structure of reactants and the initial reaction conditions, arising from their quantum states, is crucial for gaining control over chemical reactions and their outcomes. Recent advances in physical chemistry allow for detail exploration of fundamental effects behind the reaction mechanisms by combining advanced theoretical calculations with specifically designed experiments. This dissertation presents a complementary theoretical and experimental investigation of structural and quantum-state effects in bimolecular ionic and neutral reactions. Quantum-chemical calculations of the potential energy surface (PES) were employed to examine the impact of the molecular structure (conformation) on the reaction mechanisms of three conformationally-resolved ionic reactions, experimentally studied in a gas phase using molecular beams and ion-trapping methods. In the ion-molecule reaction of s-trans- and gauche-dibromobutadiene (DBB) with Ca+^+, the theoretical results revealed a conformer-specific reaction mechanism likely originating from the anisotropic interactions in the entrance channel. For the polar cycloaddition of propene ions with DBB, the PES calculations showed the competition of a stepwise mechanism with a concerted process for the reaction with gauche-DBB, whereas a pure stepwise mechanism was identified for DBB in its s-trans conformation. Combining this with experimental kinetic data, it was found that both conformers are reactive with capture-limited reaction rates, indicating that the s-trans pathway is not inhibited by steric hindrance, which is typical in concerted mechanisms. Lastly, for the polar cycloaddition reaction of propene ions with methyl vinyl ketone, the stepwise reaction pathways were identified for both s-cis and s-trans conformers. A novel crossed-molecular-beam experiment was employed to study the effects of rotational quantum states in the neutral-neutral chemi-ionisation reaction between carbonyl sulfide (OCS) and metastable neon. It was found that the branching ratio between the Penning- and dissociative-ionisation channels strongly depends on the initial rotational state of the OCS molecule. These results were compared to the analytical interaction potential constructed using semi-empirical methods to explain differences in reactivity between the rotational states, and a tentative explanation was proposed. As a first step towards the investigation of radical reactions, two pulsed discharge sources, a high-voltage plate discharge and a dielectric-barrier discharge, generating supersonic beams of fluorine radicals, were developed. Finally, a potential reaction system for examining steric effects in hydrogen-atom abstraction, a mechanism typical for radical reactions, is suggested

    Advancing models of semantic representation: empirical study designs, network analysis methods, and computational tools

    People learn an astonishing amount of things about the world throughout life. This knowledge is retained in semantic representations, the cognitive manifestation of factual information in memory. Modeling semantic representations is an important ingredient to understanding how people acquire knowledge from their environment, how this knowledge is organized in memory, and how it affects behavior. Given the wide-ranging implications of semantic representations, research questions address multiple aspects, including their structure and content. Various methodological approaches have been used to successfully model semantic representations from text and behavioral data. However, much is still unknown about the optimal ways to model semantic representations, particularly regarding structural differences of representations among people. In addition, further investigation is needed to connect the structure of semantic representations to environments and behavior, as well as to map the content of individual concepts’ semantic representations. This dissertation aims to improve the methods and tools used for modeling semantic representations across various applications while also establishing links between semantic representation and cognitive performance. The first two manuscripts explore the relationships between environment and semantic representation as well as between semantic representation and cognitive performance of younger and older adults in a proof-of-concept study. The third manuscript presents a comprehensive recovery simulation study that evaluates which empirical study designs yield accurate individual-level semantic networks based on free associations and relatedness judgments. The fourth manuscript introduces the R package associatoR and includes a tutorial on how to use it to map the content of semantic representation. Together, this dissertation contributes to the understanding of individual differences in semantic representation by linking it to cognitive performance, guiding study design for assessing individual semantic networks, and facilitating the analysis of free association based models of semantic representation

    Beliefs zur Zeitgeschichte: eine Untersuchung der Überzeugungen von Geschichtslehrer*innen an Gymnasien des Ruhrgebietes in der Berufseinstiegsphase

    Das im Folgenden vorgestellte Dissertationsprojekt lässt sich an der Schnittstelle von Fachdidaktik und Professionsforschung verorten. Im Fokus stehen die sog. Beliefs von Geschichtslehrpersonen hinsichtlich der für das Fach Geschichte besonderen Epoche, der Zeitgeschichte. Ausgangspunkt der Untersuchung waren drei theoretische Annahmen: Zum einen, dass Lehrperso-nen als Expert*innen für das „Lernen und Lehren in der Schule betrachtet“ (Bromme 2008, 159) werden. Daraus lässt sich schlussfolgern, dass Geschichtslehrpersonen als Schlüsselfaktor für erfolg-reichen Geschichtsunterricht gelten (vgl. Messner/Buff 2007, 144). Zum anderen ist davon auszu-gehen, dass aus berufsbezogenen Überzeugungen im Zusammenspiel mit den Kompetenzaspekten Wissen, Selbstregulation und Motivation das professionelle Handeln von Lehrpersonen entsteht (vgl. Baumert/Kunter 2011, 33). Dabei verweisen Reusser et al. auf die wichtige Rolle der „subjektiv geprägten berufsbezogenen Überzeugungen (beliefs) von Lehrkräften“ (Reusser et al. 2011, 478). Schließlich kommt der Zeitgeschichte für den Geschichtsunterricht eine besondere Rolle zu. Daraus resultierte die für die Untersuchung leitende Frage, welche Überzeugungen Geschichtslehrpersonen bezüglich dieser Epoche im Allgemeinen und einer historischen Zäsur („1989“) im Besonderen auf-weisen. Dabei fokussiert die Studie Berufseinsteiger*innen. Das Projekt verfolgt zur Untersuchung dieser Fragestellung eine triangulativen Ansatz: Neben ei-nem eher deskriptiv-heuristisch eingesetzten Begleitfragebogen, wurden Leitfadeninterviews mit 20 Geschichtslehrpersonen geführt und diese mittels Mayrings qualitativer Inhaltsanalyse ausgewer-tet. Außerdem wurden von den Proband*innen mitgebrachte Bildquellen in Anlehnung an die Vi-sual History untersucht. Um darüber hinaus eine wissenschaftsbasierte Interpretationsfolie zu er-halten, mit Hilfe derer die Deutungen der Geschichtslehrpersonen zum ausgewählten historischen Geschehenszusammenhang 1989 rückgebunden und miteinander verglichen werden können, wurde ferner ein Korpus des Akademischen Diskurses zu 1989 analysiert. Schließlich wurde die Ge-schichtstheorie Hayden Whites zur narrativen Systematisierung der Ergebnisse wirksam gemacht. Die Studie gelangte auf diesem Wege zu folgenden Erkenntnissen: Die Geschichtslehrpersonen definieren Zeitgeschichte als Kombination aus fluiden und zäsuralen Begrenzungen, womit ihr Verständnis anschlussfähig an die aktuelle Forschung ist. Auffallend ist dabei, dass von ihnen der Nationalsozialismus nicht mehr zur Zeitgeschichte gezählt wird. In fach-didaktischer Hinsicht wird deutlich, dass die Berufseinsteiger*innen zwar einerseits auf viele Prinzi-pien und Argumente Bezug nehmen. Andererseits aber wird kaum auf die Integration von aktuel-len geschichtskulturellen Phänomenen oder die Einbeziehung von Zeitzeug*innen im Geschichtsun-terricht verwiesen. Hinsichtlich des ausgewählten historischen Geschehenszusammenhangs 1989 dominieren die Erklä-rungsfaktoren „Protestbewegung“ und „Fehler im System“, während viele weitere mögliche Erklä-rungsmuster ungenannt bleiben. Deutlich wird hier, dass dies mit in Schulbuchanalysen herausgear-beiteten Basisnarrativen korrespondiert. Diese Konventionalität der Proband*innen wird ebenso in der Strukturierung ihrer Erzählungen deutlich. Hier dominiert das „Revolutions- und Freiheitsnarra-tiv“
