Superconducting quantum devices with germanium nanowires
This thesis investigates the potential of germanium (Ge)-based nanowires as a platform for quantum information processing, particularly focusing on superconductor-Ge hybrid devices. In the past five years, hole spin qubits in Ge quantum wells have seen rapid advancements. Long coherence times, rapid operations and high gate fidelity have been achieved. Yet, challenges remain in establishing long-distance interconnections for semiconductor spin qubits. In the meantime, superconducting qubits stand out as another leading quantum computing platform. Their larger physical size and macroscopic quantum states facilitate remote coupling. Can we combine the advantages of both platforms? This is the central question that this thesis seeks to address. Our goal is to define and coherently manipulate qubits in Ge/Si core/shell nanowires (NWs), leveraging Ge’s unique properties and integrating Ge into circuit-QED architectures.
On the path of achieving this goal, several crucial steps must be undertaken. First and foremost, we established reliable and reproducible recipes to making superconducting contact to Ge NWs. To gain a deeper insight of the Andreev bound states (ABSs) in Ge/Si nanowire Josephson junctions (NW JJs), we conducted DC measurements. Our study included tunnelling spectroscopy of ABSs, and the current-phase relation measurements of Ge/Si NW JJs using superconducting quantum interference device (SQUID).
To develop qubits in the Ge hybrids system, it is necessary to integrate Ge/Si NW JJs into microwave circuits. In this context, we explored two types of qubits: superconducting qubits, which utilize collective bosonic modes, and Andreev qubits, defined in fermionic quasiparticles of the ABSs spectrum. In Chapter 7, the NW JJ is shunted by a large capacitor, forming a gatemon qubit. In Chapter 8, we embedded the NW JJ in a RF SQUID loop, aiming for a Andeev qubit
Postmortem imaging using CT and MRI for process automation and biomarker development
Several methods exist for the forensic identification of unknown deceased, while radiologic identification (RADid) might be the method of choice. Typically, antemortem (AM) and postmortem (PM) computed tomography (CT) data are manually compared to assess similarity. However, this approach is time-consuming, examiner-dependent, and qualitative, therefore, the outcome cannot be supported by statistical evidence. Similarly, several challenges are faced in the diagnosis and prognosis of neurodegenerative diseases, such as Amyotrophic Lateral Sclerosis (ALS). Reasons for these challenges arise due to the heterogeneity of symptoms within one disease and the overlap of symptoms between different diseases. Therefore, there is a great need for magnetic resonance imaging (MRI) biomarkers to aid in diagnosis and prognosis. However, potential new MRI biomarkers need to be validated. Validation usually incorporates ex-situ MRI acquisitions of formalin fixed tissue along with histological evaluation. Nevertheless, performing ex-situ MRI scans is methodologically challenging and histology is fundamentally a qualitative technique, limiting a correlation with MRI findings. The aim of this thesis was therefore to develop forensic and diagnostic techniques based on PM imaging, specifically by automating the identification process of unknown deceased using postmortem computed tomography (PMCT) and by exploring potential biomarkers for ALS via postmortem magnetic resonance imaging (PMMRI).
The process of identifying unknown deceased individuals was automated by developing scripts that perform registration and assess the similarity between AM and PM CT data. In an initial study, two-dimensional (2D) maximum intensity projection (MIP) of CT data from the sternal bone was used, similarly to the current manual procedure (Publication 1; status accepted). In this work, an accuracy of 91.2 % was achieved for the identification process. In a second study, to take advantage of the benefits of CT, three-dimensional (3D) data was used, including both the sternal bone and the fifth thoracic (T5) vertebra (Publication 2; status submitted). This resulted in an accuracy of 97.8 %. These automated approaches are easy to use, examiner-independent, reliable, and efficient. Regarding the MRI biomarker development, an effective method was developed for ex-situ PMMRI including brain-specific 3D printed support plates and the effect of brain extraction and formalin fixation on MRI parameters was assessed by comparing ex-situ to in-situ data (Publication 3; status accepted). Changes in volume, diffusivity, and relaxation parameters were observed. This method along with the findings are crucial prerequisites in biomarker development and validation. In-situ PMMRI was further used to compare MRI parameters between brains of patients with ALS and of controls, exploring potential MRI biomarkers (Publication 4; status submitted). Moreover, a tool was developed to automatically quantify the myelin density in histological data, which was used for the validation of MRI findings. Differences in diffusivity and relaxometry in various brain regions were detected. However, larger sample sizes along with histological analyses are required to confirm the current results.
The tools and techniques developed in this thesis are a valuable contribution to process automation and biomarker development, improving efficiency, reliability, and reproducibility. Moreover, the observed MRI parameter alterations in patients with ALS might lead to the development of novel diagnostic and prognostic biomarkers
«NOW!plus» - Konzept und Trainingshandbuch zur Steigerung der körperlichen Leistungsfähigkeit und Freude an der Bewegung bei übergewichtigen Jugendlichen
Hintergrund: Technologische und gesellschaftliche Veränderungen befördern einen zunehmend inaktiven Lebensstil. Insbesondere Adipositas und daraus resultierende chronische Erkrankungen sind ein wachsendes Phänomen unserer Zeit. Besonders besorgniserregend ist die Anzahl übergewichtiger Kinder und Jugendlicher. Die körperliche Aktivität im Jugendalter ist entscheidend für die Entwicklung eines gesunden Lebensstils. Somit sind gezielte Interventionen in dieser Phase besonders wichtig. Im Rahmen dieser Arbeit und des Programms «NOW!» wurde ein Trainingshandbuch entwickelt, das die Trainer/innen des «NOW!plus»-Kleingruppentrainings bei der Planung und Durchführung eines ganzheitlichen, abwechslungsreichen und wissenschaftlich fundierten Trainings für übergewichtige Jugendliche (11-14 Jahre) unterstützt.
Methodik: Grundlage für diese Arbeit war eine umfassende Literaturrecherche zu Trainingsprinzipien, Interventionserfolgen und Bedürfnissen der Zielgruppe. Zusätzlich wurde eine achtwöchige Intervention mit vier übergewichtigen Jugendlichen durchgeführt. Neben den wissenschaftlichen Erkenntnissen flossen auch die subjektiven Bedürfnisse und Rückmeldungen der Jugendlichen ein. Die objektive Wirksamkeit des Trainings wurde mittels Tests zur Körperzusammensetzung, Kraft- und Ausdauerleistung sowie Blutdruckmessungen vor und nach der Intervention geprüft.
Ergebnisse: Die achtwöchige Intervention lieferte vielversprechende Ergebnisse. Die getesteten Jugendlichen konnten trotz der recht kurzen Interventionsphase sowohl ihre Ausdauer- als auch ihre Kraftleistungen verbessern. Neben den messbaren körperlichen Fortschritten vermerkten die Jugendlichen subjektiv eine gesteigerte Freude am Training und ein verbessertes allgemeines Wohlbefinden.
Schlussfolgerung: Das Trainingshandbuch bietet einen wissenschaftlich und empirisch fundierten und flexiblen Leitfaden, um das Training möglichst effektiv, abwechslungsreich und teilnehmerspezifisch zu gestalten. Die Durchführung eines Jahresprogramms wird die Nachhaltigkeit des Konzeptes weiter prüfen. Das Trainingshandbuch soll in diesem Prozess weiter optimiert werden. Langfristiges Ziel ist es, übergewichtigen Jugendlichen zu einem gesunden Lebensstil zu verhelfen und damit einen Beitrag zu einer nachhaltig gesünderen Gesellschaft zu leisten
Erfassung und Analyse der körperlichen Aktivität von Patienten mit einer nicht-stoffgebundenen Abhängigkeitserkrankung und Impulsvermittlung zur nachhaltigen Verbesserung der körperlichen Aktivität
Hintergrund: Die Verhaltenssüchte sind ein wachsendes Phänomen, das zum besseren Verständnis noch weiterer wissenschaftlicher Forschung bedarf. Zu herkömmlichen Therapien liegt bereits umfangreiche Literatur vor, während zu sportlichen Interventionen noch einige Lücken bestehen. Diese Studie untersucht, ob das Pilotprojekt «MOVE ME HOME» das sitzende Verhalten von stationären Patienten der Universitären Psychiatrischen Klinik (UPK) Basel wirksam reduziert.
Methoden: Patienten nehmen an einer wöchentlichen Bewegungsintervention nach dem MoVo-Konzept teil. Das Bewegungsverhalten wird mit dem SIMPAQ- Fragebogen gemessen. Zusätzlich werden Daten zur psychischen Gesundheit mit den Fragebögen SVF-18, BDI, FKK-32 und PSS erhoben. Jede Messung wird dreimal wiederholt (Baseline, Post, Follow-up). Die Daten wurden mittels ANOVA mit Messwiederholung und Spearman-Korrelationen ausgewertet.
Ergebnisse: Das sitzende Verhalten nimmt um 160 min/Tag ab, das stehende Verhalten nimmt um 56 min/Tag zu und die leichte körperliche Aktivität nimmt um 128 min/Tag zu 8 Wochen nach der Entlassung aus der Klinik (Follow-up). Mehr Beteiligung korreliert mit mehr Zeit in intensiver körperlicher Aktivität im Follow-up (rs=0.384 [M]; p=0.017). Eine stärkere Abnahme der sitzenden Tätigkeit korrelierte mit einer stärkeren Verbesserung des Depressions- (rs=0.447 [M]; p=0.006) und Stressempfindens (rs=0.336 [M]); p=0.032) zwischen Post und Follow-up.
Schlussfolgerung: Die Teilnehmer von «MOVE ME HOME» zeigen eine langfristige Verbesserung ihres Bewegungsverhaltens, und die Verringerung der sitzenden Tätigkeit korreliert moderat mit einer Verbesserung der Depressions- und Stresssymptome. Das Pilotprojekt erweist sich somit als wertvolles Interventionsprogramm für Patienten mit Verhaltenssüchten
Non-communicable diseases in people living with HIV in rural Africa: tackling the double burden
In 2019, there were around 38 million people living with HIV (PLHIV) worldwide. For the past decade, new HIV infections have been reduced by 23% and AIDS-related deaths have been reduced by 39% worldwide. All these achievements were possible mainly due to discovery and global rollout of antiretroviral therapies (ART), reduction in stigma and a global effort for funding to tackle the HIV epidemic. Sub-Saharan Africa is the most affected WHO region with around 20.7 million (54%) PLHIV. Since 2010, in the region, new HIV infections have been reduced by 38% and AIDS-related deaths have been reduced by 49%. This accomplishment was possible mostly due to extensive vertical HIV testing and treatment programs.
Each year, more than 41 million people die from non-communicable diseases (NCDs) worldwide. More than 15 millions of these deaths are premature deaths (people aged between 30-69 years) and 85% occur in lower- and middle-income countries. The most common NCDs that cause deaths are cardiovascular diseases, cancers, respiratory diseases, and diabetes mellitus. The risk factors for most NCDs can be grouped as behaviour or metabolic risk factors. The most common modifiable behaviour risk factors are harmful tobacco use, excessive alcohol consumption, eating unhealthy diet, physical inactivity, overweight and obesity. Other metabolic risk factors are raised blood cholesterol, and raised fasting blood glucose.
In sub-Saharan Africa, with the rapid demographic changes, an epidemiological transition from infectious diseases to non-communicable and chronic diseases can be observed. By 2030, mortality due to NCD is projected to surpass the mortality from combined infectious, neonatal, maternal and nutritional diseases. Therefore, there is an urgent need for innovative ways to tackle this double burden of diseases on the African continent.
The overall goal of this doctoral thesis was to summarise the burden of non-communicable diseases in sub-Saharan Africa. Additionally, data from rural Tanzania was used to estimate the burden of renal impairment, excessive weight gain and different comorbidities among HIV-infected patients receiving antiretroviral treatment.
The first study described the burden of the most relevant etiologies for NCDs in sub-Saharan Africa, which are arterial hypertension, heart diseases, diabetes mellitus and chronic kidney diseases. The review emphasized the importance of using a multifactorial approach, with emphasis on strengthening the existing health care systems to facilitate early diagnosis, treatment and management of NCDs. Furthermore, the review highlighted an urgent need for robust high-

quality evidence directed at informing national stakeholders on the key drivers of NCDs across Africa.
The second study assessed the prevalence, incidence and predictors of renal impairment among PLHIV receiving ritonavir-boosted protease inhibitors (bPI). Renal impairment was present in 7.6% of participants at the switch from first-line to bPI. Among participants with normal kidney function at the time of switch, 7.4% developed renal impairment after a median time of 3.5 years. The study reported risk associated with renal impairment at switch to be older age, low body mass index (BMI) <18.5 kg/m2 and arterial hypertension. Hence, this study highlighted the high burden of renal impairment among patients who are switched from first-line to bPI in rural sub-Saharan Africa and stress the importance of clinical monitoring of renal function for patients using ART.
The third study evaluated the weight change among newly diagnosed PLHIV starting treatment with either dolutegravir-based or efavirenz-based regimen in rural Tanzania. Additionally, it assessed risk factors associated with increased body weight ≥10% after 12 months of treatment. Initiating a dolutegravir-based ART regimen was associated with more weight gain compared to efavirenz-based regimens during the first 12 months of treatment, confirming observations from industrialized settings. Weight gain of ≥10% was associated with use of dolutegravir, level of immunosuppression, BMI at ART initiation and being female.
The fourth article is a letter to the editor that discussed the rollout of dolutegravir-based antiretroviral therapy in sub-Saharan Africa and its public health implications. The letter highlighted the lack of studies to evaluate long-term side effects of dolutegravir-based regimens among PLHIV living in sub-Saharan Africa. At the time, it was vital since there were studies that demonstrated excessive weight in PLHIV either starting treatment with dolutegravir-based regimens or those who were switched from other regimens to dolutegravir-based regimen. Therefore, this letter set the basis for the third study to evaluate weight gain in patients starting ART in rural Tanzania.
The fifth study investigated the age-related causes of morbidity and mortality of people living with HIV in rural Tanzania. Prevalence and incidence of different comorbidities in patients starting antiretroviral treatment was evaluated. The results showed that anaemia, arterial hypertension and undernutrition are the most relevant comorbidities with different age-associated frequencies and impact on death/LTFU in this population.
The sixth study is a study protocol for an open-label, three-arm, parallel randomized controlled trial conducted at two rural hospitals in Lesotho and Tanzania. The clinical trial compares the

efficacy and cost-effectiveness of three antihypertensive treatment strategies among HIV-positive and negative participants. The primary endpoint is the proportion of participants reaching the target blood pressure at 12 weeks. The results from this trial will help to identify the most effective and cost-effective treatment strategies for uncomplicated arterial hypertension among people of African descent living in rural sub-Saharan Africa.
The seventh article is an editorial that discussed the challenges for HIV-infected adolescents during transition from paediatric to adult HIV clinics in Africa. The editorial addressed the challenge of high HIV prevalence and incidence among adolescents and young adults in sub-Saharan Africa. The editorial stressed an urgent need to ensure that the transition policy from paediatric to adult-centred clinics is part of health care management for HIV-infected adolescents and young adults in Africa.
In conclusion, the studies included in this thesis highlight the double burden of infectious diseases and non-communicable diseases in sub-Saharan Africa. The lessons learnt will help to inform future guidelines on the importance of clinical monitoring of people living with HIV once they start antiretroviral treatment. The results highlighted the urgent need to improve pharmacovigilance systems in the region to monitor side effects of new drugs. Finally, the results from this thesis stress the importance that future research from local context to tackle the double burden of diseases in the region
Die Praxis des Heiratens: über die Anerkennung verbindlicher Liebesbekenntnisse
Eine Heirat ist meist ein besonderer Grund zum Feiern. Doch wie wird dieser emotional und rechtlich bedeutungsvolle Schritt heute zelebriert? Was wird durch die Praxis des Heiratens für das Paar und die Gesellschaft hergestellt? Und wie ist dabei das eigentümliche Verhältnis zwischen den romantisierten Hochzeiten und der vielfach konstatierten Bedeutungsreduktion der Institution Ehe zu verstehen? Die mikrosoziologische Analyse der Praxis des Heiratens begegnet durch die Differenzierung von Eheschließung und Hochzeit sowie die Berücksichtigung hetero- und homosexueller Brautpaare mehreren Forschungsdesideraten und zeigt aus affekt- und geschlechtertheoretischer Perspektive auf, wie Liebesbeziehungen heute durch Versprechen von Glück und Freiheit regiert werden.
Ausgangspunkt der Studie ist die statistisch gesehen markante Reduktion der Bedeutung der Institution Ehe in den letzten Jahrzehnten und die zugleich beobachtbare Hochkonjunktur emotional aufgeladener und romantisierter Hochzeitsinszenierungen. Durch den ethnographischen Zugang zu den aktuellen Praktiken des Heiratens, worunter sowohl der zivilrechtliche Akt der Eheschliessung als auch der symbolische Akt der Hochzeit gefasst wird, trägt die Untersuchung zu einem vertieften Verständnis der Bedeutung und Kontinuität des Heiratens bei und zeigt die Notwendigkeit auf, Statistiken und quantitative Daten durch qualitative Analysen zu ergänzen. Zugleich erweist sich der Einbezug von homosexuellen Paaren – die im Zeitraum der Untersuchung in der Schweiz noch keine Ehe schliessen dürfen, anlässlich ihrer Partnerschaftseintragungen aber dennoch Hochzeiten feiern – als äusserst produktiv für ein umfassendes Verständnis der Praxis des Heiratens.
Anhand von drei Spannungsverhältnissen, die den Themenkomplex der Heirat durchziehen, werden in der Einleitung drei Desiderate der soziologischen Forschung zur Ehe aufgezeigt und Fragestellung, Design sowie Anliegen der Studie vorgestellt. Kapitel 2 nimmt anhand historischer und soziologischer Studien eine Rekonstruktion von drei verschiedenen Phasen der Ehe im 20. Jahrhundert vor. In Kapitel 3 werden die subjekt- wie affekttheoretischen Ansätze vorgestellt, die für das Verständnis der Gleichzeitigkeit von Individualisierung und Konformität in der Praxis des Heiratens grundlegend sind. Diese Grundannahmen zum Zusammenhang von Machtverhältnissen und performativer Subjektivierung sowie zur Verschränkung von Allerpersönlichstem und Überpersönlichem stehen dabei in engem Verhältnis zu den soziologischen Praxistheorien, die wiederum den methodischen Zugang zu den Praktiken des Heiratens inspiriert haben. Abschliessend wir der ethnographische Datenkorpus vorgestellt, der durch die teilnehmende Beobachtung von elf Hochzeiten und die leitfadengestützten Interviews mit 24 frisch verheirateten Einzelpersonen erstellt wurde.
In den folgenden fünf empirischen Kapiteln werden unterschiedliche Praktiken des Heiratens beleuchtet: In Kapitel 4 der Entschluss der Paare für die Heirat und die zentrale Bedeutung der ebenso romantisierten wie vergeschlechtlichten Praktik des Heiratsantrags; in Kapitel 5 die Wahl der Form der Heirat, die typischerweise eine feierliche und rechtliche Anerkennung sowie eine paarinterne Bestätigung der Liebesbeziehung umfasst; Kapitel 6 beleuchtet die theatrale Praxis der Hochzeit, die auch als schönster Tag im Leben (insbesondere von Frauen) bekannt ist und nicht nur mit hohen Ansprüchen an ein perfektes Gelingen, sondern auch einer Romantisierung von Geschlechterdifferenz und -komplementarität einhergeht; Kapitel 7 dreht sich um die Frage, wie die hohen Scheidungsraten von den Paaren gedeutet werden und wie sie ihre verbindlichen Liebesbekenntnisse gegenüber der gesellschaftlich ‘normalisierten’ Scheidung positionieren; in Kapitel 8 schliesslich wird am Beispiel der Namenswahl diskutiert, wie sich die Gesetzesnormen des Eherechts zu den individuellen Bedürfnissen der Paare verhalten und aufgezeigt, dass eine formale Gleichstellung nicht unmittelbar zu einer gleichberechtigten Praxis führt.
Im Fazit werden die zentralen Einsichten aus der empirischen Analyse auf einer Metaebene weitergedacht und aufgezeigt: Es wird ausgeführt, warum es für das Verständnis der Praxis des Heiratens zentral ist, eine Differenzierung von Eheschliessung und Hochzeit im Begriff der Heirat vorzunehmen, in der Analyse aber zugleich die vorherrschende Verschränkung des zivilrechtlichen und zeremoniellen Akts in den Blick zu nehmen. Durch diese analytische Trennung und gleichzeitige Betrachtung von Hochzeit und Eheschliessung lassen sich ebenso wie durch die vergleichende Analyse der rechtlich voneinander unterschiedenen Realitäten der hetero- und homosexuellen Hochzeitspaare wesentliche Erkenntnisse gewinnen und offene Fragen in der soziologischen Forschung zur Ehe schliessen.
Durch die affekt- und subjekttheoretische Annahme einer grundlegenden Abhängigkeit der Subjekte von Anerkennung kann weiter gezeigt werden, inwiefern die Heirat eine wirkmächtige Regierungstechnik ist: Liebesbeziehungen werden heute kaum mehr durch Verbote, sondern vielmehr durch Versprechen von Glück und Freiheit regiert, wobei deren erfolgreiche Gestaltung in der Verantwortung der Einzeln liegt.
Vor diesem Hintergrund ist die Öffnung der Ehe für homosexuelle Paare, die in der Schweiz 2022 vollzogen wurde, nicht nur im Sinne eines emanzipatorischen Wandels hin zu mehr Gleichberechtigung zu verstehen. Für eine wirkliche Gleichberechtigung im Sinne der gleichwertigen Anerkennung von Differenz bedürfte es, so der Ausblick der Studie, einer Entkoppelung der Verschränkung von Liebe und Recht, also von Hochzeit und Eheschliessung, die die hegemoniale Praxis des Heiratens auszeichnet und ihre Kontinuität trotz allen Wandels im persönlichen Zusammenleben wesentlich begründet
Measuring and understanding user experience: current state and future needs
Over the last two decades of Human-Computer Interaction, the research focus has shifted from usability to the more holistic concept of user experience (UX). Given the subjective nature of UX, emphasis is placed on psychological constructs which are not directly observable. Thus, methods are needed to adequately measure these complex experiences, among which self-reported survey scales are some of the most popular. However, the psychometric quality of those scales is not always known or (re-)investigated, which would be essential to have confidence in the findings obtained from them. Consequently, this thesis consists of four manuscripts investigating the measurement of UX from different perspectives and through the investigation of various survey scales. In particular, the first manuscript studies one crucial question from UX research, namely, how interface aesthetics affect users' subjective experience and their objective performance in the to-date understudied but increasingly popular context of smartphone devices. The second manuscript looks at the quality of a particular scale for measuring website aesthetics and considers the effect negatively formulated items have on scale quality by offering an alternative positive version. The third manuscript investigates how survey scales are currently used and reported on in a particular area of research, namely player experience (PX). Finally, the fourth manuscript examines the quality of one particular scale for measuring PX. Overall, this thesis provides insights into the current state of scale usage as a method in UX research while highlighting areas for future improvement
Hierarchical tensor approximation of high-dimensional functions of isotropic and anisotropic Sobolev smoothness
In this article, we study the hierarchical tensor decomposition of functions from both smoothness classes, isotropic Sobolev spaces and anisotropic Sobolev spaces. For this purpose, we consider the known rank estimates in case of bivariate approximation, which can be found in [14] and [16], and successively apply them to analyze the truncated hierarchical tensor decomposition. In comparison to the isotropic case, we obtain improved results with respect to anisotropic Sobolev spaces. Indeed, the associated ranks of the truncated hierarchical tensor decomposition stay essentially bounded which beats the curse of dimension that is observed in the isotropic case
