A Bioprocess Engineering Approach to Boost Selection of Fully Segregated Transformants in Cyanobacteria
Cyanobacteria are photoautotrophic microorganisms with significant applications in biotechnology. Although many cyanobacteria, including Picosynechococcus sp. (formerly called Synechococcus sp.) PCC 11901 (Picosynechococcus) and Synechocystis sp. PCC 6803 (Synechocystis), are readily and naturally transformable, their polyploidy poses a major challenge. To obtain a stable phenotype, transgenic strains must be fully segregated, i.e. mutations must appear in all chromosome copies. Traditional protocols rely on re-streaking of colonies on increasingly selective plates, a time-intensive laboratory procedure that requires continuous intervention from the operator. This study proposes an alternative protocol that combines transformation in a batch system in liquid culture with transformant selection in a continuous-flow stirred-tank reactor system. This protocol led to the successful selection of homoplasmic transformants of Picosynechococcus containing, alternatively, an antibiotic resistance alone (construct "SmR") or a more complex construct ("bKT") that leads to the accumulation of a ketocarotenoid. The stability of SmR transformants under semi-continuous cultivation in the absence of antibioticsf was tested for 42 days, proving their potential fitness to industrial cultivation conditions. The selection process was also validated on the model species Synechocystis, demonstrating its applicability to other cyanobacterial strains

Rare Localization of Actinomyces israelii: Clinical, Radiological, and Histopathological Insights from a Lingual Case
Actinomycosis is a rare, chronic infection caused by Actinomyces israelii, a Gram-positive anaerobic bacterium that commonly inhabits the human oral cavity. Lingual involvement is extremely uncommon, accounting for less than 3% of reported cases, and poses a diagnostic challenge due to its ability to mimic both benign and malignant lesions. The present paper reports the case of a 38-year-old male presenting with a six-month history of swelling in the posterior region of the tongue, which exhibited cyclical size variation and fistula formation. Clinical and radiological evaluation revealed a solid mass extending into the mylohyoid and genioglossus muscles. Surgical excision was performed, followed by histopathological analysis, which confirmed the diagnosis of lingual actinomycosis. Postoperative antibiotic therapy was administered, and the patient showed complete recovery without recurrence. Lingual actinomycosis is a rare but important differential diagnosis for submucosal tongue lesions. A multidisciplinary approach involving imaging, surgical intervention, histological examination, and appropriate antibiotic therapy is essential for accurate diagnosis and effective treatment. Surgical excision combined with antibiotic therapy provides optimal outcomes, with long-term follow-up recommended to monitor for recurrence.
 
Adaptive multi-domain uncertainty quantification for digital twin water forecasting
Machine learning (ML) models are often used to predict demand in digital twins (DTs) of water distribution systems (WDS). However, most models do not provide uncertainty estimation, and this makes risk evaluation limited. In this work, we introduce the first systematic framework for hierarchical uncertainty transfer in regional water networks, because until now no method existed for DT of regional water systems. We propose Adaptive Multi-Village Conformal Prediction (AMV-CP), a method that keeps theoretical guarantees and also allows transfer of uncertainty information between villages that are similar in structure but different in operation. The main ideas are: (i) village-adaptive conformity scores that capture local patterns, (ii) a meta-learning algorithm that reduces calibration cost by 88.6%, and (iii) regime-aware calibration that keeps 94.2% coverage when seasons change. We use eight years of data from six villages with 6174 users in one regional network. The results show a theoretical basis for cross-village transfer and 95.1% empirical coverage (target was 95%), with real-time speed of 120 predictions per second. Early multi-step tests also show 93.7% coverage for 24-hour horizons, with controlled trade-offs. This framework is the first systematic method for controlled uncertainty transfer in infrastructure DTs, with theoretical guarantees under φ-mixing and practical deployment. Our multi-village tests demonstrate the value of meta-learning for uncertainty estimation and make a base method that can be used in other hierarchical infrastructure systems. The system is validated in a Mediterranean rural network, but generalization to other climates, urban settings, and cascading systems needs further empirical study

In sixteenth- and seventeenth-century England, ideas of predetermination and divine election constitute, especially in the gradual establishment of Puritan culture, the main frame of reference of the theology and ethics of evil, which, on principle, removes from the self any potential source of pleasure or seduction by excluding free will and, therefore, individual choice. Yet the experience of evildoing does not end in this closure of the subject within a fideistic perspective. Contemporary theatrical culture problematises the question by exploring in tragic play and participation in theatrical gameplay the limits and potential of freedom as an act of will recognisable in the assumption of guilt-as-doing as the foundation of one’s being. Shakespeare explores a whole range of possibilities of doing evil as cases of conscience as well as in relation to the reactions to evil done (or being done), from fear to terror and horror. It has been noted that the experience of horror threatens not just individual human lives, but the very essence of what it means to be human – rooted in the singular vulnerability of each embodied person. This encompasses the full range of violence, from overt atrocities like massacres and torture to more subtle, insidious forms of harm. In this sense, violence on the body has been interpreted as a violation of the human condition. This article explores how Shakespeare probes the boundaries of the human condition through the experiences of horror in Macbeth – distinct from fear and terror – focusing on the psychological, ethical, and symbolic dimensions at the outer limits of tragic catharsis
