    Protective Effects of Spirulina platensis, Voluntary Exercise and Environmental Interventions Against Adolescent Stress-Induced Anxiety and Depressive-Like Symptoms, Oxidative Stress and Alterations of BDNF and 5HT-3 Receptors of the Prefrontal Cortex in Female Rats

    Background and Purpose: Ample evidence indicates that chronic adolescence stress is associated with an increased risk of developing neuropsychiatric disorders in adulthood. Given the importance of the effective therapeutic ways to overcome adolescent stress-related deficits, the present study investigated the effects of Spirulina platensis (SP), environmental enrichment (EE), and voluntary exercise (EX) and their combination on anxiety or depression- like behaviors, oxidative stress, and alterations of BDNF and 5HT-3 receptors in the prefrontal cortex (PFC) induced by adolescent stress in adult female rats. Methods: During the adolescent period (PNDs30-40), rats were subjected to restraint stress. Then, the animals were subjected to SP treatment (200 mg/kg/day), EX, EE, and the combined treatments (SP+EX, and SP+EE) for 15 days between PNDs41-55. Subsequently, anxiety or depression-like behaviors, BDNF levels, oxidative stress markers and mRNA expression of BDNF and 5HT3 in the PFC were assessed. Results: Stressed rats demonstrated enhanced anxiety levels and depression-like behaviors in adulthood. Regarding the oxidative stress markers, stressed rats exhibited significantly higher levels of malondialdehyde, a lipid peroxidation product, higher activities of antioxidant enzymes (glutathione peroxidase and superoxide dismutase) and significantly lower total antioxidant reactivity capacity in the PFC. Additionally, adolescent stress significantly increased 5HT3 receptor mRNA expression and decreased BDNF content and its mRNA expression in the PFC. Treatments with SP, EX, EE, and the combined interventions alleviated these deficits. Conclusion: Our findings indicate that appropriate interventions during the adolescent period can protect against adolescent stress-induced behavioral, and biochemical defects and oxidative stress damage in adulthood

    Introduction: The most important goal of the dental education program, especially pediatric dentistry, is to train high-performance graduates, and in addition to its knowledge goals, this field has significant performance goals that should be reflected in the highest quality in its learning knowledge; Therefore, this study was conducted to investigate the achievement of practical educational goals of the Pediatrics Department of Ardabil Dental School. Materials and methods : In this descriptive cross-sectional study, a census of all students who had passed all pediatric dentistry units in Ardabil Dental School were included in the study (n = 59). For data collection, the standard questionnaire of Hori et al. (2014) was used to analyze the data. The SPSS software version 22 and Chi-square, Mann-Whitney U and Kruskal-Wallis tests were analyzed at a significant level of 0.05. Results: The results showed that the degree of agreement in achieving the learning objectives in Pediatrics lessons as a whole was relatively favorable; According to the students, the level of agreement in achieving the learning objectives was relatively favorable in Pediatrics lessons 1 and 3 and desirable in Pediatrics lessons 2. The results also showed that there is no significant difference in the achievement of learning objectives in Pediatrics lessons based on gender, interest in the field, grade point average (P> 0.05); But there was a significant difference in the achievement of learning goals in Pediatrics lessons based on the entrance quota (P <0.05) Conclusion: Based on the obtained results, students expressed an acceptable level of practical skills in the form of self-assessment, but the curriculum could not achieve its educational ideals.     Spatial distribution and ecological niche model of most intestinal parasites in Ardabil provinceby using of GIS and MaxEnt

    Background and Objective: Intestinal parasites are a health problem in many countries, especially in developing countries. Today, despite the high costs incurred annually by the World Health Organization and governments in eliminating, preventing, and treating these parasites, their existence is still evident. Intestinal parasites commonly affect children in low-income areas and developing countries. According to the World Health Organization, more than 30% of the population in developed countries suffers from parasitic diseases each year. More than 2 million deaths occur in developing countries due to this cause. A wide variety of worms and protozoa can cause infection or colonization in the digestive system of humans and animals. These organisms are usually transmitted indirectly through the fecal-oral route, including contaminated food, water, and soil; However, direct human-to-human and animal-to-human transmission is also possible in many cases Methods: In this study, we first extracted the characteristics of all intestinal parasites that have been detected and reported by all private sector laboratories in health laboratories and hospitals in the cities of the province in rural and urban areas, extracted their geographical coordinates and saved them in Excel format. Then it was produced using GIS10.4.1 software in the form of distribution maps in cities and then using MaxEnt3.3 software one of the intestinal parasites that has a high dispersion. Its ecological niche was determined using climatic and environmental factors. Results: The prevalence of different types of intestinal parasites was as follows: Giardia lamblia (79.4%), Entamba coli (10.5%), blastocystis hominis (4.2%), Ascaris (2.5%), Heminolpis nana (2.1%), enterobius vermicularis (0.8%) and Entamoeba histolytica (0.4%). The prevalence of intestinal parasites was higher in men than women. The prevalence was higher in rural area than in urban. Conclusion: In this study, risk areas are the center of the province and the most important parasite that is prevalent is Giardia, which should be planned to study pathogenicity, epidemiology and other factors affecting the prevalence of these parasites in the province, especially in the central part of the province

    Evaluation of Mothers' Self-efficacy in Caring of Children with Asthma Referred to the Subspecialty Clinic and Ardabil Educational-Medical Center of BuAli in 2018

    Background & Aim: Asthma is one of the most common chronic diseases in childhood and the first causes of admission of children in the emergency room and hospitalization. Families, especially mothers, play a key role in improving the child with asthma, and high self-efficacy of mothers is associated with improved control of asthma symptoms. Therefore, main aim of this study was to Assessment of self-efficacy of mothers of children with asthma referred to Ardebil-Bu-Ali educational center in 2018 and its relation with demographic variables. Methods: This descriptive-correlational study was performed on all children with asthma (6-11 years old) referring to Ardabil Ardabil University of Medical Sciences and Health Services, a non-random and non-randomized sampling method. In a year 2018 was done. The instrument for data collection was a two-part questionnaire including demographic characteristics, and maternal self-efficacy questionnaire. Data analysis was performed using descriptive statistics and analytical statistics using spss18 software. Results: The results of the study showed that 51.9% (n = 138) of mothers had good self-efficacy and 48.1% (128) of mothers had undesirable self-efficacy. Also, there is a significant relationship between the high self-efficacy of mothers and their educational level, job, history and frequency of admission of their child and mothers’ education source (p<0.001). Conclusion: The findings of the present study indicate that mothers of asthmatic children have high self-efficacy. Therefore, mothers' self-efficacy should be considered with regard to their demographic variables in order to improve and control asthma symptoms. Key words: self-efficacy, asthma, mother, chil

    The efficiency of BrucellaCapt method in diagnosis of human brucellosis

    Abstract Background: Brucellosis is a zoonosis of great and worldwide public health concern that can cause a severe febrile illness in humans. Aims: This study aims to evaluate the efficiency of the TaqMan Real-time PCR and serological methods in detecting Brucella spp in clinical specimens that have been collected from suspected patients in Ardabil, Iran. Materials and Methods: In this cross-sectional study, a total of 113 consecutive patients suspected of brucellosis who were referred to the three hospitals in Ardabil province were selected. In the first step, the diagnosis of brucellosis was performed by serological methods including the Rose Bengal slide agglutination test, Wright test, 2-ME test, and BrucellaCapt test. In the next step, TaqMan real-time PCR with primer and probe targeting the bcsp31 gene was used for the detection of Brucella spp. Specificity, sensitivity, and positive and negative predictive values of the TaqMan real-time PCR assay were calculated. Results: Among 113 suspected patients with different clinical manifestations, the Rose Bengal slide agglutination test, Wright test, and 2ME test were positive in 60 cases; however, the BrucellaCapt test titer was 1:160 for one patient. Seven patients had high initial serum antibody titers. Among positive cases, no correlation was observed among gender, age, and life (residence) in urban or rural areas. The TaqMan real-time PCR was positive in 35% of all 60 positive cases. The comparison of the results of the BrucellaCapt and TaqMan real-time PCR methods revealed that 19 out of 54 (35.2%) and 2 out of 6 (33.4%) BrucellaCapt positive cases with titers of > 1:320 and ≤ 1:320 were positive, respectively. The sensitivities and specificities of the TaqMan real-time PCR assay were 100% and 49.1%, respectively. Conclusion: The sensitivity of the TaqMan real-time PCR assay was low in the diagnosis of brucellosis, while the BrucellaCapt test turned out to be a very valuable, sensitive and specific test for the diagnosis of brucellosis in suspected patients and, thus, can provide reliable results in medical laboratories

    Evaluation of parental awareness about the presence of permanent first molars and its relation to DMFT index in 6_9_year_old children in year 2019_2020

    Background and Objective: The first permanent molars erupt in 6-7-year- old children in distal surface of primary second molar, without any side effects, therefore in most cases children and parents are not aware of their eruption. However these teeth are predisposed to caries and need close sanitary care and parental notice. Parent's awareness about existence of first molar as a permanent tooth in children's mouth could lead to health preservation of this tooth. This study aimed to determine the level of parental awareness about the presence of PFMs and its relation to DMFT index in 6- 9-year-old children in Ardabil. Methods: This cross-sectional study was performed on 366 students aged 6-9 in Ardabil and their parents. The sampling method in this study was cluster sampling. After sample selecting the schools and students, the checklist was completed by the researcher with an interview with the students' parents. DMFT was determined as the first permanent molar for each child Results: In this research 80/3./. parents knew about the first permanent molar Eruption in their childs mouth. The mean of DMFT of the first permanent molar in students was 1/13_+./92 . the correlation between DMFT6 and parental knowledge (P=0.523) and gender (P=0.235) was not significant. There was a significant correlation between awareness about the presence of permanent first molars and Father’s education and job.     A Comparative Study of pH and Immunoglobulin A in Unstimulated Saliva in Children with Early Childhood Caries and Caries Resistant Children between the ages 3-6 in Ardabil Faculty of Dentistry during 2018-2019

    Background and Aim Early childhood caries (ECC) is one of the most common chronic childhood diseases. Furthermore, saliva as a host factor plays an important role in keeping the integrity of oral structures and some salivary characteristics and components have been suggested as factors in children for development of ECC. This study compared pH and immunoglobulin A levels of unstimulated saliva in 3-6 years old children with and without early childhood caries in patients referred to Ardabil Dental School on 2018-2019. Materials and Methods In this descriptive cross-sectional trial, 82 children of 3-6 years old with and without involvement of early childhood caries (41 for each group) were selected. Following examination and diagnosis of the children, they were assigned into 2 groups regarding incidence or not involving early childhood caries. Unstimulated saliva of the children were collected by spitting method at 9-10 p.m. The collected saliva were freezed at -70°C temperature and the temperature were reached up to the room temperature when measuring the variables. Salivary pH levels were estimated by electronic pH meter and the A Immunoglobulin levels were estimated by ELISA method and specialized kits. Salivary pH and A immunoglobulin levels for both groups were statistically analyzed by independent T-Test. Results Salivary A immunoglobulin levels in the children with and without early childhood caries were 1.46±0.27mg/dl and 2.63±0.37mg/dl respectively while salivary pH levels of children with and without early childhood caries were 7.05±0.42 and 7.69±0.37 respectively. Salivary A immunoglobulin (p=0.001) and pH levels (p=0.001) in children with early childhood caries were significantly lower than children without early childhood caries. Conclusion Then, incidence of early childhood caries in 3-6 years old children decreased salivay pH and immunoglobulin A levels significantly as compared to the children without early childhood caries. Then, the decreased levels of these parameters are suggested as risk factors for early childhood caries

    In Vitro Anticancer Effects of All-trans Retinoic Acid in Combination with Dacarbazine against CD117+ Melanoma Cells

    Background Malignant melanoma is a common form of skin cancer that contains different cell types recognized by various cell surface markers. Dacarbazine-based combination chemotherapy is frequently used for the treatment of melanoma. Despite its potent anticancer properties, resistance to dacarbazine develops in malignant melanoma. Here, we aim to improve response to dacarbazine therapy by pretreatment with all-trans retinoic acid (ATRA) in CD117+ melanoma cells. Methods The CD117+ melanoma cells were sorted from A375 malignant melanoma cell line using magnetic-activated cell sorting (MACS). The cell viability was examined by cell proliferation assay (MTT). Apoptosis was determined by acridine orange/ ethidium bromide staining. Indeed, we performed flow cytometry to evaluate the cell cycle arrest. Results Here, the CD117+ melanoma cells were incubated with various concentrations of ATRA, dacarbazine, and their combination to determine IC50 values. We found that 20 µM ATRA treatment followed by dacarbazine was found to be more effective than dacarbazine alone. There was an indication that the combination of ATRA with dacarbazine (ATRA/dacarbazine) caused more apoptosis and necrosis in the melanoma cells (P<0.05). Furthermore, ATRA/dacarbazine treatment inhibited the cell at the G0/G1 phase, while dacarbazine alone inhibited the cells at S phase. Conclusion Collectively, combined treatment with ATRA and dacarbazine induced more apoptosis and enhanced the cell cycle arrest of CD117+ melanoma cells. These results suggested that ATRA increased the sensitivity of melanoma cells to the effect of dacarbazine
