Sr. Joanita Koors
In this interview Sr. Joanita Koors reflects on her life in community, specifically the influences that helped shape her ongoing decision to join the convent in Oldenburg, in particular the warmth and trust of Sr. Joel Franks. Self-described as an common person, an ordinary farm girl from Greensburg, Sr. Joanita details her bargaining with God, her time as an educator in Evansville, New Albany, North Vernon, and Indianapolis, as well as her mission to help all students realize their worth and potential. Additionally, she discusses the change of her prayer life from communal prayer to adopting the the more personal contemplative gaze , with coffee. Leveraging LMS Integration with LibGuides
This project was an attempt to better integrate library resources within the campus LMS (Canvas), specifically through the application of the LibGuides widget tool linking feature. With content directly discovered in the LMS, students have seamless access to library-related course content, primarily created in LibGuides. Linking was done without an LTI tool, and proved useful in embedding whole guides, specific pages, content boxes as well as our discovery layer (OCLC\u27s Discovery). Limitations included embedded media (streaming) or other objects with more complicated coding structure
Anti-Inflammatory and Anti-Angiogenic Effects of Black Raspberry Extract on Human Esophageal and Intestinal Microvascular Endothelial Cells
Polyphenolic compounds (anthocyanins, flavonoid glycosides) in berries prevent the initiation, promotion, and progression of carcinogenesis in rat’s digestive tract and esophagus, in part, via anti-inflammatory pathways. Angiogenesis has been implicated in the pathogenesis of chronic inflammation and tumorigenesis. In this study, we investigated the anti-inflammatory and anti-angiogenic effects of black raspberry extract (BRE) on two organ specific primary human intestinal microvascular endothelial cells, (HIMEC) and human esophageal microvascular endothelial cells (HEMEC), isolated from surgically resected human intestinal and donor discarded esophagus, respectively. HEMEC and HIMEC were stimulated with TNF-α/IL-1β with or without BRE. The anti-inflammatory effects of BRE were assessed based upon COX-2, ICAM-1 and VCAM-1 gene and protein expression, PGE2 production, NFB p65 subunit nuclear translocation as well as endothelial cell-leukocyte adhesion. The anti-angiogenic effects of BRE were assessed on cell migration, proliferation and tube formation following VEGF stimulation as well as on activation of Akt, MAPK and JNK signaling pathways. BRE inhibited TNF-α/IL-1β-induced NFκB p65 nuclear translocation, PGE2 production, up-regulation of COX-2, ICAM-1 and VCAM-1 gene and protein expression and leukocyte binding in HEMEC but not in HIMEC. BRE attenuated VEGF-induced cell migration, proliferation and tube formation in both HEMEC and HIMEC. The anti-angiogenic effect of BRE is mediated by inhibition of Akt, MAPK and JNK phosphorylations. BRE exerted differential anti-inflammatory effects between HEMEC and HIMEC following TNF-α/IL-1β activation whereas demonstrated similar anti-angiogenic effects following VEGF stimulation in both cell lines. These findings may provide more insight into the anti-tumorigenic capacities of BRE in human disease and cancer
Characterization of Spatio-Temporal Expression of Novel Hypoxia-Induced Genes in Zebrafish
Hypoxia is a critical stress that can affect the normal development of a mammal embryo. Normally, Hypoxia is buffered by the Utero-placental apparatus in human infants; however, prematurely delivered infants may encounter developmental defects due to the loss of the controlled environment of the uterus. Therefore, understanding the function of the genes that are hypoxia-induced is critical to understanding developmental injury in order to attempt to prevent or treat it. Why zebrafish as a model? Experimenting with zebrafish has many advantages: Approximately 2/3 of its genome is conserved with Humans, Zebrafish embryos can tolerate a wide range of hypoxia and the embryos develop ex-utero, which allows precise manipulation of the hypoxic environment and observation of phenotypes
The Effect Of a “Micronegotiation” Technique on Team Interactions
Conflict can have detrimental effects on team interaction, performance, and member satisfaction, so research on tools and techniques aimed at reducing or resolving conflict is crucial. This study trained the leaders of teams made up of health profession students on a micronegotiation technique (Rogers & Lingard, 2006) to measure its effect on levels of task conflict, relationship conflict, team performance, and team member satisfaction. The research was conducted at a mid-size, Midwestern university and included 148 students from Radiology, Physiology, and Microbiology courses divided into 47 teams. No statistically significant differences were identified for any of the dependent variables between the groups whose leaders were trained in the micronegotiation technique and the groups whose leaders were not trained on the technique. While this may seem to indicate a shortcoming on the part of the technique, low levels of emotional investment in the process may have led to little group conflict to address. Additionally, the real value of the technique may be as a way for team leads to assess conflict strategies rather than as a resolution tool

Sr. Mildred Millie Speed
In her oral history, Sister Mildred Millie Speed discusses how she learned the importance of love and the true value of family from seeing the relationship between her mother and father at a young age. She reflects on how her vocation to the Sisters of Saint Francis was based on a letter from her dear friend, Sister Charlene Wolf, and how her time teaching at various levels of education in Illinois, Indiana, Montana, New Mexico, and Ohio taught her to follow the vow of obedience—how to, ultimately, become “deeply Franciscan.” Sister Speed also describes what it was like to work with Native Americans in New Mexico, a group that had “a knowledge of our God and a love of the Great Spirit, as they refer to God,” and her starting a Kairos retreat for high school students in Middletown, Ohio. Sr. Robbie Pentecost
Equipped with degrees in finance and accounting and influenced by a college course on social issues in New York, Sr. Robbie Pentecost decided to enter the community of the Sisters of Saint Francis, Oldenburg more than 31 years ago. Her desire to volunteer, serve the poor, and a trust in the Lord also guided her decision to receive her MBA and start a restaurant in St. Louis that hired those who were unemployed. After 10 years working for the Christian Appalachian Project, Sr. Pentecost decided her call was to work for the Sisters of Saint Francis as a planned and major gifts coordinator. She describes her life as “A wonderful journey of mystery, adventure, and possibilities” while always remembering that “God is in charge.” Throughout her oral history, Sr. Pentecost continues to reveal important advice on discernment including how the process involves intuition, all of the senses, logic, and “sitting in silence with the Lord.” She highlights how listening to your heart, making clear versus emotional decisions, and talking with others are also important for successful discernment. Building a Community of Experts
Librarianship is becoming more complex with a broader range of services to support with little or no new people to take on these additional roles. The group of libraries in the Private Academic Library Network of Indiana consortium are working together to grow their expertise by expanding their role beyond their single library. With initial and projected ongoing training from OCLC and coordination from PALNI, a team of experts in acquisitions, cataloging, serials, circulation, and discovery systems from multiple PALNI libraries provide consultation and site visits to provide in-depth, in-person support to libraries. Future areas of expert development will include license and knowledgebase collaboration. By pooling our resources and experience together, we are creating space for innovation
Construction and Validation of a Novel Pathway Ratio Reporter System (PARRES)
The Transforming Growth Factor-beta (TGFβ) superfamily is a large group of signaling molecules whose members play conserved roles in embryo specification, organogenesis, and tissue homeostasis. Structural considerations delineate the TGFβ superfamily into ligands that interact with ALK1/2/3/6 or ALK4/5/7, with the former being mostly BMPs and the latter being mostly Activins and TGFβs. Reports of distinctly opposing effects between the BMP/GDF and Activin/TGFβ pathways are becoming increasingly common. A major focus of our laboratory is elucidating the molecular mechanisms that allow the BMP/GDF and Activin/TGFβ pathways to antagonize one another. To accomplish this goal, we must create a first-of-its-kind biosensor reporter system (called the Pathway Ratio Reporter System, or PARRES) that will allow us to examine the real-time ratio of BMP/GDF vs. Activin/TGFβ in vivo. PARRES is a novel biosensor system that uses two independent fluorescent reporters that are linked through reciprocal RNA interference so that, through mutual antagonism, fluorescent signal occurs only when transcriptional activation through one pathway is dominant. Here, we focus on the crucial, initial characterization of PARRES – the maturation and half-life kinetics of the fluorescent proteins. Using HEK293 cells, we demonstrate that the maturation kinetic of the Emerald variant of Green Fluorescent Protein is acceptable but the half-life is far too long for our needs. We subsequently destabilize Emerald to an approximate half-life of 29 hours by appending a validated degradation domain. Current studies involve generating point mutations in the destabilized Emerald protein to further reduce its half-life. Once completed, this biosensor will be especially useful to our lab as a tool for examining the influence of canonical signaling by skeletal mesenchymal progenitors, a population known to rely on BMPs, GDFs, TGFβ and Activins to instruct their differentiation into tissue-specific cell types such as osteoblasts
