Subversive Content Design User Experience Survey Data
This study explores the use of subversive game content in critical game design to address sensitive social, political, and personal topics while avoiding the risk of undermining their significance. Critical game design is focused on conveying messages and addressing real-world issues, with the goal of fostering thoughtful engagement and critical thinking among players as they interact with the game. However, despite these serious intentions, critical games retain fundamental elements such as rules, mechanics, and in-game objectives. A seamless gaming experience may unintentionally shift players' focus towards entertainment rather than the intended message, due to typical expectations of the medium. This unintended diversion risks trivializing the serious messages embedded in the games. To address this challenge, several subversive game content design strategies are proposed, including leveraging familiarity, the early introduction of subversive elements, justifying subversion, and promoting player agency. Two video games, Border Politics and Get Good: Grinding Chronicles are developed to test the effectiveness of the proposed design strategies. These strategies are intended to challenge conventional gaming norms and intentionally induce player discomfort, which may be necessary to safeguard against the trivialization of sensitive topics. The user experience results (N = 54) show that the proposed design strategies are effective in maintaining player engagement with the intended messages, successfully balancing the need for critical reflection with the inherent entertainment aspects of gaming. The data demonstrates that these strategies work well in preventing the trivialization of sensitive topics, thereby validating their use in critical game design
The Impact of Colour Vision Deficiency on the Ability of Stroke Clinicians to Read Perfusion Brain Scans
Colour vision deficiency (CVD), more colloquially known as colour blindness, impairs the perception and differentiation of various colours. CVD affects 1-6% of the population including medical professionals. CVD has been negatively associated with how medical staff recognise abnormalities in various disciplines, including histopathology and microscopy.
Recent advances in treatment delivery have seen the increasing use of CT perfusion and colour maps in the management of acute stroke. The impact of CVD on interpretation of these colour perfusion maps has not been explored. It is possible that certain colour palettes may pose a problem for clinicians with CVD and if clinicians with CVD are less good at interpreting certain colour maps, this could directly impact on patient care. The colour palettes used in perfusion maps could be adjusted to accommodate professionals with CVD if a significant difference in interpretation is identified.

Classical observations suggest a connection between 3D gene structure and function, but testing this hypothesis has been challenging due to technical limitations. To explore this, we developed e-HiP-HoP, a model based on genome organisation principles to predict the 3D structure of human chromatin. We defined a new 3D structural unit, a “topos”, which represents the regulatory landscape around gene promoters. Using GM12878 cells, we predicted the 3D structure of over 10,000 active gene topoi and stored them in the 3DGene database. Data mining revealed folding motifs and their link to gene ontology features. We computed a structural diversity score and identified influential nodes—chromatin sites that frequently interact with gene promoters, acting as key regulators. These nodes drive structural diversity and are tied to gene function. e-HiP-HoP provides a framework for modelling high-resolution chromatin structure and a mechanistic basis for chromatin contact networks that link 3D gene structure with function. Plasticity in malaria parasite development: mosquito resources influence vector-to-host transmission potential
Parasites rely on exploiting resources from their hosts and vectors for survival and transmission. This includes nutritional resources, which vary in availability between different hosts and changes during infections. For malaria (Plasmodium) parasites, sexual reproduction (sporogony) and subsequent development of oocysts, which produce sporozoites infectious to the vertebrate host, occurs in the mosquito vector. Mosquitoes in the field exhibit diversity in the amount and type of food they acquire, directly impacting the nutrients available for the replication and development of parasites. While the rate of parasite transmission from vector to host is influenced by the nutritional state of mosquitoes, whether this is due to resource limitation mediating parasite development and productivity is poorly understood. We use the rodent model parasite P. chabaudi and the vector Anopheles stephensi to ask how variation in the amount of sugar and blood provided to malaria-infected mosquitoes affects parasite transmission potential. We show that parasites in well-resourced mosquitoes grow faster and have a 1.7-fold higher sporozoite burden those whose vectors only receive sugar. However, this increase in productivity is only partly explained by oocyst development, suggesting that resource availability also impacts the ability of sporozoites to reach the salivary glands. As well as challenging the assumption of a simple relationship between the number or size of oocysts and transmission, our findings suggest malaria parasites may actively adjust within-vector development to best exploit nutritional resources; while parasites in low-resourced mosquitoes exhibited a reduction in oocyst burden during sporogony, the remaining oocysts developed more rapidly, reaching a similar size to those in well-resourced mosquitoes. Understanding the impacts of resource availability for malaria transmission is urgent given that parasites encounter increasingly variable vectors as consequences of climate change and vector control tools
