NuA4 interaction with RNA polymerase II is stimulated by phosphorylation of CTD serines 2 and 5
Transcription of protein-coding genes is carried out by RNA polymerase II (Pol II). The largest of Pol II’s 12 subunits (Rpb1) has a long C-terminal domain (CTD). The CTD in yeast is composed of 26 repeats of the heptapeptide Y1S2P3T4S5P6S7. Post-translational modification of the CTD is important for recruiting protein complexes to gene bodies. We investigated the importance of serine 2 and serine 5 phosphorylation (Ser2p/Ser5p) for interaction between the CTD and NuA4. NuA4 is the only essential lysine acetyltransferase complex in yeast and has been shown to be important for both transcription initiation and elongation. We have previously proposed that NuA4 occupancy in gene bodies is stimulated by interaction with the CTD phosphorylated on serine 5, but were not able to observe binding to Ser5p peptides in vitro. In this study, we examined NuA4 binding to Pol II in mutants affecting the CTD kinases Kin28, Bur1, and Ctk1. Kin28 phosphorylates serines 5 and 7, while Bur1 and Ctk1 phosphorylate serine 2. We found that NuA4 interaction with Pol II as measured by coimmunoprecipitation was reduced in kin28 mutant cells. Consistent with this, NuA4 occupancy at the ADH1 3’ ORF was also reduced in kin28 cells. Interestingly, loss of Ser2p in either bur2Δ (the cyclin for Bur1) or ctk1Δ led to reduced NuA4 binding to Pol II as well as decreased occupancy across GAL1. These results provide the first evidence that NuA4 may be able to bind both the Ser2p and Ser5p forms of the Pol II CT
Decision Regret and Correlation with Results Amongst Parents of Children who have Undergone Whole Exome Sequencing
Whole exome sequencing (WES) is pushing its way to the forefront of genetic testing strategies amongst both adults and children with complex disease. For affected individuals that have phenotypes that do not point to a specific causative gene, or have had previous inconclusive test results, WES is proving itself to be extremely useful in a clinical setting. WES is the most comprehensive genetic testing currently available, leading to a conclusive test result for approximately 25- 30% of individuals, many of whom have had prior, inconclusive evaluations. However, since this test is new in the clinical realm, little research exists examining the psychological implications of WES, specifically decision regret. This is important to understand as the clinical utility of this testing becomes more popular. We surveyed parents of children who have undergone WES to determine whether decision regret correlated with their test result. Results demonstrated no statistically significant difference in total regret between those with a positive, negative, or uncertain result. This study indicates that type of results may be a poor predictor of decision regret amongst those undergoing whole exome sequencing, and highlights the need for future research to identify potential factors that may influence decision regret amongst these patients

Income plays an important role in day to day life. Many factors affect the employment status of an individual. While some factors are personal traits such as self motivation and perseverance, where a person lives can also have an impact on their employability. Living in a state with a high median income can be beneficial to a person’s ability to earn a wage to support a standard of living. A state’s median income can be influenced by a variety of factors. Some factors are environmental, such as work place infrastructure and comfort, and geographical conditions such as location, climate and weather conditions. A state’s ability to have a strong economy which helps provide jobs for its residents also depends on the educational attainment of it residents and how welcoming the state is to industry. In this paper, we will examine five attributes that help determine the level of median income in each state. We will estimate a regression model for these variables and examine how these variables affect the median- income
The Effects of Caffeine and Body Change.
The Effects of Caffeine and Body Change. Recent studies show that black coffee is good for controlling weight, dilating airways, and decreasing chronic fatigue. The purpose of this study was to test the relationship between different roasting level of coffee beans and body change. Mice were placed in a 2 bottle-choice cage; one bottle contained water while the 2nd bottle contained a type of coffee (light/ medium/ dark). Additionally a separate 4th group of mice will receive a 200mg caffeine/water solution.All mice were given same amount caffeine, which is about one cup of coffee. The solutions were given 5 times a week, and body weights were taken each day to check for any dehydration. Bottle weights and body weights of the mice were checked daily. Lastly, as a means of incentive to drink the coffee or caffeinated solutions, water bottles were removed from cages for 2-3 hours every morning. Although there was no significant difference in water consumption between the groups, there was a significant difference in amount of caffeine consumed by the different roasting groups with the dark roast group having the highest level of consumption over all 3 weeks. Finally, there was no significant change in body weights among all groups negating prior evidence of caffeine as a means for weight control
Age and size at first reproduction in the Channeled Whelk population of the Great South Bay, Long Island, New York.
The channeled whelk, Busycotypus canaliculatus, is a commercially valuable gastropod found on the east coast of the United States. In New York, the channeled whelk fishery is largely unregulated, due to a lack of knowledge regarding the life history of the species. This study focuses on determining the age and size at first reproduction of the channeled whelk population in the Great South Bay, on the southern shore of Long Island. Whelks were obtained from a commercial fisherman and individually dissected to determine maturity; correlations to shell length and shell width were then examined. The operculum of each individual dissected was then examined for growth striae. Previous findings have confirmed that growth striae, dark rings on the operculum, are laid annually and can be used to determine approximate age in gastropod species (Kreuter 1985). Male channeled whelks of the Great South Bay population were found to reach sexual maturity at around 7 years of age, with shells generally at least 135mm in length and 70mm in width. Females were found to mature more slowly; first reaching sexual maturity at around age 8 with shells approximately 155mm in length and 90mm in width. The long-lived nature of the species and late onset of maturity makes the species especially vulnerable to being harvested before having the opportunity to reproduce and replenish the fishery. Therefore, the need for regulation is imperative to avoid a future fishery collapse; the goals of this study are to provide scientific backing in support of future management decisions
What Parts of the Nucleosome Does NuA4 Bind To?
The DNA in the nuclei of eukaryotic cells is organized into a structure called chromatin. The basic unit of chromatin is the nucleosome, which contains DNA tightly wrapped around eight histone proteins, two each of histones H2A, H2B, H3, and H4. Chromatin prevents other proteins from accessing the DNA and must be disassembled before transcription can take place.The NuA4 lysine acetyltransferase (KAT) complex acetylates histones H4 and H2A, weakening the histone-DNA interactions, and stimulating chromatin disassembly. NuA4 is the only essential KAT in yeast and is involved in transcription, DNA repair, and cell cycle control. While NuA4 acetylates nucleosomes, it is largely unknown how NuA4 actually binds to the nucleosome. We investigated the parts of the nucleosome bound by NuA4 by screening a library of histone point mutants for defects in H4 acetylation. We have so far analyzed H2A and parts of H2B and H4. We found mutations in all three histones that increased and decreased H4 acetylation. As expected, mutation of NuA4 targets H4K5 and K8 dramatically reduced acetylation. H2A L66A reduced acetylation. The most surprising mutations affecting H4 acetylation were threonine 118 and serine 125 in H2B, which both reduced H4 acetylation. We will continue to confirm these results and examine how the mutations affect NuA4-nucleosome binding. Our preliminary finding suggests that NuA4 may be able to interact with multiple parts of the nucleosome
A Study on The Positives and Negatives of Using Technology In The Classroom
As the growth of technology application in the classroom continues to grow, educators and researchers speculate as to how effective the use of assistive technology, and educational technology aids are. Over time, several researchers have done studies, and conducted observations on technological aids and devices being used in the classroom. Some results were measurable, and others observable, but mixed results were found. This study explores the different research conducted on the subject, and several of the factors that contribute to the use of, and success of technology in the classroom. Of the research examined, most of the responses were positive, supporting the idea that incorporating technology into the classroom is beneficial for both the students and the teachers. However, with the concerns expressed by teachers, the success of technology in school settings is dependent on the self-efficacy and professional development, or training, that the teachers receive. Using a) changes made to the Federal and state government policy on education, b) examples of implementation of technology in the classroom, both positive, negative and questionable, c) the use of technology with diverse groups of students, d) the perceptions of students, teachers/school staff, and parents, and e) research on the self-efficacy and professional development and technology training of teachers, this study concluded that the use of technological devices, programs and aids in the classroom should improve students’ assessment performance, level of engagement, and academic success. However, these results depend directly on the self-efficacy, and continuous training of classroom teachers
