    Developing a Protocol for Bacteriophage Guided Evolution

    Bacteriophage therapy is being explored as an alternative to antibiotic use due to an increase in antibiotic resistance. Expansion of phage host ranges is important for future phage therapy. A protocol was developed based upon the Appelmans Protocol to expand the host range of selected Gordonia bacteriophage. A spectrophotometer was used to determine lysis in a 96-well plate and confirm the success of the methods. A 100 phage to 1 bacteria concentration ratio was found to be effective. Two methods of bacteria concentration calculations were compared, one using a countable plate and the other using a spectrophotometer. The findings of the experiments were applied to a final protocol, which is followed by instructions on publishing research

    Quantification of Gordonia terrae Bacteriophages using Lysate qPCR

    Bacteriophages are viruses that infect and replicate only in bacterial cells. Quantitative PCR was used in this experiment to count the number of DNA copies in the phage lysate sample. In this project, the more rapid lysate qPCR method was compared to standard plate count titer assays using the double layer agar method, to match the number of genomic copies with the plaque forming units in the lysate. Initially, multiple primer sets were designed to amplify the best primers and reaction conditions to use for phage detection. Because qPCR is sensitive to contamination, there were multiple controls to ensure no contamination. Subsequently, a standard curve was generated by comparing the phage concentration determined from titer assays with that measured using SYBR Green qPCR. The optimized lysate qPCR method will be a faster method to determine the number of bacteriophage DNA copies in phage lysate, allowing the rapid determination of phage concentration for optical density-based infection growth curve experiments

    Synthesis of ZnTPP-IL as a Potential Photodynamic Therapy Agent

    Photodynamic Therapy is a new technique used in cancer treatment involving the use of a photosensitizer—a porphyrin—and light exposure to kill malignant cells. In this research, ZnTPP-IL was used as a photosensitizer with white light as a possible source of treatment of NIH 3T3 fibroblast and A549 lung cancer cell lines, with preliminary DNA testing on MYC-22 and G-quadruplex samples. ZnTPP-IL was synthesized by the reaction of the porphyrin ZnTPPC with isoleucinol. The new porphyrin was then purified by column chromatography using Sephadex LH-20 and G-50. ZnTPP-IL was characterized through NMR, IR, UV-vis, and fluorescence spectroscopies. The purity of the compound was then determined using HPLC. Additionally, ROS production using ZnTPP-IL was determined. The cytotoxicity of ZnTPP-IL was then examined by analyzing MTT assay with A549 lung cancer cell lines when paired with white and red light

    Dr. Wesley Kluck did not have a restful 2020. As university physician and vice president for student development at Ouachita, he led the university’s health efforts in response to COVID-19, chairing Ouachita’s Health Monitoring and Action Team (HMAT). “The worst thing was waking up in the middle of the night, thinking of things,” he said. “There was a lot of that. Sometimes I would get up and make a PowerPoint real quick, just type it all down and go back to sleep.” HMAT, a group of 13 faculty and staff members as well as the Student Senate president, worked through a planning guide of more than 625 items to prepare Ouachita for on-campus learning in the fall, collaborating with colleagues across campus to check off each one. It’s no wonder he couldn’t sleep
