Graphene-Gold Hybrid Nanomaterial Based Impedimetric Immunosensor for H3N2 Influenza A Virus Detection
H3N2, known as "swine flu" is a kind of influenza A virus that can also infect humans. Therefore, rapid, practical and accurate techniques for H3N2 detection is needed. In this study, we fabricate Graphene-gold hybrid nanomaterial modified hemagglutinin-3 antibody (anti-H3) based electrochemical immunosensor for practical, sensitive, and selective detection of H3N2 virus. The specific interaction between anti-H3 and H3 protein on H3N2 virus was monitored with electrochemical impedance spectroscopy. After experimental parameters were optimized, analytical characteristics were searched. A linearity in the range of 1.0-17.5 mu g/mL with limit of detection value of 1.01 mu g/mL were obtained for the virus. Relative standard deviation value was found as 4.60 % for 15 mu g/mL H3N2 virus (n=3). By using synthetic saliva solution that was supplemented with the virus, matrix effect was also investigated. Lastly, the selectivity of developed anti-H3 based electrochemical influenza A immunosensor was investigated by applying it for H1N1 virus detection
Cross-cultural adaptation, validation, and the reliability of the Sleep-Related Behaviors Questionnaire in patients with multiple sclerosis
Purpose Examining sleep-related behavioral disorder strategies in detail for MS patients provides an essential assessment to address specific disease findings. The aim of the study was to demonstrate the reliability and validity of the Turkish Sleep-Related Behaviors Questionnaire in patients with multiple sclerosis (MS).
Methods A total of 100 MS patients were filled Sleep-Related Behaviors Questionnaire (SRBQ), Beck Depression Inventory (BDI), Insomnia Severity Index (ISI), and Pittsburgh Sleep Quality Index (PSQI). One week later, 30 randomly selected patients were refilled the SRBQ. The test-retest reliability, internal consistency, and contruct validity were determined.
Results The mean age of the individuals included in the study was 38.9 +/- 11.0 years. The ICC value of the reproducibility of SRBQ was calculated as 0.877 (CI: 0.74-0.94). The test-retest reliability of the SRBQ was excellent (ICC > 0.80). Cronbach's alpha value of the SRBQ was 0.877, which indicates excellent consistency. The SRBQ had correlation coefficients of 0.573, 0.509, and 0.523 with ISI, PSQI, and BDI, respectively. The SRBQ had high validity (r > 0.50, p < 0.01).
Conclusion The Turkish SRBQ was found to be valid and reliable in patients with MS. Owing to the long but comprehensive nature of the SRBQ, a low-cost detailed measurement could be obtained in clinical practice
TE[KO]E U DIJAGNOZI HbS-BETA TALASEMIJE: ZAISTA BLAGA BOLEST?
Background: HbS/beta cases having clinical, hematologic and electrophoretic similarities cannot be sufficiently distinguished from sickle cell anemia cases and are misdiagnosed as sickle cell anemia. This study will investigate the congruence between the HPLC thalassemia scanning tests and the laboratory findings compared to the DNA sequence analysis results of the patients diagnosed with SCA between 2016 and 2020. This study also aims to indicate the current status to accurately diagnose sickle cell anemia and HbS/beta in the light of hematologic, electrophoretic and molecular studies.
Methods: Fourteen patients who were diagnosed with SCA in hospitals at different cities in Turkey and followed by the Thalassemia Diagnosis, Treatment and Research Center, Mugla Sitki Kocman University were included in this retrospective study. The socio-demographic characteristics, hemogram, hemoglobin variant analysis results and DNA chain analysis results of the patients were taken from the database of the centre and then examined. The informed consents were taken from the patients. The patients were administered a survey containing questions about transfusion history and diagnostic awareness. The Beta-Thalassemia mutations were analysed using a DNA sequencer (Dade Behring, Germany) based on the Sanger method.
Results: According to the DNA sequence analysis, the results of these patients diagnosed with SCA in hospitals in different cities of Turkey were the following: of 14 patients, 8 had HbS/beta(0), and HbS/beta + and one had HbS carrier, and one had Hb-O, and three had SCA. The patient with HbS carrier status also contains three additional mutations, all of which are heterozygous. We discovered that although two of three mutations, which are c.315+ 16G> C and c.316-185C>T, are previously reported as benign, at least one of the two mentioned mutations, when combined with HbS, causes transfusion-dependent HbS/beta.
Conclusions: Briefly, HbSS and HbS/beta thalassemia genotypes cannot be definitely characterized by electrophoretic and hematologic data, resulting in misdiagnosis. c.315+16G>C and c.316-185C> T are previously reported as benign; at least one of the two mentioned mutations, when combined with HbS, causes transfusion-dependent HbS/beta. In undeveloped or some developing countries, molecular diagnosis methods and genetic analyses cannot be used. If mutation analyses could be performed, then such differential diagnosis errors would reduce. However, if mutation analysis cannot be performed, other methods such as HPLC, capillary electrophoresis absolutely be sought to have insight into the parental carriage status
First case with RANBP2 biallelic mutation and severe acute necrotizing encephalopathy phenotype
Familial acute necrotizing encephalopathy (ANE) is a rapidly progressive encephalopathy that can occur after common viral infections at different stages of life. The clinical findings of 2 siblings diagnosed with ANE were shared and the whole-exome-sequencing study of the index case was performed. It was confirmed by the Sanger method. We found the RANBP2 gene p.I656V variant homozygous in the index case. We found the variant in the parents as heterozygous. We argue that biallelic mutations in the RANBP2 gene may result in ANE with early onset and severe prognosis by increasing penetrance
Evaluation of the Clinical Characteristics of Patients with Food Protein-Induced Enterocolitis Syndrome: A Multicenter Study
Introduction: Food protein-induced enterocolitis syndrome (FPIES) is a rare non-IgE, cell-mediated food allergy disorder. We aimed to report the demographic characteristics, clinical features, and management of pediatric patients with FPIES. Methods: This retrospective study included all children diagnosed with FPIES at the pediatric allergy departments of the participating twelve study centers from January 2015 to November 2020. Results: A total of 73 patients (39 males, 53.4%) with a male/female ratio of 1.1 were included in the study. The median (interquartile ranges) age at symptom onset was 6 months (0.5-168, 4-9.5). The most frequent offending foods were cow's milk, egg's yolk, fish, and egg's white, identified in 38.4% (n = 28), 32.9% (n = 24), 21.9% (n = 16) and 20.5% (n = 15) of the patients, respectively. The total number of reported FPIES episodes was 290 (3.9 episodes per child). Oral food challenge (OFC) was performed in 54.8% (n = 40) of the patients, and tolerance was detected in 17 OFCs (42.5%) at a median age of 15 months (range 8-132 months). Conclusion: FPIES is a non-IgE-mediated food hypersensitivity that commonly affects infants and is often misdiagnosed. The pathophysiology of the disease remains unclear and the low awareness of FPIES among physicians and parents highlights the need for more education
Morphological and molecular notes of Gypsophila pilulifera
A detailed study of the Turkish endemic Gypsophila pilulifera were presented by both morphological and molecular investigations of several sites (each one divided in four biotopes). The characteristics were investigated using statistic analyses, i.e. t-Test and One-way ANOVA tests, as well as Discriminant analysis (a=0.05). Some differences were detected among the individuals in the locations stufied where the species was distributed in terms of morphological characteristics and obvious similarities/differences were determined among the biotopes as a result of the discriminant analysis. Morphological characterization was supported by molecular data. Four individuals from two different locations were analyzed by PCR using twenty-four ISSR primers. Analysis of PCR products on agarose gel yielded three primary reproductive band profiles, and the similarity frequencies calculated with the Jaccard coefficient were found to be among 0.229 and 0.769. In this context, according to the results, there was no difference in species, subspecies or variety level between individuals between the two locations. But the obtained molecular differences support the morphological differences. It is predicted that this may change towards speciation as ecological changes increase in the locations where the species is distributed
Accuracy of emergency physicians' interpretation of computed tomography for urgent-emergent diagnoses in nontraumatic cases
Objective: The aim of this study is to evaluate the accuracy levels of the emergency physicians (EPs) managing the patient in the interpretation of the urgent-emergent pathological findings in thoracic and abdominal computed tomography (CT) scans.
Methods: The EPs interpreted the CT scans of patients who visited the emergency department because of nontraumatic causes. Then, a radiology instructor made final assessments of these CT scans. Based on the interpretation of the radiology instructor, the false-positive rate, false-negative rate, sensitivity, specificity, positive predictive value, negative predictive value, and kappa coefficient (κ) of the EPs' interpretations of the CT scans were calculated.
Results: A total of 268 thoracics and 185 abdominal CT scans were assessed in our study. The overall sensitivity and specificity of the EPs' interpretation of the thoracic CT scans were 90% and 89%, respectively, whereas the abdominal CT interpretation was 88% and 86%, respectively. There was excellent concordance between the EPs and the radiology instructor with regard to the diagnoses of pneumothorax, pulmonary embolism, pleural effusion, parenchymal pathology, and masses (κ: 0.90, κ: 0.87, κ: 0.71, κ: 0.79, and κ: 0.91, respectively) and to the diagnoses of intraabdominal free fluid, intraabdominal free gas, aortic pathology, splenic pathology, gallbladder pathology, mesenteric artery embolism, appendicitis, gynecological pathology, and renal pathology (κ: 1, κ: 0.92, κ: 0.96, κ: 0.88, κ: 0.80, κ: 0.79, κ: 0.89, κ: 0.88, and κ: 0.82, respectively).
Conclusion: The EPs are successful in the interpretation of the urgent-emergent pathological findings in thoracic and abdominal CT scans
In vitro biological activities of potassium metaborate; antioxidative, antimicrobial and antibiofilm properties
Antioxidant, antimicrobial and antibiofilm activities of potassium metaborate (KBO2) was investigated within the present study. Antioxidant capacity of potassium metaborate was determined by β-carotene bleaching (BCB) assay and hydroxyl radical scavenging activity. Potassium metaborate was evaluated for its antimicrobial effects against selected Gram-positive bacteria, Gram-negative bacteria and a yeast via broth dilution method. The inhibition capability of potassium metaborate on the microbial biofilm formation of tested microorganisms was measured by microplate biofilm method using MTT (3- [4, 5- dimethyl-2-thiazolyl]-2, 5-diphenyl-2H-tetrazolium-bromide). Biofilm inhibition capacity of potassium metaborate was also observed by Scanning Electron Microscope (SEM). Potassium metaborate was found to have the ability to scavenge hydroxyl radicals with an inhibition rate of 71.13% at 100 mM concentration. Antioxidant activity of potassium metaborate as determined by BCB assay gave higher result with an inhibition rate of 86.96% at the same concentration. According to the MIC (minimum inhibition concentration) values, the potassium metaborate inhibited the growth of C. albicans, S. aureus and E. coli at 62.5 mM concentrations while it was 31.25 mM for B. subtilis and 125 mM for P. aeruginosa. The highest antibiofilm activity was determined at the MIC of potassium metaborate with the reduction rate of 90.18% against C. albicans. It was concluded that, potassium metaborate have strong biological activities and can be effectively used for biomedical and environmental solutions
Semiparametric Time-Series Model Using Local Polynomial: An Application on the Effects of Financial Risk Factors on Crop Yield
This paper proposes a semiparametric local polynomial estimator for modelling agricultural time-series. We consider the modelling of the crop yield variable according to determined financial risk factors in Turkey. The derivation of a semiparametric local polynomial estimator is provided with its fundamental statistical properties to estimate the semiparametric time-series model. This paper attaches importance to precision agriculture (PA) and therefore a local polynomial technique is considered due to some advantages it has over alternative methods. The introduced estimator provides less estimation risk, involving both parametric and nonparametric components that allow the estimator to represent the data structure better. From that, it can be said that the proposed estimator and model is beneficial to agricultural researchers for financial decision-making processes
A device that facilitates screwing at an appropriate angle in quadrilateral surface fractures: 105-degree drill attachment
Background/aim: Within this study, we aimed to investigate the radiological and functional outcomes of acetabular fractures involving quadrilateral surface using 105 degrees drill attachment in the anterior intrapelvic approach. Materials and methods: The 35 patients who underwent surgical treatment from January 2016 and January 2020 for acetabular fractures involving quadrilateral surface with anterior intrapelvic approach using 105 degrees drill attachment and a minimum of 12 months of postoperative follow-up were included. Perioperative complications, operation duration, and the quality of reduction were evaluated. Reduction quality was classified as poor, imperfect, and anatomic. Functional evaluation was performed according to the Harris Hip Score (HHS) and Merle d'Aubigne Score. Results: Among 35 patients (median age 36 (21-80)), radiological results of the acetabular fixations were anatomic, imperfect, and poor in 28 (80%), 5 (14.3%), and 2 (5.7%) patients, respectively. Postoperative 1-year functional outcomes with Merle d'Aubigne scores and HHS were median 18 (10-18) and 90 (60-96), respectively. The clinical outcomes of the patients showed concordance with reduction quality. The median operation duration was 180 minutes (range 125-270). Iatrogenic neurovascular damage was not noted in any patients. Conclusion: Reduction and fixation of deep intrapelvic fractures are risky and difficult due to the narrow anatomy and adjacent crucial neurovascular structures. As the 105-degree drill application is safe and easy to intervene in, short surgery duration and satisfactory results with minimum complications can be obtained with a 105 angulated drill in the deep pelvic region
