Stronger Binders, Better Probiotics: Evaluating Mucin Affinity in Yeast Strains for Probiotic Potential
The goal of our lab is to determine if the yeast, Pichia pastoris, can survive and secrete recombinant proteins in the murine gut. To possibly increase the retention of P. pastoris in the gut two strains were created. Strain yJGGA contains an agglutinin protein and strain yJGG33 contains a fusion protein of agglutinin and mucin binding domain. Both agglutinin and mucin binding domain have affinity to mucin. Mucin is the major protein found in the mucus linings of the gut.
The ability of these strains to bind mucin was determined by incubating the strains with mucin-coated glass coverslips. Coverslips were washed to remove unbound cells and bound cells were collected through vigorous pipetting. Bound cells were then counted by plating onto a selective medium. This binding assay suggested that yJGGA had higher affinity to mucin than yJGG33. Immunofluorescence microscopy was then used to examine the presence of agglutinin and mucin binding domain on the cell wall of the two P. pastoris strains. The results showed that only yJGGA expressed agglutinin on the cell wall, further supporting the results of the binding assay.
The affinity of yJGGA to mucin suggests that this P. pastoris strain would have higher binding to the mucus lining of the gut. This adhesiveness may extend the time yJGGA lives in the gut compared to yJGG33 and wildtype P. pastoris
The Complexity of a Mother’s Life: An analysis of Claudia Llosa’s film, Distancia de Rescate.
This research will examine the theme of a mother’s fear and desires and the techniques of sound and use of nature in Claudia Llosa\u27s film Distancia de Rescate, 2021. Located in a rural town of Argentina, Llosa includes the country\u27s personal attributes that play a part in the story\u27s narrative, expanding on the themes of fear and desire. In my paper I talk about the complexities that come with being a mother. Examining the two mothers in the film, Amanda and Carola, I explore their growing relationship that begins to develop between them and their two children, Nina and David. Llosa highlights this new riveting relationship between Amanda and Carola, and it becomes known to the audience that something deeper is happening beyond the surface. I build upon prior analysis of this film, from scholar Andrea Meador Smith’s “Monstrous Desire in Samanta Schweblin’s and Claudia Llosa’s Distancia de rescate”. In the analysis by Smith, she writes; “Amanda’s memories, however, repeatedly return to her fascination with Carla and her constant preoccupation with Nina’s safety” (Smith 110). I identify the monstrous desires and unprecedented dangers that are following this mothers children. I seek to identify how a mothers own personal desires play a role in the safety of their children. Furthermore, analyzing how the mother’s in the film are in a constant state of measuring their children’s rescue distance to be there to save them from their own death
Meat Cogito
Meat Cogito is a single-player, narrative-driven psychological horror game that explores themes of identity, perception, and reality through the lens of Cartesian philosophy. The player begins as a disembodied brain in a surreal, organic world where flesh, thought, and simulation intersect. As the game progresses, players make choices that alter their evolving body—slowly constructing what they believe to be a “true” human form. These choices influence how the world reacts to them, raising existential questions inspired by Descartes: If I think, am I? and Do I exist, or do I merely perceive?
Gameplay takes place in a grotesquely immersive 3D environment—walls pulse, flesh breathes, and grotesque NPCs whisper riddles. Players interact with their environment and other malformed beings, gradually shaping themselves through psychological and moral choices. The game\u27s atmosphere is dense with tension, inviting players to reflect on what it means to be real in a world that may not be.
The game culminates in three potential endings: the player becomes a horrific “True Being,” ascends as a disembodied mind alone in the void, or rejects both paths and breaks the simulation entirely. Inspired by Mouthwashing by Wrong Organ, Meat Cogito uses horror not just to frighten, but to provoke introspection.
The project aims to be playable on PC, developed using Unity, with all assets, programming, and design executed by a solo developer. The final deliverables include a fully playable experience, development documentation, and a presentation analyzing the game’s themes and systems
Rain: Eternity Untold
Role-playing games, especially games considered JRPG , have long kept to the same visual and gameplay style since their inception over 50 years ago. To help further along the progress of this genre of games, we have combined modern game engine rendering quality with a classic pixel art style and a refreshing style of combat-- blending together both action and turn system-- to enhance the player experience. In “Rain: Eternity Untold”-- a JRPG inspired 2.5D game with modern physics based rendering influences-- solve the mysteries of Rain and the world around her. Against various foes, use powerful skills and exciting items to overcome any obstacle. Use the circle to your advantage; gain boosts beyond belief. To those who want more out of the classic turn-based RPG, to those who want an unique setting, or to those who want to enjoy a story, take up the mantle of an adventurer, and explore the world of Rain: Eternity Untold
Understanding Institutional Factors That Hinder Black Students’ Ability to Persist at Community Colleges
Community colleges face a significant challenge with high attrition rates, which disproportionately impact Black students compared to other student groups. Hope Community Colleges reflect this trend, with a notable percentage of students dropping out before achieving their academic goals. The implications of this high dropout rate extend beyond individual students, affecting the local, state, and national economies.
Although existing research primarily focuses on students’ personal characteristics that influence their ability to stay in college, there is a noticeable gap in understanding the institutional factors that negatively affect student persistence, particularly for Black students in community colleges. This study sought to address this gap by identifying institutional factors, as perceived by Black students, that contribute to their decision to discontinue their studies before completing their program. The insights gained from this research aimed to equip Hope Colleges with valuable information to recognize and address the institutional barriers influencing students’ decisions to drop out.
The significance of this research lies in addressing the alarming rates of student departures, especially among Black students. The primary research questions focus on identifying institutional factors that negatively impact the success of Black students at Hope Colleges, and the support systems that could have helped them persist in their studies.
The conceptual framework for this study revolves around three essential elements: (a) transition, (b) engagement, and (c) support services. Situated in Hope Community College District, which encompasses four community colleges in Northern California, this qualitative study employs a general qualitative approach using semistructured interviews to understand the experiences of Black students who dropped out. The goal was to provide a nuanced understanding of the institutional factors influencing their decision to leave before completing their academic programs
