The process of thicket encroachment in semi-arid savanna: community patterns and biotic interactions
Bush encroachment in savannas is widespread in South Africa and is concerning, from both socio-economic and conservation viewpoints, as it affects ecosystem services, functioning and productivity. This phenomenon depends on multiple factors such as history, vegetation, management and environmental conditions, and their interplay. Encroachment into savannas has been relatively well-documented, however understanding of the different roles of tree-tree interactions between species that occur during this process is still limited. This includes the interactions causing spatial patterning, or how interactions and outcomes change over time in terms of encroachment succession from open savanna to closed-canopy thicket. The main objectives of this research are to document thicket establishment in a savanna ecosystem and consider the ecological roles of the key woody species and the abiotic properties of their micro-sites. Determining interactive effects of species co-occurrence is critical to understanding or predicting patterns and changes in biodiversity, nutrient distribution and available water resources. It is also imperative in determining correct and effective land management practices, particularly for reducing bush encroachment and its negative effect on rangelands. All data were collected on Endwell farm, located in the Smaldeel region of the Eastern Cape, South Africa. Endwell farm is a semi-arid savanna with a mean annual rainfall of 730 mm. First, I examine and describe the thicket encroachment process by exploring the associations between species and their size classes in the field. This was done by using plot-based belt transects and looking at changes in species size-class compositions from early to late successional stages. Association rules (market basket) analysis was used to identify the most common species size-class association patterns. The association between the savanna tree Vachellia karroo and the thicket pioneer Scutia myrtina was the most prevalent at all stages, with V. karroo being central to all associations in the first stage of encroachment; during later stages of encroachment, associations shift to incorporating other thicket pioneer species. The demography and clump formation of S. myrtina was strongly linked to associations with V. karroo to initiate bush clump formation. Results suggest that mature V. karroo facilitate the establishment and growth of S. myrtina. These two species were the focus of more detailed investigations to explore the nature and magnitude of their interspecific interactions. I then examined the effects of pairwise tree interactions between V. karroo and S. myrtina on soil and leaf nutrient content. I measureddifferences betweeninter-canopy and sub-canopy soil nutrient content, and the effect of associations on plant leaf nutrients, between pair-size combinations and individual controls. Results confirmed that pair-size tree interactions affected both soil nutrient and leaf nutrient content. All individuals increased soil K, N and organic C in the sub-canopy, while association with V. karrooincreased S. myrtinafoliar N, Pand K. In contrast, association with S. myrtinaloweredV. karroofoliar N, P and K. Small S. myrtina individuals werefound to benefit most from establishing and growing next to a large V. karroo individual, through mechanisms affecting soil and foliar nutrients. Scutia myrtina individuals establishing in association with smaller size classes of V. karroo showed no significant effects. I tested for positive and negative effects of pairwise tree interactions between Vachellia karroo and Scutia myrtina on available soil water and plant water potential (Ψ). This was done by looking at differences betweeninter-canopy and sub-canopy soil moisture and bulk density and associations on plant water stress (pre-dawn and mid-day leaf Ψ), between pair-size combinations and individual controls. I also selectively removed large V. karroo individuals from pairs to confirm the effects of competition andfacilitation. Similar to other studies, results confirmed positive and negative effects of pairwise tree interactions. Small S. myrtina individuals weremost facilitated by establishing and growing up next to a large V. karroo individual, through mechanisms affecting soil water content, bulk density and leaf Ψ. Scutia myrtina establishing in association with other size classes of V. karroo were much less facilitated, showing no significant effects. Effect of Helicosporidium sp. (Chlorophyta; Trebouxiophyceae) infection on Cyrtobagous salviniae Calder and Sands (Coleoptera: Curculionidae), a biological control agent for the invasive Salvinia molesta D.S. Mitchell (Salviniaceae) in South
The effectiveness of established biological control agents depends on biotic and abiotic interactions in the introduced range. The weevil, Cyrtobagous salviniae Calder and Sands (Coleoptera: Curculionidae), was released as a biological control against Salvinia molesta D.S. Mitchell (Salviniaceae) in South Africa in 1985. This agent has been highly successful against S. molesta and has significantly reduced the weed’s populations around the country. However, in 2007, the parasitic alga, Helicosporidium sp. (an undescribed species), was detected in field-collected C. salviniae adults in South Africa. The distribution and impacts of this disease on the weevil and its efficacy as a control agent were not known. In this thesis, the prevalence, infection load, and impact of Helicosporidium sp. on C. salviniae was determined. In 2019, adult weevils were collected from 10 sites across the Eastern Cape, KwaZulu-Natal, Limpopo, and Western Cape provinces and screened to determine the occurrence, infection load, and geographic distribution of Helicosporidium sp. Transmission mechanisms of this disease in C. salviniae were then evaluated. The possible impact of Helicosporidium sp. was assessed by comparing the feeding rates and the reproductive output of the diseased and healthy adults of C. salviniae. An attempt was then made to eliminate the disease in C. salviniae through the application of the antibiotic, ketoconazole. Further, the role of temperature on infection load in C. salviniae was also assessed. Finally, recommendations for the long-term biological control programme against S. molesta in South Africa were made. The disease covers the entire distribution range of C. salviniae in South Africa, with the disease occurrence rate ranging from 92.15% to 100% insects infected per site. Helicosporidium sp. was found to transmit vertically within the populations of C. salviniae. Infection by the Helicosporidium sp. disease reduced the reproductive output of C. salviniae as well its impact on biomass reduction of S. molesta when a diseased culture was compared to a healthy culture from the USA. 98.44 to 98.55% of Helicosporidium sp. loads were reduced through multiple applications of ketoconazole concentrations under in vitro trials. In vivo treatments resulted in 70% control of Helicosporidium sp. in the adults of C. salviniae that were fed ketoconazole three times over a 21 day period. Adult C. salviniae feeding and survival performances were similar when fed fronds of S. molesta inoculated with ketoconazole and water. The lowest and highest disease loads of Helicosporidium sp. were recorded when the weevils were reared at 30°C and 14°C, respectively. As expected, the highest impact and reproductive output of C. salviniae were at 30°C. The evaluations discussed in this thesis highlight the role of diseases in biological control agents, and gaps in both the pre-release and post-release monitoring that should integrate screening of diseases in these studies. Although the combined application of the antibiotic and temperature will reduce Helicosporidium sp. loads and impact, this technology is most likely only applicable where the weevils are reared in small numbers in a rearing facility and not really applicable to the field situation. It is important to release healthy agents that will cause efficient control of the target weed plant species, therefore, when introducing new biological control agents, the health status of such agents needs to be understood. Therefore, long-term field monitoring and assessment of the impact of C. salviniae on S. molesta should be conducted to track all the changes that may result due to the presence of Helicosporidium sp. The impact of Attention-deficit/hyperactivity disorder on the well-being of adults diagnosed with the disorder: a systematic review
The aim of this research is to explore how adults who were diagnosed with Attention-Deficit/Hyperactivity Disorder (ADHD) experience the impact that the disorder has on their wellbeing. Since this research is an exploration of human experience the qualitative paradigm was used to guide the research. The research design was a Systematic Review. Studies for this research were found in the Fort Hare University Databases. The search terms used included a combination of the following and/or their synonyms Positive Psychology, well-being, life satisfaction, impact on life, life experience, coping strategies, and ADHD. There were 10 articles that met the selected inclusion criteria that became the sample. These articles were then further appraised by using the Critical Appraisal Skills Programme (CASP) appraisal tool. Data was extracted from the studies by using Thematic Analysis to provide the results. The themes for the research were identified as: the impact of an ADHD diagnosis, the impact ADHD has on identity, the impact ADHD has on performance and coping with ADHD. Colistin utilisation and clinical outcomes at a public hospital in Bloemfontein, South Africa
Background Colistin is an antibiotic used as the last resort in the treatment of multi-drug resistant Gram-negative bacteria. Its use started in the 1950s but was decreased in the 1980s owing to its nephrotoxic side effects. The re-emergence of Colistin utilisation in 2012 in South Africa followed the emergence of multi-drug resistant Gram-negative bacteria. There is a dearth of information on the rationale use of Colistin in South Africa. Aim To describe the use of Colistin and its clinical outcomes at a tertiary hospital in Bloemfontein, South Africa. Methodology A retrospective cross-sectional study was conducted at a tertiary hospital in Bloemfontein between 2015 and 2019. Relevant data was extracted from the medical records of patients treated with Colistin. Stratified random sampling was used in selecting 50percent of the eligible medical records of patients treated with Colistin per stratum. Data was analysed using simple descriptive and inferential statistics. Results Of the total sample (N=69), the majority were neonates (43.5percent), while children constituted the lowest number of patients (18.8percent). The highest contributor to the top diagnosis, septicaemia, were neonates (44.2percent). Adherence to policy and Colistin treatment guidelines was suboptimal, more so in neonates (45.3percent) than in adults (73.7percent) and children (72.3percent). Colistin was used as a last resort in 68.1percent of the participants. Cure was achieved in 26.9percent, 46.2percent and 80percent of adults, children and neonates, respectively. The highest rate of nephrotoxicity was seen in adults (57.7percent). In the multivariate logistic regression model analysis, both adults [adjusted odds ratio (AOR)=25.54, 95percent confidence interval (CI) 2.73-238.65] and children (AOR=8.56, 95percentCI 1.06 – 69.10) had a higher risk of death than neonates. However, there was no significant difference in the odds for mortality by gender, co-morbidities, illness duration prior to admission and adherence to treatmen Conclusions This study found a suboptimal level of compliance with policy and recommended guidelines on the use of Colistin in a South African public sector tertiary hospital. In addition, there were variations in the level of compliance by age categories, with lower levels of compliance in neonates than in children and adults. The odds for mortality by gender, co-morbidities, illness duration prior to admission and adherence to treatment guidelines and policies were found to be insignificant, and age was the only predictor of mortality found in the study. The findings of the study highlight the need for improved clinical governance on antibiotic stewardship and monitoring of use of Colistin across all categories of patients in the hospital. Future studies should examine the contributing factors for suboptimal compliance, with evidence-based recommendations on the use of Colistin in the study setting as well as factors contributing to high mortality in adults.Thesis (MPA) -- Faculty of Health Sciences, 202
An investigation into water and sanitation in the Eastern Cape Province and potential for implementation of biotechnology platforms
In contemporary South Africa, a country in transition, destruction rather than reconstruction seems commonplace. Electricity supply is at an all-time low and ‘load shedding’ is an almost daily occurrence. Similarly, more fragility of water delivery and sanitation service is being reported with the likelihood of ‘water shedding’ a soon-to-be reality. In view of the ever-increasing reported mismanagement of South Africa’s water and sanitation infrastructure, which is likely nearing the point of collapse if not already collapsed, this thesis set out to interrogate at a provincial and municipal level the status of water and sanitation in Chris Hani District Municipality (CHDM), Eastern Cape Province. One major driver behind this study was to evaluate provincial and municipal water/wastewater infrastructure as a platform to support implementation of locally developed bioprocess technologies. First, publicly available historical data was used to derive a baseline or benchmark from which to determine gains/losses in compliance, water quality and efficiency. The period 2009-2013/4 during which Blue and Green Drop reporting was routine, was used as an appropriate start point. Data thus derived indicated that, at provincial and district municipality (DM) level, potable water supply was by bulk water schemes and, sanitation was typically by waste stabilization pond (WSP) treatment of municipal sewage. The derived benchmark for the period 2009-2013/14 indicated that most plants were not compliant (~75% of the Eastern Cape Province water treatment plants (WTP) operating between average performance to critical state), operated either at- or above design capacity (Eastern Cape Province, 16%; CHDM, 19%). A large number of plants for the province (62.4%) and district (81.25%) had hydraulic design capacity or average daily flows that were unknown (information not provided in available literature or plant reports) and final effluent did not always meet the general standard (70.3% of the CHDM wastewater treatment works (WWTW)). To determine the contemporary state (2020 - 2022) of water and sanitation within CHDM, a scoping exercise of WTP and WWTW in selected local municipalities was carried out. This was done along a west-east transect by appraising WTP and WWTW in the towns of Cradock, Tarkastad, Komani, Tsomo, Cacadu, and Ngcobo. Furthermore, targeted assessment of the Queenstown WTP and the Cradock WWTW was used to derive real-time data on the status of water and sanitation infrastructure. Results showed that water and sanitation services within CHDM had, in the years between 2009-2013/14 to date, deteriorated. Freshwater demand significantly exceeded capability of water supply schemes, where demand was as much as three times greater than reported available supply. For most WWTW, operation was in excess of capacity and between 1.52 and 12 times installed hydraulic loading. The targeted scoping exercise revealed that Queenstown WTP is a moderate risk plant (Cumulative risk rating, CRR = 11 and maximum risk rating, MRR = 47.8%), whereas Cradock WWTW was in working condition but with challenges and some infrastructural dysfunction. Assessment of water/effluent quality revealed that turbidity and TOC were above SANS 241 general limit. Non-compliance in terms of nitrate/nitrite-N, ammonium-N, phosphate-P, TSS, total coliforms, E. Coli, and free chlorine was evident at Cradock WWTW. Unstructured interviews with plant operators corroborated these findings. The overwhelming support for bulk schemes for potable water provision and WSP for sanitation indicated a partially water secure municipal district but derelict in terms of its sanitation services. The later, it was concluded, in particular provides an ideal opportunity for implementation of platform technologies to support bioprocesses for entrepreneurship, employment, economic benefit and to secure a closed circular economy for regional water and sanitation through valorisation of co-product streams. The impacts of anthropogenic fires in West African savanna woodlands and parklands: the case of the Guinea savanna, Ghana
Fire is recognised as an important factor influencing the structure and function of tropical savannas. Despite the extensive studies conducted on the effects of fire on global savannas, there are relatively few studies focusing on fire-assisted land use practices in the agroforestry parkland of the Sudano-Guinean savannas of West Africa. The region experiences recurrent fires in the dry season which begins from November to April. The fires are anthropogenic and are mainly caused through rural livelihoods and cultural practices such as farming - to remove debris from crop fields and to improve soil fertility and hunting to flush out animals, among other reasons. This study therefore sought to: 1. contribute to the understanding of fire-assisted traditional land use practices, people’s knowledge and perceptions of fire use and fire regime: 2. analyse the effects of fire on vegetation, and 3. determine the effects of fire on soils in the Guinea savanna woodlands and parklands of Ghana. Study sites (burnt and unburnt land use types) were selected based on five-year daily fire counts (2013-2017) data, obtained from the Earth Observation Research and Innovation Centre (EORIC), Ghana in collaboration with the Council for Scientific and Industrial Research (CSIR) Meraka Institute, South Africa. Fire densities were calculated for the 18 districts in the Guinea savanna (Northern Region of Ghana). The districts were stratified into low, medium and high fire frequency areas. The East Gonja district recorded the highest fire density (1.0 fires km-2) while Tamale recorded the lowest fire density (0.3 fires km-2). Of the eighteen districts, six districts were purposively selected and ten communities sampled. Firstly, I investigated the frequency of fire use and control, perceptions of fire regime for selected livelihood and socio-cultural activities in the six districts. The majority of respondents (83%) across the study districts indicated that they used fire once a year for at least one of the following activities: land preparation, weed/grass/pest control, burning stubble after harvest, bush clearing around homesteads, firebreaks, charcoal burning and hunting. The study showed the highest frequency of fire use in the dry season was for land preparation for cropping. However, less than a fifth of the respondents (17%) indicated that they did not use fire for any of the above activities. The results of a multiple regression predicted fire activities in the dry season from gender, age, level of education, occupation and household size. Secondly, the study examined how fire influenced the population structure and abundance of two economically important woody species Vitellaria paradoxa C. F. Gaertn. (Shea tree) and Anogeissus leiocarpa (DC.) Guill. & Perr. (African Birch) in burnt and unburnt land-use types. Stand basal area, mean densities of juveniles and adult trees and Simpson’s index of dominance were determined. Eight diameter size classes of each species were analysed by comparing their observed distributions to a three-parameter Weibull distribution across the land use types. A total of 3,366 individuals of A. leiocarpa (n = 1,846) and V. paradoxa (n = 1,520) were enumerated. The highest total basal area of Anogeissus leiocarpa (16.9 m2/ha) was estimated in sacred groves whereas Vitellaria. paradoxa (20.6 m2 /ha) was found in unburnt woodland. The highest mean densities of A. leiocarpa (22.7±29.7 stems/ha) and V. paradoxa (15.3±2.2 stems/ha) were found in sacred in groves. Anogeissus leiocarpa was, however, absent in fallows and burnt crop fields. A somewhat inverse J-shaped distribution was found in sacred groves for both species. Thirdly, the study compared species composition in early burnt, late burnt and unburnt plots in a protected area where fire was regulated. Vitellaria paradoxa C. F. Gaertn., Terminalia avicennioides Guill. & Perr., Combretum adenogonium Steud. ex A. Rich. and Combretum molle R. Br. Ex. G. Don. were the most common and abundant in all treatments. Late burnt and unburnt plots recorded the lowest diversity amongst the three treatments. Unburnt plots had higher tree density than burnt plots. A Detrended Canonical Analysis showed a changing trend, indicating a moderately strong positive association between burning time and species composition. The first and second axes contributed 53% and 12% variation, respectively. Most of the species found in axis one had fairly strong positive association to early and late burnt treatment than unburnt treatments. Finally, the effects of fire on soil properties in burnt and unburnt crop fields and woodland in the ten communities were also investigated. A total of 151 composite samples in selected burnt and unburnt land use types (burnt crop field n=20; unburnt crop field n=27; burnt woodland n=53, unburnt woodland n=51) were analysed. The variables analysed were pH, soil organic carbon (SOC), total nitrogen (TN), available phosphorous (P), Exchangeable bases - potassium (K), magnesium (Mg), calcium (Ca), cation exchange and soil texture. Total N, SOC, pH and Ca differed significantly across the burnt and unburnt land-use types. A principal component analysis showed a stronger association and more positive gradient in woodlands than in crop fields. Total N showed a positive association with SOC, whereas silt showed a negative association to sand and clay. Traditional knowledge and perceptions of users of savanna agroforestry parklands can inform the formulation of local by-laws for community fire management as well as national policy regulation on fire use in the savanna through the interlinked analysis of social and ecological systems as have been elucidated in this study. The results on vegetation assessment revealed that fire practices and land uses influenced size class distribution of the two study species as well as the densities of woody species in traditional crop fields and the National Park. The unstable populations observed in most land uses and the absence of A. leiocarpa in crop fields and fallows call for education and policy actions on the use of fire in parklands of West Africa. Furthermore, fires positively influenced some soil properties in both woodlands and crop fields confirming one of the emphasised reasons why rural subsistence farmers use fire. As an environmental management decision and land use policy intervention, early dry season burning could be an option to curbing the indiscriminate and unplanned fire use. Also, the protection of trees on farm lands could check the unstable population structure of economically important woody species leading to the different structures that deviated from the recommended reverse J-shaped distribution curve observed in these agroforestry parklands. Photodynamic anticancer and antimicrobial activities of π-extended BODIPY dyes and cationic mitochondria-targeted porphyrins
Cancer is among the most devastating diseases and is mainly caused by gene mutation. This could be hereditary, or the mutation could be stimulated due to a lifestyle one lives, such as smoking, which induces lung cancer. The high morbidity rates of cancer are attributed to it being metastatic. The relatively poor physicochemical properties of existing drugs have caused treatment to be ineffective. Photofrin®, Foscan®, and Photogem® are some of the porphyrin-based derivatives approved by the Food and Drug Administration (FDA) for use in photodynamic therapy (PDT). Despite having such drugs, the quest to find better cancer drugs is still ongoing and 4,4-difluoro-4-bora-3a,4a-diaza-s-indacene (BODIPY) dyes are among the molecules that are being studied as potential photosensitisers (PS) in PDT. However, these molecules suffer from poor solubility and ineffective generation of singlet oxygen, the main ingredient in PDT treatment. Furthermore, photosensitisers used in PDT face a problem with hypoxic conditions associated with cancer cells, which causes the generation of singlet oxygen to be relatively low. The PS also suffer from the untargeted treatment, increasing their toxicity. Therefore, the main aim of this study was to improve the bioavailability of BODIPY dyes. Thus, a series of BODPIY dyes were synthesised with hydrogen bond accepting atoms and heavy atoms that enhance singlet oxygen generation. Additionally, to override hypoxia conditions, porphyrins with mitochondria targeting properties were synthesised since it has been well established that the mitochondria will always have a decent amount of oxygen in cancerous cells. When employed as PS in PDT studies, these molecules have better cytotoxic abilities than BODIPY dyes, and this potency was credited to their mitochondria targeting ability and efficient singlet oxygen generation. Finally, this study reports the synthesis of di- and mono-substituted BODIPY dyes with improved solubility and porphyrins substituted with triphenyl phosphine, a mitochondria targeting moiety. On the other hand, the work further illustrates the synthesis of β-substituted cationic porphyrin with mitochondria targeting properties.Thesis (MSc) -- Faculty of Science, Chemistry, 202
Older persons’ perceptions of services that support positive ageing in the Khayelitsha and Gugulethu districts of the Western cape
Globally people are growing older, and nearly every country is experiencing an increase in their ageing populace. In 1960 older people made up 5 % of the world population, while in 2018, they were about 9% of the overall population. Projections are that in 2050 people over the age of 65 will make up 16% of the overall population. To respond to an ageing population, a government must ensure that its policy framework can respond to the needs of its citizens. A society that supports a positive ageing environment improves older people’s well-being. In our complex society, each person is dependent on others in various ways, and social, emotional and practical interdependence is a feature of the human social condition. This means that older people do not function well in isolation but need support for positive ageing. This research project focused on older people’s perceptions of services that support their positive ageing in the Khayelitsha and Gugulethu districts of the Western Cape, South Africa. The theoretical framework employed in the study is Systems Theory. The research approach utilised in the study is qualitative with an explorative and descriptive research design. A non-probability purposive, criterion-based sampling technique was employed. The researcher recruited eleven older persons aged sixty years and above in the Khayelitsha and Gugulethu areas. An NGO that works with older persons’ served as gatekeepers. Semi-structured telephonic interviews were conducted. The data collected was analysed using thematic data analysis. The trustworthiness of the research process was enhanced using Guba’s model. Ethical principles as set out in the Belmont report were also adhered to. The key findings of the current study indicated that in Khayelitsha and Gugulethu areas, there are available services that support positive ageing, such as clubs, excursions and social grants. Environmental aspects challenging positive ageing included unfavourable home conditions such as financial burden and ill-treatment, poverty and unemployment, health problems, and COVID-19 effects. Older people mentioned that there are available but inadequate services such as municipal services and care facilities. Thus, they recommended that there should be poverty alleviation schemes, community development projects such as crafting, gardening, sewing, and more institutions and home-based care for older people.Thesis (MA) -- Faculty of Health Sciences, 202
Business cycle synchronization in SACU countries: a wavelet coherence analysis
The discussion on the viability of the Southern African Customs Union (SACU) as a monetary union has been ongoing for years. SACU is the oldest Customs Union in history, formed in 1910. Employing the Optimum Currency Areas (OCA) theory, the study examined SACU’s readiness for a single currency. There are several technical requirements needed to form an OCA, however, one of the most vital of these with regards to a country’s suitability to enter a currency union is Business Cycle Synchronization. Therefore, SACU’s readiness was tested by establishing Business Cycle synchronization between the 5 member states. Although Business Cycles Synchronization have not been proven, SACU countries have moved towards a complete monetary union because of increasing macroeconomic convergence. Our study using Wavelet analysis sets out to establish if there is a synchronization of Business cycles with SACU members. Cyclical components were isolated, from the real GDP growth rate of each country from 1981 to 2020, using the Hodrick-Prescott filter (HP), Band-Pass filter (BP), and the Christiano-Fitzgerald Filter (CF). In the 2 by 2 analysis, our wavelet coherence showed that SACU countries are synchronized with each other, while South Africa displayed its dominance in the union by showing significant synchronization with all the members of the union both in the short and long run. The Vector wavelet analysis results concluded that, as a group, SACU shows much higher degrees of synchronization. The group synchronization is statistically significant throughout the period of study. Furthermore, there is an indication of consistent synchronization, in the long and short run. Overall, the results of our study conclude that there is synchronization in the SACU countries.
