Kenyan comedy: transmogrifying stereotypes and fashioning a ‘Kenyanness’ of aesthetic escapism
Despite the proliferation of stereotypes in Kenyan comedy, little research exists to show the two intersect to shape a peculiar notion of ‘Kenyanness’. Indeed, Kenyan comedy, besides reproducing and playing on the quotidian and the historical, is heavily invested with ethnic and gender stereotypes that reflect the Kenyan milieu as it intersects with the global. This comedy is oftentimes seen by some critics as detrimental to Kenyan society because it supposedly shapes and reinforces ethnic and/or gender relations in the country. However, this study contends that Kenyan comedy is open to multiple interpretations and meaning contestations that are not necessarily clear to the comedians and audiences due to the asymmetry that abounds between the production and consumption ends of the comedy’s spectrum. The comedy is therefore ambivalent. This research seeks to demonstrate that Kenyan comedy—as a form of entertainment and critique of society—does not seek to fix and reify Kenyan identities. Instead, the comedy frees these identities from the presumed vice-hold, constricting world of stereotypes by disrupting the linearity of the stereotypes, thereby unsettling the hierarchical structure of hegemonic ideology embedded in them through postmodern humour: a form of humour amenable to postmodern sensibilities. To this end, the comedy offers Kenyans momentary escape into a comedic utopia, through which, they address pertinent issues affecting their nationhood, even as they endeavor to fashion a ‘Kenyanness’ of aesthetic escapism that is celebratory of the country’s rich socio-cultural diversity. The study employs Jörg Schweinitz’s (2011) stereotype theory and the postpositivist realist theory of identity in its study of stand-up, scripted episodic and topical comedy as transposed on to YouTube. Characterization of Trypanosoma brucei Sti1 and its interactions with Trypanosoma brucei Hsp83 and human Hsp90
Neglected tropical diseases continue to pose global concern due to their impact on health and socio-economic status of developing countries in sub-Saharan Africa. African trypanosomiasis is one of the neglected tropical diseases caused by the kinetoplastid flagellate parasite Trypanosoma brucei (T. brucei). The disease is fatal if untreated and the toolbox to combat the disease has been plagued with many difficulties such as drug resistance, toxic chemotherapeutics, and cumbersome drug delivery processes. In recent years, the disease has received attention from organizations such as the Drugs for Neglected Diseases initiative (DNDi) in partnership with WHO as well as academia and industry to provide alternatives to the existing drugs as part of a targeted approach to eliminate human African trypanosomiasis by 2030. The life cycle of the T. brucei parasite requires that it transitions between a cold-blooded vector (the tsetse fly) and a human host. To survive this extreme environmental change and maintain its infectious cycle, the parasite has evolved an arsenal of tools which include a strong immune evasion technique and a robust molecular chaperone system. Heat shock protein 90 (Hsp90) is one of the most abundant eukaryotic molecular chaperones that has been extensively studied in many organisms. It is indispensable for maintaining proteostasis in some organisms and its inhibition is currently being explored as a drug target for cancer and other parasitic diseases. In T. brucei, cytosolic Hsp90 is specifically referred to as Hsp83 due to variations in the sizes amongst different orthologues. Hsp90 is present in high levels in all stages of the T. brucei cell cycle both constitutively and on exposure to stress. To function in the cell, Hsp90 is dependent on co-chaperones, one of which can be found in most organisms, namely, the stress-inducible protein 1 (Sti1). The Hsp90-Sti1 interaction was shown to be crucial for growth in the intracellular kinetoplastid parasite, Leishmania donovani. However, this partnership has not been explored in the extracellular parasite T. brucei. To analyse the interaction of Hsp90 with Sti1 in T. brucei, this study combined in silico, in vitro and in vivo tools. In silico analyses of the Hsp90 complement in T. brucei revealed the presence of twelve putative Hsp90 genes, ten of which code for the cytosolic protein and are arranged in tandem in a head to tail fashion on the same chromosome. One gene each was found for the mitochondrial and ER paralogues of Hsp90, similar to all other species analysed. Eight putative co-chaperones specific to T. brucei were also discovered: six tetratricopeptide repeat domain (TPR) containing co-chaperones and two non-TPR containing co-chaperones. Structural and evolutionary analysis also confirmed that the domains were conserved across the species analysed. T. brucei Sti1 (TbSti1), T. brucei cytosolic Hsp90 (TbHsp83) and human cytosolic Hsp90 (hHsp90) were heterologously overproduced in E. coli and purified using nickel affinity chromatography. With specific antibodies, the expression and localization of the proteins were confirmed. TbSti1 showed strong affinity to the Hsp90s in the nanomolar range, with higher affinity for hHsp90 compared to TbHsp83. TbHsp83 and hHsp90 showed typical chaperone properties by suppressing the aggregation of thermolabile substrate MDH at equimolar concentrations and both chaperones had potent ATP hydrolysis activity. TbSti1, on the other hand, showed no MDH suppression activity and did not affect the ATP hydrolysis activity of TbHsp83 or hHsp90. Ex-vivo experiments using HeLa CRISPR Hop knockout (KO) human cell lines transfected with pcDNA3.1(+)HA-TbSti1 revealed TbSti1 also localized to the cytoplasm. The transfected cells showed a distinct fibroblast-like morphology which was different from the circular morphology seen in the Hop KO untransfected and wild type untransfected cells. Finally, co-immunoprecipitation studies revealed that TbSti1 co-immunoprecipitated with hHsp90. These results show the first characterization of the TbHsp83-TbSti1 partnership in T. brucei. The strong association between both proteins suggests a functional role for this partnership in T. brucei and could provide an updated context for understanding Trypanosome brucei biology.Thesis (PhD) -- Faculty of Science, Biotechnology and Innovation Centre, 202
The development of ionic zinc(II) phthalocyanines for sono-photodynamic combination therapy of cervical and breast cancer
This study focuses on the development of the sono-photodynamic combination therapy (SPDT) activity of phthalocyanines (Pcs) on the cervical and breast cancer cell lines in vitro. The SPDT technique utilizes ultrasound in combination with light to elicit cytotoxic effects for cancer eradication. In this work, a selection of tetra-peripherally substituted Zn(II) cationic and zwitterionic Pcs were prepared. The photophysical parameters of the Pcs were determined including their fluorescence behaviours and efficiency of the triplet excited state population. The effects of the ultrasonic parameters (frequencies (MHz) and power (W.cm-2)) on the stability of the Pcs were evaluated. Four parameters were evaluated: Par I (1 MHz: 1 W.cm-2), Par II (1 MHz: 2 W.cm-2), Par III (3 MHz: 1 W.cm-2) and Par IV (3 MHz: 2 W.cm-2). The stability of the Pcs reduced with the increase in the ultrasonic power (for Par II and Par IV). The Par I showed the least degradation compared to the other parameters and was therefore used for the SPDT treatments. The sonodynamic (SDT), photodynamic (PDT) therapy activities of the Pcs were studied and compared to their SPDT efficacies. The Pcs showed reactive oxygen species generation during the SDT, PDT and SPDT treatments. For the SDT and SPDT, singlet oxygen (1O2) and hydroxyl radicals (•OH) were detected. For PDT, only the 1O2 were detected. The cell cytotoxicity studies for the Pcs showed relatively higher therapeutic efficacies for the SDT treatments compared to the PDT treatments, where the SPDT showed higher therapeutic efficacies compared to both the SDT and PDT monotreatments on both the cell lines in vitro. Overall, the combination treatments were better compared to the monotreatments. The activities of the Pcs were compared by their differences in structures, including the type of R-group, type of quaternizing agent and type of nanoparticle conjugates.Thesis (PhD) -- Faculty of Science, Chemistry, 202
Distribution and conservation status assessment of the freshwater fishes in the Krom River system in the Eastern Cape Province, South Africa
The Cape Fold Ecoregion (CFE) contains the highest number of endemic freshwater fishes in Africa, south of the Zambezi River system. The CFE’s unique fish fauna has, however, suffered severe decline in historical distribution ranges and population sizes due to multiple impacts, with the two main threats being introduced invasive piscivores and habitat degradation. Growing evidence shows that biodiversity is being lost at unprecedented levels due to increased human pressure and demand for limited resources. Future projections also indicate high extinction risk, particularly for freshwater ecosystems, as a result of global climate change. Yet for many regions, including the CFE, there is still limited biodiversity knowledge to guide decision making processes. The Krom River system in the eastern Cape Fold Ecoregion (CFE) contains three native freshwater fish species. These are Pseudobarbus senticeps, which was recently revalidated, Sandelia capensis and a galaxiid lineage currently informally referred to as Galaxias zebratus sp. ‘Joubertina’. Two of these taxa are listed under threatened categories of the International Union for Conservation of Nature (IUCN). These are P. senticeps, which is listed as Critically Endangered and Galaxias zebratus sp. ‘Joubertina’, which is listed as Endangered. Although S. capensis is currently listed as Data Deficient, the most recent information indicates that the species comprises three allopatric lineages, with the Krom River population belonging to a lineage that is distributed across a number of river systems on the south coast. Two non-native fishes, Micropterus salmoides and Lepomis macrochirus, have also been introduced into this river system. Updated information on distribution patterns and habitat utilisation is required to guide authorities to develop effective management plans and conservation of the threatened fishes of the Krom River system. The aim of this study was to determine the current distribution of the fishes of the Krom River system and to provide recommendations for in situ conservation measures to protect remnant populations and promote recovery and range expansion of the native fishes. The primary objective of the study was to use historical and present data to map the distributions patterns. Data on the distribution of the fishes of the Krom River system were obtained from various sources, including the NRF-SAIAB database, studies published in peer reviewed literature and unpublished reports. Systematic sampling was conducted in the Krom River system and several of its tributaries to provide a snapshot of the current distribution of both native and non-native fishes in this system at the time of this study. The presence of instream physical barriers was recorded, and habitat and water quality were visually assessed. Only four species of freshwater fishes were recorded during the surveys. These were two native species, the Krom River redfin P. senticeps and the Cape kurper Sandelia capensis, and two introduced species, the black bass M. salmoides and bluegill sunfish L. macrochirus. Pseudobarbus senticeps was the most common and widely distributed species in the system, and was recorded at 20 localities (four mainstem and 16 tributary localities). Sandelia capensis was less common and was recorded at only eight localities (four mainstem and four tributary localities). These two native species co-occurred at all the eight localities where S. capensis was recorded, but P. senticeps was always more abundant than Sandelia. Micropterus salmoides was recorded at six localities (four mainstem and 2 tributary localities) while L. macrochirus was recorded at five localities (four mainstem and one off-stream dam site). Native fishes were not recorded at sites where non-native fishes were present except at two localities in the Wit Els River where juvenile M. salmoides were found amongst the native fish samples. However, unlike the other localities where all size classes (i.e., young of the year, juveniles, subadults and adults) were present, only adult redfins were found in the Wit Els River where juveniles of largemouth bass occurred. A comparison of the past and present distribution patterns of the native fishes indicates a considerable decline in distribution range, and remnant populations are now fragmented compared to past observations. The major threats and impacts on the Krom River system are the presence of non-native piscivores, construction of instream physical barriers, and agricultural activities. The information from this study could form a basis for establishing long-term conservation measures that should focus on preventing the spread of non-native fishes, and rehabilitating critical habitats for the future persistence of remnant populations of native fishes. The study also evaluated the implications of incomplete taxonomy on conservation status assessments and prioritisation, by evaluating case studies of species complexes of freshwater fishes whose taxonomy has been recently resolved, as well as two complexes with lineages that await formal recognition as distinct species. The aim was to demonstrate how incomplete knowledge of taxonomy affects the assessment of extinction risk and can potentially misdirect conservation prioritisation. Specifically, the study examined how change in taxonomy or recognition of undescribed genetic lineages in faunal listings affects range size and IUCN Red List risk category. The study taxa were Pseudobarbus afer sensu lato (sl), Enteromius anoplus sl, Amphilius natalensis sl, Sandelia capensis and S. bainsii, and the study assessed the species complexes as Least Concern or Near Threatened. The majority of these taxonomically revised species and genetic lineages were determined to be Critically Endangered (CR), Endangered (EN) and Vulnerable (VU), as the extent of occurrence was estimated to be < 100 km2; < 5000 km2 and < 20 000 km2 respectively. The species and lineages have been observed in fewer than ten locations, and the populations have been observed to be in decline. The results of this study indicate that the current taxonomic status of the native species in the Krom River system obscures the diversity of these fishes and affects conclusions for conservation assessments. Load shedding and performance of small-scale resturants in Mthatha, South Africa
Small businesses in South Africa are recognized for creating jobs and increasing economic growth. Ever since South Africa experienced load shedding in 2008, small businesses, especially those in the restaurant sector, have been hugely affected by load shedding as they heavily rely on electricity for most of their business operations. Load shedding results in a loss of sales and a decrease in productivity, which reduces their profitability and eventually, sustainability. The purpose of the study was to establish the extent to which load shedding affects the performance of small-scale restaurants in Mthatha, South Africa. A positivist research paradigm was adopted for the study, which was anchored on a quantitative approach using an explanatory research design. A fully structured self-administered questionnaire was sent to 53 small-scale restaurant owners or managers and 51 responded. Data analyzed using SmartPLS4 revealed that load shedding has a negative but insignificant impact on both the profitability and productivity of small-scale restaurants. Based on the results load shedding does not impact the performance of small-scale restaurants in Mthatha as both performance proxies showed a statistically insignificant impact. Looking at the current situation of electricity in South Africa, things are getting worse as it is common for Mthatha people to go the entire day without electricity. This study helps in identifying the extent of the effects load shedding has on small-scale restaurants. This study adds to the findings of prior research on the effects of load shedding on the performance of small-scale restaurants and the study contributed to the body of iii knowledge on the effects of load shedding on the performance of small-scale restaurants and mitigates on existing literature gap where the populace is not aware of the extent to which load shedding has affected the performance of small-scale restaurants.Faculty of Economics and Financial Sciences, 202
Diet and lifestyle changes during the Covid-19 pandemic among health care professionals in the Eastern Cape province
Background: The unprecedented COVID-19 era is one that posed drastic changes and challenges to healthcare professionals and healthcare system at large. During disastrous events such as the pandemic, psychosocial stress is expected among health care professionals with the development of different copying mechanisms. It is therefore important to ensure preparedness among these individuals in the event of future pandemics. It is essentials for healthcare professionals to practice healthy dietary habits and regular physical activity which has a positive impact on the immune system by improving its response to viral and bacterial infections and supports mental health. Aim: The aim of the study was to determine dietary and lifestyle changes among healthcare professionals during the COVID-19 pandemic. Methodology: A quantitative, descriptive, and explorative research approach was followed. Data was collected through an online survey and analysed using MS Excel® version 2209, SPSS version 27, cross tabulation and Pearson’s chi-square test (p-value<0.05) The questionnaire consisted of four sections that had demographics, lifestyle, dietary and COVID-19 related questions. The study sample were Eastern Cape healthcare professionals. Results: The findings indicated that health care professionals’ dietary habits and lifestyle were negatively affected by the COVID-19 pandemic. A total of 37.5% (n=131) had indicated that their weight had increased, while 20.4% (n=71) indicated a decrease in weight during the pandemic. It is understandable that there was weight gain during the pandemic as these professionals consumed more alcohol and fast foods with less exercise. There was a high caffeine consumption and decreased sleep time, which can affect productivity, resilience and mental health. The negative impacts posed by the pandemic need to be mitigated as it has the potential to lead to non-communicable diseases and cause even more of a burden to the health system. Authorities need to work with different healthcare professionals to develop programmes relating to coping mechanisms during any state of disaster and support wellness initiatives. The moderation and mediation influence of dynamic capabilities and decision-making dimensions on South African family business performance
Family businesses are fundamental for any world economy where they contribute significantly to economic prosperity. However, family firms are faced with new obstacles that are directly affecting their business performance due to the current economic uncertainties and the rapidly changing environments. The implementation of dynamic capabilities is a fundamental strategic tool family firms can exercise to improve their competitive advantage. Although family business research has proliferated over the last 10 years, there are few studies in South Africa investigating the extent to which dynamic capabilities influence family business performance. This study, therefore, had a primary objective to investigate the determinants of family business performance and the role that dynamic capabilities play as mediators and moderators. Studying such a research theme has not received sufficient attention within the family business field, which emphasises the need for such a topic to be investigated. The independent variables for the study entailed dynamic capabilities and decision-making constructs while the dependent variable comprised family business performance. Consequently, the mediating variables included strategic foresight, strategic flexibility, and strategic creativity while the moderating variables included strategic creativity and strategic flexibility. This investigation followed a quantitative methodological logic by employing the questionnaire as the research instrument. In total, 347 family business managers and owners from the eight districts of the Eastern Cape participated in the study. The family businesses were accessed through a convenience sampling technique to identify if their business suited the inclusion criteria of the study. Data analysis was executed through SPSS where the moderation-mediation analysis followed the Hayes process macro through multiple regression analysis. The research findings revealed that there is both a significant positive and mediation relationship between the dynamic capabilities constructs entailing environmental scanning, scenario planning, knowledge creation, culture, and formal organisation with the decision-making constructs of strategic foresight, flexibility, and creativity with family business performance. Interestingly, a moderation relationship does not exist between these constructs and the strategic foresight of the family firm. However, a moderation relationship does exist between the dynamic capabilities constructs of knowledge creation and culture and strategic foresight when moderated by strategic creativity. Thus, family business performance is positively influenced by the said dynamic capabilities and decision-making constructs. The results ultimately provided implications for family businesses with practical advice on the successful implementation and monitoring of dynamic capabilities and decisionmaking in the family business. Lastly, this study makes recommendations for family and non-family business owners, government, and academics. Foreign direct investment, institutions and economic growth in the selected Southern African Development Community (SADC) countries
Examining the relationship between economic growth and foreign direct investment (FDI) has been a subject of discussion for many researchers, economists, and policy analysts mainly in developing regions. It is important to note that, recent literature highlights that there are other country-specific conditions such as state of institutions which are important in attracting FDI inflows into a country. Given this, the study analysed the relationship between FDI, institutions and economic growth in the Southern African Development Community (SADC) for the period 1990- 2020. The analysis was carried out at both cross- country (2010 to 2018) and individual country level (1990 to 2018). The main objectives of the study were to review the trends of FDI inflow into the region, institutional framework, and trends economic growth in the region as well as member countries. Secondly, to analyse the impact of FDI inflow and institutions on economic growth in the selected SADC countries. Thirdly, to examine how institutions and other factors determine the amount of FDI inflow to the selected SADC countries an provide policy recommendations. Existing literature has documented the relationship between FDI and economic growth. However, the significance of this study is that it provides an analysis of the impact of FDI inflows on economic growth in the SADC region at both cross-country and country specific level. At cross- country level, the Generalized Methods of Moments (GMM) was utilized as the estimation technique. The empirical results revealed that there exists a positive relationship between FDI and economic growth both in the short run and long run. The results also revealed that institutions in combination with financial sector development have a positive effect on economic growth in the SADC region. This gives support to the complimentary view of the importance of institutions and financial sector development as important factors determining the extent to which FDI influences economic growth. Guided by economic theory which suggests that there is a two-way relationship between FDI and economic growth, granger causality tests were performed to check the direction of effect between the two variables. The empirical results revealed that there is a bi-directional relationship between FDI, institutions and economic growth. This in a way suggest that the past values of each of the variables, explains the current values of the other variables. On the other hand, at country level, utilising the Autoregressive Distributed Lag model, empirical results revealed that the effects of FDI and institutions on economic growth is positive and significant. However, this was not found to be the case for Mauritius and Namibia. Given the significant role played by FDI in promoting economic growth, the study also investigated the factors determining the inflow of FDI into the SADC region focusing on the role played by institutions and other factors utilising GMM technique. The empirical results revealed that, in addition to institutions, financial development, infrastructure, and education also play an important role in determining the inflow of FDI into these countries. To a greater extent the same findings were also established at country level. Of great importance the study recommends that at a country level, countries should develop and adopt policies that strengthen good governance and sound institutions. The analysis of the impact of financial integration on financial development and economic growth in the Southern African development community
The study investigated the impact of financial integration on financial development and economic growth in the Southern African Development Community using annual data for the period 2000 to 2018. Literature shows that there is no universally accepted measure of financial integration hence the study utilized the Lane and Milesi-Ferretti measure, foreign direct investment as a percentage of GDP and Chinn-Ito (KAOPEN) index in achieving the objectives of the study. One of the main objectives of the study was to examine the impact of financial integration on financial development in the SADC community. The study utilized first difference GMM to achieve this objective and the results showed that Lane and Milesi-Ferretti measure and foreign direct investment as a percentage of GDP significantly affect financial development whilst Chinn-Ito (KAOPEN) index displayed an insignificant effect. The next step was to investigate the impact of financial integration on economic growth in the SADC region. To accomplish this objective, the study investigated both the direct channel as well as the possibility of financial integration indirectly influencing economic growth through financial development, trade openness, and institutional quality. The Three Stage Least Squares (3SLS) technique was utilized on a system of five simultaneous equations in examining the effect of financial integration on economic growth. The results revealed that financial integration affects economic growth both directly and indirectly. Regarding the indirect channels, only the financial development and institutional quality channel proved to be significant. To ensure the robustness of the results the study checked if the impact of financial integration was sensitive to the measure of financial integration and financial development used. The study employed KAOPEN to proxy financial integration while broad money supply (% of GDP) measured financial development. Robustness results confirmed that financial integration affects economic growth both directly and indirectly. This time, the trade openness channel was significant at 10% which shows the benefits through this channel are significant. This shows that the results of the trade openness channel are inconclusive. When the Lane and Milesi-Ferretti measure of financial integration was employed the impact of the trade openness channel was insignificant whilst a significant impact was found based on KAOPEN. The inconclusive findings for the trade openness channel may indicate the impact of financial integration on economic growth depends on the proxy of financial integration utilized. In addition, the study also gathered some interesting results where financial development, institutional quality, trade openness, and government size (government spending) are significant drivers of financial integration. The study makes some fundamental contributions to literature on financial integration, financial development, and economic growth. Initially, the study provides empirical evidence on the nature of the impact of financial integration on financial development in the SADC. In particular, this study contributes to the body of knowledge by showing that the impact of financial integration on financial development is linear. Secondly, this study makes an original contribution to the literature on the channels through which financial integration affect economic growth in the SADC, providing a more subtlety understanding of the mechanisms at play. Finally, the study provides important policy implications for policymakers and financial regulators in the SADC who seek to promote economic growth through financial integration. The findings of the study imply that deeper financial integration is crucial in the SADC region as it can potentially increase the rate of economic growth in the region. Not only economic growth will be boosted but also the institutional quality and the development of the financial sector of the countries in the region. Based on these findings, the study recommends that the governments of the member countries in the region continue to come up with policies that boost regional and international financial integration. The study suggests that to ensure that they continuously reap positive benefits from financial integration, member countries of the SADC should appoint a board that deals with implementation and accountability. This board must be responsible for ensuring that member countries implement the formulated policies and should also hold member countries accountable in case of failure to implement the formulated policies. Since the region is pursuing financial integration, the region must come up with policies that prioritize domestic developments in the form of financial development, improving domestic institutional quality and reducing trade restrictions in advance in order to ensure that preconditions for financial integration are met. Validation and adaptation of statistical models based on the SAPS III score to predict in-hospital mortality in a South African ICU
In-hospital mortality prediction remains an important task in Intensive Care Units (ICUs). In particular, the estimated in-hospital mortality risk is essential to describe case-mix, for research and clinical auditing purposes. Furthermore, in settings with limited hospital resources (e.g beds) such as the South African public health care system, the estimated in-hospital mortality risk is essential for resource allocation and to inform local patient triage guidelines. Commonly used models for prediction of in-hospital mortality in ICU patients includes, but not limited to, the Simplified Acute Physiology Score III (SAPS III). The SAPS III model was developed in 2005. Notably, the SAPS III model was developed without data collected from African based hospitals. Given the general application of the SAPS III model, including benchmarking and quality control, the development of such a model based on local data is of paramount importance. To this end, this study developed a model for prediction of in-hospital mortality based on data collected in a hospital in South Africa. Logistic regression modelling was used to develop the proposed mortality risk assessment model. The results indicated that the proposed model exhibited superior discrimination and classification abilities compared to the SAPS III model.
