    Whisking Behaviour Reveals Stronger Evidence of Habituation in Homozygous Reeler Mice Compared to Controls

    Reeler mice have a mutation in the reelin gene. As a result, Reeler mice lack cortical layers, yet their brains are still largely functional. However, Reeler mice display strong motor phenotypes, including ataxic gait and tics, and we posit that their whisking behaviour might also be disrupted. We used high-speed video to film and track whisker movements in 9 adult Reeler mice and 9 age-matched controls in three whisker movement assessment tasks, including our established novel object exploration and open field tasks, as well as a new open field habituation task. Overall, whisker movements in Reeler mice were highly conserved during the novel object exploration and open field tasks, and they demonstrated all behaviourally relevant whisking features during exploratory contact with an object, including contact-induced asymmetry, spread reduction, and decreasing whisker speeds following object contact. In the habituation task, whisker angular position and whisker spread decreased between the first consecutive sessions in all mice, suggesting that the animals were less focused on sampling the area as they got more familiar with the environment. However, only Reeler mice were affected by more extensive habituation. We suggest that whisker-dependent sensory function is surprisingly intact in Reeler mice. However, our observed habituation-related changes in Reeler mice whisker movements suggest some behavioural differences in these mice, which is a likely result of their disorganised cortex due to reelin deficiency

    The application of Artificial Intelligence (AI) in Cultural Heritage surveying has gained notable interest, particularly in enhancing the classification and segmentation of 3D point clouds. Recent research focuses on automating processes to manage surveying data and integrating it into the BIM environment. The present research proposes a semi-automated AI-based pipeline to semantically classify archi-tectural features in 3D point clouds and match them with an existing BIM library. The case study examines the school and theatre of the Crespi d’Adda industrial village, a UNESCO Cultural Heritage site. Data includes a 3D point cloud generated via terrestrial photogrammetry, with windows selected as the feature to model in the BIM library. The methodology encompasses three phases: (i) creating a Crespi d’Adda window dataset aligning with BIM library parameters; (ii) applying Machine Learning classifiers for semantic categorization; and (iii) using prediction algorithms to recognize windows in the point cloud, matching them to the BIM library, and calcu-lating accuracy. This research bridges architectural representation and data mining, streamlining BIM reconstruction. Anticipated results include automated detection and labeling of elements with accurate placement in the BIM environment, enhancing efficiency and interdisciplinary integration

    From clinical inertia to therapeutic optimization in patients with atherosclerotic cardiovascular disease: A Monte Carlo simulation within the ITACARE-P registry

    BACKGROUND Despite the intensive approach recommended by the 2019 European Society of Cardiology/European Atherosclerosis Society guidelines, low-density lipoprotein cholesterol (LDL-C) target attainment (<55 mg/dL or <40 mg/dL for patients with recurrent events within 2 years) in atherosclerotic cardiovascular disease (ASCVD) patients remains low, with clinical inertia and lack of lipid-lowering therapy (LLT) optimization as major barriers. METHODS We analyzed real-world LLT patterns in the ITACARE-P registry, enrolling 1909 Italian ASCVD patients referred to cardiovascular rehabilitation or secondary prevention programs. Baseline LLT and LDL-C levels were recorded. For patients not at LDL-C target, a Monte Carlo simulation with 10,000 iterations was performed using efficacy data from pivotal randomized trials to model sequential addition of ezetimibe, bempedoic acid, proprotein convertase subtilisin/kexin type 9 (PCSK9) inhibitors, and inclisiran to estimate potential LDL-C goal attainment rates. RESULTS Among 1909 patients (mean age 66 ± 10 years, 26% women), 41.3% were at LDL-C target. Most (90%) were on statins, predominantly at moderate or high intensity, whereas only 3% were untreated. Among patients not at LDL-C target, the Monte Carlo simulation predicted a stepwise increase in goal attainment from 43% to 50% after ezetimibe, 63% after bempedoic acid, 95% after PCSK9 inhibitors, and 90% after inclisiran. A baseline percentage distance of 23.66% from the LDL-C target was identified as a threshold beyond which the addition of bempedoic acid alone was rarely sufficient (<5% success), supporting direct escalation to injectables. CONCLUSION A structured, guideline-based intensification strategy in secondary prevention could close the treatment gap and enable near-universal LDL-C target achievement, supporting early implementation of combination therapy

    Atomized inversion of 4D gravity data for risk management of CO2 leakage

    We propose a new approach for monitoring CO2 storage sites and to assess leakage patterns through inversion of time-lapse gravity data. Our focus is on leakage scenarios involving permeable faults and the formation of secondary plumes at shallow depths. The methodology is based on the concept of Extremely Compact Sources, which provides a subsurface representation in terms of atomized sources. This framework enables the successful determination of plume locations, net mass changes, and CO2 leakage rates. A case study using realistic multiphase simulations demonstrates the effectiveness of the method for reservoir management, including forecasting, operational planning, and safe management of the injection and post-injection phases
