    Real-time malaria detection in the Amazon rainforest via drone-collected eDNA and portable qPCR

    Zoonotic malaria risk at human-wildlife-environment interfaces requires surveillance that integrates signals from reservoirs, vectors and the environment. We coupled a drone-based environmental DNA (eDNA) canopy swabbing approach with portable quantitative PCR (qPCR) to detect Plasmodium DNA in situ during a 24-h field exercise in the Amazon rainforest. Drone-lowered sterile swabs into the canopy, which were then extracted and subjected to a multiplex pan-Plasmodium assay targeting five human-infecting Plasmodium species (limit of detection 0.2 parasites μL−1). Of 12 samples (10 canopy swabs, 2 field blanks; 13 total runs including repeats), one canopy swab amplified in duplicate (Ct = 28.7 and 29.23), while positive controls amplified as expected (Ct = 30.82 and 31.11) and all other environmental samples and blanks were negative. Passive acoustics confirmed co-occurring howler monkeys (Alouatta spp.), a known reservoir, whereas Anopheles mosquitoes were not recovered from concurrently deployed insect canopy traps. The end-to-end workflow, from drone deployment to qPCR diagnostic readout, averaged 1.5 h per assay, without requiring cold-chain logistics. This proof-of-concept demonstrates that intracellular parasite DNA can be recovered from canopy surfaces and read out in real-time, providing upstream, landscape-level intelligence to guide targeted vector surveillance in remote settings. Our approach operationalizes One Health by integrating environmental, wildlife, and vector signals within a single technological platform, representing a paradigm shift from reactive, sector-specific surveillance to proactive, integrated pathogen intelligence across the human-animal-environment interface. © 2025 The Author

    Equity in unilateral value chain policies: A monitoring framework for the EUDR and beyond

    Unilateral value chain policies have recently emerged as a key strategy of international land use governance. They\u27re part of a broader trend towards trade-based environmental policies, from corporate due diligence to sustainability certification and trade moratoria, that has been critiqued for reinforcing inequities in global trade. Such critique has been heightened by the current rise of unilateralism, whereby states impose environmental rules on imported commodities. Debates have ensued over the political legitimacy of unilateralism, the unequal distribution of its socio-economic impacts, and the need to safeguard local producers and communities. This paper informs these debates by developing and applying a framework for monitoring equity across scales and phases of the policy process. The framework is applied to the 2023 EU Deforestation-free Regulation (EUDR), which aims to stop EU imports of commodities linked to deforestation. We find that EUDR policy references equity as a desired outcome, but excludes affected actors from the design process. Drawing on the case of cocoa in Ghana, we identify diverse potential impacts on smallholder farmers and economies. Opportunities for the EUDR to improve equity include embedding non-EU stakeholders in international decision-making processes, enhanced and equitable partnerships with producing countries and major investments in farmer support. The paper concludes by providing an equity checklist and agenda for monitoring progress, adaptable to a wide range of unilateral and trade-based policies. © 2025 The Author

    Study protocol for the Rainbow Austrian Longitudinal Family (RALF) study: a longitudinal, multi-method, multi-rater investigation of risk and resilience factors in Austrian LGBTQ+ parent families

    Background: Research on LGBTQ+ parent families is evolving to include a growing range of family systems, identities, methodologies, and topics. However, studies that examine minority-specific risk and resilience factors and their associations with within-family processes remain scarce, particularly outside a US-American context. Addressing these research gaps quantitatively poses challenges for researchers, because traditional modelling techniques based on (generalized) linear models are not ideally suited to capture the complexity and intersectionality of family experiences. Within this study protocol, we introduce the Rainbow Austrian Longitudinal Family (RALF) study. Its main goal is to comprehensively investigate general and minority-specific factors that affect the well-being of LGBTQ+ parent family members in Austria. Methods: RALF is a three-wave, longitudinal study over two years that examines risk and resilience factors at the individual, couple, and family level using a multi-method, multi-rater approach. We will assess child adjustment outcomes across three child cohorts, parental mental health, and family processes across three annual data waves through online questionnaires. We aim to recruit N = 150 LGBTQ+ parent families from a variety of family forms and with various identities residing in Austria. A focal sample (target n = 60) will additionally participate in observational, video-based assessments. Our participatory research approach aims to actively engage community members and stakeholders throughout the study. A community advisory board ensures that the study reflects the lived experiences of LGBTQ+ parent families adequately, while community events and accessible dissemination strategies for study results, such as the open access data explorer ExploRALF, support community engagement and facilitate the dissemination and discussion of results. Data will be analyzed using machine learning-based approaches designed to capture complex, non-linear interactions, which are ideally suited to model intersectional experiences of LGBTQ+ parent families. Discussion: The RALF study is the first prospective study to comprehensively investigate minority-specific risk and resilience factors in Austrian LGBTQ+ parent families. Findings have the potential to fill key research gaps, inform policy, and guide clinical practices that support LGBTQ+ parent families. © The Author(s) 2025

    Rational ellipticity of G-manifolds from their quotients

    We prove that if a compact, simply connected Riemannian G-manifold M has orbit space M/G isometric to some other quotient N/H with N having zero topological entropy, then M is rationally elliptic. This result, which generalizes most conditions on rational ellipticity, is a particular case of a more general result involving manifold submetries. © The Author(s), 2025

    Metal mining is a global driver of environmental change

    Global metal extraction is increasing, owing to rising mineral demands from infrastructure development and the growing need for metal-intensive renewable energy technologies to mitigate climate change and phase out coal mining. However, extraction of metal ores also drives impacts on land use, water resources and biodiversity. In this Review, we evaluate mining trends of 47 metal ores between 1970 and 2022 and explore the environmental consequences. Global extraction of crude metal ores has nearly quadrupled, from 2.7 gigatonnes (Gt) in 1970 to almost 9.4 Gt in 2022, with the greatest increases in Oceania (+1,222%), South America (+929%) and Asia (+285%). Ore-specific mining activities are generally concentrated, with the top-five producers contributing on average 82.7% of the global supply in 2022. The impacts of mining are also concentrated. In 2022, about 50% of the 100,000 km2 global mining areas were located in Russia, China, Australia, the United States and Indonesia. Mining-induced water consumption, pollution and biodiversity loss substantially affect local ecosystems, with tropical rainforests and deserts being especially vulnerable. Around 70% of global metal extraction is linked to international supply chains. Enhanced environmental assessments, stricter implementation of policies, and coordinated actions across sectors throughout supply chains (mining, processing, consumers and financial markets) can help to mitigate the environmental impacts of mining. © Springer Nature Limited 2025
