Umkämpfte Erinnerungskultur: Historikerstreit 2,0, das Humboldt Forum und die neue deutsche Geschichte
The Humboldt Forum, a reconstructed Prussian palace, is uniquely entangled in the ongoing debates around Germany’s relationship with its colonial past. As early as 2007, NGOs such as “NoHumboldt21!” formed to protest the palace, which would house the state’s ethnography collection, entailing an estimated 500,000 stolen artifacts. Upon opening in 2020, the Forum had come to embody the controversy surrounding its construction. Words adorned the façade of the building contrasting enlightenment ideals with colonial exploitation and a permanent exhibition—“Berlin Global”—sought to address the nation’s colonial past.
The Humboldt Forum can thus be conceptualized as a reactive object, one that resituates critique from engaged postcolonial activist groups in an institutional context, therefore synthesizing societal discourses on Germany’s relationship with colonialism. Given the Humboldt Forum’s embodiment of current societal discourses on colonialism, my research project asks: How does imagery in the Humboldt Forum’s “Berlin Global” exhibition integrate colonial atrocity into Germany’s historical narrative? Because historical narratives serve constitute their subjects’ understanding of self in a linear temporal order, my project therefore also asks: What effects does this narrative have on the German nation’s modern identity? Which trajectories does this narrative imply for the Republic? Which futures are enabled or precluded
Mechanism of PTH CA2+ Sensing on G-Protein Interactions with PTH1R
Parathyroid hormone 1 receptor (PTH1R) is a family-B GPCR that plays a crucial role in bone remodeling. Previous studies show extracellular Ca2+ is a positive allosteric modulator for one PTH1R ligand, parathyroid hormone (PTH), which is approved by US FDA to treat severe osteoporosis. Moreover, PTH residues E19 & E22 have shown to be involved in Ca2+ sensing. However, the effects of PTH Ca2+ sensing on intracellular G-protein binding are unknown. Here, we used FRET-based SPASM sensors to study the interaction between PTH1R and different Ga peptides. SPASM sensors, which are isolated in native HEK293T membranes through optimized protocol, contain PTH1R followed by the acceptor fluorophore, a flexible linker, the donor fluorophore, and a peptide from a Ga subunit that mimics the interaction of the full G-protein heterotrimer. In the current study, two SPASM sensor preparation methods, Giant Plasma Membrane Vessiculation and native membrane preparation, were employed. The quality and integrality of the SPASM sensors isolated through each method were evaluated and compared to one another. We performed FRET experiments to quantify the activation of different Ga isoforms by PTH and its Ca2+ sensing mutant, PTHE19AE22A. PTH binding to PTH1R SPASM sensors causes differential interactions between PTH1R and the Gs, Gq and Gi peptides. PTHE19AE22A activation of PTH1R-SPASM sensors leads to distinct interaction profiles between each G-peptide isoforms, which were further modulated by the presence of extracellular Ca2+. Quantifying the differential activation of the specific Ga isoforms by PTH and PTHE19AE22A in the presence and absence of Ca2+ will delineate mechanistic details of PTH1R activation and its role in bone-related diseases. Further, understanding the extracellular Ca2+ modulation of PTH signaling will provide insight for developing treatments for chronic hypocalcemia associated with hypoparathyroidism, while uncovering PTH1R novel regulation in bone remodeling
Excavation of the Augustin Grignon Home in the Grignon Trading Post Site, OU-0072, Kaukauna
Between September and November 2022 Lawrence University conducted excavations of the structure identified previously as the Augustin Grignon Home, located within the Grignon Trading Post Site (OU-0072) in the City of Kaukauna, Wisconsin. Excavations were conducted to determine if the early 19th century Augustin Grignon Home incorporated an 18th century cabin, purportedly used by Dominique DuCharme as a trading post. Excavations determined that there are intact archaeological deposits within the Grignon Trading Post Site, which indicate that it is potentially eligible for inclusion in the National Register of Historic Places. Artifacts recovered from the excavations suggest that the Augustin Grignon Home is an entirely 19th centuryconstruction, with the only possible 18th century material being window glass. The presence of possibly 18th century glass can, however, be reasonably explained in other ways. Thus the excavations support Augustin Grignon’s autobiographical account that he built the structure around 1813 when he moved to the site and that, in doing so, he did not incorporate the earlier DuCharme cabin
