Identification and validation of SSR markers for Xanthomonas axonopodis pv. punicae an incitant of bacterial blight of pomegranate
This study reports genome wide characterization and development of first set of microsatellite markers through in silico analysis of eight sequenced Xanthomonas axonopodis pv. punicae strains available in the public database. SSR survey resulted in identification of ~ 4638 perfect SSRs, with mean marker frequency 901 SSRs/Mb and densitiy of 11,006 bp/Mb aross the eight genomes. Frequency distribution graphs revealed hexa-nucleotide repeats were more prominent fowllowed by tri-, tetra-, di- and penta-nucleotides in the analysed genomes. We desinged 2927 SSR primers that are specific to the strain LMG 859 and ePCR confirmed on seven other Xap genomes. This resulted in identification of 542 informative SSRs that are producing single amplicons, from which 66 primers were successfully validated through wet lab experiments on eight Xap isolates of pomegranate. Furthermore, utility of these SSRs were demostrated by analysing molecular diversity among 22 Xap isolates using 20 Xap_SSR primers. SSRs revealed moderate genetic diversity among Xap isolates (61%) and grouped 11 isolates that are repersenting six different states into one cluster. This proved the earlier evidence of wider spread of ST3 type Xap acoss India using Multi locus Sequence Typing (MLST) technique. In summary, Xap_SSR will serve as powerful genomics tools that would helps in monitoring of population dynamics, taxonomy, epidomology and quarantine aspects in bacterial blight pathogen through development of microsatellite based Multilocus Variable number of Tandem repeat analysis (MLVA) in future
The impact of the COVID-19 pandemic on physical activity and associated technology use in persons with multiple sclerosis: an international RIMS-SIG Mobility survey study.
Objective
To investigate the impact of the COVID-19 pandemic on physical activity (PA) in persons with multiple sclerosis (PwMS).
Design and Setting
A multi-centre international online survey study was conducted within 11 participating countries. Each country launched the survey using online platforms from May to July 2021.
Participants
This was an electronic survey study targeting PwMS.
Intervention
Not applicable.
Outcome measures
The survey ascertained PA performance and its intensity, the nature of the activities conducted and the use of technology to support home-based physical activity before and during the pandemic.
Results
3725 respondents completed the survey. Pre-pandemic, the majority (83%) of respondents reported being physically active, and this decreased to 75% during the pandemic. This change was significant for moderate and high intensity activity (p<.0001). Activities carried out in physiotherapy centres, gyms or pools decreased the most. Walking was the most frequently performed activity pre-pandemic (27%) and increased during the pandemic (33%). 24% of those inactive during the pandemic had no intention of changing their PA behaviour post-pandemic. 58% of the respondents did not use technology to support PA during the pandemic. Of those who did use technology, wearables were most used (24%). Of those currently non-active (25%) expressed a preference for an in-person format to conduct PA post-pandemic.
Conclusion
PA performance, especially activities at moderate and high intensities, decreased during the pandemic in PwMS compared to pre-pandemic. Walking and using wearables gained popularity to stay active. As we move towards an endemic-COVID-19, a call for action to develop interventions focused on walking programmes, with specific emphasis on increasing PA of persons with MS is proposed
Leverage point in high-performance work systems
Drawing on systems theory, this paper aims to search for a leverage point in a high-performance work system (HPWS) wherein a small change of a constituent part significantly enhances the effect of the whole system on organizational performance (OP). Based on meta-analysis of 59,207 firms and establishments from 240 sample studies up to December 2021, the paper examines the effect of HPWS composition, coupled with country of origin and industrial affiliation, on the HPWS–OP relationship. The paper finds that training and development serves as a leverage point to significantly strengthen the synergy of HPWS. However, this leverage point works in advanced countries rather than developing countries, and in service industries rather than manufacturing industries. The finding indicates that a leverage point is not omnipresent, but contingent on country of origin and industrial affiliation. This study has practical implications for managers, highlighting the importance of a leverage point to the HPWS–OP relationship and the contingency nature of the leverage point
Exploring Teaching and Learning experience during COVID-19 pandemic in engineering education
The education system is continuously modernizing by accommodating the need due to the industrial revolution. Various teaching modes are also introduced including a diverse range of students, particularly in engineering education. The COVID-19 pandemic has disrupted normal education worldwide, forced to shut down campus activity for an extended period which forced Universities to adopt alternative approaches to continue student’s academic year. Engineering education faced significant challenges to find a realistic substitution for lab-based hands-on activity as well as group or team-based learning experiences. It is therefore very important to know the challenges and ways to address them. This paper evaluates the teaching and learning experiences observed in engineering education in Australia and abroad during the COVID-19 period compared to the pre-COVID period. The key motivation of this study is to identify key challenges arises due to COVID-19, develop Teaching & Learning (T & L) approaches to address these challenges and evaluate the effectiveness of the applied changes in the T & L approach, identify shortcomings, and find ways to improve them. The student feedback on selected engineering units have been collected from Deakin and Murdoch university in Australia to evaluate the performances of the applied changes. This data is considered as an authentic source of information to compare and identify the key challenges and effectiveness for students’ learning in pre-COVID and during COVID condition. This study later explored various literatures to gather experiences from other universities across the globe and by analysing all findings including academic experiences finally developed constructive recommendations for improvement. It is found that the current form of online mode of teaching has room to improve further as one segment of students finds it challenging and some others like a few approaches. It is also found that the online infrastructure, staff skills to innovate new unit designs, and motivating students are the other challenging areas. Therefore, a new teaching and learning framework is required to overcome all the challenges for future learning
Bread wheat productivity in response to Humic acid supply and supplementary irrigation mode in three northwestern coastal sites of Egypt
Drought stress is a major factor limiting wheat crop production worldwide. The application of humic acid (HA) and the selection of the appropriate genotype in the suitable site is one of the most important methods of tolerance of wheat plants to drought-stress conditions. The aim of this study was achieved using a three-way ANOVA, the stress tolerance index (STI), the Pearson correlation coefficient (rp), and principal component analysis (PCA). Three field experiments in three sites (Al-Qasr, El-Neguilla, and Abo Kwela) during the 2019/21 and 2020/21 seasons were conducted, entailing one Egyptian bread wheat variety (Sakha 94) with three HA rates (0, 30, and 60 kg ha−1) under normal and drought-stress conditions (supplemental irrigation). According to the ANOVA, the sites, supplemental irrigation, HA rates, and their first- and second-order interactions the grain yield and most traits evaluated (p ≤ 0.05 or 0.01) were significantly influenced in both seasons. Drought stress drastically reduced all traits registered in all factors studied compared with normal conditions. The wheat plants at the Al-Qasr site in both seasons showed significantly increased grain yield and most traits compared with that of the other sites under normal and drought-stress conditions. HA significantly promoted all studied traits under drought stress, and was highest when applying 60 kg HA ha−1, regardless of the site. The greatest grain yield and most traits monitored were observed in wheat plants fertilized with 60 kg HA ha−1 at the Al-Qasr site in both seasons under both conditions. Grain yield significantly (p ≤ 0.05 or 0.01) correlated with water and precipitation use efficiency as well as the most studied traits under normal and drought-stress conditions. The results of STI, rp, and PCA from the current study could be useful and could be used as a suitable method for studying drought-tolerance mechanisms to improve wheat productivity. Based on the results of statistical methods used in this study, we recommend the application of 60 kg HA ha−1 to improve wheat productivity under drought conditions along the north-western coast of Egypt
Depression and antidepressant treatment in the development of hypertensive disorders of pregnancy: Results from a prospective cohort study
Background:
Hypertensive disorders of pregnancy are associated with longer term cardiovascular risk. Understanding if depression or antidepressant use in pregnancy is associated with HDP is important in identifying those potentially vulnerable to poorer health in later life. This study examines if depression and antidepressants are associated with HDP.
Methods:
In all, 815 pregnant women were recruited within an Australian pregnancy cohort study at less than 20 weeks of pregnancy, all undertook the Structured Clinical Interview for the Diagnostic and Statistical Manual of Mental Disorders, Fourth Edition, and were assigned to four groups for this paper: those with unmedicated depression meeting criteria for current depression (n = 97), those taking selective serotonin reuptake inhibitors in early pregnancy (n = 101), those taking serotonin and noradrenaline reuptake inhibitors in early pregnancy (n = 31), and those without depression or taking antidepressant medication (control; n = 586). Women were then assessed again following birth. Hypertensive disorders of pregnancy were diagnosed according to the Society of Obstetric Medicine in Australia and New Zealand Guidelines.
Results:
Use of serotonin and noradrenaline reuptake inhibitors (SNRIs) (adjusted risk ratio = 9.10, 95% confidence interval = [3.82, 21.67]) and unmedicated depression (adjusted risk ratio = 3.11, 95% confidence interval = [1.32, 7.35]) were independently associated with significantly higher risk for developing hypertensive disorders of pregnancy compared to controls. Selective serotonin reuptake inhibitors (SSRIs) use did not confer any increased risk. Higher doses of SNRIs, but not selective serotonin reuptake inhibitors, were associated with significantly higher risk for developing HDP (adjusted risk ratio = 4.83, 95% confidence interval = [1.50, 15.58]).
Conclusions:
Our findings suggest that those with depression in pregnancy and/or on an serotonin and noradrenaline reuptake inhibitor should have closer surveillance for the development of hypertensive disorders of pregnancy. These findings support treatment of depression in pregnancy, however, also the consideration of class of antidepressant
A non‐randomised pragmatic trial for the early detection and prevention of surgical wound complications using an advanced hydropolymer wound dressing and smartphone technology: The EDISON trial protocol
Total hip or knee arthroplasty is a highly effective intervention for treating the symptoms of degenerative joint disease or osteoarthritis (OA), often an unwelcome consequence of obesity. A safe and common surgical procedure, hip and knee arthroplasty procedures are not immune to the occurrence of postoperative complications such as surgical site infection (SSI) or surgical wound dehiscence (SWD). While published rates of SSI following hip or knee arthroplasty are low, 1% to 2% in some cases, it is the resulting wound complication and its clinical management and the impact on patient well-being and return to daily life for the 1% to 2% that is of concern. Postoperative complications such as SSI are a major cost driver to the health care system following arthroplasty and often result in extended lengths of stay, readmission for further surgery, primary and community nursing visits, and are a costly burden to health care settings. Early identification of a wound complication through post-discharge surveillance using a fully transparent dressing and smartphone technology and patient education may ameliorate contributing factors or reduce the likelihood of a complication occurring in the first instance. This clinical trial is a non-randomised pragmatic convenience sample carried out in 200 participants of both sexes receiving either a TKA or THA. There will be equal allocation to two groups (100 hips and 100 knees), with 50 in each allocation receiving the interventional dressing and 50 as control. The dressing will be applied prior to discharge and participants will be provided with education on postoperative wound care, when to contact home care nursing for a potential wound problem, and use of their smartphone to capture and send images of their incision site to the study nurses. Participants will also be followed up by home care nursing services at day 14 for suture removal and wound assessment. Participants will complete a patient-reported outcomes survey on day 14 and followed up on day 30 after surgery for wound assessment. The results of this trial may provide a novel pathway using a fully transparent dressing and digital technologies for the prevention of acute readmissions because of wound complications through early detection and intervention
Asexual cultivation techniques of the red macroalgae Asparagopsis taxiformis for commercial application
Asparagopsis is a genus of red macroalgae (seaweed) endemic to coastal waters in Australia and characterised by its complex heteromorphic life history. Previous research discovered Asparagopsis metabolites such as bromoform reduce methanogenesis in livestock rumen by up to 99%. Agriculture accounts for 14% of Australia's total greenhouse gas emissions, predominantly derived from enteric fermentation from ruminant livestock. Cultivating this macroalga is of environmental significance, given the urgency of emission reductions for climate change mitigation while meeting market demand for meat. Understanding manual cultivation techniques is necessary due to limitations of abundance in the wild. There is limited research into cultivation of the asexual lifecycle of Asparagopsis.
This research aimed to identify if fragmentation of Asparagopsis is viable, and if growth can be sustained for nearshore cultivation, by mimicking naturally occurring conditions in lab-based cultivation. Light exposure, salinity, temperature, and a lack of nutrients experienced in ocean environments was emulated. Fragments of Asparagopsis taxiformis gametophytes were collected from Perth coastal waters in June and September and cultivated in aquaria at varying temperatures, substrates, and fragment types.
Growth rates were overall negatively correlated, with marginal growth in some conditions, concluding that asexual cultivation using these parameters is not recommended. The viability of fragments for up to three months in some conditions gives rise to questions of the potential for asexual cultivation in other environments. The optimum conditions across these aquaria were based around a temperature of 21°C, 25μmol photons m-2 s-1 light exposure and holdfast fragments attached to a substrate such as rope. Of particular significance were tip fragments that retained a dark red pigment, which may provide an ideal setting for continued viability and eventual growth due to the reliance of pigment in photosynthesis processes.
Further testing is required to analyse the specific ideal temperature, light exposure and fragmentation origin, alongside other factors such as nutrient levels, to assess if asexual fragmentation is possible for use in commercial cultivation
Improving irrigation, potassium and sulphur use efficiency of Peanut (Arachis hypogaea) in tropical sandy soils
Increasing the productivity of irrigated crops on low fertility sands depends on jointly improving water and nutrient use efficiencies. The aim of this study was to investigate the hypothesis that improved water scheduling and delivery would increase K and S use efficiency of irrigated peanut crops on deep sands. Factorial experiments (irrigation management and K, S rates) were conducted on two deep sands in Binh Dinh province, South Central Coastal Vietnam (SCV). Peanut yield increased 17% in response to switching from the farmers’ irrigation practice (FI) to sprinkler irrigation scheduled according to evaporation rates (ER), combined with increasing fertilizer K and S from 50 kg K ha−1 + 20 kg S ha−1 to 75 kg K ha−1 + 30 kg S ha−1. Nutrient loss with ER relative to FI decreased by 52–64% for K and by 20–24% for S. The irrigation water use efficiency of peanut (kg pod kL−1 of water) increased by 44% under the ER approach. Integrated water and nutrient management practices substantially improved use efficiency of K and S fertiliser, achieved substantial irrigation water savings and increased profits for peanut on infertile deep sands
Bidding strategy for a virtual power plant for trading energy in the wholesale electricity market
Virtual power plants (VPPs) are an effective way to increase renewable integration. In this PhD research, the concept design and the detailed costs and benefits of implementing a realistic VPP in Western Australia (WA), comprising 67 dwellings, are developed. The VPP is designed to integrate and coordinate an 810kW rooftop solar PV farm, 350kW/700kWh vanadium redox flow batteries (VRFB), heat pump hot water systems (HWSs), and smart appliances through demand management mechanisms.
This research develops a robust bidding strategy for the VPP to participate in both load following ancillary service (LFAS) and energy market in the wholesale electricity market in WA considering the uncertainties associated with PV generation and electricity market prices. Using this strategy, the payback period can be improved by 3 years (to a payback period of 6 years) and the internal rate of return (IRR) by 7.5% (to an IRR of 18%) by participating in both markets. The daily average error of the proposed robust method is 2.7% over one year when compared with a robust mathematical method. The computational effort is 0.66 sec for 365 runs for the proposed method compared to 947.10 sec for the robust mathematical method.
To engage customers in the demand management schemes by the VPP owner, the gamified approach is adopted to make the exercise enjoyable while not compromising their comfort levels. Seven gamified applications are examined using a developed methodology based on Kim’s model and Fogg’s model, and the most suitable one is determined. The simulation results show that gamification can improve the payback period by 1 to 2 months for the VPP owner.
Furthermore, an efficient and fog-based monitoring and control platform is proposed for the VPP to be flexible, scalable, secure, and cost-effective to realise the full capabilities and profitability of the VPP
