Pyrolysis of sewage sludge under conditions relevant to applied smouldering combustion
Pyrolysis of sewage sludge under conditions relevant to applied smouldering combustion was carried out in this study to investigate the influences of gas flow rate, oxidative atmosphere, and inert porous medium involvement on the properties of products. The experiments were carried out at 300–600 °C under atmospheres of N2, 5% O2/95% N2, 10% O2/90% N2, and 15% O2/85% N2, with Darcy flow rates of 1.0 and 3.5 cm/s, respectively, with dried sewage sludge loaded individually or as a mixture with sand. As a result, both the increment of gas flow rate and involvement of sand leaded to lower yields of char and higher yields of bio-oil and gas under N2 at temperature of ≤500 °C, due to the enhanced efficiency of pyrolysis reaction and gas transportation. However, when temperature increased to 600 °C, the influencing trends on product distributions changed due to the mechanisms of secondary cracking reaction and volatile-char interaction. The involvement of oxygen in fraction of ≤15 vol% at temperatures of 400–500 °C would lead to the intense decreasing yields of char and bio-oil, and increasing yield of the gaseous (dominated by CO2 and CO), due to the involved oxidation reaction during pyrolysis. Both increment of temperature and oxygen fraction would lead to the delay of ignition and the increase of activation energy of the produced char, except for that of char produced at 400 °C under 5% O2/95% N2, whose calculated activation energy was lower and volatile content was higher compared to that of char produced from pyrolysis at 400 °C under N2. The bio-oil from pyrolysis under N2 was dominated by aliphatic acids, phenols, steroids, amides, and indoles, etc., and the involvement of partial oxidation would lead to the weakened formation of aromatics, phenols, and S/Cl/F-containing compounds in bio-oil
Cytochrome P450 Gene, CYP6CX3, is involved in the resistance to Cyantraniliprole in Bemisia tabaci
Bemisia tabaci is an important agricultural sucking pest, and it develops serious resistance to various insecticides. Although cytochrome P450 was involved in the resistance to cyantraniliprole, limited studies have been conducted on B. tabaci. In the present study, piperonyl butoxide significantly increased the toxicity of cyantraniliprole. P450 activities in two resistant populations were 1.97- and 2.17-fold higher than that in the susceptible population. Among 79 P450 genes, CYP6CX3 expressions in two resistant populations were 3.08- and 3.67-fold higher than that in the susceptible population. When CYP6CX3 was knocked down, the toxicity of cyantraniliprole increased significantly. The LC50 value of cyantraniliprole to the Drosophila melanogaster line overexpressing B. tabaciCYP6CX3 increased 7.34-fold. The content of cyantraniliprole was decreased by 25.74 ± 4.27% after mixing with CYP6CX3 and CPR for 2 h. These results suggested that the overexpression of CYP6CX3 was likely involved in the resistance to cyantraniliprole in B. tabaci
Novel miRNA-SSRs for Improving Seed Hardness Trait of Pomegranate (Punica granatum L.)
Present research discovered novel miRNA-SSRs for seed type trait from 761 potential precursor miRNA sequences of pomegranate. SSR mining and BLASTx of the unique sequences identified 69 non-coding pre-miRNA sequences, which were then searched for BLASTn homology against Dabenzi genome. Sixty three true pri-miRNA contigs encoding 213 pre-miRNAs were predicted. Analysis of the resulting sequences enabled discovery of SSRs within pri-miRNA (227) and pre-miRNA sequences (79). A total of 132 miRNA-SSRs were developed for seed type trait from 63 true pri-miRNAs, of which 46 were specific to pre-miRNAs. Through ePCR, 123 primers were validated and mapped on eight Tunisia chromosomes. Further, 80 SSRs producing specific amplicons were ePCR-confirmed on multiple genomes i.e. Dabenzi, Taishanhong, AG2017 and Tunisia, yielding a set of 63 polymorphic SSRs (polymorphism information content ≥0.5). Of these, 32 miRNA-SSRs revealed higher polymorphism level (89.29%) when assayed on six pomegranate genotypes. Furthermore, target prediction and network analysis suggested a possible association of miRNA-SSRs i.e. miRNA_SH_SSR69, miRNA_SH_SSR36, miRNA_SH_SSR103, miRNA_SH_SSR35 and miRNA_SH_SSR53 with seed type trait. These miRNA-SSRs would serve as important genomic resource for rapid and targeted improvement of seed type trait of pomegranate
Building-up leaders for challenges through vicarious learning: Reflections from Australian leaders
Organizational leaders are essential for dealing with various workplace challenges, such as VUCA (volatility, uncertainty, complexity, and ambiguity) circumstances, and leadership development (LD) is a common way to build-up leaders. However, leadership practitioners are sometimes strongly dissatisfied with LD programs, which has been identified as ineffective in developing leaders for challenging circumstances. Thus, this research aims to discover how to prepare leaders for challenging work environments.
This inductive research investigates open-ended reflections collected from 42 Australian leaders in LD programs to achieve the objective in regard to how crucial leadership capabilities are demonstrated in dealing with challenges and how to develop them. This research includes one methodological study and two empirical inductive studies. The methodological study develops a transparent step-by-step research framework through which the two qualitative studies are conducted. This qualitative research found that leadership resilience is essential in dealing with challenges, and it can strongly motivate others to learn. By observing leadership resilience demonstrated in challenging situations, leaders can improve their capabilities of dealing with challenges (e.g., leadership self-efficacy and selfawareness).
These findings enrich the theoretical understanding of social cognitive theory by integrating ‘Resilient Attitude’ and ‘Coping Skills’ dimensions into the triadic schematization and contributes to better preparing leaders for challenges from two aspects. Firstly, the vicarious learning mechanism can explain ‘How to develop leaders’. Secondly, this research provides a theoretical contribution to psychological resilience by presenting a three-dimension model (resilient attitude, resilient behaviour, and coping skills), which answers ‘What is to be developed’. This research outlines that leaders can improve their capabilities of dealing with challenging circumstances by observing others' challenges and their resilient responses. This finding provides practical contributions to leadership development programs and other human resource management, e.g., talent management. In addition, the author-designed research framework may assist other new researchers in their studies
Therapeutic (Sub)stance: Current practice and therapeutic conduct in preparatory sessions in substance-assisted psychotherapy—A systematized review
Background:
Clinical trials are currently investigating the potential of substance-assisted psychotherapy (SAPT) as treatment for several psychiatric conditions. The potential therapeutic effects of SAPT may be influenced by contextual factors including preparation prior to and integration after the substance-assisted therapy sessions.
Aims:
This systematized review outlines recommendations for current practice in preparatory sessions in SAPT including safety measures and screening procedures, preparation of set and setting, session contents, methods, and roles, prerequisites, and appropriate conduct of therapists.
Methods:
A systematized review of the literature was conducted based on PRISMA guidelines. MEDLINE (OVID), PsycINFO (OVID), and Cochrane Library were searched and clinical trials, treatment manuals, study protocols, case studies, qualitative studies, descriptive studies, theoretical papers, reviews, book chapters, and conference proceedings published until February 1, 2022 were retrieved.
Results:
The final synthesis included k = 83 sources. Information about safety measures including screening of participants, set and setting, contextual-, physiological-, and psychological preparation, roles, competencies, prerequisites, and characteristics of the therapists, and the establishment of a therapeutic relationship were summarized and discussed.
Conclusion:
It is concluded that there is a consensus in the literature about the importance of adequate preparation before the administration of psychoactive substances in SAPT. However, the extent and approaches for these sessions vary across different models and there is a need for timelier and more rigorous qualitative and quantitative investigations assessing different approaches and techniques for the optimal preparation of clients in SAPT
Evidence of male alliance formation in a small dolphin community
The photo-identification of uniquely marked individuals has revealed much about mammalian behaviour and social structure in recent decades. In bottlenose dolphins (Tursiops spp.), for example, the long-term tracking of individuals has unveiled considerable variation in social structure among populations and various spatio-temporal aspects of group formation. In this study, we investigated associations among individual males in a small community of Indo-Pacific bottlenose dolphins (T. aduncus) residing in an urbanized estuary in southwestern Australia. Given the relative proximity of our study area to other populations in which complex male alliances form for the purpose of mate acquisition, we used long-term photo-identification records and social analyses to assess whether such alliances also occur in smaller and more isolated settings. Our work revealed strong social bonds and long-term, non-random associations among individual males, suggesting the occurrence of male alliances. Behavioural observations of alliances interacting with potentially receptive adult females from the estuary community and from adjacent communities, and exhibiting sexual display behaviours near females, suggest that these alliances occur in a reproductive context. As the first formal analysis indicating the occurrence of male alliances outside Shark Bay along the vast western coastline of Australia, this study complements previous research and extends our understanding of the evolutionary and ecological processes that drive alliance formation
Understanding post-crisis trauma recovery for the past decade in uniformed services: A narrative review
Purpose
The purpose of this study is to explore mental health difficulties, including risk and protective factors, which may impact on symptom severity after exposure to crisis situations (war, terrorism and/or natural disasters), among first responders from uniformed services.
Design/methodology/approach
Peer-reviewed journal articles published in English between January 2012 and March 2022 were searched in ProQuest, Wiley, Google Scholar and PubMed databases. In total, 12 articles were obtained from an original screening of 94,058 articles. Full article texts were screened for content and quality by two reviewers, with high agreeability.
Findings
Post-traumatic stress disorder and depression were the most common diagnoses. Risk factors identified were pre-deployment factors of overweight, low cognitive ability and social support, existing emotional difficulties, negative childhood experiences and stressful life events; during crisis situations factors of higher frequency and subjective severity of combat, increased rates of combat stress reaction, high levels of concerns for life and family, more stressful mission position, threat of death/severe injury and high rate of killing the enemy; and post-deployment factors, such as low social support and physical health, lack of coping mechanisms and use of avoidance strategies and social stigma. Protective factors increasing resilience and lessening symptom severity were reported as pre-deployment cognitive ability, high social support, stable physical health, effective coping, post-traumatic growth and high levels of perceived adequacy in pre-deployment preparation and training. In addition to main findings, data about author(s), publication type, population, age, type of crisis and evaluation measures were extracted. Key findings and related theories, gaps in literature and recommendations are discussed.
Originality/value
As yet, however, research into the factors that could serve as risk and/or protective factors are not clearly indicated in terms of post-crisis recovery. As per the authors’ knowledge, this study is an initial approach to considering this area
Characterization of glossy spike mutants and identification of candidate genes regulating cuticular wax synthesis in barley (Hordeum vulgare L.)
Cuticular waxes comprise the hydrophobic layer that protects crops against nonstomatal water loss and biotic and abiotic stresses. Expanding on our current knowledge of the genes that are involved in cuticular wax biosynthesis and regulation plays an important role in dissecting the processes of cuticular wax metabolism. In this study, we identified the Cer-GN1 barley (Hordeum vulgare L.) mutant that is generated by ethyl methanesulfonate mutagenesis with a glossy spike phenotype that is controlled by a single recessive nuclear gene. A physiological analysis showed that the total cuticular wax loads of Cer-GN1 were one-third that of the progenitor wild-type (WT), and its water loss rate was significantly accelerated (p < 0.05). In addition, Cer-GN1 was defective in the glume’s cuticle according to the toluidine blue dye test, and it was deficient in the tubule-shaped crystals which were observed on the glume surfaces by scanning electron microscopy. Using metabolomics and transcriptomics, we investigated the impacts of cuticular wax composition and waxy regulatory genes on the loss of the glaucous wax in the spikes of Cer-GN1. Among the differential metabolites, we found that 16-hydroxyhexadecanoic acid, which is one of the predominant C16 and C18 fatty acid-derived cutin monomers, was significantly downregulated in Cer-GN1 when it was compared to that of WT. We identified two novel genes that are located on chromosome 4H and are downregulated in Cer-GN1 (HvMSTRG.29184 and HvMSTRG.29185) that encode long-chain fatty acid omega-monooxygenase CYP704B1, which regulates the conversion of C16 palmitic acid to 16-hydroxyhexadecanoic acid. A quantitative real-time PCR revealed that the expression levels of HvMSTRG.29184 and HvMSTRG.29185 were downregulated at 1, 4, 8, 12, and 16 days after the heading stage in Cer-GN1 when it was compared to those of WT. These results suggested that HvMSTRG.29184 and HvMSTRG.29185 have CYP704B1 activity, which could regulate the conversion of C16 palmitic acid to 16-hydroxyhexadecanoic acid in barley. Their downregulation in Cer-GN1 reduced the synthesis of the cuticular wax components and ultimately caused the loss of the glaucous wax in the spikes. It is necessary to verify whether HvMSTRG.29184 and HvMSTRG.29185 truly encode a CYP704B1 that regulates the conversion of C16 palmitic acid to 16-hydroxyhexadecanoic acid in barley
The troublesome ticks research protocol: Developing a comprehensive, multidiscipline research plan for investigating human tick-associated disease in Australia
In Australia, there is a paucity of data about the extent and impact of zoonotic tick-related illnesses. Even less is understood about a multifaceted illness referred to as Debilitating Symptom Complexes Attributed to Ticks (DSCATT). Here, we describe a research plan for investigating the aetiology, pathophysiology, and clinical outcomes of human tick-associated disease in Australia. Our approach focuses on the transmission of potential pathogens and the immunological responses of the patient after a tick bite. The protocol is strengthened by prospective data collection, the recruitment of two external matched control groups, and sophisticated integrative data analysis which, collectively, will allow the robust demonstration of associations between a tick bite and the development of clinical and pathological abnormalities. Various laboratory analyses are performed including metagenomics to investigate the potential transmission of bacteria, protozoa and/or viruses during tick bite. In addition, multi-omics technology is applied to investigate links between host immune responses and potential infectious and non-infectious disease causations. Psychometric profiling is also used to investigate whether psychological attributes influence symptom development. This research will fill important knowledge gaps about tick-borne diseases. Ultimately, we hope the results will promote improved diagnostic outcomes, and inform the safe management and treatment of patients bitten by ticks in Australia
Potential of variegated lady beetle Hippodamia variegata in management of invasive tomato potato psyllid Bactericera cockerelli
BACKGROUND
The tomato potato psyllid, Bactericera cockerelli (Šulc) is a new invasive pest in Western Australia, which may disperse across the whole of Australia within a few years and cause significant economic losses. Chemical control is the most widely used approach to manage B. cockerelli, but insect resistance, chemical residue and effects on non-target species have become an increasing concerned. Therefore, in this study, the biocontrol potential of variegated lady beetle, Hippodamia variegata (Goeze) was investigated. The impact of utilizing B. cockerelli as a food source on the predator's development and reproduction was assessed by formulating age-stage, two sex life tables. The predatory potential of H. variegata on B. cockerelli nymphs was assessed in a closed arena and the effects of releasing H. variegata for the control of B. cockerelli were then evaluated.
RESULTS
H. variegata could successfully develop and oviposit when feeding on B. cockerelli. However, both survival and the rate of development were higher for H. variegata feeding on Myzus persicae (Sulzer) than B. cockerelli or a mixed population of B. cockerelli and M. persicae. A type II functional response was observed for H. variegata. In the greenhouse, the releases of H. variegata larvae reduced the number of B. cockerelli nymphs by up to 66% and adults by up to 59%, which positively influenced the plant chlorophyll content and biomass.
CONCLUSIONS
This study demonstrated the potential of the resident generalist predator, H. variegata as a biocontrol agent for the invasive pest, B. cockerelli, which may help improving current management strategies
