    Combined training improves the diagnostic measures of sarcopenia and decreases the inflammation in HIV‐infected individuals

    Background HIV-related sarcopenia is an emerging health issue that often remains undiagnosed and can lead to reduced quality of life, independence, and premature death if untreated. This study investigated the effects of a 6 month combined training (resistance plus aerobic exercise) (CT) intervention on diagnostic measures of sarcopenia, including grip strength, appendicular lean mass index (ALMI), and gait speed. Methods Forty participants were randomized into either a CT group (n = 20; age = 38.3 ± 4.9 years) or a control group (CON; n = 20; age = 37.9 ± 5.1 years). Participants in the CT group performed three supervised sessions per week for 6 months, consisting of weekly reverse linear periodized resistance training followed by 20 min aerobic training. Participants in the CON group were instructed to continue with their current lifestyle habits. Assessments were completed at baseline and after 6 months. Statistical analyses were performed using a two-way analysis of covariance (ANCOVA) adjusted for sex and preintervention values. Primary outcomes included grip strength, ALMI, and gait speed. Secondary outcomes were changes in levels of pro-inflammatory cytokines (IL-6 and TNF-α), IGF-1, and myostatin. Associations were explored between changes in inflammatory markers (IL-6 and TNF-α), gait speed, and ALMI with grip strength. Results A significant increase in ALMI was found for CT compared with CON (0.29 ± 0.13 kg/m2 vs. −0.11 ± 0.14 kg/m2, respectively; P 85%). Conclusions Combined training appears to be an effective means to counteract sarcopenia and improve various inflammatory markers and growth hormones in people living with HIV

    Ridinilazole: a novel, narrow‐spectrum antimicrobial agent targeting Clostridium ( Clostridioides ) difficile

    Clostridium (Clostridioides) difficile infection (CDI) remains an urgent threat to patients in health systems worldwide. Recurrent CDI occurs in up to 30% of cases due to sustained dysbiosis of the gut microbiota which normally protects against CDI. Associated costs of initial and recurrent episodes of CDI impose heavy financial burdens on health systems. Vancomycin and metronidazole have been the mainstay of therapy for CDI for many years; however, these agents continue to cause significant disruption to the gut microbiota and thus carry a high risk of recurrence for CDI patients. Treatment regimens are now turning towards novel narrow spectrum antimicrobial agents which target C. difficile while conserving the commensal gut microbiota, thus significantly reducing risk of recurrence. One such agent, fidaxomicin, has been in therapeutic use for several years and is now recommended as a first-line treatment for CDI, as it is superior to vancomycin in reducing risk of recurrence. Another narrow spectrum agent, ridnilazole, was recently developed and is undergoing evaluation of its potential clinical utility. This review aimed to summarize experimental reports of ridinilazole and assess its potential as a first-line agent for treatment of CDI. Reported results from in vitro assessments, and from hamster models of CDI, show potent activity against C. difficile, non-inferiority to vancomycin for clinical cure and non-susceptibility among most gut commensal bacteria. Phase I and II clinical trials have been completed with ridinilazole showing high tolerability and efficacy in treatment of CDI, and superiority over vancomycin in reducing recurrence of CDI within 30 days of treatment completion. Phase III trials are currently underway, the results of which may prove its potential to reduce recurrent CDI and lessen the heavy health and financial burden C. difficile imposes on patients and healthcare systems

    A reinforcement learning-based approach for imputing missing data

    Missing data is a major problem in real-world datasets, which hinders the performance of data analytics. Conventional data imputation schemes such as univariate single imputation replace missing values in each column with the same approximated value. These univariate single imputation techniques underestimate the variance of the imputed values. On the other hand, multivariate imputation explores the relationships between different columns of data, to impute the missing values. Reinforcement Learning (RL) is a machine learning paradigm where the agent learns by taking actions and receiving rewards in response, to achieve its goal. In this work, we propose an RL-based approach to impute missing data by learning a policy to impute data through an action-reward-based experience. Our approach imputes missing values in a column by working only on the same column (similar to univariate single imputation) but imputes the missing values in the column with different values thus keeping the variance in the imputed values. We report superior performance of our approach, compared with other imputation techniques, on a number of datasets

    Role of melatonin in Alzheimer’s disease: From preclinical studies to novel melatonin-based therapies

    Melatonin and novel melatonin-based therapies such as melatonin-containing hybrid molecules, melatonin analogues, and melatonin derivatives have been investigated as potential therapeutics against Alzheimer’s disease (AD) pathogenesis. In this review, we examine the developmental trends of melatonin therapies for AD from 1997 to 2021. We then highlight the neuroprotective mechanisms of melatonin therapy derived from preclinical studies. These mechanisms include the alleviation of amyloid-related burden, neurofibrillary tangle accumulation, oxidative stress, neuroinflammation, apoptosis, mitochondrial dysfunction, and impaired neuroplasticity and neurotransmission. We further illustrate the beneficial effects of melatonin on behavior in animal models of AD. Next, we discuss the clinical effects of melatonin on sleep, cognition, behavior, psychiatric symptoms, electroencephalography findings, and molecular biomarkers in patients with mild cognitive impairment and AD. We then explore the effectiveness of novel melatonin-based therapies. Lastly, we discuss the limitations of current melatonin therapies for AD and suggest two emerging research themes for future study

    Improving pH control and carbon dioxide utilisation efficiency in microalgae cultivation systems with the use of a Proportional-integral + dead-zone control strategy

    This study investigated the use of an innovative Proportional Integral (PI) + dead-zone control strategy to improve pH control and to overcome the negative environmental impact associated with the currently employed on/off pH control schemes for microalgae cultivation. Scenedesmus sp. grown in anaerobically digested abattoir effluent (ADAE) was used in this study to evaluate the effectiveness of the alternative/advanced control strategy with respect to the on/off control strategy. Results obtained showed that the PI+dead-zone control strategy significantly improved the regulation of the pH level in the pond as evident by noteworthy reductions of key controller performance indicators with respect to the on/off control strategy. Additionally, the advanced control scheme was able to reduce the CO2 usage by a remarkable 59.21% (highest recorded in related literature), resulting in a reduction in CO2 associated costs (approximately 60%) and most importantly, reduction in the amount of greenhouse gas lost to the atmosphere

    Myths and misconceptions about university student volunteering: Development and perpetuation

    This paper examines myths and misconceptions about university student volunteering. Our study explored the experiences of students, host organisations and universities participating in volunteering in Australia, identify good practice, and discover barriers to success. A qualitative approach involved 60 semi-structured interviews with stakeholders. Students were often seen as being energetic, having flexible time and having skills associated with their studies. Some organisations, however, viewed students as unreliable, hard to manage and requiring specific programs. Some hosts were viewed as not valuing student volunteers, or not having the capacity to supervise. These perceptions were found to be nuanced. Erroneous myths were seen to develop from a single event, later confirmed by a ‘related’ event; in scenarios with multiple players, motivations, and complexities. The potential for misconceptions to undermine the true value of student volunteering for all stakeholders is ameliorated when there is common understanding, clear expectation setting, and ongoing dialogue

    This chapter focuses on key national reform agendas and recent policy decisions since 2011 – when the Australian Institute for Teaching and School Leadership initially developed requirements for Initial Teacher Education (ITE) in Australian universities. Universities providing ITE are required not only to comply with national policy, but also with the additional policies of the governments within the states or territories where the universities are located. Australia has a complex arrangement of responsibilities for aspects of school education between its national and state governments, with the Commonwealth in recent years increasingly intervening in what had traditionally been a policy area largely for the states. Strategies for attracting ‘higher-quality’ graduates into the profession which were discussed in the literature at that time – and occasionally advocated for – included salary considerations, teacher education scholarships and performance incentives in the profession itself. The chapter outlines the chronology of the national reforms to give a sense of the snowballing of regulation and compliance

    Integrated application of bacterial carbonate precipitation and silicon nanoparticles enhances productivity, physiological attributes, and antioxidant defenses of wheat (Triticum aestivum L.) under semi-arid conditions

    The use of calcium carbonate-precipitating bacteria (CCPB) has become a well-established ground-improvement technique. However, the effect of the interaction of CCPB with nanoparticles (NPs) on plant performance is still meager. In this study, we aimed at evaluating the role of CCPB and/or silicon NPs (Si-NPs) on the growth, physio-biochemical traits, and antioxidative defense of wheat (Triticum aestivum L.) under semi-arid environmental conditions. A 2-year pot experiment was carried out to determine the improvement of the sandy soil inoculated with CCPB and the foliar application of Si-NPs on wheat plants. We tested the following treatments: spraying plants with 1.0 or 1.5 mM Si-NPs (control = 0 mM Si-NPs), soil inoculated with Bacillus lichenforms (MA16), Bacillus megaterium (MA27), or Bacillus subtilis (MA34), and the interaction of individual Bacillus species with Si-NPs. Our results showed that soil inoculation with any of the three isolated CCPB and/or foliar application of Si-NPs at the rates of 1.0 or 1.5 mM significantly improved (p ≤ 0.05) the physiological and biochemical attributes as well as the enzymatic antioxidant activities of wheat plants. Therefore, the combined treatments of CCPB + Si-NPs were more effective in enhancing physio-biochemical characteristics and enzymatic antioxidant activities than the individual treatments of CCPB or Si-NPs, thus achieving the best performance in the treatment of MA34 + 1.5 mM Si-NPs. Our results demonstrated that the co-application of CCPB and Si-NPs, particularly MA34 + 1.5 mM Si-NPs, considerably activated the antioxidant defense system to mitigate the adverse effects of oxidative stress, thus increasing tolerance and enhancing the production of wheat plants in sandy soils under semi-arid environmental conditions

    A function-based typology for Earth’s ecosystems

    As the United Nations develops a post-2020 global biodiversity framework for the Convention on Biological Diversity, attention is focusing on how new goals and targets for ecosystem conservation might serve its vision of ‘living in harmony with nature’1,2. Advancing dual imperatives to conserve biodiversity and sustain ecosystem services requires reliable and resilient generalizations and predictions about ecosystem responses to environmental change and management3. Ecosystems vary in their biota4, service provision5 and relative exposure to risks6, yet there is no globally consistent classification of ecosystems that reflects functional responses to change and management. This hampers progress on developing conservation targets and sustainability goals. Here we present the International Union for Conservation of Nature (IUCN) Global Ecosystem Typology, a conceptually robust, scalable, spatially explicit approach for generalizations and predictions about functions, biota, risks and management remedies across the entire biosphere. The outcome of a major cross-disciplinary collaboration, this novel framework places all of Earth’s ecosystems into a unifying theoretical context to guide the transformation of ecosystem policy and management from global to local scales. This new information infrastructure will support knowledge transfer for ecosystem-specific management and restoration, globally standardized ecosystem risk assessments, natural capital accounting and progress on the post-2020 global biodiversity framework

    An explicit feature of the World Health Organisation’s response to the COVID-19 pandemic has been to ensure that naming conventions, both for the disease itself and for the variants of its underlying virus, should not have a stigmatising effect on any one population or region. An implicit feature of this undertaking is the recognition that the relation between ‘what is said’ and ‘what is heard’ involves an ongoing and even generative tension that cannot be mapped following a defined set of coordinates. The reason for this, the following paper claims, employing the work of Barbara Cassin, is that there is a uniquely performative (epideictic) aspect to language – one that takes place in a uniquely non-globalised space whose extension remains unscripted or extemporised, a space that is often distended and sometimes localisable but ultimately indivisible. Such a space, this paper shows, is one oriented by an understanding of rhetoric whereby utterances obtain according to the actual use [chreia] or decisive moment [kairos] – as opposed to any definitive place [topos] – of what is said
