    Anthelmintic resistance of gastrointestinal nematodes in dairy calves within a pasture‐based production system of south West Western Australia

    The objective of this study was to determine the prevalence of gastrointestinal nematodes among post-weaned calves aged between 4 and 12 months old within a pasture-based system of south west Australia and quantify the level of anthelmintic resistance. Pre-treatment FECs were monitored on 14 dairy farms. Anthelmintic resistance was assessed on 11 of the farms. Control FECs were compared with anthelmintic FECs at 14 days post-treatment with doramectin (injectable), levamisole (oral), fenbendazole (oral) and a levamisole/abamectin combination (pour-on). Results demonstrate a strong level of anthelmintic resistance, with at least one class of anthelmintic failing to achieve a 95% reduction in FEC in one or more gastrointestinal nematode species. Doramectin was fully effective against Ostertagia, but C. oncophora displayed resistance in 91% of the farms. Conversely, levamisole was fully effective against C. oncophora, but Ostertagia displayed resistance in 80% of the farms. Fenbendazole resistance was present in both C. onocphora and Ostertagia in 64% and 70% of the farms, respectively. Trichostrongylus showed low resistance, occurring in doramectin (14%) and levamisole/abamectin combination (14%). This study confirms that anthelmintic resistance is common. Regular FEC reduction testing is recommended to monitor and guide decision-making for appropriate anthelmintic usage

    Defining pain-validation: The importance of validation in reducing the stresses of chronic pain

    Purpose: To validate an individual's feelings or behaviour is to sanction their thoughts or actions as worthy of social acceptance and support. In contrast, rejection of the individual's communicated experience indicates a denial of social acceptance, representing a potential survival threat. Pain-invalidation, though ill-defined, appears to be a fundamental component of psychosocial stress for people with chronic pain. As such, the aim of this paper was to define pain-validation and outline its importance for those with chronic pain. Methods: The pain-validation construct was defined using themes inherent in the narratives of those with chronic pain, as identified in a previously published systematic search and thematic analysis, together with examination of additional literature on pain-validation in the clinical context. Results: We present a construct definition, proposing that pain-validation must necessarily include: (i) belief that the pain experience is true for the individual, (ii) acceptability of the individual's expressions of pain, and (iii) communication of belief and acceptability to the individual experiencing pain. Further, we outline the importance of pain-validation as a protective factor and means of reducing many of the psychosocial stresses of chronic pain; for example, by indicating social support for pain-coping, buffering negative emotions, and re-enforcing unity and shared identity. Implications: The role of pain-validation in the current era of pain management intervention is discussed. Adhering to interventions that involve cognitive and behavioural change is often difficult. Acknowledging and validating the acceptability of the patient's pain experience in the early stages of pain management may, therefore, be a key component of intervention that encourages compliance to the treatment plan and achieving therapeutic goals

    Multi-omic phenotyping reveals host-microbe responses to bariatric surgery, glycaemic control and obesity

    Background Resolution of type 2 diabetes (T2D) is common following bariatric surgery, particularly Roux-en-Y gastric bypass. However, the underlying mechanisms have not been fully elucidated. Methods To address this we compare the integrated serum, urine and faecal metabolic profiles of participants with obesity ± T2D (n = 80, T2D = 42) with participants who underwent Roux-en-Y gastric bypass or sleeve gastrectomy (pre and 3-months post-surgery; n = 27), taking diet into account. We co-model these data with shotgun metagenomic profiles of the gut microbiota to provide a comprehensive atlas of host-gut microbe responses to bariatric surgery, weight-loss and glycaemic control at the systems level. Results Here we show that bariatric surgery reverses several disrupted pathways characteristic of T2D. The differential metabolite set representative of bariatric surgery overlaps with both diabetes (19.3% commonality) and body mass index (18.6% commonality). However, the percentage overlap between diabetes and body mass index is minimal (4.0% commonality), consistent with weight-independent mechanisms of T2D resolution. The gut microbiota is more strongly correlated to body mass index than T2D, although we identify some pathways such as amino acid metabolism that correlate with changes to the gut microbiota and which influence glycaemic control. Conclusion We identify multi-omic signatures associated with responses to surgery, body mass index, and glycaemic control. Improved understanding of gut microbiota - host co-metabolism may lead to novel therapies for weight-loss or diabetes. However, further experiments are required to provide mechanistic insight into the role of the gut microbiota in host metabolism and establish proof of causality

    Profiling of shelter campers, their attitudes, and perceptions towards environmental impacts of campsite use and management: Evidence from national parks of Sri Lanka

    There is a dearth of research describing the campsite user segment and in assessing camp-site users’ perceptions of the bio-physical impacts of campsite use in Sri Lanka. Using a structured questionnaire, we evaluated campsite user characteristics, visitor motivations, behaviors, and how visitor experience is affected by visible biophysical impacts at campsites. Based on underlying visitor motivations derived through a Factor Analysis and Cluster Analysis, we identified three distinct segments of campsite users: Eco-tourists, Prestige seekers, and Adventure seekers. Results indicate that the overall cleanliness of the campsite is an important attribute that affects the visitor experience. The Ecotourist segment reported environmentally desired motives, attitudes and behaviours. However, majority of campsite users belong to the Prestige-seeker and Adventure-seeker segments. Prestige seekers in particular demand better facilities and proper maintenance of campsites. Additionally, Prestige seekers are more attractive as a market segment to target because they are high-income, well-educated individuals with pro-environmental attitudes and behaviors. Recommended management actions include increasing visitor awareness, monitoring, zonation of activity areas, and enforcement of park campsite user policies to ensure enhanced visitor experience

    Integrated generalized zero-shot learning for fine-grained classification

    Embedding learning (EL) and feature synthesizing (FS) are two of the popular categories of fine-grained GZSL methods. EL or FS using global features cannot discriminate fine details in the absence of local features. On the other hand, EL or FS methods exploiting local features either neglect direct attribute guidance or global information. Consequently, neither method performs well. In this paper, we propose to explore global and direct attribute-supervised local visual features for both EL and FS categories in an integrated manner for fine-grained GZSL. The proposed integrated network has an EL sub-network and a FS sub-network. Consequently, the proposed integrated network can be tested in two ways. We propose a novel two-step dense attention mechanism to discover attribute-guided local visual features. We introduce new mutual learning between the sub-networks to exploit mutually beneficial information for optimization. Moreover, we propose to compute source-target class similarity based on mutual information and transfer-learn the target classes to reduce bias towards the source domain during testing. We demonstrate that our proposed method outperforms contemporary methods on benchmark datasets

    Optimal Design of a hybrid solar PV/BG-powered heterogeneous network

    The increased penetration of renewable energy sources (RESs) along with the rise in demand for wireless communication had led to the need to deploy cellular base stations powered by locally accessible RESs. Moreover, networks powered by renewable energy sources have the ability to reduce the costs of generating electricity, as well as greenhouse gas emissions, thus maintaining the quality of service (QoS). This paper examines the techno-economic feasibility of developing grid-tied solar photovoltaic (PV)/biomass generator (BG)-powered heterogeneous networks in Bangladesh, taking into account the dynamic characteristics of RESs and traffic. To guarantee QoS, each macro and micro-base station is supplied through a hybrid solar PV/BG coupled with enough energy storage devices. In contrast, pico and femto BSs are powered through standalone solar PV units due to their smaller power rating. A hybrid optimization model for electric renewables (HOMER)-based optimization algorithm is considered to determine the optimum system architecture, economic and environmental analysis. MATLAB-based Monte-Carlo simulations are used to assess the system’s throughput and energy efficiency. A new weighted proportional-fair resource method is presented by trading power consumption and communication latency in non-real-time applications. Performance analysis of the proposed architecture confirmed its energy efficiency, economic soundness, reliability, and environmental friendliness. Additionally, the suggested method was shown to increase the battery life of the end devices

    Using an incomplete block design to allocate lines to environments improves sparse genome‐based prediction in plant breeding

    Genomic selection (GS) is a predictive methodology that trains statistical machine-learning models with a reference population that is used to perform genome-enabled predictions of new lines. In plant breeding, it has the potential to increase the speed and reduce the cost of selection. However, to optimize resources, sparse testing methods have been proposed. A common approach is to guarantee a proportion of nonoverlapping and overlapping lines allocated randomly in locations, that is, lines appearing in some locations but not in all. In this study we propose using incomplete block designs (IBD), principally, for the allocation of lines to locations in such a way that not all lines are observed in all locations. We compare this allocation with a random allocation of lines to locations guaranteeing that the lines are allocated to the same number of locations as under the IBD design. We implemented this benchmarking on several crop data sets under the Bayesian genomic best linear unbiased predictor (GBLUP) model, finding that allocation under the principle of IBD outperformed random allocation by between 1.4% and 26.5% across locations, traits, and data sets in terms of mean square error. Although a wide range of performance improvements were observed, our results provide evidence that using IBD for the allocation of lines to locations can help improve predictive performance compared with random allocation. This has the potential to be applied to large-scale plant breeding programs

    Prediction of lamb carcase C-site fat depth and GR tissue depth using a non-invasive portable microwave system versus body condition scoring

    This study compared portable ultra-wide band microwave system (MiS) versus body condition score to predict C-site fat depth, GR tissue depth and eye muscle depth (EMD) in lambs. Experiment 1 assessed MiS and condition score to predict ultrasound measured C-site and EMD (n = 1549). Precision and accuracy was greatest for the MiS measurement with liveweight included in the model, with a C-site predicted RMSEP of 0.58 mm, R2 0.60 and bias of 0.021 mm and an EMD predicted RMSEP of 2.27 mm, R2 0.72 and bias of 0.088 mm. Experiment 2 (n = 900) assessed pre-slaughter MiS scanning and condition scoring to predict carcase GR tissue depth, C-site fat depth and EMD. MiS performed better than condition score for all three carcase trait predictions, regardless of the inclusion of liveweight, with the highest precision and accuracy for GR tissue depth determination with a RMSEP of 3.68 mm, R2 0.63 and bias 0.072 mm

    Fast reconstruction of centre of mass and foot kinematics during a single-legged horizontal jump: A point-cloud processing approach

    Horizontal jumps are discrete, fast, over-ground movements requiring coordination of the centre of mass (CoM) and base of support and are routinely assessed in sports settings. There is currently no biomechanics-based system to aid in their quick and objective large-scale assessment. We describe a practical system combining a single low-cost depth-sensing camera and point-cloud processing (PCP) to capture whole-body CoM and foot kinematics. Fourteen participants performed 10 single-leg horizontal jumps for distance. Foot displacement, CoM displacement, CoM peak velocity and CoM peak acceleration in the anterior-posterior direction of movement were compared with a reference 15-segment criterion model, captured concurrently using a nine-camera motion capture system (Vicon Motion Systems, UK). Between-system Pearson’s correlations were very-large to near-perfect (n = 140; foot displacement = 0.99, CoM displacement = 0.98, CoM peak velocity = 0.97, CoM peak acceleration = 0.79), with mean biases being trivial–small (-0.07 cm [0.12%], 3.8 cm [3.5%], 0.03 m·s−1 [1.6%], 0.42 m·s−2 [7%], respectively) and typical errors being trivial–small for displacement (foot: 0.92 cm [0.8%]; CoM: 3.8 cm [3.4%]) and CoM peak velocity (0.07 m·s−1 [4.3%]), and large for CoM peak acceleration (0.72 m·s−2 [15%]). Limits of agreement were −1.9 to 2.0 cm for foot displacement, −11.3 to 3.6 cm for CoM displacement, −0.17 to 0.12 m·s−1 for CoM peak velocity and −2.28 to 1.43 m·s−2 for CoM peak acceleration. The practical system captured CoM and foot kinematics during horizontal jumps with acceptable precision. Further work to improve estimates of CoM accelerations and different populations are warranted

    Moving beyond DNA sequence to improve plant stress responses

    Plants offer a habitat for a range of interactions to occur among different stress factors. Epigenetics has become the most promising functional genomics tool, with huge potential for improving plant adaptation to biotic and abiotic stresses. Advances in plant molecular biology have dramatically changed our understanding of the molecular mechanisms that control these interactions, and plant epigenetics has attracted great interest in this context. Accumulating literature substantiates the crucial role of epigenetics in the diversity of plant responses that can be harnessed to accelerate the progress of crop improvement. However, harnessing epigenetics to its full potential will require a thorough understanding of the epigenetic modifications and assessing the functional relevance of these variants. The modern technologies of profiling and engineering plants at genome-wide scale provide new horizons to elucidate how epigenetic modifications occur in plants in response to stress conditions. This review summarizes recent progress on understanding the epigenetic regulation of plant stress responses, methods to detect genome-wide epigenetic modifications, and disentangling their contributions to plant phenotypes from other sources of variations. Key epigenetic mechanisms underlying stress memory are highlighted. Linking plant response with the patterns of epigenetic variations would help devise breeding strategies for improving crop performance under stressed scenarios
