Molecular Signals Controlling the Inhibition of Nodulation by Nitrate in Medicago truncatula
The presence of nitrogen inhibits legume nodule formation, but the mechanism of this inhibition is poorly understood. We found that 2.5 mM nitrate and above significantly inhibited nodule initiation but not root hair curling in Medicago trunatula. We analyzed protein abundance in M. truncatula roots after treatment with either 0 or 2.5 mM nitrate in the presence or absence of its symbiont Sinorhizobium meliloti after 1, 2 and 5 days following inoculation. Two-dimensional gel electrophoresis combined with mass spectrometry was used to identify 106 differentially accumulated proteins responding to nitrate addition, inoculation or time point. While flavonoid-related proteins were less abundant in the presence of nitrate, addition of Nod gene-inducing flavonoids to the Sinorhizobium culture did not rescue nodulation. Accumulation of auxin in response to rhizobia, which is also controlled by flavonoids, still occurred in the presence of nitrate, but did not localize to a nodule initiation site. Several of the changes included defense- and redox-related proteins, and visualization of reactive oxygen species indicated that their induction in root hairs following Sinorhizobium inoculation was inhibited by nitrate. In summary, the presence of nitrate appears to inhibit nodulation via multiple pathways, including changes to flavonoid metabolism, defense responses and redox changes
Pollution assessment of organochlorine pesticides in urban agricultural soils of Jilin City, China
The content, source, and pollution level of organochlorine pesticides (OCPs) in soils are necessary to assess potential risks to eco-environment and human health, and to target environment-friendly policies. A total of 50 surface soil samples were collected from urban vegetable fields of Jilin City and thirteen OCPs were analyzed. The concentrations were in the ranges of 3.16-48.35ngg(-1) for Dichloro-Diphenyl-Tricgloroethanes (DDTs, sum of o, p-DDT, p, p-DDT, p, p-DDD, and p, p-DDE), 4.37-44.77ngg(-1) for Hexachlorocyclohexanes (HCHs, sum of -HCH, -HCH, -HCH, and -HCH), 1.19-13.17ngg(-1) for Chlordanes (sum of heptachlor, t-chlordane, and c-chlordane), 0.24-2.60ngg(-1) for aldrin, and nd-3.43ngg(-1) for dieldrin, respectively. The different compositions indicated that the residues of DDTs and HCHs originated mainly from the historical application, while chlordanes were mostly from recent input. On the basis of soil quality standards of China and the Netherlands, DDTs and chlordanes in this study were categorized as light pollution, and HCHs were classified as no pollution for the majority of soil samples. There are a variety of OCPs residues in urban vegetable soils of Jilin City, but it is still safe and suitable for agricultural production
A proteomic analysis of mushroom polysaccharide-treated HepG2 cells
The anti-tumor properties of fungal polysaccharides have gained significant recognition in Asia and tropical America. In this study, the differential expression of proteins in normal HepG2 cells and those treated with polysaccharides that had been isolated from Phellinus linteus (PL), Ganoderma lucidum (GL) and Auricularia auricula (AA) was investigated. Using two-dimensional electrophoresis (2DE), a total of 104 protein spots were determined to be overexpressed in these cells compared with noncancerous regions. A total of 59 differentially expressed proteins were identified through MALDI-TOF-MS. In addition, 400 biological processes (BP), 133 cell components (CC) and 146 molecular functions (MF) were enriched by Gene Ontology (GO) analysis, and 78 KEGG pathways were enriched by pathway enrichment. Protein-Protein Interaction (PPI) analysis demonstrated the interaction networks affected by polysaccharides in HepG2 cells. Then, DJ-1 and 14-3-3 were identified as the key proteins in the networks, and the expression of the mRNA and proteins were evaluated using Real-time quantitative PCR (qRT-PCR) and Western blotting (WB), respectively. The results were in agreement with the 2DE. These results provided information on significant proteins of hepatocellular carcinoma (HCC) and form an important basis for the future development of valuable medicinal mushroom resources
Rhizobium acidisoli sp nov., isolated from root nodules of Phaseolus vulgaris in acid soils
Two Gram-negative, aerobic, non-motile, rod-shaped bacterial strains, FH13(T) and FH23, representing a novel group of Rhizobium isolated from root nodules of Phaseolus vulgaris in Mexico, were studied by a polyphasic analysis. Phylogeny of 16S rRNA gene sequences revealed them to be members of the genus Rhizobium related most closely to Whizobium anhuiense' CCBAU 23252 (99.7 % similarity), Rhizobium leguminosarum USDA 2370(T) (98.6 %), and Rhizobium sophorae CCBAU 03386(T) and others (<= 98.3 %). In sequence analyses of the housekeeping genes recA, glnll and atpD, both strains formed a subclade distinct from all defined species of the genus Rhizobium at sequence similarities of 82.3-94.0 %, demonstrating that they represented a novel genomic species in the genus Rhizobium. Mean levels of DNA-DNA relatedness between the reference strain FH13(T) and the type strains of related species varied between 13.0 +/- 2.0 and 52.1 +/- 1.2 %. The DNA G+C content of strain FH13(T) was 63.5 mol% (T-m). The major cellular fatty acids were 16 : 0, 17 : 0 anteiso, 18 : 0, summed feature 2 (12 : 0 aldehyde/unknown 10.928) and summed feature 8 (18 : 1 omega 7c). The fatty acid 17 : 1 omega 5c was unique for this strain. Some phenotypic features, such as failure to utilize adonitol, L-arabinose, D-fructose and D-fucose, and ability to utilize D-galacturonic acid and itaconic acid as carbon source, could also be used to distinguish strain FH13(T) from the type strains of related species. Based upon these results, a novel species, Rhizobium acidisoli sp. nov., is proposed, with FH13(T) (=CCBAU 101094(T)=HAMBI 3626(T)=LMG 28672(T)) as the type strain
Simulation of the Impact of a Sensor's PSF on Mixed Pixel Decomposition: 1. Nonuniformity Effect
The nonuniformity of the spatial response to surface radiation is a fundamental characteristic of all airborne and spaceborne sensors that inevitably introduces uncertainty into the estimation of object proportions by the spectral unmixing of mixed pixels. Simulated data of the surface radiation distribution and a TM (thematic mapper) response matrix were developed and utilized to imitate the generation of mixed pixels and the extraction of the object proportion via a Monte Carlo simulation, and then, the nonuniformity effect of a sensor's PSF (point spread function) was explored. The following conclusions were drawn: (1) given a nonuniform spatial response of a sensor to a surface scene with a constant object proportion and various object distribution patterns, the mixed pixel DN (digital number) of a remotely-sensed image becomes a random variable, which causes a PSF nonuniform effect on the object proportion extraction; (2) for the estimated object proportion, the corresponding true object proportion appears with a random variation; its upper and lower bounds take on an asymmetrical spindle shape; and models of these bound curves at any probability level were established; (3) there exists a negative linear relationship between the bias of the spectral unmixing and the estimated proportion; the bias is zero at an estimated proportion of 50%, and when the estimated proportions are approximately 100% and 0%, the object proportion is overestimated by 0.78% and underestimated by 0.78%, respectively; (4) the relationship between the standard deviation of the spectral unmixing and the estimated proportion follows a symmetrical polynomial function opening downward; the standard deviation reaches a maximum of 4.4% at the estimated proportion of 50%, and when the estimated proportion is approximately 100% or 0%, the standard deviation is a minimum, 1.05%. The above findings contribute to a comprehensive understanding of the PSF nonuniformity effect, have the potential to compensate for the bias of proportion estimation and present its confidence interval at any probability level
Mapping paddy rice distribution using multi-temporal Landsat imagery in the Sanjiang Plain, northeast China
Information of paddy rice distribution is essential for food production and methane emission calculation. Phenology-based algorithms have been utilized in the mapping of paddy rice fields by identifying the unique flooding and seedling transplanting phases using multi-temporal moderate resolution (500 m to 1 km) images. In this study, we developed simple algorithms to identify paddy rice at a fine resolution at the regional scale using multi-temporal Landsat imagery. Sixteen Landsat images from 2010-2012 were used to generate the 30 m paddy rice map in the Sanjiang Plain, northeast China—one of the major paddy rice cultivation regions in China. Three vegetation indices, Normalized Difference Vegetation Index (NDVI), Enhanced Vegetation Index (EVI), and Land Surface Water Index (LSWI), were used to identify rice fields during the flooding/transplanting and ripening phases. The user and producer accuracies of paddy rice on the resultant Landsat-based paddy rice map were 90% and 94%, respectively. The Landsat-based paddy rice map was an improvement over the paddy rice layer on the National Land Cover Dataset, which was generated through visual interpretation and digitalization on the fine-resolution images. The agricultural census data substantially underreported paddy rice area, raising serious concern about its use for studies on food security
Measuring the sustainable urbanization potential of cities in Northeast China
Sustainable urbanization is not only an important research topic in the field of urbanization, but also the development direction of new-type urbanization. In this paper, we construct an index system to evaluate sustainable urbanization potential with the entropy method. Results show that potential values of sustainable urbanization in most cities are not high. Cities with higher sustainable urbanization potential values are mainly located in the central part of Northeast China. Environmental potential of sustainable urbanization is the main contributor to sustainable urbanization potential in Northeast China. There is no absolute relationship between city size and potential value, large city does not always mean greater potential. Correlation analysis shows that urbanization rate cannot reflect the sustainable urbanization potential of a region. Population urbanization is not the ultimate goal of sustainable urbanization. Unilateral pursue urbanization rate cannot improve the potential of sustainable urbanization. Towards sustainable urbanization, governments in Northeast China should revitalize local economy, pay more attention to the rural areas and develop low-carbon economy or ecological economy. Finally, this paper highlights the importance of choosing more integrated methodology or new models for measuring sustainable urbanization potential in view of the shortcomings of one method
Variations of Dissolved Iron in the Amur River During an Extreme Flood Event in 2013
As a key factor limiting primary productivity in marine ecosystem, dissolved iron (DFe) export from fluvial systems has increased recently. There is particular concern about discharges of DFe during extreme flooding, when they are thought to increase considerably. An extreme flood event that caused inundation of extensive areas of Far Eastern Russia and Northeastern China occurred in the basin of the Amur River during summer and autumn 2013. During this event, water samples were collected in the middle reaches of the Amur River and the lower reaches at Khabarovsk City and analyzed for DFe concentrations and other aquatic parameters. The results show that the average DFe concentrations in the middle reaches of the Amur River (right bank) and at Khabarovsk were 1.11 mg/L and 0.32 mg/L, respectively, during the extreme flood in 2013. The total discharge of DFe during the flood event was 6.25 × 10~4 t. The high discharge of DFe during the flood reflects the elevated discharge of the river, hydrologically connected riparian wetlands, vast quantities of terrestrial runoff, and flood discharges from the Zeya and Bureya reservoirs. These results show that long-term monitoring is needed to identify and assess the impacts of DFe transport on the downstream reaches, estuarine area, and coastal ecosystems of the Amur River
基于共享资源的灌区三维地理场景构建
在三维虚拟场景中进行灌区日常任务的模拟优化、设计分析更符合人们的认知习惯,能提供更丰富的展现手法与更多的信息量,可以弥补传统方法难以表达复杂、宏大场景、资金投入大、持续周期长等不足。综述了用来构建三维虚拟场景的技术,并提出综合利用GIS(Geographic Information System)、RS(Remote Sensing)技术、虚拟现实技术,基于共享资源,采用数据融合、混合建模、交互式一体化建模等方法来构建灌区三维虚拟环境的技术路线。该技术路线较为简单、快捷,能充分利用、发挥共享资源的作用,不需要构建特殊硬软件环境就能满足灌区日常运行与管理的需求。构建的灌区三维虚拟场景对提升灌区服务管理水平与科学决策能力具有借鉴意义
