长江中游典型河段底栖动物的物理栖息地模型构建与应用
考虑水生生物生境需求的物理栖息地模型被认为是评估河流流量变化对水生态系统影响的最可信的方法之一。本研究选取长江干流含有河漫滩的监利江段为实例,建立底栖动物各类群的物理栖息地模型,计算变化流量下栖息地适宜面积的时间序列,并据此进行生态流量决策。结果显示长江中游底栖动物最敏感的环境参数是流速,适宜范围为0 ~ 0.2 m/s;其次是水深,适宜范围为0 ~ 6 m。在考虑敏感环境参数的前提下,得出监利江段底栖动物的最佳生态流量为20 000 m~3/s。三峡大坝蓄水后枯水期和平水期底栖动物适宜面积的低值部分减小,丰水期适宜面积增加。为了保护底栖动物栖息地,建议三峡大坝在防洪蓄水的同时能兼顾底栖动物的生态流量需求,调节枯水期和平水期的流量,让监利江段接近4000 m~3/s,丰水期接近20 000 m~3/s。在枯水年增大枯水期和平水期的流量,平水年增大枯水期的流量,丰水年减少丰水期的流量。本研究方法可以供长江其他河段目标物种的生态流量决策和生态修复方案设计参考
水域生态学中生物稳定同位素样品采集、处理与保存
稳定同位素分析技术由于能够刻画复杂的食物网结构并追踪食物网中的能量流而成为水域生态学研究中的重要手段。但是当水生生物样品采集、处理和保存过程中存在不确定性时,营养关系分析中的同位素结果可能会产生误导性解释。文章采用数据模拟分析和文献总结的方法,研究了水域生态系统中样品采集、处理和保存对于稳定同位素的影响,概括性地建议了水域生态系统中适合应用稳定同位素分析技术开展生态学研究的样品采集、处理和保存的注意事项。但今后仍需进一步评估样品采集、处理和保存对稳定同位素比值的影响效果,确定化学动力学在水生生物样品采集、处理和保存中的作用,以进一步完善水生生物样品的采集、处理和保存稳定同位素生态学研究规范
长江岸线宜昌江段五种不同类型水样对稀有鲫胚胎的毒性
为了检测和评估长江岸线宜昌江段五种不同类型水样的生物毒性,研究于2019年3月在宜昌江段采集五种典型水样,采用稀有鲫胚胎进行了暴露试验。经检测发现3#水样(工业生产污水)、4#水样(生活污水)常规污染物(氨氮、化学需氧量、高锰酸盐指数)等指数超标。胚胎毒性试验结果显示3#和4#水样的生物毒性最大,其畸形率和死亡率等指标均显著高于其他组(P<0.05)。2#水样(长江白洋港江段水样)对稀有鲫胚胎发育无显著影响;其中3#和4#两组水样处理的胚胎出现诸如脊椎弯曲、心率下降、死亡率上升、孵化率下降和发育迟缓等发育异常现象;同时, 4#水样处理的胚胎还出现大量心包水肿现象。研究表明氨氮含量等常规水质指标超标会对稀有鲫胚胎产生较大毒性效应。研究可为长江岸线宜昌江段水体的综合评价及后续治理提供依据

基于环境DNA宏条形码的洱海鱼类多样性研究
研究使用环境DNA宏条形码(eDNA metabarcoding)检测洱海鱼类多样性,探索适用于洱海鱼类多样性监测和保护的新方法。通过水样采集、过滤、eDNA提取、遗传标记扩增、测序与生物信息分析的环境DNA宏条形码标准化分析流程,从洱海16个采样点中获得可检测的9个采样点数据,共检测出17种鱼类,其中土著种5种、外来种12种;鲫(Carassius auratus)、鳙(Hypophthalmichthys nobilis)、麦穗鱼(Pseudorasbora parva)、泥鳅(Misgurnus anguillicaudatus)和食蚊鱼(Gambusia affinis)为优势种。研究结果表明虽然环境DNA宏条形码无法完全替代传统的鱼类监测方法,但作为一种新兴的生物多样性监测手段,其可用于快速检测洱海鱼类多样性及其空间分布
The NanDeSyn database for Nannochloropsis systems and synthetic biology
Nannochloropsis species, unicellular industrial oleaginous microalgae, are model organisms for microalgal systems and synthetic biology. To facilitate community-based annotation and mining of the rapidly accumulating functional genomics resources, we have initiated an international consortium and present a comprehensive multi-omics resource database named Nannochloropsis Design and Synthesis (NanDeSyn; ). Via the Tripal toolkit, it features user-friendly interfaces hosting genomic resources with gene annotations and transcriptomic and proteomic data for six Nannochloropsis species, including two updated genomes of Nannochloropsis oceanica IMET1 and Nannochloropsis salina CCMP1776. Toolboxes for search, Blast, synteny view, enrichment analysis, metabolic pathway analysis, a genome browser, etc. are also included. In addition, functional validation of genes is indicated based on phenotypes of mutants and relevant bibliography. Furthermore, epigenomic resources are also incorporated, especially for sequencing of small RNAs including microRNAs and circular RNAs. Such comprehensive and integrated landscapes of Nannochloropsis genomics and epigenomics will promote and accelerate community efforts in systems and synthetic biology of these industrially important microalgae
Understanding the Differences in the Growth and Toxin Production of Anatoxin-Producing Cuspidothrix issatschenkoi Cultured with Inorganic and Organic N Sources from a New Perspective: Carbon/Nitrogen Metabolic Balance
Cyanotoxins are the underlying cause of the threat that globally pervasive Cyanobacteria Harmful algal blooms (CyanoHABs) pose to humans. Major attention has been focused on the cyanobacterial hepatotoxin microcystins (MCs); however, there is a dearth of studies on cyanobacterial neurotoxin anatoxins. In this study, we explored how an anatoxin-producing Cuspidothrix issatschenkoi strain responded to culture with inorganic and organic nitrogen sources in terms of growth and anatoxins production. The results of our study revealed that - alanine could greatly boost cell growth, and was associated with the highest cell productivity, while urea significantly stimulated anatoxin production with the maximum anatoxin yield reaching 25.86 mu g/mg dry weight, which was 1.56-fold higher than that in the control group (BG11). To further understand whether the carbon/nitrogen balance in C. issatschenkoi would affect anatoxin production, we explored growth and toxin production in response to different carbon/nitrogen ratios (C/N). Anatoxin production was mildly promoted when the C/N ratio was within low range, and significantly inhibited when the C/N ratio was within high range, showing approximately a three-fold difference. Furthermore, the transcriptional profile revealed that anaC gene expression was significantly up-regulated over 2-24 h when the C/N ratio was increased, and was significantly down-regulated after 96 h. Overall, our results further enriched the evidence that urea can stimulate cyanotoxin production, and -alanine could boost C. issatschenkoi proliferation, thus providing information for better management of aquatic systems. Moreover, by focusing on the intracellular C/N metabolic balance, this study explained the anatoxin production dynamics in C. issatschenkoi in response to different N sources
A holistic assessment of water quality condition and spatiotemporal patterns in impounded lakes along the eastern route of China's South-to-North water diversion project
Water quality is one of the key determinants for assessing effectiveness and success of water diversions, but rarely studied at a spatial scale that crosses large river basins. Multiple statistical methods and the water quality index (WQI) were used to assess overall condition and detect spatiotemporal patterns of water quality in a series of impounded lakes along the Eastern Route of China's South-to-North Water Diversion Project. Principal components analysis and analysis of variances identified three groups with distinct water quality characteristics: upstream Gaoyou Lake and Hongze Lake showing relatively higher nutrients, turbidity, and total suspended solids; downstream Dongping lake and Donghu Lake showing higher conductivity, total hardness, and chloride; and Luoma Lake and Nansi Lake intermediate between the two former groups. The WQI indicated overall "Good" water quality with an improving trend from upstream to downstream lakes. The upstream Gaoyou Lake had over 55% of the monitoring sites with "Moderate" water quality in all the seasons. Management should focus on preventing high nitrogen, phosphorus, turbidity, and total suspended solids in upstream lakes, high chloride in downstream lakes, high nitrogen during water diversion seasons, and high phosphorus during non-water diversion seasons. These findings greatly improved our understanding of the spatiotemporal water quality patterns of the impounded lakes, and can be used to develop water quality management strategies. 共球藻纲丝状藻中国新记录属种———洪泛拟寇丽藻
从甘肃省张掖市黑河流域分离了一株不分枝的丝状绿藻,对其进行纯培养并保藏于中国科学院淡水藻种库,编号FACHB-2451。形态观察表明,该藻株具有相对较长的营养细胞(细胞长为宽的3~8倍)、双叶形的片状叶绿体、末端细胞顶端钝圆或略尖细等特征。结合形态比较和基于18SrDNA基因序列的系统发育分析,确定该藻株为中国淡水共球藻纲新记录属种———洪泛拟寇丽藻(Koliellopsis inundata Lokhorst)。分子系统发育研究结果表明,所分离的藻株与分离自比利时与荷兰边境农地的洪泛拟寇丽藻模式藻株序列相似度极高,且拟寇丽藻属与寇丽藻属、针丝藻属和拟针丝藻属等不分枝丝状拟寇丽藻科成员亲缘关系密切,可以较好地聚成一个亚支
Comparative transcriptome analyses and identification of candidate genes involved in vertebral abnormality of bighead carp Hypophthalmichthys nobilis
Body deformity occurs both in wild and farmed fishes, which is one of the most challenging problems for aquaculture industry. In most cases, such body deformities are linked to skeletal deformities. Currently, very limited information is available on skeletal deformities of farmed fish species which may be caused by genetic factor. In this study, we performed muscle and vertebra transcriptome analyses in body deformity and normality of bighead carp Hypophthalmichthys nobilis (from one meiotic gynogenesis family) using RNA-Seq. A total of 43,923 and 44,416 unigenes were predicted in muscles and vertebrae, respectively. Based on these data, we further explored the gene expression profiles in gynogenetic normal and abnormal bighead carp. No differen-tially expressed gene (DEG) was found in transcriptome data of muscles. Totally, 20 key DEGs were identified in transcriptome data of vertebrae, such as low density lipoprotein-related protein 2 (lrp2), bone morphogenetic protein 2B (bmp2b) and collagen alpha-1(IV) (col4a1). 12 potential pathways were also identified in vertebra transcriptome data, which were mainly involved in development, growth, cytoskeleton and energy metabolism, such as MAPK signaling pathway, regulation of actin cytoskeleton and TGF-beta signaling pathway. Results of this study will be informative for the understanding of genetic mechanisms for body shape formation and also provide potential candidate genes for selection program involved in body shape and skeletal development in H. nobilis
