腐殖酸对沉积物中纳米银的释放及其毒性的影响研究
以有机质腐殖酸(Humic acid, HA)为例,研究沉积物中不同HA含量(0、1%、5%和10%)对沉积物其中纳米银(Silver nanoparticles, AgNPs)浓度为100 μg/g dw中AgNPs释放的影响,以及银释放对斑马鱼(Danio Rerio)的毒性效应。结果显示,暴露15d和30d后,水柱中总银浓度为(0.01±0.01)-(2.53±0.52)μg/L,且高HA含量(5%和10%)的沉积物组中释放到水柱中的银浓度高于低HA含量(0和1%)的沉积物组,表明HA的存在促进了沉积物中AgNPs向水柱释放。释放的AgNPs能够积累在斑马鱼组织中,且主要积累在肝脏和肠道中,并可导致斑马鱼组织氧化应激效应。另外, 16S rRNA高通量测序结果显示,受释放到水柱银影响,斑马鱼肠道中微生物群落结构也会发生改变。研究初步探明了沉积物中AgNPs的释放规律及其环境风险,可为全面评价AgNPs的环境行为及生态安全性提供重要依据
Foundation species across a latitudinal gradient in China
Foundation species structure forest communities and ecosystems but are difficult to identify without long-term observations or experiments. We used statistical criteria--outliers from size-frequency distributions and scale-dependent negative effects on alpha diversity and positive effects on beta diversity--to identify candidate foundation woody plant species in 12 large forest-dynamics plots spanning 26 degrees of latitude in China. We used these data (1) to identify candidate foundation species in Chinese forests, (2) to test the hypothesis--based on observations of a midlatitude peak in functional trait diversity and high local species richness but few numerically dominant species in tropical forests--that foundation woody plant species are more frequent in temperate than tropical or boreal forests, and (3) to compare these results with data from the Americas to suggest candidate foundation genera in northern hemisphere forests. Using the most stringent criteria, only two species of Acer, the canopy tree Acer ukurunduense and the shrubby treelet Acer barbinerve, were identified in temperate plots as candidate foundation species. Using more relaxed criteria, we identified four times more candidate foundation species in temperate plots (including species of Acer, Pinus, Juglans, Padus, Tilia, Fraxinus, Prunus, Taxus, Ulmus, and Corlyus) than in (sub)tropical plots (the treelets or shrubs Aporosa yunnanensis, Ficus hispida, Brassaiopsis glomerulata, and Orophea laui). Species diversity of co-occurring woody species was negatively associated with basal area of candidate foundation species more frequently at 5- and 10-m spatial grains (scale) than at a 20-m grain. Conversely, Bray-Curtis dissimilarity was positively associated with basal area of candidate foundation species more frequently at 5-m than at 10- or 20-m grains. Both stringent and relaxed criteria supported the hypothesis that foundation species are more common in mid-latitude temperate forests. Comparisons of candidate foundation species in Chinese and North American forests suggest that Acer be investigated further as a foundation tree genus
PELP1在胃癌中的致瘤作用
目的探讨谷氨酸-脯氨酸-亮氨酸富集蛋白1(proline-,glutamic acid-,and leucine-rich protein 1,PELP1)在胃癌中的生物学功能及临床意义。方法通过生物信息学方法分析PELP1在胃癌中的表达,并在胃癌细胞系和胃癌临床样本中予以验证,Kaplan-Meier生存分析探讨PELP1表达与胃癌患者生存预后的关系。以siRNA敲降胃癌细胞AGS中的PELP1表达,以CCK-8实验、平板克隆形成实验、EdU细胞增殖实验检测细胞增殖活性;流式细胞术测定细胞周期;细胞划痕实验、Transwell小室检测细胞迁移、侵袭能力。Western blot和qRT-PCR检测敲降PELP1表达的AGS细胞中Src-Erk通路信号分子表达情况。结果PELP1在胃癌组织中表达相对较高,生存分析显示PELP1高表达的患者生存期更短;敲降PELP1表达后的胃癌细胞增殖、迁移、侵袭等恶性生物学行为均受到抑制;敲降PELP1表达的AGS细胞,其Src-Erk通路关键信号分子c-Src、PI3K和Erk的表达均降低。结论PELP1在胃癌的发生发展中起致瘤作用,可能是胃癌潜在的治疗靶点
FishDB: an integrated functional genomics database for fishes
Background: Hundreds of genomes and transcriptomes of fish species have been sequenced in recent years. However, fish scholarship currently lacks a comprehensive, integrated, and up-to-date collection of fish genomic data.
Results: Here we present FishDB, the first database for fish multi-level omics data, available online at . The database contains 233 fish genomes, 201 fish transcriptomes, 5841 fish mitochondrial genomes, 88 fish gene sets, 16,239 miRNAs of 65 fishes, 1,330,692 piRNAs and 4852 lncRNAs of Danio rerio, 59,040 Mb untranslated regions (UTR) of 230 fishes, and 31,918 Mb coding sequences (CDS) of 230 fishes. Among these, we newly generated a total of 11 fish genomes and 53 fish transcriptomes.
Conclusions: This release contains over 410,721.67 Mb sequences and provides search functionality, a BLAST server, JBrowse, and PrimerServer modules
Establishment and characterization of a cold-sensitive neural cell line from the brain of tilapia (Oreochromis niloticus)
The aquaculture of tilapia (Oreochromis sp.) is adversely affected by the sensitivity to cold stress. A large number of genes in tilapia were found to be regulated by cold stress, but their functions and mechanisms in cold tolerance remain largely unknown, partially due to the lack of a suitable in vitro model. An immortal neural cell line designated as tilapia brain neural (TBN) was established from brain tissue of the genetically improved farmed tilapia strain of Nile tilapia (Oreochromis niloticus). The TBN cells show a neuron-like morphology at low density and form a fibroblast-like monolayer at high density. Transcriptome profiling through RNA-sequencing revealed that a total of 15,011 genes were expressed in the TBN cells. The TBN cells express a wide array of marker genes for neural cells. A comparative analysis of the featured genes among the 17 cell clusters isolated from the subventricular zone of mouse brain revealed the highest transcriptome similarity between the TBN cells and the transient amplifying progenitors (TAPs). The TBN cells tolerate relatively high culture temperatures, and the highest growth rate was observed for the cells cultured at 32 degrees C compared with those at 30 degrees C, 28 degrees C and 26 degrees C. Nonetheless, this cell line is cold sensitive. Exposure of the cells to 16 degrees C or lower temperatures significantly decreased cell confluences and induced apoptosis. The TBN cells were more sensitive to cold stress than the ZF4 cells (embryonic zebrafish fibroblasts). Moreover, the TBN cells can be efficiently transfected through electroporation. This study provides an invaluable research tool to understand the nature of cold sensitivity of tilapia and to dissect the function and mechanism of genes in regulating cold tolerance of fish
Spatial and seasonal variation of water parameters, sediment properties, and submerged macrophytes after ecological restoration in a long-term (6 year) study in Hangzhou west lake in China: Submerged macrophyte distribution influenced by environmental variables
Submerged macrophyte restoration is the key stage in the reestablishment of an aquatic ecosystem. Previous studies have paid considerable attention to the effect of multiple environmental factors on submerged macrophytes. Meanwhile, few studies have been conducted regarding the spatial and seasonal characteristics of water and sediment properties and their long-term relationship with submerged macrophytes after the implementation of the submerged macrophytes restoration project. On a monthly basis, we monitored the spatial and seasonal variation in water parameters, sediment properties, and the submerged macrophyte characteristics of West Lake in Hangzhou from August 2013 to July 2019. From these measurements, we characterized the relationship between environmental factors and submerged macrophytes. Water nutrient concentrations continuously decreased with time, and the accumulation of sediment nutrients was accelerated as the submerged macrophyte communities developed on a long-term scale. The results indicated that the difference in water parameters was due to seasonal changes and land-use types in the watershed. The differences in the sediment properties were mainly attributed to seasonal changes and changes in the flow field. Redundancy analysis showed that the influence of water nutrients on the submerged macrophyte distribution was greater than that of sediment nutrients. The result also suggested that the developed root system, high stoichiometric homeostasis coefficients of P, and compensation ability of substantial leaf tissue may lead to a large distribution of Vallisneria natans in West Lake in Hangzhou. The correlation of water parameters and sediment properties with submerged macrophytes for a long time was very important as the restoration was achieved. To ensure the stability of the aquatic ecosystem after performing the submerged macrophyte restoration, a greater emphasis must be placed on reestablishing the entire ecosystem, including the restoration of aquatic animals and fish stocks. Expression and antibacterial analysis of galectin-8 and-9 genes in mandarin fish, Siniperca chuatsi
Galectin-8 and galectin-9 belong to tandem repeat-type galectins, and in the present study, these two genes were cloned in mandarin fish Siniperca chuatsi. The open reading frame (ORF) of the mandarin fish galectin-8 and galectin-9 contains 942, and 1008 bp, encoding 313 and 335 amino acids, respectively. As a conserved feature, an N-terminal carbohydrate recognition domain (CRD), and a C-terminal CRD were observed in each of the two galectins in mandarin fish. In healthy fish, galectin-8 and -9 were constitutively expressed in all organs/tissues examined, and their expression can be induced following the stimulation of LPS and poly(I:C). It is obvious that galectin-8 had a higher increase at mRNA level following the stimulation of poly(I:C). It is further demonstrated that mandarin fish galectin-8 inhibited the growth of Flavobacterium columnare and Streptococcus agalactiae, and in addition to the two species of bacteria, galectin-9 inhibited also the growth of Edwardsiella piscicida, which provides the basis for further understanding their antibacterial role in immune response of mandarin fish
Development of Novel SNP Assays for Genetic Analysis of Rare Minnow (Gobiocypris rarus) in a Successive Generation Closed Colony
The complex genetic architecture of closed colonies during successive passages poses a significant challenge in the understanding of the genetic background. Research on the dynamic changes in genetic structure for the establishment of a new closed colony is limited. In this study, we developed 51 single nucleotide polymorphism (SNP) markers for the rare minnow (Gobiocypris rarus) and conducted genetic diversity and structure analyses in five successive generations of a closed colony using 20 SNPs. The range of mean Ho and He in five generations was 0.4547-0.4983 and 0.4445-0.4644, respectively. No significant differences were observed in the Ne, Ho, and He (p > 0.05) between the five closed colony generations, indicating well-maintained heterozygosity. The F-statistics analysis revealed a relatively stable genetic structure of the closed colony. Furthermore, the genetic distance between the newer and older generations increased with the breeding generations in closed colonies. Our results confirmed previous findings in the same samples using microsatellite markers. The results will be beneficial for establishing genetic variability monitoring criteria and restoration of the wild population of the rare minnow and other laboratory fish
Analysis of constructed wetlands technology performance efficiency for the treatment of floriculture industry wastewater, in Ethiopia
In the present study subsurface flow constructed wetlands treatments (SSFCWT) for the treatment of floriculture industry wastewater in Ethiopia was studied. The objective of the study was to assess the efficiency of SSFCWT in treating floriculture industry wastewater. Seven SSFCW was assessed for physiochemical and heavy metal removal efficiency using standard methods (APHA) and compared to its treatment efficiency. The results showed that treated water quality significantly improved from the inlet to the outlet, with TSS, TDS and TP reduced from 247 +/- 0.02 to 31.66 +/- 5.8, 391.67 +/- 6.18 to 80.66 +/- 3.86, 6.7 +/- 0.92 to 2.06 +/- 0.38, respectively. Similarly, NH 4 +-N, NO3 -N, and TN, reduced from 22 +/- 2.94 to 9.8 +/- 1.96,73.67 +/- 4.99 to 27.0 +/- 7.12, and 94 +/- 4.08 to 38.33 +/- 4.11, respectively. The result showed that highest performance variability was observed among the studied SSFCWs. The heavy metal concentration in the influent to effluent for Pb, Cu and Mn was reduced from 0.63 +/- 0.09 to 0.36 +/- 0.11, 0.49 +/- 0.08 to 0.18 +/- 0.05 and 0.70 +/- 0.07 to 0.43 +/- 0.15, respectively. This assessment has proved that a significant reduction of pollutants was observed using SSFCW treatments. There is a need to encourage other floriculture industry to adopt and implement constructed wetland technology as a primary option to protect the environment and public health
Identification and characterization of two mannan-binding lectin associated proteins in lectin complement pathway of grass carp
The lectin pathway of complement activation is an important component of the innate immune response, which must be tightly controlled to maintain immune homeostasis. However, its control mechanisms have not been investigated in detail in bony fish. In this study, we identified and characterized two novel, phylogenetically conserved mannan-binding lectin (MBL)-associated proteins (MAps) of grass carp (Ctenopharyngodon idella), CiMAp27 and CiMAp39, which were truncated, alternatively-spliced forms of grass carp MBL-associated serine proteases (MASPs), CiMASP1 and CiMASP2, respectively. Gene expression profiling showed that both CiMAp27 and CiMAp39 were upregulated by low doses of Aeromonas hydrophila, and inhibited by high doses, which lead to the inference that these genes acted as immune factors in antibacterial defense. Sequence analysis showed that CiMAp27 lack a catalytic domain but retains two domains (CUB1-EGF) involved in the association with MBL, while CiMAp39 retained four domains (CUB1-EGF-CUB2-CCP1). Not only the two CiMASPs but also the CiMAps were detected in grass carp serum. Furthermore, both recombinant CiMASPs (rCiMASPs) and recombinant rCiMAps (rCiMAps) interacted with recombinant MBL and the two CiMAps competed with CiMASPs for binding to MBL, and hence inhibited downstream C4 binding. These results indicated that CiMAps acted as competitive inhibitors in the lectin complement pathway of grass carp.</p
